Production of interleukin 6 from human liver cell lines: production of interleukin 6 is not concurrent with the production of alpha-fetoprotein.

The production of interleukin (IL) 6 from six human liver cell lines, including Chang liver, HLF, HLE, HepG2, PLC/PRF/5, and HuH-7, was investigated using enzyme-linked immunosorbent assay and Northern blot analysis. When cells were cultured in the presence of 12-O-tetradecanoylphorbol-13-acetate, significant amounts of IL6 were detected in the culture supernatants of Chang liver cells, HLF cells, and HLE cells. However, IL6 was not detected in the culture supernatants from HepG2 cells, PLC/PRF/5 cells, or HuH-7 cells which had been treated similarly. To further investigate the production of IL6, expression of the IL6 gene was studied. Results of Northern blot analysis using IL6 complementary DNA as a probe showed that the induction was initiated at the mRNA level. Moreover, IL6 mRNA was also induced by IL1 beta and tumor necrosis factor but not by a calcium ionophore (A23187) or IL6 itself in Chang liver cells. This is the first study to demonstrate the production of human IL6 in liver cells. Furthermore, when the production of alpha-fetoprotein (AFP) from the liver cell lines was examined, the three that were able to produce IL6 failed to produce AFP, whereas the other three cell lines succeeded in producing AFP. These observations may indicate the heterogeneous origin of the liver cell lines.

Muroctasin [MDP-Lys(18)] augments the production of granulocyte colony-stimulating factor (G-CSF) from human peripheral blood mononuclear cells in vitro.

N2-[(N-acetylmuramoyl)-L-alanyl-D-isoglutaminyl]-N6-stearoyl-L-Lysine (MDP-Lys(L18), muroctasin) is an immunopotentiating substance. Neutrophilia and elevated levels of colony-stimulating factor (CSF) in peripheral blood were previously found after the administration of this compound in both mice and humans. To specify the type of CSF and to elucidate the mechanisms of the neutrophilia, we cultured human peripheral blood mononuclear cells (PBMC) in the presence of muroctasin and measured the levels of granulocyte CSF (G-CSF) in the culture supernatants using our sensitive enzyme-linked immunosorbent assay. G-CSF is an active hematopoietic growth factor specific for cells of a neutrophilic lineage, and muroctasin was found to significantly augment the G-CSF production from PBMC in vitro (P less than 0.01). Furthermore, production of G-CSF from human PBMC in the presence of muroctasin was also supported by the Northern blot analysis using cDNA encoding G-CSF as a probe.

[A case of retroperitoneal cavernous lymphangioma with the content of chylous fluid].

We report a case of retroperitoneal lymphangioma in a 26-year-old man whose chief complaint was lumbar discomfort. Ultrasonography revealed pararenal cyst on his left side. At the operation, we discovered a cystic tumor between peritoneum and Gerota's fascia. Pathological study indicated cavernous lymphangioma containing chylous fluid. There has been no recurrence for a year after the operation.

Both granulocyte-macrophage colony-stimulating factor and monocytic colony-stimulating factor are produced by the human T-cell line, HUT 102.

We present the first report of a T-helper cell line, HUT 102, constitutively producing both granulocyte-macrophage colony-stimulating factor (GM-CSF) and monocytic colony-stimulating factor (M-CSF), as detected by sensitive enzyme-linked immunosorbent assays and Northern blot analysis. However, neither amplification nor structural change of the GM-CSF and M-CSF genes was detected by Southern blot analysis. In the case of HUT 102, in which human T-lymphotropic leukemia virus type I (HTLV-I) is integrated, the viral protein, which acts as a trans-acting transcriptional activator, may induce the production of both GM-CSF and M-CSF.

Expression of cytokine genes in hematological malignancies.

A substantial number of leukemic blast colonies were formed when conditioned medium of human bladder carcinoma cell line 5637 was added as a stimulator. Recombinant colony-stimulating factor (CSF) also stimulated leukemic blast cell proliferation, leading to colony formation. Furthermore, serum CSF levels in some patients with acute myelogenous leukemia (AML), as detected by sensitive enzyme-linked immunosorbent assay (ELISA), were high. These observations prompted us to study further the expressions of hematopoietic growth factor genes. Granulocyte-macrophage CSF (GM-CSF) mRNA was detected in the leukemic blast cells from about 30% of patients with AML by Northern blot analysis using strict hybridization conditions with or without in vitro blast cell enrichment. These findings suggest that the expression of cytokine genes including the GM-CSF gene reflects in vivo phenomena, although no clear relationship between expression of the genes and serum CSF level has been established. Gene encoding tumor necrosis factor alpha (TNF-alpha), lymphotoxin (LT) and transforming growth factor beta (TGF-beta) were sometimes expressed in some malignant hematological cell lines and also some fresh leukemic cells.

[Acute epididymitis in Schoenlein-Henoch purpura: a case report of emergency exploration].

A case of acute scrotal swelling with pain in a 4-year-old boy suffering from Schoenlein-Henoch purpura is reported. Immediate scrotal exploration revealed epididymal ecchymosis and slightly edematous spermatic cord on the right side. Spermatic cord torsion was not recognized. The testis had a normal appearance. Convalescence was uneventful. Scrotal lesion disappeared within a week. Schoenlein-Henoch purpura is systemic vasculitis, which is noticed on any site of the body including male external genitalia. Several reports mainly on the testicular lesion have been made, but the epididymal findings were obscure. This case showed the prominent lesion on the epididymis. The anatomical architecture of the epididymis suggested that the principal inflammatory change of scrotal signs of Schoenlein-Henoch purpura is epididymitis.

[Morphologic study of microscopic hematuria using phase contrast microscopy].

Phase contrast microscopic examination of the urine has been recently reported to be valuable in predicting whether hematuria is due to glomerulonephritis or not. Phase contrast microscopy can differentiate glomerular red cells from nonglomerular ones since the former varies in size and shape with distortion, whereas the latter is uniform in size and shape. Urinalysis of 217 cases of microscopic hematuria was performed at our Hospital. Red blood cells were classified into two types, glomerular and nonglomerular type. Ninety percent of the cases of bladder leukoplakia as well as all cases of glomerulonephritis had glomerular type red blood cells.

[A case of metastatic renal cell carcinoma with complete remission achieved by interferon therapy].

A case of metastatic renal cell carcinoma in which complete remission was achieved using Interferon therapy is reported. A 67-year-old male was found to have extensive paraaortic lymph node metastasis while undergoing left nephrectomy following diagnosis of 1-renal cancer. Pathological diagnosis of the node was metastasis of clear cell type renal cell carcinoma. Interferon (Hu IFN-alpha 2: Sch 30500; Yamanouchi Pharmaceutical Co.) therapy was started 34 days after surgery. The Interferon was administered i.m. at a dose of 1 X 10(6) units per injection, 3 times a week, and was continued up to a total dose of 236 X 10(6) units. Response of the metastatic lymph nodes was examined using CT scan. Marked response was observed 4 weeks after starting the Interferon therapy, and complete disappearance of the tumors was obtained 5 months after Commencement of therapy. The duration of complete response has lasted 4 months. The toxic manifestations of the Interferon therapy were fever and loss of appetite.

Three-dimensional holographic display of images of otological specimens.

Three-dimensional displays of anatomical structures and clinical findings are very persuasive and instructive. Using multiplex holograms, we designed a display of three-dimensional images of otological specimens. Multiplex holograms, reported by Cross of the United States in 1975, enable reconstruction of three-dimensional moving images and are used for artistic display as well as for teaching in medicine and general education. Multiplex holograms were recorded in a two-step process. The first step is to make a series of original cine-pictures of an object from different horizontal directions, rotating it on a turntable. In the second step, one frame of the original film is recorded on a narrow strip hologram. All frames of the original film are recorded one after another and a complete multiplex hologram can be synthesized. In the reconstruction stage, the multiplex hologram is formed into a cylinder and illuminated from below by a small white light source. Reconstructions of the three-dimensional bright images of the object inside the cylindrical holographic screen are shown.
