The corticotrophic cells of the canine pituitary gland in pituitary-dependent hyperadrenocorticism.

The distribution of specifically stained corticotrophic cells has been studied in the pituitary glands of 11 dogs with pituitary-dependent hyperadrenocorticism. The results suggest that the disease is not a single entity, and that some cases are caused by primary abnormality of the pituitary gland whereas others appear to be the result of dysfunction of the hypothalamus or central nervous system. The patterns correspond closely to those demonstrated in the human pituitary gland in Cushing's disease, and confirm that the canine disease is a useful model for the study of the pathogenesis of the variants of the condition.

Interaction of long-chain acyl-CoA analogs with pig kidney general acyl-CoA dehydrogenase.

The interaction of two long-chain acyl-CoA analogs with pig kidney general acyl-CoA dehydrogenase (EC 1.3.99,3) was examined. The effect of S-heptadecyl-CoA and heptadecan-2-onyl-dethio-CoA on the flavo-protein was observed spectrophotometrically using the flavin as an active-site probe. The S-heptadecyl thioether analog bound strongly to the enzyme (Kd = 17 nM) and was a powerful competitive inhibitor (Ki less than 40 nM). In contrast to the thioether analog, the dethiocarba derivative, heptadecan-2-onyl-dethio-CoA, was a substrate inthe standard assay system being dehydrogenated at about 60% of the rate shown by palmitoyl-CoA. These results support the proposal that alpha-carbanion formation is an early event in the dehydrogenation of acyl-CoA substrates.

Patterns of menopause: a study of certain medical and physiological variables among Caucasian and Japanese women living in Hawaii.

Comparisons were made between menopausal women and nonmenopausal controls among Caucasians and Japanese living in Honolulu, to investigate the extent of physical changes and clinical symptoms associated with menopause. The analysis was conducted using the multiphasic screening records of 170 menopausal cases and 162 nonmenopausal controls in Caucasians, and of 159 menopausal women and 187 nonmenopausal controls in Japanese. Discriminant function analysis was employed with relevant anthropometric, medical, and physiological variables. After adjusting for the linear and non-linear effects of age, only surgery and medication were retained as significant discriminant variables. Discriminant functions for Caucasian and Japanese groups were not found to be significantly heterogeneous. With regard to the discrimination of the menopausal and nonmenopausal groups, the data suggested that, while no clinical conditions other than those attributable to the effects of aging were significantly associated with the menopausal state, medication and surgical procedures for female disorders were significantly related to menopause.

Tuberculosis infection in a paediatric department.

Sputum-positive tuberculosis was diagnosed in a member of the medical staff of a paediatric department. Four children were infected, all suffering from debilitating diseases. Three of them had evidence of pulmonary tuberculosis. Eighty-two infants in the baby care unit during the eight weeks before the diagnosis of the index case were given insurance isoniazid treatment. None developed tuberculosis. Whereas nearly all the non-medical adult contacts were traced and examined, fewer than half the medical contacts attended for chest radiography. All babies in the pre-allergic phase of contact, and all children whose natural immunity is likely to be depressed, should receive antituberculosis insurance chemotheraphy.

Determination of the optimum dose of subcutaneous salbutamol in asthmatic patients.

Saline and salbutamol (250 mug, 500 mug, 750 mug, and 1000 mug) injections were administered under double-blind conditions to seven chronic stable asthmatic patients. The response on the respiratory system (FEV1, FVC) and cardiovascular system (pulse rate, blood pressure and ECG) was monitored for 3 hours. Increasing the dose of salbutamol increased the peak and weighted average effects of FEV1 and pulse rate. The optimum dose, a titration of desired increases in FEV1 and undesirable increases in pulse rate, is 500 mug but doses up to 1000 mug may be administered if necessary.

Gas chromatographic determination of beta-phenethylbiguanide and its metabolite p-hydroxy-beta-phenethylbiguanide in serum and urine.

A gas chromatographic method is presented for the detection of beta-phenethylbiguanide (PEBG) and its metabolite, p-hydroxy-beta-phenethylbiguanide (p-OHPEBG). The procedure is applicable for the determination of the drug and its metabolite in the serum and urine of rats. The detection limit is 0.2 mug PEBG and 0.5 mug p-OH PEBG per ml of serum or urine. A time course study of blood concentration and elimination rate following intraperitoneal injection of 100 mg/kg of PEBG to normal rats was performed. Beta-PEBG was found to be present in the blood and the urine, p-OH PEBG was only detected in the urine. Twenty-four hours following intraperitoneal injection, the urine contained 32% of the administered dose, 20% as unaltered PEBG and 12% as p-OH PEBG.

13C NMR or coenzyme A and its constituent moieties.

Natural abundance 13C nuclear magnetic resonance spectra, in D2O, were obtained for the following: pantoyl lactone, beta-alanine, cysteamine hydrochloride, cystamine dihydrochloride, calcium pantothenate, beta-aletheine oxalate, pantetheine, pantethine, pantetheine 4'-phosphate, oxypantetheine 4'-phosphate, desulfopantetheine 4'-phosphate, N-acetyl-aminodesthiopantetheine 4'-phosphate, adenosine 2',5'-diphosphate, adenosine 3',5'-diphosphate, and coenzyme A. A complete assignment of the 13C chemical shifts in the NMR spectrum of CoA is reported. Comparison of spectra indicates that CoA most likely exists in an extended conformation.
