A type-specific antiserum induced bya major herpesvirus type 1 glycoprotein.

A type-specific antiserum was prepared against VP 7/8, the major HSV-1 type-specific glycoprotein. The specificity of this antiserum was demonstrated by both neutralization of HSV-1 infectivity and immunoprecipitation followed by SDS-polyacrylamide gel electrophoresis of the immunoprecipitates. Only minimal cross-reactivity with HSV-2 was observed. This major VP 7/8 glycorprotein was also purified from a host of virus-induced proteins with the specific immunoadsorbent prepared with anti-VP 7/8 serum. The one-step purification of this type-specific neutralizing antigen of HSV-1 should allow the development of a sensitive and specific radioimmunoassay.

Genetic variability of Hong Kong (H3N2) influenza viruses: spontaneous mutations and their location in the viral genome.

The genetic heterogeneity of five influenza A (H3N2) strains isolated between 1968 and 1977 has been estimated by T1-oligonucleotide fingerprinting of 32P-labeled viral RNA. Assuming that the large T1-resistant oligonucleotides represent a random sample of the viral RNA, the genetic differences observed would affect 0.3 to 10.7% of the RNA positions of the genes studied, depending on the pair of viruses considered. A smaller degree of genetic heterogeneity was observed when six coetaneous viral samples were compared. The distribution of spontaneous mutations among the viral genes was studied by fingerprinting individual RNA segments isolated either by gel electrophoresis or hybridization with plasmids containing influenza-specific DNA sequences. No statistically significant differences were detected in the distribution of mutations among the viral genes studied. The mutation frequency at the hemagglutinin RNA region coding for the HA1 subunit was found to be two times higher than that at the region encoding that HA2 subunit. Our results suggest that the antigenic variability of influenza viruses may be a consequence of a general genetic variability which effects many of the viral genes.

[Salmonella infection in children. Epidemiological and clinical considerations (author's transl)].

An analysis of data on Salmonella infection treated at the Children's Hospital "La Fe", in Valencia, from 1974--75 is presentd. A patient population of 211 selected cases were divided into two groups: Gastroenteritis (GEC), 155 cases, and typhoid-paratyphoid fever (T-P F), 56 cases. Hospitalization was required in 79% of the cases. The following parameter were studied: Locality and district of origin with respect to residence, conditions of hygiene, size of family: age, sex, seasonal incidence, previous time of illness, presenting symptoms and physical sings and complementary studies. The following conclusions were obtained: Salmonella infection are and endemic problem in Valencia. Their incidence is maximal during the months of June to October. Epidemiological environment was positive in 15% of the cases. Higher morbidity in children less than two years of age. Most frequent presenting symptoms: Diarrhea, fever and vomiting as often in GEC as in T-P F. In 38.7% of GEC cases, the diarrhea was bloody; 21.9% of GEC cases began with fever. Salmonella paratyphi B was the agent most frequently responsible. Data concerning sex, family size, conditions of hygiene and white blood cells offer little discriminatory information. Results obtained are concordant with those described in the literature.

Degradation of benzothiophene and related compounds by a soil Pseudomonas in an oil-aqueous environment.

Pseudomonas aeruginosa PRG-1, an isolate from oil-contaminated soil, degrades benzothiophene (BT) and other related compounds in a 5% oil-basal medium system. The organism cannot grow on BT alone; 0.05% yeast extract is a suitable substrate for its growth and for its attack on BT. Although BT is partially toxic to the bacteria, toxicity is reduced when BT is added in this oil system. The oil phase is emulsified by bacterial action during the process. Oxygen uptake studies with washed cell suspensions show increased respiration in the presence of BT. Endogenous respiration is markedly decreased by p-hydroxy-mercuribenzoate, whereas respiration due to BT is scarcely affected, suggesting that oxygen is added directly to BT. Results obtained both in direct degradation and in respiration studies indicate that 3-methyl-thiophene is more rapidly and extensively degraded than BT and other related compounds.

Atrial natriuretic factor in cirrhosis with ascites: plasma levels, cardiac release and splanchnic extraction.

The aim of this study was to determine the plasma levels, cardiac release and splanchnic extraction of atrial natriuretic factor in cirrhosis with ascites. The plasma concentration of immunoreactive atrial natriuretic factor in samples obtained from an antecubital vein was measured in 18 healthy volunteers and in 35 cirrhotics with ascites. In 11 of these cirrhotics and in 11 patients admitted to the hospital for the study of a thoracic pain who had no clinical or hemodynamic signs of cardiac failure (control group), the plasma levels of immunoreactive atrial natriuretic factor in samples from the coronary sinus, right atrium, pulmonary artery, hepatic vein and femoral vein were determined and the coronary sinus blood flow measured by thermodilution. Cirrhotic patients showed significantly higher plasma levels of immunoreactive atrial natriuretic factor in each vascular territory studied than did control subjects (coronary sinus: 101.2 +/- 10.6 vs. 26.1 +/- 4.7 fmoles per ml; right atrium: 32.5 +/- 5.8 vs. 9.4 +/- 3.5; pulmonary artery: 36.8 +/- 10.1 vs. 7.5 +/- 2.4; hepatic vein: 10.7 +/- 2.0 vs. 2.7 +/- 0.8; femoral vein: 18.2 +/- 2.4 vs. 3.1 +/- 0.9; antecubital vein: 14.7 +/- 1.6 vs. 4.0 +/- 0.8). The coronary sinus blood flow was also higher in cirrhotics (200 + 22 ml per min) than in controls (105 +/- 7 ml per min). Consequently, the estimated cardiac release and cardiac production of immunoreactive atrial natriuretic factor were strikingly increased in cirrhotics (13,334 +/- 2,007 and 5,484 +/- 1,734 fmoles per min, respectively) as compared to control subjects (1,669 +/- 338 and 1,431 +/- 350 fmoles per min, respectively; p less than 0.01).(ABSTRACT TRUNCATED AT 250 WORDS)

Liposarcoma of the stomach.

We present a case of gastric liposarcoma, a very uncommon pathology; only nine cases have been reported in the literature. We describe the radiologic findings of this neoplasm and emphasize the correlation between computed tomography (CT) and the macroscopic morphology of the tumor, which is conditioned by its histology. In our case, CT demonstration of fatty areas within the gastric mass facilitated the diagnosis. This finding has not been described for liposarcomas of the stomach.

[Video fiber endoscopy during induced sleep].

Video-fiberoscopy is the exploration of upper respiratory airways used in our diagnostic protocol in patients with chronic snoring and obstructive sleep apnea syndrome (OSAS). We perform a fibroscopy during propofol induced sleep without tracheal intubation; video recording and monitoring of level of sleep, pulse oximetry, blood pressure, ECG, etc. is used. This exploration allows us to asses upper respiratory airways when the patient is asleep as well as the areas of collepse and vibration both from a functional and anatomical points of view.

Distribution of HCV genotypes in patients infected by different sources.

To determine the distribution of hepatitis C virus (HCV) genotypes in our population within the Basque country, 58 HCV-infected patients were analysed. The predominant HCV genotype among subjects infected by blood transfusions (n = 9) and surgical procedures (n = 14) was 1b (88 and 50%, respectively). On the other hand, genotype 3a was the most common among intravenous drug users (n = 16), with data statistically significant (p < 0.0001). There were 18 patients (31%) with unknown risk factors, twelve of whom (66.7%) were infected with genotype 1b. Patients infected with genotype 1b were older than patients infected with genotype 3a (p < 0.0001). There existed a relationship between the HCV genotype and the source of infection.

Oxidative degradation of azo dyes by manganese peroxidase under optimized conditions.

The application of enzyme-based systems in waste treatment is unusual, given that many drawbacks are derived from their use, including low efficiency, high costs and easy deactivation of the enzyme. The goal of this study is the development of a degradation system based on the use of the ligninolytic enzyme manganese peroxidase (MnP) for the degradation of azo dyes. The experimental work also includes the optimization of the process, with the objective of determining the influence of specific physicochemical factors, such as organic acids, H(2)O(2) addition, Mn(2+) concentration, pH, temperature, enzyme activity and dye concentration. A nearly total decolorization was possible at very low reaction times (10 min) and at high dye concentration (up to 1500 mg L(-)(1)). A specific oxidation capacity as high as 10 mg dye degraded per unit of MnP consumed was attained for a decolorization higher than 90%. Among all, the main factor affecting process efficiency was the strategy of H(2)O(2) addition. The continuous addition at a controlled flow permitted the progressive participation of H(2)O(2) in the catalytic cycle through a suitable regeneration of the oxidized form of the enzyme, which enhanced both the extent and the rate of decolorization. It was also found that, in this particular case, the presence of a chelating organic acid (e.g., malonic) was not required for an effective operation. Probably, Mn(3+) was chelated by the dye itself. The simplicity and high efficiency of the process open an interesting possibility of using of MnP for solving other environmental problems.
