Studies on the inhibitory effect of tetrandrine on silica-induced fibrogenesis in vitro.

Two cell culture systems composed of rat peritoneal macrophages and 2BS fibroblasts were used to study the inhibitory action of tetrandrine on collagen and glycosaminoglycan synthesis. The activation of silica-treated macrophage supernatants stimulated collagen synthesis by 2BS fibroblast cells. Tetrandrine had an inhibitory effect on the biosynthesis of collagen and glycosaminoglycans. The increase in the content of hydroxyproline, representing collagen, was verified by comparing it with that of DNA.

Resibufogenin-induced delayed afterdepolarization in rabbit heart in situ.

The pharmacological effects of resibufogenin (RGB), an effective cardiotonic obtained from a famous traditional Chinese medicine, on the electrical activity of the rabbit heart (n = 12) in situ were studied by using a new technique of recording monophasic action potential (MAP). RBG (0.3 mg/kg IV) induced DAD in 3/12 animals. DAD appeared within the first 0.5 to 1 min after RBG and lasted about 20-30 min. The position of DAD was in the end of phase 3 of MAP. The DADs of rabbit epicardium showed a transient, regular and phasic depolarization process. The triggered arrhythmias were not recorded in these experiments, as amplitudes of DAD were lower and did not reach the threshold. The results indicate that DADs are induced by RBG at first time in situ in rabbits. The characteristics of DAD were similar to those previously described in single cardiac fibers in vitro.

Characterization of hemopoietic activities in media conditioned by a murine marrow-derived adherent cell line, B.Ad.

The hemopoietic activities present in medium conditioned by a murine bone marrow-derived adherent cell line (B.Ad) have been studied. B.Ad-conditioned medium stimulated neutrophil, neutrophil-macrophage, and macrophage colonies in agar cultures of bone marrow cells and 90% of this activity was neutralized by antimacrophage colony-stimulating factor (anti-M-CSF). The conditioned medium supported the generation and/or maintenance of spleen colony-forming units (CFU-S) in liquid cultures and synergized with multilineage colony-stimulating factor (Multi-CSF; IL-3) to stimulate colony formation by day-3 post-5-fluorouracil (FU)-treated bone marrow cells. When used as feeder layers, B.Ad cells stimulated erythroid colony-forming units (CFU-E) and markedly enhanced erythroid burst-forming units (BFU-E) stimulation more than did maximal Multi-CSF (IL-3) and Epo stimulation. No CFU-E- or BFU-E-stimulating activities were detected in medium conditioned by B.Ad cells. Similarly, B.Ad-conditioned medium was unable to stimulate Multi-CSF (IL-3) or granulocyte-macrophage (GM)-CSF-dependent cell lines. The data suggest that medium conditioned by this bone marrow-derived adherent cell line contains M-CSF and other factors not detectable as CSFs that either directly or by means of a synergistic mechanism are able to stimulate CFU-S and colony-forming cells (CFC).

Release of platelet activating factor and its involvement in the first phase of carrageenin-induced rat foot edema.

Platelet activating factor (PAF), a potent lipid-like vasoactive agent, induced rat foot edema when it was injected subplantarly. The edema reached its maximum 1 h after PAF challenge. Indomethacin did not inhibit the peak edematous response whereas both PAF antagonists, kadsurenone and L-652,731, inhibit the PAF-induced rat foot edema (PFE). Both PAF antagonists also partially block the first phase of the carrageenin-induced rat foot edema (CFE). Using the inhibition of [3H]PAF receptor binding to prepared rabbit platelet membranes, release of PAF or PAF-like materials in carrageenin-injected rat hindpaw was observed. These results suggest that the released PAF or PAF-like materials together with the released histamine and kinin evoke the first phase hindpaw edema in the rats. Indomethacin or PAF antagonist, administered alone, does not block the first phase or the second phase of CFE, respectively. However, PAF antagonist potentiated the inhibitory effects of indomethacin suggesting that the released PAF may also be involved in the biosynthesis of prostaglandins to initiate the second phase of rat CFE.
