Transformation frequencies of 4 x 10(-5) were obtained in chromosome-mediated gene transfer experiments using human cell line HeLa S3 as donor and mouse cell line A9 as recipient. This high frequency of interspecific transformation was achieved by treating the recipient cells with dimethylsulfoxide in addition to other facilitators. The high frequency of transformation correlated positively with transgenome size on the basis of both co-transfer of linked markers and chromosome analysis. The syntenic human markers glucose-6-phosphate dehydrogenase (D-glucose-6-phosphate:NADP(+) 1-oxidoreductase, EC 1.1.1.49) and phosphoglycerate kinase (ATP:3-phospho-D-glycerate 1-phosphotransferase, EC 2.7.2.3) were sometimes transferred together with the selected X-linked prototrophic marker hypoxanthine phosphoribosyltransferase (IMP: pyrophosphate phosphoribosyltransferase, EC 2.4.2.8) into murine somatic cells. Donor human chromosome material could be demonstrated cytologically in some of the transformed cell lines. Transformants exhibited various rates of loss of the human hypoxanthine phosphoribosyltransferase marker when grown under nonselective conditions. These results reveal a broader range of possible interspecific transgenome sizes than has been recognized in the past. The largest transgenomes consist of cytologically detectable donor fragments and contain syntenic markers that are not closely linked to the selected marker.

Changes in local cerebral blood flow during sodium nitroprusside hypotension were measured using the hydrogen electrode technique. At mean arterial pressures from 90% to 50% of control values, local cerebral blood flow showed a significant decrease by 20%. When blood pressure was reduced below 50%, the local cerebral blood flow increased significantly and approached control levels. It is suggested that the flow increase may be due to local hypoxia. Associated with this increase were electrocorticogram changes indicative of hypoxia. The significant decrease in mean local cerebral blood flow and the large range of flows seen during mild hypotension indicate that autoregulation is impaired. During hypotension, intracranial pressure increased by, at most, 3 mm Hg. Cardiac output was usually unaffected and was never decreased by more than 20%.
Non-transformed human fibroblasts, strain PA-2, were treated with polyethylene glycol (PEG) in monolayer culture to produce multinucleate fibroblast homokaryons. Antibodies to tubulin or actin were used to monitor cytoplasmic microtubule and actin filament patterns immediately after cytoplasmic fusion, as well as after the fused cells had been in culture for varying amounts of time. The cytoplasmic microtubule complex as increased for a short time after cell fusion and then decreased to resemble the complex seen in control cells. The actin stress fibers were similarly enhanced for a comparable period of time. However, this initial enhancement of the actin stress fibers gradually diminished for approximately one month in culture after which the fibers were greatly reduced in both size and number. Concurrent with the changes in cytoplasmic microtubule and actin fiber complexes, the PEG-treated cells began to show alterations in growth parameters which progressively resembled those characteristic of transformed cell populations. Fusion of normal cells may be an initial step in the transformation of such cells to malignancy.

Tearing was the initial symptom in two patients who subsequently developed a mass over the anterior lacrimal crest area caused by adenoid cystic carcinoma of the antrum. In a 76-year-old woman, a paranasal sinus X-ray film revealed an antral mass, and the histopathologic diagnosis was made by incisional biopsy. She refused further examination and treatment. In a 62-year-old woman, a paranasal sinus X-ray film and computed axial tomography examination revealed an antral mass. Epiphora caused by obstruction of the nasolacrimal sac was shown by dacryocystography. Bony destruction of the medial orbital floor was shown by paranasal sinus X-ray film, and confirmed at the time of initial biopsy. Orbital exenteration and radical maxillectomy and ethmoidectomy were performed, and the histopathologic diagnosis and primary site of the tumor were confirmed. There was no recurrence 24 months postoperatively.

Patterns of cytoplasmic microtubules in somatic cell hybrids between transformed mouse cells and nontransformed human skin fibroblasts were examined using antitubulin antibodies as an immunofluorescent probe. Nontransformed cells have been shown to exhibit an extensive cytoplasmic microtubule complex (CMTC), while in transformed cells, this complex is greatly diminished. The hybrid populations contained both types of cells. In addition, they contained cells with previously undescribed intermediate CMTC phenotypes. The percentage of each phenotype present in hybrid populations was determined for sixteen hybrid clones. Seven clones were found which appeared transformed on the basis of their CMTC pattern. The others were comprised of various proportions of all the cell types described. Repeated quantitation of the proportions of these types in the hybrid populations showed them to be stable with time in culture. Growth in vitro of the hybrid clones was assayed by determining their saturation densities, their plating efficiencies on plastic and their colony-forming abilities in soft agar. In vitro growth of a cell population was found to be directly proportional to the percentage of cells in the population which showed the greatly diminished CMTC pattern which has been described for transformed cells. This is strong evidence that a greatly reduced CMTC is associated with transformed behavior, especially the increased capacity of transformed cells for in vitro growth.
The relationship between verbal report and cardiac orienting response measures of speech discrimination in adult listeners was examined in two experiments using stimuli and paradigms previously employed in studies of infant speech perception. The results of Experiment I revealed that all listeners, those who reported discrimination of a synthetic [ba-ga] change (Group D) as well as those who did not (Group ND), demonstrated cardiac discrimination of the stimulus shift. However, this pattern of cardiac activity, both at stimulus onset and the shift, was found to be different in these two groups of listeners. Experiment II replicated the Group D results using a slightly different cardiac paradigm and quasinatural speech syllables. The implications of these findings for developmental research on speech perception with older infants, children, and populations with language disorders are discussed.

Mouse embryo cells can be plated directly from single-cell suspensions of fresh embryos. For randomly bred Swiss mice, there is an increase in plating efficiency as a function of gestational age. Colony-forming units appear at about day 10 and there is an exponential increase in colony-forming ability up to day 16, after which there is no further increase.
Boys in a boarding-school given inactivated influenza-A or influenza-B vaccine have been observed during a simultaneous outbreak of influenza due to A/Port Chalmers, B/Hong Kong, and B/Intermediate strains. Influenza-B vaccine conferred substantial protection, the attack-rates in boys given B vaccine being 24% compared with 45% in a control group. A/Hong Kong vaccine, by preventing infection during the previous influenza A/England outbreak, left the boys vulnerable to influenza A/Port Chalmers. Influenza-A haemagglutination-inhibiting (H.I.) antibody acquired by natural infection was associated with more protection than H.I. antibody induced by vaccination. This difference was not seen with influenza-B H.I. antibody.
An analysis was made of the severity of over 8000 cases of whooping cough notified from October 1974 to March 1975. Ten per cent of these patients were admitted to hospital and there were 10 deaths. Severity was directly related to age, the disease being most severe in children under 1 year, especially those under 6 months. There was some evidence that previous vaccination reduced the severity of the disease, but the vaccination programme does not cover very young children, and for them whooping cough is a dangerous disease.

Categorical discrimination of place of articulation in the stimuli [dae] and [gae] was investigated in three- to four-month-old infants using a 20/20 cardiac orienting response paradigm. In this procedure infants were presented with 20 tokens of one syllable followed immediately by 20 tokens of a change stimulus. Each infant received a between-category shift, a within-category shift, and a no-shift control condition, with the order of conditions completely counterbalanced across subjects. Discrimination was indexed by an orienting response to the onset of the second 20 stimuli. The data were analyzed for evidence of categorical discrimination both within subjects (each condition pooled across orders of presentation) and between subjects, as is typical of infant-categorical discrimination studies. Both sets of results revealed that infants discriminate the cues for place of articulation categorically. These findings demonstrate the generality of the infant categorical discrimination observed with the nonnutritive operant sucking paradigm and indicate the usefulness of the heart rate paradigm employed in the present study for the collection of within-subject speech discrimination data across a wide-range of subject populations.
