Synthesis of optically active beta-lactams using the photochemical reaction of 3-deazauridines.

The photochemical reaction of 3-deazauridines was investigated with the intention of obtaining chiral beta-lactams. Irradiation of appropriately protected 3-deazauridines at greater than or equal to 300 nm gave 2-(protected D-ribofuranosyl)-3-oxo-2-azabicyclo[2.2.0]hex-5-enes as a mixture of diasteromers, which were chromatographically separable. Treatment of each isomer with acid, such as trifluoroacetic acid, afforded the 5-oxo derivatives which by methanolysis gave rise to the corresponding chiral 4-methoxy-carbonylmethyl-1-(protected D-ribofuranosyl)azetidin-2-ones in good yields.

Synthesis of 1-beta-D-ribofuranosyl-1,2-dihydropyrimidin-2-one derivatives and their biological activities.

Several alkyl substituted 1-beta-D-ribofuranosyl-1,2-dihydropyrimidin-2-one derivatives were synthesized by the method of stannic chloride-catalyzed glycosidation method to elucidate their inhibitory activity of cytidine deaminase and also their antitumor activities in vitro and in vivo. Alkyl substitution at position 4 or 6 of the derivatives decreased their inhibitory activity for cytidine deaminase and also decreased antitumor activity against L1210 cells in vitro.

Synthesis of desmosterol and epidesmosterol from hyodeoxycholic acid.

A new synthesis of desmosterol was described using hyodeoxycholic acid (3alpha,6alpha-dihydroxy-5beta-cholanic acid) as a starting material. Epidesmosterol (3alpha-hydroxycholesta-5,24-diene) was also synthesized for the first time from the same starting material.

Curative effects of 1alpha-hydroxycholecalciferol on calcium metabolism and bone disease in patients with chronic renal failure.

Synthetic 1alpha-hydroxycholecalciferol (1alpha-OH-D3) was given intravenously in a dose of 2.5-10 mug per day to three patients with chronic renal failure. As little as 10 mug of 1alpha-OH-D3 daily for a week improved intestinal calcium absorption to a normal level, raised serum calcium, and reduced serum alkaline phosphatase. Severe rickets which had not responded to large amounts (greater than 200 mg in total) of vitamin D2 was markedly cured with 2.5 mug of 1alpha-OH-D3 given daily for 3 weeks. These clinical data hold promise that is certainly useful in the improvement of intestinal malabsorption of calcium and bone diseases in renal failure.
