[Culture medium optimization and primary kinetics analysis for nisin production].

Response surface methodology was used to optimize a medium for nisin production of Lactococcus lactis. In the first optimization step the influence of sucrose, soybean peptone, yeast extract, potassium dihydrogen phosphate, sodium chloride, and magnesium sulfur on nisin production was evaluated using a fractional factorial design. Potassium dihydrogen phosphate influenced nisin production positively while soybean peptone affected nisin production negatively. The other components had no significant effect on nisin production. The path of steepest ascent was used to approach the optimal region of the medium composition. In the third step the optimal concentrations of KH2PO4 and soybean peptone were determined by a central composite design and response surface analysis. The optimized medium allowed nisin production to be increased from 1074 IU/mL to 2150 IU/mL. The kinetic analysis showed that nisin production fashion at optimized and non-optimized media was not changed and maintained partially growth-associated. But the specific growth rates and the specific nisin production rates for the strain at the optimized medium were bigger than the ones at the non-optimized medium after the cells entered the middle of exponential phase.

[QTL analysis of anther length and ratio of stigma exsertion, two key traits of classification for cultivated rice (Oryza sativa L.) and common wild rice (O. rufipogon Griff.)].

A genetic map including 120 RFLP markers covering 1418.2 cM has been constructed using a BC1 population derived from a cross between common wild rice (Oryza rufipogon Griff.) from Dongxiang, Jiangxi Province, China, and an elite Indica variety Guichao 2. The linkage orders and genetic distances of markers in this map are consistent with the map constructed by Rice Genome Program of Japan except the markers of short arm of Chromosome 1. The QTLs analysis of anther length and ratio of stigma exsertion, two key traits of classification for cultivated rice and common wild rice indicated that there are two QTLs controlling anther length located at the region of C424-G39 of Chromosome 2, and C2807-C1263 of Chromosome 9 respectively. It was also found that there are two QTLs controlling ratio of stigma exsertion, and located at the region of C2289-R1553 of Chromosome 5 and G1149-R1963 of Chromosome 8 respectively. The detected QTLs affecting the key traits of classification for wild rice and cultivated rice will be useful for study of molecular evolution mechanism of cultivated rice.

Conjugation with (111)In-DTPA-poly(ethylene glycol) improves imaging of anti-EGF receptor antibody C225.

UNLABELLED: Significant liver uptake often limits the clinical application of radiolabeled antibodies in radioimmunodetection. The purpose of this study was to evaluate the gamma-imaging properties of an antiepidermal growth factor receptor (EGFR) antibody, C225, conjugated with heterofunctional poly(ethylene glycol) (PEG) with 1 terminus of the polymer attached to a radiometal chelator, diethylenetriaminepentaacetic acid (DTPA). METHODS: Two preparations of PEG-modified C225, one with 20% and the other with 60% amine substitution, were labeled with (111)In. The conjugates, (111)In-DTPA-PEG-C225, were injected intravenously into nude mice with EGFR-positive A431 tumors. For comparison, C225 directly labeled with (111)In was also injected. In a competitive study, mice with A431 tumors were pretreated intravenously with 100-fold excess of native C225, followed by an injection of (111)In-DTPA-PEG-C225 30 min or 20 h later. In addition, (111)In-DTPA-PEG-C225 was injected into mice with EGFR-positive MDA-MB-468 tumors and EGFR-negative MDA-MB-435 tumors. Images were acquired at 5 min and at 2, 6, 24, and 48 h after injection of the radiotracers. Regions of interest (ROIs) were drawn on the computer images around the whole body, liver, muscle, and tumor. The counts per pixel in the tumor and normal tissues were calculated. At 48 h, the mice were killed and dissected. Blood, liver, muscle, and tumor samples were removed and the radioactivity of each sample was measured. RESULTS: In A431 tumor xenografts, the tumor uptake of C225 modified with PEG was not significantly different than the uptake of unmodified (111)In-DTPA-C225. Uptake in the liver, however, was reduced by 38%-45%, and the reduction increased with increasing degree of PEG substitution. Tumors of A431 and MDA-MB-468 xenografts were clearly visualized with (111)In-DTPA-PEG-C225, whereas tumors of the MDA-MB-435 xenograft, which expresses low levels of EGFR, were not as readily visible. The tumor-to-blood ratios of (111)In-DTPA-PEG-C225 in A431 and MDA-MB-468 xenografts were about 3 fold higher than in MDA-MB-435 xenografts. Blocking EGFR by pretreatment with native C225 significantly reduced the uptake of (111)In-DTPA-PEG-C225 in the liver. The tumor-to-blood ratios in mice with A431 tumors were decreased 2.5-2.7 fold after pretreatment with a large excess of C225. Similar results were obtained with MDA-MB-468 tumor xenografts. In contrast, the tumor-to-blood ratios in mice with MDA-MB-435 tumor xenografts were not significantly different in C225-pretreated mice than in nonpretreated mice. CONCLUSION: These findings indicate that (111)In-DTPA-PEG-C225 selectively localized to the tumors expressing high levels of EGFR. PEG-modification of C225 significantly reduced its liver uptake, resulting in improved visualization of EGFR-positive tumors. Using PEG as a linker between the monoclonal antibody and metal chelator is a useful strategy to optimize the imaging characteristics of antibody-based scintigraphic agents.

[A preliminary study of the combination of noninvasive parameters in the diagnosis of liver fibrosis].

OBJECTIVE: To explore the combination of noninvasive parameters and its significance in the diagnosis of liver fibrosis. METHODS: Liver biopsy was performed on 200 patients with chronic liver diseases, and the biochemical parameters, serum fibrotic markers, and B ultrasound, CT, and MRI were detected in the corresponding period. Then, by the regression, we obtained the combination of the noninvasive parameters and evaluated the significance in the diagnosis of liver fibrosis. RESULTS: Three combinations have been obtained. The parameters in the first combination which was used to differentiate fibrosis and no fibrosis(S0 vs S1+S2+S3+S4) were the parameter of blood flow of portal vein per minute by B ultrasonography, age, the maximum oblique line of right liver by B ultrasound, the wave expression of the surface of liver by CT/MRI and GGT. The parameters in the second combination which was used to evaluated the degree of fibrosis(S1+S2 vs S3+S4) were HA, A/G and the longest length of spleen by B ultrasound. The third combination which was used to distinguish fibrosis with cirrhosis (S1+S2+S3 vs S4) was composed by HA, the little cyst-like change near the intrahepatic duct, thickness of liver capsule by B ultrasound, age and the index of symptoms. All the three combinations were sensitive, specific, and accurate. CONCLUSIONS: The combinations of noninvasive parameters is more valuable than single parameter in the diagnosis of liver fibrosis.

The cAMP-mediated protein kinase signal transduction pathway is involved in the pyrogenic effect of CRH in rats.

OBJECTIVE: To determine whether the cyclic adenosine monophosphate (cAMP) mediated protein kinase signal transduction pathway is involved in the pyrogenic action of corticotropin releasing hormone (CRH) in rats. METHODS: Corticotropin releasing hormone, 2',3'-dideoxyadenosine (DDA) and adenosine-3',5'-(cyclic) monophosphorothionate, Rp-Isomer (Rp-cAMPS), were administered intracerebroventricularly (i.c.v.). The colonic temperature was measured using a thermistor, and the content of cAMP in the hypothalamus was determined by radioimmunoassay. Hypothalamic incubation was used to assess the effects of CRH on the content of cAMP in the hypothalamus in vitro. RESULTS: Microinjection (i.c.v.) of CRH (2.5 micrograms, 5.0 micrograms and 10 micrograms) caused increases in colonic temperature and the hypothalamus cAMP level in conscious rats. CRH increased hypothalamus cAMP level in vitro. The pyrogenic effects of CRH were abolished or markedly inhibited by prior injection (i.c.v.) of an adenylate cyclase inhibitor, DDA (30 micrograms), or an inhibitor of cAMP-dependent protein kinase, Rp-cAMPS (15 micrograms). CONCLUSION: cAMP mediates the pyrogenic action of centrally administered of CRH in rats, and protein kinase A may play an important role in the central CRH-induced fever. The cAMP-dependent protein kinase signal transduction pathway may be involved in the central mechanisms of the pyrogenic action of CRH in rats.

[Effects of fungal elicitor on cell status and taxol production in cells suspension cultures of Taxus chinesis var. mairei].

Effects of fungal elicitor on cell redox status and taxol production were studied in suspension cultures of Taxus chinesis var. mairei in the late exponential stage. The results show that fungal elicitor induced oxygen burst, changes of the cell redox status, alkalinization of medium and the fluctuation of the activity of redox enzymes with a sequence. The content of protein representing the quantity of enzymes increased. The activity of SOD increased quickly after treatment by fungal elicitor and that of POD could be kept at a higher level in contrast to the control. The activity of CAT was inhibited at first and followed by an obvious increase, while the activity of PAL was promoted. The taxol yield was 5 folds of the control, reaching to 67 mg/L.

[Effects of different varieties and sowing dates on development stages of wheat].

The effects of different sowing dates and varieties on development stages of wheat were studied in this paper. The results showed that the emergence of tiller was mainly regulated by the ecological factors rather than the genetic effect. For winter-type variety(Jing 411), most variable growth duration throughout the entire growth cycle occurred from tillering to jointing. In contrast, the timing of single ridge stage was determined by genotypic effect rather than ecological factors. Single ridge to double ridge stage was most sensitive to vernalization progress. For spring sowing of winter-type variety, the vernalization process could last till floral primordium differentiation stage. The corresponding relationships existed between phenological and spike development stages in different sowing dates, although a little variable. Winter-type variety had more ecologically variable leaf primoudia, due to its stronger vernalization requirement. For winter-type variety, the primordium numbers of reproductive organs were not apparently related to the GDD accumulated prior to vernalization completion, while for spring-type variety(Yangmai 158), the numbers of different apex primordia including vegetative and reproductive organs were significantly related to the accumulated GDD during the entire phenological stages as well as before anther separation stage.

[Effect of natural zeolite on nutrient preserving capability of degraded slope soil in south China].

Soil column leaching experiment showed that after zeolite application, the content of nitrogen and potassium in leachate was reduced significantly, but no significant effect was detected for phosphate. This application had a positive effect on soil available nitrogen, phosphorus and potassium. It increased the content of soil AL-P and Fe-P, but did not increase the NO3-N content in leachate.

[Studies on the relationship between clinicopathological features and human papillomavirus types in female lower genital tract carcinoma].

OBJECTIVE: To investigate the relationship between clinicopathological features and human papillomavirus types in female lower genital tract carcinoma. METHODS: Clinicopathological features of 100 cases of female lower genital tract carcinoma (63 cervical carcinoma and 37 vulvar carcinoma) were studied retrospectively. Standard PCR (HPV type 6/11, 16, 18) was applied to formalin fixed, paraffin embedded sections. RESULTS: There were 54 cases of cervical carcinoma and 33 cases of vulvar carcinoma in the 87 cases of target DNA qualified samples. The HPV detection rate in cervical carcinoma was 83.3%. HPV16 (55.6%) and HPV 18(24.4%) were the predominant types. In vulvar carcinoma, HPVs, mainly HPV 16(70%, 7/10), were detected in basaloid (83.3%, 5/6) and warty carcinoma (83.3%, 5/6), but none in conventional type of keratinized squamous cell carcinoma (0.0%, 0/21). Three of the 6 women with basaloid carcinoma were associated with cervical squamous neoplasia, and the same HPV type was found in both lesions in 2 of the 3 patients with two primary tumors. Four patients with basaloid carcinoma recurred after simple vulvectomy or local excision, but no lymph node metastasis occurred and all were still alive at last follow-up, with a median follow-up of 6.3 years. In contrast, the majority of the women with keratinized squamous cell carcinoma were over 65 years of age, with histologic extensive keratinization and poorer prognosis. CONCLUSIONS: A high detection rate of HPV 16 and 18 was found in cervical carcinoma. However, the sensitivity of HPV in vulvar carcinoma seems to be more related to histologic type.

[Phenolic acid degradation fungi screened from successive plantation site of Chinese fir].

Successive plantation of Chinese fir (Cunninghamia lanceolata) leads an obvious decline of soil fertility. To solve the urgent problem, five fungal species (F2, F3, F4, F7, F15) which can effectively degrade phenolic acid were screened to degrade the toxic material in successive plantation site and to improve the soil bioactivity. Pot experiment showed that all test fungus, except F4, could improve plant growth individually, but not counteract the effect of phenolic acid. Inoculating five inoculated fungi mixed with equal proportion not only counteracted the inhibitory effect of phenolic acid but also improved the growth of Chinese fir uninoculated with VAM fungi but inoculated with G. mosseae.

[Brief history of cosmetology in the imperial palace of China].

The development of Chinese Palace's cosmetology is divided into three periods. It is in its embryonic stage that from the Xia to the Western Jin. The drugs and perfumes related to cosmetology were taken seriously by the Palace. The formulation of facial beauty was established. From the Eastern Jin to the end of Tang Dynasty was the forming periods. Cosmetology has been fully developed, and became the physical civilization which could enjoy by both nobels and common people. The developing and enriching time was from the Wu Dai to the end of the Qing Dynasty. The theory of cosmetology achieved a great development. The number of prescriptions of the Palace's cosmetology were increased greatly.

[CAG microsatellite polymorphisms of androgen receptor gene and the stage and grade of prostate cancer].

OBJECTIVE: To explore the relationship between the CAG microsatellite in androgen receptor (AR) gene and the stage and grade of prostate cancer(PC). METHODS: The number of CAG repeats in exon 1 of the AR gene was measured in 37 PC tissues by PCR-SSCP analysis, and its association with stages and grades of PC determined. RESULTS: The average number of CAG repeats of AR gene differed significantly between PC in stage B (25.04 +/- 1.88) and in stage C-D (24.14 +/- 2.64) (P = 0.002). The frequency of short CAG(< 22) was 13.0% (3/23) in stage B and 28.6%(4/14) in stage C-D, while that of long CAG(> or = 22) was 87.0% (20/23) and 71.4% (10/14) (P = 0.029), respectively. The average number of CAG repeats increased with the decrease in tumor cell differentiation. The frequency of short CAG repeats dropped with the increase in tumor cell differentiation while the opposite was true for that of long CAG repeats. CONCLUSION: The number of CAG repeats in AR gene may be one of the factors determining the biologic characteristics PC. Short CAG repeats may signify aggressiveness of PC.

[Application of mRNA differential display technique in screening related gene of endometrial carcinoma].

OBJECTIVE: To screen related gene of endometrial cancer by mRNA differential display (DD)-polymerase chain reaction (PCR). METHODS: Individual mRNA from normal and neoplastic endometrium obtained from the same patient were comparatively studied by DD methodology. The mRNAs were isolated from the tissues, reverse transcribed to cDNA, and then amplified by PCR. The PCR products were displayed on a polyacrylamid gel by silver staining, and the differentially expressed bands were retrieved and reamplified. Subsequently we selected four bands for sequencing and making further investigation. RESULTS: cDNA homology was analysed by searching through GenBank sequence databases and indicated that tumor fragment (T1.7) has no significant sequence homology with any sequence of known function. Acession number is AF315581. Further study showed that T1.7 expressed more abundantly in endometrial cancer than hyperplasia endometrium, but not expressed in normal endometrium. CONCLUSIONS: T1.7 is a new expressed sequence tag. It may be related gene fragment of endometrial cancer.

[A study on effective parameters to represent the heterogeneity in ventricular repolarization].

The aim of this study was to investigate whether ARI (activation-recovery interval) dispersion on the sheep epicardium was related with the ischemia induced heterogeneity in ventricular repolarization. We have developed an algorithm based on wavelet transformation to analyze ARI from a 64-channel epicardial ECG data of an ischemic sheep. The quadratic spline wavelet was used to extract QRS with 2(1) scale and T wave with 2(5) scale, and then A and R points were determined by definition. It was found that there was not a significant difference in the average ARI on the epicardium before and during ischemia. Meanwhile ARI dispersion on the epicardium was significantly increased during ischemia (126.3 ms vs 59.9 ms, P = 0.0007). Furthermore, a computer simulation showed that ARI is very close to ADP, suggesting that ARI dispersion may be an effective parameter to estimate the heterogeneity in ventricular repolarization induced by ischemia.

[Summarization of studies on Chinese marine medicinal animal Syngnthus acus].

Syngnthus acus L. is a kind of very important traditional Chinese-medicine from sea. It has plentiful amino acid, protein, trace element, poly-carbon non-saturation fat acid, etc. Syngnthus acus has sexual-hormones-like, anti-cancer and resisting fatigue effects; It can resist fatigue. It can also improve organism immunity and enhance the systolic strength of heart muscle. It can be widely used in many fields such as food field, medical field and aquatic products industry. Its classification, resources, chemical composition and its medical value have been summarized in this article.

[Significance of the CT scan in renal tuberculosis].

OBJECTIVE: To elucidate the features and clinical significance of CT scan in the diagnosis of renal tuberculosis. METHODS: The CT findings and the clinical characteristics of 42 cases with renal TB were retrospectively analyzed. RESULTS: Unilateral renal TB was found in 34 cases, bilateral in 8 cases. The typical CT features were characterized by one or more cysts surrounding the calyx with thin adjacent cortex (34/50), thickened wall of ureters and calyx, hydronephrosis (22/50) and kidney calcification (8/50). Atypical features presented with single or multiple low density nodes in the renal parenchyma (6/50). 30 patients receiving contrast enhanced CT scan showed that intensified Hu (by 20 approximately 120 HU) in the affected kidney. Multiple lymph nodes calcification in abdomen was found in 2 patients, calcification and low density nodes in spleen and liver in 4 patients, vertebral damage and paravertebral abscess in 6 patients and post-abdominal abscess in 1 patient. CONCLUSION: CT scan examination is not only useful in the diagnosis of renal tuberculosis, but also of significance in assessing the severity of the disease, renal function and the involvement of other organs in the abdomen.

[Studies on infiltration mechanism of adenoid cystic carcinoma: haptotaxis function assay of fibronectin].

OBJECTIVE: To investigate the invasion mechanism of adenoid cystic carcinoma (ACC) by ACC haptotaxis changes in fibronectin (FN). METHODS: Three strains of ACC cells were used. The haptotaxis changes in FN, and receptor changes in the cell membrane surface were determined. RESULTS: All three strains of ACC cells had remarkably greater migration ability than SCC cell. High level of alpha 2, alpha 3, alpha 6 and beta 1 receptor subunit types was found in the cell surface of ACC and SCC; and high level of alpha 5 in ACC3. High inhibition of haptotaxis reaction was found after blocked alpha 5 and beta 1 receptor subunit in ACC and a middle inhibition after blocked alpha 4. In SCC more strong inhibition was found after blocked alpha 1, alpha 4 and beta 1. CONCLUSIONS: This study indicates that ACC cells have greater invasion ability than SCC cells in FN and haptotaxis reaction of different receptor of tested cells in FN have some feature pattern.
