Granular cells associated with the enamel organ of a developing tooth.

An accumulation of granular cells associated with the enamel organ of a developing deciduous, incisor tooth was noted during light microscopy (LM) of serial sections from the right half of a mandible from a stillborn, female infant of 37 weeks gestations. There was a break in continuity of the enamel matrix associated with the focus of granular cells, which appeared to be continuous with, and arise directly from, the cells of the stratum intermedium. The granular cells were very similar histologically to those of a congenital epulis but it is impossible to say whether the lesion would have developed sufficiently to present clinically as an epulis. Less probably, the granular cells might eventually have been associated with the subsequent development of a granular cell ameloblastoma or ameloblastic fibroma. If the lesion does represent an early stage in the formation of a congenital epulis, it would support the odontogenic theory of origin of the granular cells.

Concanavalin-A-affinity molecular heterogeneity of human hepatoma AFP and cord-serum AFP.

Glycosylation of alpha-fetoprotein (AFP) by human primary hepatocellular carcinoma (PHC) is abnormal. Concanavalin A (Con A)-affinity molecular variant patterns of serum AFP from patients with PHC are different from those of cord-serum AFP. Different patients with PHC exhibit different Con-A-affinity AFP molecular variant patterns, and the pattern remains constant over time in a given individual. The degree of deviation of the AFP molecular variant pattern from the molecular pattern of AFP secreted by neonatal liver cells is independent of the total serum AFP concentration. We propose that analysis of the AFP lectin-affinity molecular heterogeneity will improve the discrimination between malignant and nonmalignant liver disease in cases when the degree of elevation of the serum AFP concentration is nondiagnostic.

Prospects for development and use of recombinant deoxyribonucleic acid techniques with ruminal bacteria.

Over the last decade, developments in recombinant deoxyribonucleic acid techniques and molecular biology have revolutionized bacterial genetics, creating vast, new potential uses of bacteria (as well as animal and plant cells) that were not even considered previously. Bacterial production of hormones is but one example. With bacterial species with well developed genetic systems, such as Escherichia coli, it is now possible genetically to "design" or "engineer" bacterial strains having specific characteristics. One reasonable future approach toward improvement of animal agriculture would be manipulation of the rumen ecosystem via the use of genetically modified ruminal bacteria, but significant obstacles exist with this approach. Genetic systems of ruminal and of anaerobic bacteria of the mammalian gastrointestinal tract, in general, have not been studied and are largely unknown. In this paper, the various criteria for possible establishment of recombinant deoxyribonucleic acid systems in ruminal bacteria are outlined. Secondly, applications for utilizing genetically engineered ruminal bacteria to control digestion of specific feedstuffs, to regulate specific fermentation products, and to control growth of specific bacterial species are discussed.

Polyamine-mediated inhibition of in-vitro cell proliferation is not due to acrolein.

The influence of acrolein or spermine on the viability and growth of phytohaemagglutinin-stimulated rat thymic lymphocytes in cultures supplemented with foetal calf serum have been investigated. Acrolein (greater than 20 microM) was cytotoxic; spermine had little effect on viability, but inhibited [3H]TdR incorporation at low concentrations (approximately 10 microM). Cells treated with greater than 8 microM acrolein 3 hr before stimulation exhibited irreversible inhibition of protein synthesis, whereas 50 microM spermine had no effect, even when cells were treated for 24 hr before stimulation. However, addition of 25 microM spermine after stimulation did inhibit both [3H]-uridine incorporation and protein synthesis: this was reversible if cells were freed of polyamine within 4 hr, but not if washed after 24 hr. These results show that, contrary to several previous reports, in-vitro inhibition of cell proliferation by spermine is not due to the formation and action of acrolein.

Inhibition of cells in culture by polyamines does not depend on the presence of ruminant serum.

Using T-lymphocyte (T-LC) and granulocyte colony (GC) assays with truly proliferating cells, the inhibitory dose-response relationships of spermine and spermidine in the presence of selected sera have been examined. In contrast to previous studies which used [3H]thymidine uptake as an index of proliferation, in vitro inhibition by polyamines was shown to require neither foetal calf serum (FCS) nor the addition of any exogenous polyamine oxidase. Cells grown in the absence of FCS were between 5-50% as sensitive to polyamines as in its presence. By using specific inhibitors of polyamine oxidase, it was shown that polyamine-elicited mitotic inhibition in the absence of FCS was still dependent on a polyamine oxidase, and evidence is presented to show that the source of the enzyme is the cells themselves.

A simple method for obtaining blood samples from mature frogs.

A technique was developed for obtaining periodic blood samples from mature frogs. Up to 150 microliter of blood was collected at weekly intervals. Anesthetization was not required. The data compiled with this technique indicated that activity-induced hyperglycemia was minimal.

Short term augmentation of amphetamine-induced rotation bias.

Using measures of stereotypy such as rearing, licking, and chewing, low dosage administration of amphetamine can be shown to produce increased stereotypy over a period of days. In the present study amphetamine-induced perservation was observed in two sessions when rotational behaviors were used as a behavioral measure. Rotational perseveration occurred in animals receiving both drug and testing experience in session one and session two. Animals receiving either drug or testing experience in session one did not show perseveration with drug and testing experience in session two.

Escherichia coli delta 1-pyrroline-5-carboxylate reductase: gene sequence, protein overproduction and purification.

The sequence of the Escherichia coli proC gene which encodes for delta 1-pyrroline-5-carboxylate (PCA) reductase was determined. Overproduction of the proC gene product via an expression plasmid carrying the bacteriophage lambda PL promoter allowed the purification to homogeneity of PCA reductase by affinity adsorption chromatography. NH2 and COOH-terminal analysis and amino acid composition of the purified proC protein is consistent with the gene sequence reported. The molecular weight of the proC monomer is 28,112.

Guar biscuits in the diabetic diet.

A new type of guar-containing biscuit has been developed and incorporated into the diabetic diet in both short- and medium-term studies. It has been found to be effective in reducing the postprandial rise in the blood glucose level and in improving glycaemic control, as shown by reduced fasting blood glucose values and decreased 24-hour urinary glucose excretion. This form of guar has proved to be palatable and acceptable to patients. It was effective in smaller quantities than have previously been tested and may prove a valuable adjunct in the treatment of diabetes.

Insulin activates a tyrosine-specific protein kinase in extracts of 3T3-L1 adipocytes and human placenta.

Insulin activates a tyrosine-specific cAMP-independent protein kinase when added directly to detergent extracts of differentiated 3T3-L1 adipocytes and humal placental membranes. The kinase is also activated by antibody to the insulin receptor and, to a lesser extent, by proinsulin. It catalyzes the phosphorylation of the 92,000-dalton component of the insulin receptor, histone, and casein; in each case, tyrosine is the principal amino acid modified. Under the conditions used to activate the kinase, insulin does not affect the rate of dephosphorylation of the receptor or of histone. The insulin-activated kinase is copurified with the human placental insulin receptor until the final elution from insulin-Sepharose. It remains to be established whether the kinase and the insulin receptor are separate molecules.

Novel antibiotic resistance transfer in Bacteroides.

Resistance to tetracycline and lincosamide antibiotics was transferred en bloc from a strain of Bacteroides fragilis (V503) to a plasmidless strain of Bacteroides uniformis (V528) during in vitro filter matings. Resistance transfer was detected at frequencies of 10(-5) to 10(-6) drug-resistant progeny per input donor cell and was dependent on cell-to-cell contact of donors and recipients. Transfer was insensitive to DNase and was not mediated by chloroform- or filter-sterilized donor broth cultures. A determinant for resistance to cefoxitin in V503 was not transferred in this system. V503 contained a 3.7 x 10(6)-dalton plasmid (pVA503). Drug-resistant progeny of V503 x V528 matings usually contained pVA503, but up to 20% of the total progeny of such crosses were plasmid free. Filter blot DNA hybridization studies (Southern method) confirmed that pVA503 was not integrated into the host chromosome of the plasmidless progeny. Drug-resistant progeny from V503 x V528 matings (with or without pVA503) conjugally transferred clindamycin resistance an tetracycline resistance to a suitable recipient strain. None of the drug resistance determinants of V503 were affected by treatment with standard plasmid curing regimens, and methods designed to detect very large plasmid molecules failed to suggest the involvement of extrachromosomal DNA in this resistance transfer system. The well-characterized Bacteroides R plasmid, pBF4 (conferring clindamycin resistance), was found to share hybridizing sequences with bulk cellular V503 DNA when examined by filter blot hybridization. Similarly sized sequences were found in drug-resistant progeny recovered from matings. Neither of the two pBF4 derivatives carrying deletions that abolished clindamycin resistance hybridized with V503 DNA.

Two independent conjugal transfer systems operating in Bacteroides fragilis V479-1.

Bacteroides fragilis V479-1 (also designated strain 92) has previously been shown to contain a conjugative plasmid, pBF4, that specifies resistance to clindamycin (Cc). A report of inducible tetracycline (Tc) resistance in this strain suggested that this phenotype was also plasmid associated (G. Privitera et al., Nature [London] 278:657-659, 1979) and prompted further investigation. Mating experiments with V469-1 and a Cc-sensitive derivative of V479-1, V598, showed that Tc resistance transfer occurred by a conjugation-like event which was insensitive to DNase, was not mediated by donor culture cell-free filtrates, and required cell-to-cell contact. Results from transfer experiments with V479-1 indicated that Tc and Cc resistance determinants were not linked and segregated independently in matings. Progeny recovered from matings with the V479-1 or V598 donor strain were able to transfer the Tc resistance marker in secondary crosses. Tc resistance transfer from V479-1 or V598 was greatly stimulated by pregrowth in the presence of Tc but not Cc. pBF4-mediated Cc resistance transfer was not affected by pregrowth in the presence of Cc or Tc. Filter blot DNA hybridization studies revealed that pBF4 sequences were not involved in either the Tc resistant phenotype or its associated conjugal transfer properties. The Tc resistance transfer element was not associated with pBF4 or any other extrachromosomal DNA element.

Hybridization studies reveal homologies between pBF4 and pBFTM10, Two clindamycin-erythromycin resistance transfer plasmids of Bacteroides fragilis.

Two clindamycin-erythromycin resistance transfer factors of Bacteroides fragilis, pBF4 and pBFTM10, were analyzed for regions of DNA homology. Although the plasmids were derived from different clinical isolates of B. fragilis and have different sizes, they showed homology in the clindamycin-erythromycin resistance region; no homology could be detected outside of this region.

Cloning of African trypanosomes in mice immunosuppressed by cyclophosphamide treatment.

The use of cyclophosphamide-treated mice for purpose of cloning the African trypanosomes was assessed. C3HeB/FeJ mice were injected with 200 mg/kg cyclophosphamide (CY) 24-72 hours prior to infection with a single trypanosome isolated from Trypanosoma rhodesiense infected blood samples. All CY-treated mice exhibited depressed parasite-exposure, which was a period of time sufficient to grow trypanosomes from a single organism to a fulminating parasitemia. Cloning efficiency was routinely 45% in these animals. Thus, our study demonstrates that CT-treated mice are a convenient and efficient vehicle for cloning African trypanosomes. Techniques which facilitate the selection of single trypanosomes from infected blood are also described in this report.

Insulin stimulates the phosphorylation of ribosomal protein S6 in a cell-free system derived from 3T3-L1 adipocytes.

Physiological concentrations of insulin stimulate the incorporation of 32P into ribosomal protein S6 in intact 3T3-L1 adipocytes (Smith, C. J., Wejksnora, P. J., Warner, J. B., Rubin, C. S., and Rosen, O. M. (1979) Proc. Natl. Acad. Sci. U. S. A. 76, 2725-2729). Extracts from cells treated with insulin also exhibit an increased ability to incorporate 32P from [gamma-32P]ATP into protein S6 in vitro (Smith, C. J., Rubin, C. S., and Rosen, O. M. (1980) Proc. Natl. Acad. Sci. U. S. A. 77, 2641-2645). We now report that the direct addition of insulin to particulate preparations containing high affinity insulin receptors and ribosomes derived from insulin-sensitive cells (not previously exposed to insulin) stimulates incorporation of 32P from [gamma-32P]ATP into ribosomal protein S6 1.5- to 3.0-fold. The stimulation occurs at the same concentrations of insulin (0.1-1.0 nM) and within the same period of time as it does in intact cells.
