Activated CD8+ T cells are involved in elimination of Candida albicans from the livers of mice.

We have investigated the course of infection of Balb/c mice with Candida albicans. After intravenous infection of mice, the yeasts were evenly distributed in the liver sections. There was a 90% reduction in the yeasts found within 7 days after infection. This reduction was accompanied by an increase in the number of phagocytic cells (Mac-1+, Gr-1+) in cellular infiltrates around the yeast cells. Furthermore, both CD4+ and CD8+ T lymphocytes were found in these pseudogranulomata, with the CD8+ cells in the periphery. Peak amounts of the cell types investigated were found at days 5 and 6 post infection, with an increased expression of receptors for interleukin 2 (IL-2R) on the surface of CD8+ cells. At day 14 of infection, the same levels as those in control mice were reached. At this time, the yeasts were virtually eliminated from the liver. By using simultaneous detection of cell surface markers and yeast cells in immunohistology, these data demonstrate the close association of fungal cells, phagocytes, TH and TC cells in pseudogranulomata during elimination of Candida albicans from the livers of mice.

Exploratory celiotomy in 200 nontraumatized dogs and cats.

Two hundred dogs and cats undergoing exploratory celiotomy for reasons other than trauma were grouped by disease classification and by body system affected. There were 61 animals in which infection or inflammation predominated, 63 animals with neoplasia, and 76 animals with noninflammatory and non-neoplastic conditions. Body systems affected were digestive, lymphatic, urinary, reproductive, other, and undetermined. Seventy-three percent of the animals survived the hospitalization period; survival rates for animals with infection or inflammation, neoplasia, and other disorders were 69%, 60%, and 86%, respectively. Within the same groups, the exploratory celiotomy provided strictly diagnostic information in 72%, 79%, and 24% of the animals, and surgical treatment was provided to 28%, 21%, and 58% of the animals, respectively. Intraoperative cytologic and histologic diagnoses were consistent in 78% of the animals; the consistency rates for animals with infection or inflammation, neoplasia, and other disorders were 81%, 88%, and 59%, respectively. Complications after surgery were observed in 30% of the animals, with 60% of the complications disease related. Complications were observed in six animals with reproductive disorders (67%), six animals with urinary disease (46%), 35 animals with digestive disease (29%), and three animals with lymphatic disease (13%).

Thoracic ultrasonography technique in newborn calves and description of normal and pathological findings.

The aim of this study was to establish whether thoracic ultrasonography is a viable screening method for detecting respiratory disorders in newborn calves. Thoracic ultrasonography was performed on 66 newborn calves. There were 10 normal control animals with healthy lungs and 56 newborn calves suffering from respiratory disorders such as lung atelectasis, aspiration of amniotic fluid, pulmonary edema, pneumonia, and lung abscesses. Ultrasonography of the lung enabled rapid classification of the severity, extent and type of pulmonary lesion found, thus supplementing the clinical examination. Processes deep to the pleura were not amenable to thoracic diagnosis by ultrasonography. This is due to the inability of sound waves to penetrate normally ventilated lung. Despite this problem, ultrasonography of the lung is suitable for assessment of clinically unclear respiratory disorders in calves.

p-Nitrophenetole deethylase activity of rat liver microsomes entrapped in polyelectrolyte capsules.

Liver microsomes from phenobarbital induced rats are entrapped in capsules prepared from polyelectrolytes. A comparative analysis of the deethylase activity against p-nitrophenetole by encapsulated and freely suspended microsomes is carried out. The pH optimum occurs at about 7.2 for encapsulated as well as free microsomes. The pH activity profile of encapsulated microsomes, however, is strongly flattened. The maximal velocity of entrapped microsomes is about a quarter of that of free microsomes. The Michaelis-Menten constants are virtually equal. Despite of the lowered activity of encapsulated microsomes this kind of immobilization of enzymes may be useful in biotechnology and medicine because mild immobilization conditions like room temperatures and aqueous solutions are realized.

Detection of Tropheryma whippelii DNA (Whipple's disease) in faeces.

To date the diagnosis of Whipple's disease is based mainly on the histopathological analysis of duodenal biopsies since Tropheryma whippelii cannot be cultured in vitro. We investigated the possibility to diagnose Whipple's disease by detection of bacterial DNA in faces. Nested polymerase chain reaction with amplification of part of the 16S rRNA gene of this bacterium in DNA extracted from faeces of a patient with Whipple's disease was performed. Sequencing of the polymerase chain reaction product revealed the sequence of Tropheryma whippelii. We conclude that Whipple's disease will be able to be diagnosed non-invasively by DNA analysis from the faeces as soon as more specific sequences of this bacteria are known.

[Cutaneous side effects of gold therapy. Clinical and histologic results].

Based on our own observations in 40 patients and a review of the literature the clinical and histological aspects of dermatitis caused by gold therapy are discussed. There is a broad spectrum of cutaneous side effects following gold therapy. In most of the patients a nonspecific dermatitis can be observed. The skin lesions may also mimic lichen planus, pityriasis rosea or various forms of eczema, obscuring early diagnosis. Histologically four types of reactions can be differentiated including a dermatitis type, vasculitis type, a lichenoid and an urticarial type.
