A pre-operative mydriatic regime using a single application of tropicamide Novel Ophthalmic Delivery System and guttae phenylephrine 10%.

Tropicamide Novel Ophthalmic Delivery System (NODS) and guttae (g.) phenylephrine 10% was applied once, 2 hours before cataract surgery, as an alternative to our standard pre-operative regime of g. cyclopentolate 1% and g. phenylephrine 10% used four times before surgery. The two treatment groups were compared using a randomised, prospective, observer-masked study design. The horizontal pupil diameter was significantly smaller using the new regime but the per-operative decrease in diameter was not significantly different, suggesting that surgical miosis was not influenced by reducing the number of mydriatic applications. There was no difference in the subjective grading of the pupil, suggesting that adequate mydriasis suitable for cataract surgery was obtained using the alternative regime. No major complications were documented in relation to the use of NODS. We discuss why less intensive mydriasis is likely to be as efficacious and describe its potential advantages.

The notochord and floor plate guide growth cones in the zebrafish spinal cord.

Elimination of the floor plate, a row of distinctive cells at the ventral midline of the spinal cord, dramatically increased the frequency of errors made by specific growth cones in the zebrafish embryo. This demonstrated that the floor plate participated in guiding specific growth cones at the ventral midline of the spinal cord. However, since a significant proportion of these growth cones followed their normal pathway despite the absence of the floor plate, we hypothesized that a second source of cues must exist at or near the ventral midline. We tested whether the notochord, which is located just ventral to the spinal cord, was an additional source of pathfinding cues by eliminating it prior to axonogenesis. Laser ablation of the notochord in wildtype embryos increased errors by spinal growth cones. Likewise, spinal growth cones made errors in no tail mutants that are missing the notochord but not the floor plate. These results demonstrate that the notochord also participates in guiding growth cones at the ventral midline. Furthermore, removal of both the floor plate and notochord resulted in a higher error rate than removal of either one alone. These results suggest that the notochord and floor plate normally act in concert to guide spinal growth cones at the ventral midline.

Axonal outgrowth within the abnormal scaffold of brain tracts in a zebrafish mutant.

The role of specific axonal tracts for the guidance of growth cones was investigated by examining axonal outgrowth within the abnormal brain tracts of zebrafish cyclops mutants. Normally, the earliest differentiating neurons in the zebrafish brain establish a simple scaffold of axonal tracts. Later-developing axons follow cell-specific pathways within this axonal scaffold. In cyclops embryos, this scaffold is perturbed due to the deletion of some ventromedial neurons that establish parts of the axonal scaffold and the development of an abnormal crease in the brain. In these mutant embryos, the growth cones projected by the neurons of the nucleus of the posterior commissure (nuc PC) are deprived of the two tracts of axons that they sequentially follow to first extend ventrally, then posteriorly. These growth cones respond to the abnormal scaffold in several interesting ways. First, nuc PC growth cones initially always extend ventrally as in wild-type embryos. This suggests that for the first portion of their pathway the axons they normally follow are not required for proper navigation. Second, approximately half of the nuc PC growth cones follow aberrant longitudinal pathways after the first portion of their pathway. This suggests that for the longitudinal portion of the pathway, specific growth cone/axon interactions are important for guiding growth cones. Third, although approximately half of the nuc PC growth cones follow aberrant longitudinal pathways, the rest follow normal pathways despite the absence of the axons that they normally follow. This suggests that cues independent of these axons may be capable of guiding nuc PC growth cones as well. These results suggest that different guidance cues or combinations of cues guide specific growth cones along different portions of their pathway.

Does topical flurbiprofen affect the pupillary response to acetylcholine?

Pre-operative topical non-steroidal anti-inflammatory drugs (NSAIDs) are known to be effective in maintaining pupillary dilation during cataract surgery. However, previous studies have suggested that pre-treatment with these agents interferes with the miosis produced by intraoperative acetylcholine injection. This study was designed to determine whether the pupillary response to acetylcholine injection is reduced following pre-operative topical flurbiprofen 0.03%. Pupil diameters were measured with a millimetre rule, and the measurements verified using computerised image analysis of digitised video images. Eyes treated with flurbiprofen were compared with controls. There was no significant difference in pupillary diameter before corneal incision, but following aspiration of soft lens matter and insertion of intraocular lens the pupil was significantly larger in the flurbiprofen group (p = 0.002). At 1.0, 2.5 and 5.0 minutes following injection of acetylcholine no significant difference in the degree of miosis was detected. This study confirms that topical flurbiprofen maintains dilation during cataract surgery, and does not find any evidence to suggest that intra-operative response to acetylcholine is reduced.

Centrally administered mu- and delta-opioid agonists increase operant responding for saccharin.

In previous reports, ICV administration of selective mu- or delta-opioid receptor agonists was found to stimulate the intake of saccharin and salt solutions in nondeprived rats. In the present study, we measured the effects of selective mu-, delta-, and kappa-agonists on operant responding for saccharin. The selective mu-agonist [D-Ala2,N-Me-Phe4,Gly5-ol]-enkephalin (DAMGO) and the selective delta-agonist [D-Thr2]-leucine enkephalin-Thr (DTLET) increased responding, whereas the kappa-agonist dynorphin A analog kappa ligand (DAKLI) had no significant effect. These results agree with previous studies on saccharin and salt intake and are consistent with the possibility that the effects of opioids on the intake of these fluids are mediated via enhancement of activity in brain reward pathways.

Cutaneous granuloma with ataxia telangiectasia--a case report and review of literature.

A patient is reported who had ataxia telangiectasia with multiple cutaneous lesions mainly on the limbs, which showed atrophy and scarring. Histopathology of these skin lesions showed tuberculoid granulomas without frank collagen necrobiosis in the dermis. Chromosomal analysis revealed a translocation between 7 and 14 as well as deletion of chromosomes 4 and 6.

Axonal trajectories and distribution of GABAergic spinal neurons in wildtype and mutant zebrafish lacking floor plate cells.

The role of the midline floor plate cells in the neuronal differentiation of the spinal cord was examined by comparing putative GABAergic neurons in wildtype zebrafish embryos with those in cyc-1 mutant embryos. The mutation produces a pleiotropic recessive lethal phenotype and is severe in rostral brain regions, but its direct effect in the caudal hindbrain and the spinal cord is apparently restricted to the depletion of the midline floor plate cells. In wildtype embryos, an antibody against the neurotransmitter GABA labeled the cell bodies, axons, and growth cones of three classes of previously identified neurons; dorsal longitudinal neurons (DoLA), commissural secondary ascending neurons (CoSA), and ventral longitudinal neurons (VeLD). A novel ventral cell type, Kolmer-Agduhr (KA) neurons, was also labeled. In the cyc-1 mutant, abnormalities were observed in some, but not all, of the GABAreactive CoSA, VeLD, and KA axons, while the axonal trajectories of DoLA neurons were not affected. Furthermore, the number of KA cells was reduced in the mutant while the numbers of the other GABAreactive cells were unperturbed. These observations corroborate our earlier hypothesis that the floor plate cells are one of several guidance cues that direct axonal outgrowth near the ventral midline of the spinal cord. They also suggest that the floor plate cells may play a role in the cellular differentiation of the spinal cord of zebrafish embryos.

Industrial ecology: concepts and approaches.

Industrial ecology is a new approach to the industrial design of products and processes and the implementation of sustainable manufacturing strategies. It is a concept in which an industrial system is viewed not in isolation from its surrounding systems but in concert with them. Industrial ecology seeks to optimize the total materials cycle from virgin material to finished material, to component, to product, to waste product, and to ultimate disposal. To better characterize the topic, the National Academy of Sciences convened a colloquium from which were derived a number of salient contributions. This paper sets the stage for the contributions that follow and discusses how each fits into the framework of industrial ecology.

Industrial ecology.

Industrial ecology addresses issues that will impact future production, use, and disposal technologies; proper use of the concept should reduce significantly the resources devoted to potential remediation in the future. This cradle-to-reincarnation production philosophy includes industrial processes that are environmentally sound and products that are environmentally safe during use and economically recyclable after use without adverse impact on the environment or on the net cost to society. This will require an industry-university-government round table to set the strategy and agenda for progress.

A specific brain tract guides follower growth cones in two regions of the zebrafish brain.

Neurons of the nucleus of the posterior commissure (nuc PC), an identifiable cluster of neurons in the embryonic zebrafish brain, project growth cones ventrally along the posterior commissure to the anterior tegmentum where the PC intersects two longitudinal tracts, the tract of the postoptic commissure (TPOC) and the medial longitudinal fasciculus (MLF). Once at the intersection, nuc PC growth cones turn posteriorly onto the TPOC in the dorsal tegmentum and follow it to the hindbrain. Previously we showed that in the absence of the TPOC, nuc PC growth cones often extended along aberrant pathways suggesting that fasciculation, that is, contact with TPOC axons is an important factor in guiding growth cones along their normal pathway. However, a significant number of nuc PC growth cones also followed their normal pathway suggesting that cues associated with the dorsolateral tegmentum, independent of the TPOC, can also guide nuc PC growth cones. We have now confirmed using electron microscopy that nuc PC growth cones fasciculate with axons in the TPOC. In the absence of the TPOC, the nuc PC growth cones that extend along their normal pathway do so in contact with dorsolateral neuroepithelial cells. This suggests that cues associated with these cells can also guide the nuc PC growth cones. Furthermore, in the absence of the TPOC axons, these growth cones now inappropriately turn onto axons that normally intersect the TPOC near the border of the midbrain and hindbrain, that is, at a second intersection of tracts. This suggests that fasciculation with TPOC axons may also guide nuc PC growth cones in this second region of the brain.

Endophotocoagulation in vitrectomy with a carbon dioxide laser.

A continuous-wave carbon dioxide laser operating at a wavelength of 10.6 microns has been applied transvitreally by means of a miniature articulating arm and intraocular probe to produce chorioretinal lesions in albino white rabbit eyes. The advantages of using a carbon dioxide laser are its lack of pigment dependence, its ability to act both as a photocoagulator and as a phototransector, and its ability to deposit its energy in a well-defined area without adverse effect on neighboring ocular tissue. The main disadvantage of the carbon dioxide laser in vitreoretinal surgery is that in a fluid medium, such as the vitreous, the laser tip must be placed in direct physical contact with the target tissue to obtain a reaction.
