Identification of endogenous outward currents in the human embryonic kidney (HEK 293) cell line.

Human embryonic kidney cells (HEK 293) are widely used as an expression system in studies of ion channels. However, their endogenous ionic currents remain largely unidentified. To characterize these currents, we performed patch clamp experiments on this expression system. In whole-cell voltage clamp mode, the HEK 293 cells showed mainly outward currents using physiological concentrations of Na+ and K+ and symmetric concentrations of Cl- (150 mM) across the plasma membranes. K+ currents contributed to a small portion of these outward currents, since a shift of the reversal potentials of only approximately 20 mV was seen with a change of extracellular K+ concentration from 3 to 150 mM. In contrast, the reversal potential shifted approximately 25 mV when extracellular Cl- was reduced to 50 mM, indicating that most of the outward currents are carried by Cl-. In inside-out patches, several distinct Cl- currents were identified. They were: (1) 350 pS Cl- current, which was voltage-activated and had a moderate outward rectification; (2) 240 pS Cl- current with a weak outward rectification; and (3) 55 pS Cl- current, which was voltage-activated, sensitive to DIDS, and showed a strong outward rectification. Activation of these Cl- currents did not require an elevation of free Ca2+ level in the cytosol. Besides these three currents, we observed two other Cl- currents with much smaller conductances (25 and 16 pS, respectively). Two different K+ currents were seen in the HEK 293 cells, with one of them (125 pS) showing inward rectification and the other (70 pS) outward rectification. Moreover, a 50 pS cation channel was recorded in these cells. The presence of a variety of ion channels in the HEK 293 cells suggests that a great precaution needs to be taken when this expression system is used in studies of several similar ion channels.

Polyploid formation created unique avenues for response to selection in Gossypium (cotton).

A detailed restriction fragment length polymorphism map was used to determine the chromosomal locations and subgenomic distributions of quantitative trait loci (QTLs) segregating in a cross between cultivars of allotetraploid (AADD) Gossypium hirsutum ("Upland" cotton) and Gossypium barbadense ("Sea Island," "Pima," or "Egyptian" cotton) that differ markedly in the quality and quantity of seed epidermal fibers. Most QTLs influencing fiber quality and yield are located on the "D" subgenome, derived from an ancestor that does not produce spinnable fibers. D subgenome QTLs may partly account for the fact that domestication and breeding of tetraploid cottons has resulted in fiber yield and quality levels superior to those achieved by parallel improvement of "A" genome diploid cottons. The merger of two genomes with different evolutionary histories in a common nucleus appears to offer unique avenues for phenotypic response to selection. This may partly compensate for reduction in quantitative variation associated with polyploid formation and be one basis for the prominence of polyploids among extant angiosperms. These findings impel molecular dissection of the roles of divergent subgenomes in quantitative inheritance in many other polyploids and further exploration of both "synthetic" polyploids and exotic diploid genotypes for agriculturally useful variation.

PPAR-gamma agonists inhibit production of monocyte inflammatory cytokines.

The peroxisome proliferator-activated receptor-gamma (PPAR-gamma) is a member of the nuclear receptor family of transcription factors, a large and diverse group of proteins that mediate ligand-dependent transcriptional activation and repression. Expression of PPAR-gamma is an early and pivotal event in the differentiation of adipocytes. Several agents that promote differentiation of fibroblast lines into adipocytes have been shown to be PPAR-gamma agonists, including several prostanoids, of which 15-deoxy-delta-prostaglandin J2 is the most potent, as well as members of a new class of oral antidiabetic agents, the thiazolidinediones, and a variety of non-steroidal anti-inflammatory drugs (NSAIDs). Here we show that PPAR-gamma agonists suppress monocyte elaboration of inflammatory cytokines at agonist concentrations similar to those found to be effective for the promotion of adipogenesis. Inhibition of cytokine production may help to explain the incremental therapeutic benefit of NSAIDs observed in the treatment of rheumatoid arthritis at plasma drug concentrations substantially higher than are required to inhibit prostaglandin G/H synthase (cyclooxygenase).

Increase in interphotoreceptor matrix gelatinase A (MMP-2) associated with age-related macular degeneration.

Matrix metalloproteinases have increasingly been shown to be associated with diseases involving neovascularization and/or abnormal cellular migration or proliferation. A number of diseases of this type affect the retina. In this study, the activity of gelatinase A (MMP-2), the most abundant matrix metalloproteinase in IPM (interphoto receptor matrix) and vitreous, was measured with respect to age in normal human donor eyes and compared to donors with age-related macular degeneration. IPM and vitreous were obtained from a total of 88 human donors. Samples for electrophoresis were normalized for protein content and subjected to quantitative gelatin zymography. The zymograms were scanned and then digitized and quantitated using the NIH 'Image' program. There was not a statistically significant change in the level of gelatinase A in IPM or vitreous as a function of age, although a slight downward trend was found in the total gelatinase A activity within the normal population. Likewise, when comparing normal and age-related macular degeneration donors, there was not a significant difference in the gelatinase A level in vitreous or in retina-associated IPM. However, the level of gelatinase A was nearly doubled specifically in retinal pigment epithelium-associated IPM from eyes with age-related macular degeneration [0.99 +/- 0.09 U mg-1 (56) vs 1.71 +/- 0.28 U mg-1 (14) (mean +/- S.E.M. (number), P < 0.0021; 1 unit = 1.0 ng gelatin cleaved h-1). Gelatinase A may be associated with the changes that occur in age-related macular degeneration, especially the neovascularization which accompanies the exudative ('wet') form of the disease.

Experimental results on mechanisms of action of electrical neuromodulation in chronic urinary retention.

Sacral foramen neuromodulation--initially applied for the treatment of urinary incontinence--has proved to be effective in patients with chronic urinary retention. Thus far, the underlying neurophysiological mechanisms have not been elucidated. In an experimental study on the neurophysiological basis of sacral neurostimulation, one objective was to investigate the mechanisms responsible for initiation of micturition in chronic urinary retention. In ten female cats anesthetized with alpha-chloralose the clinical situation of sacral foramen stimulation was experimentally reproduced by isolated S2 nerve stimulation after L6-S3 laminectomy. Stimulation responses were recorded from the bladder, peripheral nerves, and striated muscles of the foot and pelvic floor. The effect of sudden cessation of prolonged S2 stimulation, during which the bladder was completely inhibited, was evaluated in 70 stimulation sequences in 5 cats. Sacral nerve stimulation induced excitatory and inhibitory effects on the bladder, depending on the frequency and intensity of stimulation. With unilateral S2 stimulation, bladder excitation was best at frequencies of 2-5 Hz and at intensities ranging between 0.8 and 1.4 times the threshold for the M-response of the foot muscle. Inhibition was the dominating effect at frequencies of 7-10 Hz and at intensities exceeding 1.4 times the threshold. Prolonged S2 stimulation above the threshold produced complete bladder inhibition during stimulation but induced strong bladder contractions after sudden interruption of stimulation, with amplitudes being significantly higher than that of spontaneous contractions preceding the stimulation. These results confirm the hypothesis of a "rebound" phenomenon as the mechanism of action for induction of spontaneous voiding in patients with chronic urinary retention.

Further characterization of the latency-associated transcription unit of Marek's disease virus.

Previous studies have identified a large (L) and a small (S) RNA transcript antisense to the MDV homologue of the ICP4 gene of herpes simplex virus (HSV) in cells infected with Marek's disease virus (MDV) and in lymphoblastoid cell lines. In this study the 5' and 3' ends of the L RNA and of the sense ICP4 transcript of MDV were mapped by Northern hybridization and RNase protection assays. The results showed that L RNA is approximately 10.6 kb and that the ICP4 sense transcript is initiated in the region of genomic DNA where the L RNA terminates whereas L RNA is initiated where the ICP4 transcript terminates. L RNA was abundant in chick embryo fibroblasts (CEF) infected with virus strain HPRS16/attenuated whereas S RNA was predominant in CEF infected with oncogenic HPRS16 and in RPL-1 cell line. Results of cycloheximide experiments showed that the ICP4 gene of MDV was transcribed as an immediate-early gene in infected CEF whereas transcription of the L RNA required protein synthesis. Sequencing of cDNA and Northern hybridization using oligonucleotide probes showed that S RNA shared similar intron/exon boundaries as the cDNAs from several cell lines indicating that there might be a relationship between the S RNA and the antisense transcripts that generated the cDNAs.

Identification of H. pylori in saliva by a nested PCR assay derived from a newly cloned DNA probe.

A novel probe was developed from genomic DNA of Helicobacter pylori ATCC type strain 43629. It hybridized with all 73 H. pylori clinical isolates tested but not with any of 183 non-H. pylori DNAs in dot blot hybridization. Typing tests revealed 41 different HaeIII-digestion patterns from 57 H. pylori strains tested. Based on the sequence of the probe, a nested PCR was developed that detected as little as 2 fg of H. pylori DNA or approximately equivalent to one cell. No PCR products were amplified from any of 21 non-H. pylori strains tested. Using this nested PCR, H. pylori DNA was detected in 33 of 45 (73%) saliva samples collected from patients with gastric H. pylori infection. These data suggest that the probe is useful for typing H. pylori and that the nested PCR is a valuable tool for detecting H. pylori DNA in saliva.

Pathogenic characterization of 1-methyl-1-nitrosourea-induced mammary carcinomas in the rat.

The induction of mammary carcinogenesis in the rat by 1-methyl-1-nitrosourea (MNU) is widely used in experimental breast cancer research. In the experiments reported, the Ha-ras codon 12 (ras12) mutation (GGA-->GAA) was used as a molecular marker to address issues of the clonality of carcinomas induced, pathogenetic independence among multiple carcinomas within the same animal and topographic distribution of mutant ras12 carcinomas in different mammary gland chains. In order to determine whether the frequently observed morphologically distinguishable lobules within carcinomas originate from the coalescence of independent lesions or whether cancerous cells within a carcinoma share a common origin, 44 randomly selected MNU-induced mammary carcinomas were genotyped for two to four lobules each for the ras12 mutation. A total of 43 carcinomas out of 44 (97.7%) had concordant ras12 genotypes among the multiple sites within each tumor, which is consistent with the latter possibility. Next, it was observed that as carcinoma multiplicity increased, the discordance rate of ras12 genotypes among multiple carcinomas within the same animal increased in a manner that was in excellent agreement with the expected discordance rate based on an assumption of no pathogenetic association among carcinomas. Furthermore, a significant difference was observed in the occurrence of mutant ras12 carcinomas between the cervical-thoracic and the abdominal-inguinal mammary glands in that three times as many carcinomas were mutant in the former as in the latter glands, whereas the occurrence of wild-type carcinomas was approximately the same in both regions. Taken together, the data are consistent with (i) carcinomas induced by MNU and detected by palpation are monoclonal in origin, (ii) independently-initiated cells emerge as distinct mammary carcinomas in the same animal, and (iii) the anatomical location of the gland may affect the prevalence of mammary carcinomas that harbor a mutant ras12.

Adenovirus-mediated persistent cystic fibrosis transmembrane conductance regulator expression in mouse airway epithelium.

Replication-defective adenovirus (Ad) vectors have been used for gene transfer to the respiratory epithelium of experimental animals and individuals with cystic fibrosis. Studies from several laboratories have suggested that administration of first-generation Ad vectors results only in transient gene expression in the lung, due at least in part to destruction of vector-transduced cells by host cellular immune responses directed against viral proteins and/or immunogenic transgene products. We have constructed new Ad2-based, E1-deleted vectors encoding a weakly immunogenic transgene, the human cystic fibrosis transmembrane conductance regulator (hCFTR) under the control of the cytomegalovirus enhancer-promoter. These vectors contain wild-type E2 and E4 regions. These new Ad/CFTR vectors were instilled into the lungs of immunocompetent C57BL/6, BALB/c, and C3H mice. In vitro cytotoxic T lymphocyte (CTL) analysis indicated the presence of Ad-specific CTLs in treated mice. However, we were not able to demonstrate a CTL response specific for hCFTR. Reverse transcriptase PCR analysis demonstrated that hCFTR mRNA expression continued in all three strains of mice for at least 70 days, the last time point analyzed. The E3 region did not play a significant role in persistence of the Ad/CFTR vectors in the mouse lung. Functional hCFTR expression was also observed in the nasal epithelia of CF mutant mice. These results suggest that long-term expression of hCFTR is possible in the airway epithelia of immunocompetent mice without radical modification of Ad vector and in spite of the presence of CTLs.

Partial restoration of cAMP-stimulated CFTR chloride channel activity in DeltaF508 cells by deoxyspergualin.

Deletion of the codon encoding phenylalanine 508 (DeltaF508) is the most common mutation in cystic fibrosis (CF) and results in a trafficking defect. Mutant DeltaF508-CF transmembrane conductance regulator (CFTR) protein retains functional activity, but the nascent protein is recognized as abnormal and, in consequence, is retained in the endoplasmic reticulum (ER) and degraded. It has been proposed that this retention in the ER is mediated, at least in part, by the cellular chaperones heat shock protein (HSP) 70 and calnexin. We have investigated the ability of deoxyspergualin (DSG), a compound known to compete effectively for binding with HSP70 and HSP90, to promote trafficking of DeltaF508-CFTR to the cell membrane. We show that DSG treatment of immortalized human CF epithelial cells (DeltaF508) and cells expressing recombinant DeltaF508-CFTR partially restored cAMP-stimulated CFTR Cl- channel activity at the plasma membrane. Although there are several possible explanations for these results, one simple interpretation is that DSG may have altered the interaction between DeltaF508-CFTR and its associated chaperones. If this is correct, agents capable of altering the normal functioning of cellular chaperones may provide yet another means of restoring CFTR Cl- channel activity to CF subjects harboring this class of mutations.

Na+/Ca++ exchanger and myocardial ischemia/reperfusion.

Myocardial hypoxia and ischemia are characterized by the depletion of ATP and the development of intracellular acidosis, which alter cellular ionic homeostasis. Specifically, elevated cytosolic free Ca++ concentrations cause cellular injury during hypoxia/ischemia and lead to irreversible myocardial damage during reoxygenation/reperfusion. An increase in the intracellular Na+ concentration has been shown to correlate with Ca++ overload. Although inhibition of Na+/K+ exchange because of decreased ATP production may be involved, it is more likely that intracellular acidosis drives Na+ into the cells via Na+/H+ exchange. Experimental evidence supports the notion that Na+/H+ exchange is primarily responsible for Na+ influx during hypoxia/ischemia. The accumulation of intracellular Na+ may then activate the Na+/Ca++ exchanger causing Ca++ overload. Therefore, the Na+/Ca++ exchanger plays a crucial role in cellular injury during hypoxia/ischemia and in cell death during reoxygenation/reperfusion. In the past few years, the Na+/Ca++ exchanger has been cloned and the structure/function relationship studied intensively. Agents which inhibit the Na+/Ca++ exchanger may have therapeutic potential for the treatment of ischemic heart disease. These advances will greatly accelerate the understanding of the cellular and molecular mechanisms underlying the role of the Na+/Ca++ exchanger in the development of myocardial damage during hypoxia/ischemia and reoxygenation/reperfusion.

Alveolar echinococcosis in China.

DATA SOURCES: All reference data originated from related Chinese- or English-language literature in Chinese journals. STUDY SELECTION: Twenty-three original articles published in 1992-1996 were selected according to the stated purpose and 9 of them were written by myself. DATA EXTRACTION: The present paper dealt with 5 subtopics, i.e. epidemiology, parasitology, pathology, diagnosis, treatment and prognosis. RESULTS: Five hundred and eighty-four patients with alveolar echinococcosis (AE) have been reported from 7 provinces or autonomous regions. Human infection rate was 19.2% or 2.8%, and the morbidity was 2.4%-5.0%. The intermediate hosts of Echinococcus multilocularis included 7 species of rodents and 3 species of livestocks, and the final hosts of that were fox, dog or wolf. Diagnosis of AE was chiefly based on imaging examination (ultrasound and CT) and immunological test. The operative resection rate for liver AE was only 10.5% (27/258). Albendazole was a certain remedy for the chemotherapy of AE, and TCM "Xiao-Bao" pill may be a hopeful drug. CONCLUSIONS: In the past 30 years, a lot of work concerning basic and clinical studies has been done in China and some achievements have been achieved. However, some important problems such as how to conduct further research on molecular biology, how to make early diagnosis, how to improve the chemotherapeutic effect, and how to control the prevalence of AE, need to be studied more deeply in the 21st century.

[Neuroendocrine differentiation in ovarian mucinous tumors].

OBJECTIVE: To investigate the relationship between neuroendocrine differentiation of ovarian mucinous tumors and its genesis. METHODS: A morphologic study on 73 cases of ovarian mucinous tumors (32 benign, 20 borderline, 21 malignant) using immunohistochemical and immunohistochemical/histochemical double staining techniques. RESULTS: The incidences of CgA positive cell in tumors of benign, borderline and malignant were 62.5%, 75%, 76% and that of 5-HT positive cells were 31.3%, 40%, 39% respectively. But these NEC were not evenly distributed. The interesting point was that in four cases of the benign tumors the number of CgA positive cell was more than 30 per cent localizing between the glandular basement membrane and the mucinous epithelial cells. There were also many intermediate cells containing both CgA and PAS positive granules. CONCLUSIONS: The positive rate of CgA was increased corresponding to the lowering down of differentiation grades. The four cases of the benign tumors might be originating from a common stem cell, such as so-called amphocrine cell. The relationship between these four tumors and neuroendocrine differentiation in ovarian mucinous tumors remain further clarified.

[Clinical application of combined acupuncture-drug anesthesia in cerebral functional area operation].

OBJECTIVE: To reach for higher tumor resection rate and low incidence of disability by operation with combined acupuncture-drug anesthesia in cerebral functional area. METHODS: According to the different operation ways, two groups of combination of body-acupuncture and ear-acupuncture were adopted. RESULTS: Satisfactory effect was obtained in all the 80 patients, including 20 cases in language center and 60 in sensory and motor center. The anesthesia grade 1 rate was 100%. After operation, no aggravation in dysphasia or dysfunction was found, 5 cases showed aggravation on myasthenia and dysesthesia, but all recovered within 2-4 weeks. CONCLUSION: Operation with combined acupuncture-drug anesthesia might result high tumor resection rate and low incidence of disability, therefore, this method of anesthesia could be regarded as one of the routine anesthesia methods.

[Effects of extremity cuffs against physiological changes induced by head-down bedrest].

-6 degrees head down bedrest (HDBR) 10d was used to simulate weightlessness. 12 volunteers were randomed into control group and cuffs group, with 6 in each group. Inflated cuffs on the upper part of thighs and arms were performed 12 h/d in cuffs group during bedrest. No countermeasures were applied in the control group. During head up tilt (HUT) +75 degrees 20 min orthostatic tolerance test after HDBR, 5 in control group became presyncopal, while only one in cuffs group. The study indicates that the use of inflated cuffs provides a protective effect against the cardiovascular deconditioning after bedrest.

[Development and clinical application of a brain stereotactic endoscope].

OBJECTIVE: To investigate the feasibility, indications and advantages of endoscopic surgery for brain neoplasms and hypertensive haematoma. METHOD: A self-designed brain endoscope with a wide working channel was used for resection of deep-seated brain neoplasm or other cranial cerebral operations of 30 patients operated on endoscopically, 13 had intracranial tumor, 15 hypertensive intracerebral haematoma, 1 thalamic abscess, and 1 cysticercosis. RESULT: Total removal of tumor was achieved in 4 patients, subtotal or major partial removal in 7, and biopsy in 2. Completely evacuation of haematoma was achieved in 7 patients and partial evacuation in 8 (over 80%). The abscess was evacuated and the solitary cysticercosis focus was removed with our endoscope. No obvious postoperative complications were noted. CONCLUSION: The advantages of stereotactic endoscopy for deep-seated brain neoplasm include accurate intraoperative localization, minimal invasion, good recovery, no or a few complications. The indications for this intervention are small lesions deep in the brain parenchyma with well-defined boundary and light vascularity. Intracerebral haematoma can be evacuated under direct vision endoscopically, without damage to the haematoma cavity wall.

[Maisonneuve fracture of the fibula: clinical and experimental study].

OBJECTIVE: To study the traumatic mechanism and clinical treatment of Maisonneuve fracture. METHOD: 19 of 25 patients (6 - 60 months, average 16.8 months) were followed up. 11 patients were operated on and 8 were treated conservatively. Of the 25 patients, 8 had MRI. Osteotomy and serial section of the interosseous membrane (IOM) on fresh cadaveric specimens were doc carried out to observe the diastasis on stress mortise view. RESULT: Functional rehabilitation of the ankle joint: in 11 operative cases, 8 showed good results and 3 fair. In 8 conservative cases, 3 showed good results, 2 fair and 2 poor. MRI: IOM was ruptured in a range 3 - 8 cm proximal to the tibial plafond in the 8 cases. No IOM rupture was found at the high fibular fracture level. Fresh cadaveric experiment: fixation of the medial side alone can not control the syndesmotic diastasis on the artificial Maisonneuve fracture model made on cadaveric specimen. CONCLUSION: The injury level of the IOM during Maisonneuve fracture may not be consistent with the high fibular fracture. The limited external rotation of the fibula during injury by the superior tibio-fibular syndesmosis comprise to the high level fibular fracture. ORIF with syndesmotic fixation is recommended for the treatment.

Telomerase activity in the normal and neoplastic rat mammary gland.

The 1-methyl-1-nitrosourea-induced rat mammary tumor model system is well studied, reproducible, and widely used. We have investigated whether these tumors possess higher telomerase activity than normal mammary tissue. Using the telomeric repeat amplification protocol assay, we found significantly higher telomerase activity in 36 mammary carcinomas than in 72 mammary glands of virgin rats. The level of telomerase activity in virgin rats was unaffected by strain, age, stage of the estrous cycle, or ovariectomy. However, mammary glands obtained from pregnant rats exhibited telomerase activity comparable to that found in the tumors, possibly reflecting the high epithelial content of these tissues. Indeed, isolated epithelial cells from virgin and pregnant mammary glands and from carcinomas had similar telomerase activities. Thus, telomerase activity is constitutive in the rat mammary epithelium and is not a unique characteristic of malignant transformation in this tissue. These results underscore the importance of attributing biochemical properties to specific cell types in a tissue, a situation not paralleled in the interpretation of data from in vitro models.
