The anticoccidial efficacy of arprinocid in broiler chickens under floor pen conditions.

The efficacy of arprinocid was tested against artificial infections of mixtures of Eimeria spp. in broiler chickens under floor pen conditions in 3 experiments. Treatment with arprinocid at 60 ppm over a 56 d period significantly increased the live mass gain and feed efficiency of broiler chickens. This increase compared favourably with that obtained by treatment with lasalocid, robenidine and halofuginone. Birds treated with arprinocid had substantially reduced numbers of sporulated oocysts in their litter, and less severe lesion scores than non-mediated birds.

Reactivity of amniotic fluid alpha-fetoprotein with concanavallin A in diagnosis of neural tube defects.

Alpha-fetoprotein (AFP) concanavallin-A-affinity molecular variant patterns were determined in amniotic fluid samples from 10 pregnancies complicated by anencephaly (6), spina bifida (1), Turner's syndrome (1), osteogenesis imperfecta congenita (1), and fetal death (1) and 20 normal pregnancies between 14.6 and 25.5 weeks of gestation. With the exception of one anencephalic pregnancy, the AFP concentrations in samples from women with a fetal abnormality were more than 5 SD above normal for gestational age. In every case, however, the proportion of total amniotic fluid AFP that was not reactive with concanavallin A was significantly smaller in the presence of a fetal abnormality (mean 2%) than when the fetus was normal (mean 20%). The results suggest that measuring the amount of concanavallin-A-non-reactive amniotic fluid AFP will be a valuable test for diagnosing fetal abnormality, especially when an increase in total amniotic fluid AFP concentration is equivocal or gestational age is uncertain.

Superior colliculus lesions and environmental experience: nonvisual effects on problem solving and locomotor activity.

Bilateral lesions of the superior colliculus were produced in rats reared in either a restricted or complex environment. Problem solving ability in a Hebb-Williams closed field and activity in an open field were subsequently observed in conditions of either bright or dim illumination. Animals with superior colliculus lesions were deficient in problem solving ability and were hyperactive in the open field. Complex environment exposure during development increased problem solving ability and initial ambulation scores in all groups. Extent of pretectal damage and behavioral measures were significantly related for animals reared in the complex, but not in the restricted environment. There were no interactions with illumination level, suggesting that the deficits resulting from collicular lesions are not dependent upon the availability of visual cues.

Insulin-stimulated protein phosphorylation in 3T3-L1 preadipocytes.

A protein of molecular weight 31,000 became labeled with 32P within 5 min after addition of insulin to differentiated 3T3-L1 preadipocytes previously incubated for 55 min with 32Pi. The effect was mimicked by antisera directed against the insulin receptor and was eliminated by anti-insulin antiserum. Incorporation of 32P into this protein was more than 20-fold greater in insulin-treated cells than in cells not exposed to the hormone. At concentrations greater than required with insulin, epidermal growth factor and serum (1-5%) also stimulated phosphorylation whereas 1-isoproterenol, a beta-adrenergic agonist that increases intracellular accumulation of cyclic AMP, was without effect. The 31,000-dalton protein has been tentatively identified as ribosomal protein S6 by two-dimensional polyacrylamide gel electrophoresis. Incorporation of 32P into S6 could be detected within the same time period (5 min) and at the same insulin concentrations (0.1-1.0 nM) as are required to stimulate hexose uptake in both 3T3-L1 cells and mature mammalian adipocytes. The mechanism by which this phosphorylation either mediates or reflects the intracellular actions of insulin remains to be elucidated.

Evaluating outcome measures for deinstitutionalization programs.

Since deinstitutionalization has become a feature of many state and county mental health programs, suitable and effective measures are needed for evaluating the success of patients in adjusting to community life. This study attempted to identify the factors linked with satisfactory adjustment or return to the hospital and evaluated the usefulness of various measures of success in the community.

Quantitative analysis of histological parameters in giant cell lesions of the jaws and long bones.

Stereological techniques have been used to compare histological parameters of the giant cell component in 10 giant cell tumours of long bones, 10 central giant cell granulomas of the jaws and 10 peripheral giant cell granulomas of the jaws. The giant cell parameters investigated were: mean profile diameter, mean diameter, mean profile axial ratio, volume-to-surface ratios, nuclear numerical density, profile area and cell volume. Serial sections were cut at 4 micron from routinely fixed, paraffin-embedded specimens of each case and two sections, separated by 100 micron, were stained with haematoxylin and eosin. Random fields were photographed until at least 25 giant cell profiles had been recorded from each of the two sections of each lesion. The processed film was projected for point and intersect counting, the entire procedure ensuring double-blind assessment of the cases under investigation. In general there was no significant difference for any of the parameters between the giant cells of central and peripheral jaw lesions. There was a significant difference, however, between the giant cells of central jaw lesions and long bone tumours in respect of both nuclear numerical density and mean absolute cell volume.

Inhibition of lymphocyte proliferation by polyamines requires ruminant-plasma polyamine oxidase.

Spermine and spermidine in vitro are potent inhibitors of proliferation of phytohaemagglutinin-stimulated rat thymic lymphocytes, lymphoma cells and human lymphoblastic leukaemia cells, but only in media supplemented by foetal calf serum. This inhibition is shown to be due to a bovine plasma polyamine oxidase, with a high specificity for these polyamines. Spontaneously dividing lymphocytes are not subject to this inhibition. This, plus direct evidence from synchronous cultures of EB2 cells demonstrates that the inhibition is expressed in the late G1 or G1/S interface of the cell cycle. Putrescine was not an inhibitor in the presence of foetal calf serum but became so in the presence of human pregnancy serum, possibly due to the action of diamine oxidase.
