[Targeting studies of humanized scFv25 fusing to TNFalpha against hepatocellular carcinoma].

OBJECTIVE: To obtain humanized engineering bifunctional antibody, which has potentialities for clinical application. METHODS: Humanized anti-human hepatocellular carcinoma (HCC) single chain fragment (hscFv25) was linked with human TNF-alpha gene to form anti-HCC bifunctional antibody, then it was subcloned into prokaryotic GST fusion expression vector pGEX 4T-1 and expressed in the host E.coli. Indirect immunofluorescent staining was performed on HCC cell smearing slides in order to evaluate the activity of the purified aim protein, then MTT trial to evaluate the cytotoxicity of hscFv25-TNFalpha to SMMC-7721, finally primary tumor regression trial in nude mice bearing HCC to evaluate the targeting therapeutic value of hscFv25-TNFalpha. RESULTS: The hscFv25-TNFalpha had the similar specificity to parental antibody HAb25 for SMMC-7721 antigen. One hour predisposed MTT trial in control with parental antibody HAb25 affirmed that hscFv25-TNFalpha was cytotoxic to targeted cell SMMC-7721 with the IC(50) to be 7.1 microg/ml. The cytotoxicity can be inhibited by parental antibody HAb25. This indicated that the cytotoxicity of hscFv25-TNFalpha to targeted cell is antibody-mediated selective cytotoxicity. The tumor regression trial to the 3mm HCC xenografts in nude mice showed that hscFv25-TNFalpha had assured targeting cytotoxicity and the efficiency was nearly up to 3/3 (1/3 complete remission, 2/3 partial remission). The cytotoxicity of the hscFv25-TNFalpha was better than that of TNFalpha, whose efficiency was only 2/3 and without complete remission. CONCLUSION: hscFv25-TNFalpha is an anti-HCC bi-functional antibody which has potentialities for clinical application.

[Study on the biocompatibility and toxicology of biomaterials--poly(epsilon-caprolactone)].

Four observation indexes were selected to evaluate the biocompatibility and toxicology of biodegradable materials poly(epsilon-caprolactone)(PCL), which is synthesized in our laboratory using Ti(OBu)4 as an initiator. The results showed that PCL films had a mild inflammatory reaction in the early days of planting and after 3 months the inflammation basically disappeared. We conclude that PCL material possesses good biocompatibility and has a good developmental prospect.

[Analysis of the design principle and mechanics nature of adjustable semi-circular planum external fixator].

This thesis presents our studies of an adjustable semi-circular planum external fixator and offers a relatively detailed introduction and analysis of the structure principle and mechanics nature. Because dual-direction joining pole with active joints was used, by adjusting the length of each pole, it can not only make the two semi-circular steel plates more parallely along the axis line to stabilize the fracture and lengthen the extremity, but it also can change the planum angle of the steel plate, and thus achieve the effect of orthopaedic function.

[Detection of anti-HIV-1/2 and HIV nucleic acid in population of non-remunerated blood donors in Shenzhen].

OBJECTIVE: To study the prevalent status of HIV-1 in population of non-remunerated blood donors in Shenzhen. METHODS: 46,095 non-remunerated blood donors were tested for anti-HIV-1/2 by ELISA. The donors of anti-HIV positive were further detected for HIV DNA from peripheral blood mononuclear cells (PMBCs) by nested-PCR and HIV RNA from plasma by RT-PCR. The partial genome of env of 2 blood donors were sequenced. RESULTS: The anti-HIV-1 was tested positive in 7 of 46 095 voluntary blood donors and the positive rate was 0.015%. In these 7 non-remunerated blood donors of anti-HIV-1 positive,7 were positive for HIV DNA in PMBCs and 5 positive for HIV RNA in plasma. The sequence analysis showed that 2 donors were infected by HIV-1 subtype E strains. CONCLUSIONS: There exists HIV-1 subtype E infection in population of non-remunerated blood donors in Shenzhen. It is essential to detect and monitor strictly the population of non-remunerated blood donors.

[Light effect of gaps in Huangshan pine community and regeneration of Huangshan pine].

This paper dealt with the gaps of Huangshan pine(Pinus taiwanensis) forest developed on the meadow of 1400-1600 m above sea level of Huangshan Mountain. The method of analyzing annual ring growth of pine trees surrounding gaps was employed to estimate the time of gap formation, which was almost the same as the regeneration occurred there. The dynamic variations of total radiation and photosynthetic available radiation(PAR) at different points in gaps were measured, and a regression model for their estimation based on gap size, expanded gap size and canopy closure situation overhead the extended gap was established. The results indicated that the total radiation and PAR were respectively 90% and 71% of full sunlight in gap center, and 30-40% and 25-33% in expanded gaps, while the light under canopy was less than 10% of full sunlight. In the gaps with an area of 40 m2 or in the extended gaps with an area of 110 m2, the light might basically satisfy the growth of the pine, and the gap regeneration could occur. It is suggested that man-made gaps could be made to facilitate the regeneration of Huangshan pine, which could be used as a management approach for Huangshan pine forests, and also for maintaining the landscape in beauty spots of Huangshan Mountain.

[Relationship between light and regeneration character of Huangshan pine].

The growth and physiological characters of 1-3 years old Huangshan pine seedlings in pure and mixture plots with four different transparancy were determined, and the germination percentage of seeds and their complex stressing vigour were tested. Meanwhile, the regeneration mechanism of Huangshan pine in natural forest was studied in this paper. The results showed that the plumpness of seeds was 65%, the germination percentage was 27%, and the complex stressing vigour was 26%. The lower germination percentage affected the reproduction of Huangshan pine. The photosynthetic rate was more affected by solar energy, but less by humidity and surface temperature. With high light composation, Huangshan pine's regeneration under forest was difficult. The seedlings of Huangshan pine in mixed coniferous and broadleave tree plots were at a disadvantage because of slow growth and insufficient light.

[The effect of antisense hsp90 beta on the malignant phenotype and sensitivity of HeLa cells to chemotherapeutic drugs].

OBJECTIVE: To study the effects of hsp90 beta on tumorigenesis and sensitivity of HeLa cells to chemotherapeutic drugs. METHODS: A recombinant plasmid expressing either sense or antisense hsp90 beta RNA, and a vector pLXSN were transfected into HeLa cell line by lipofectin method. The transfected cells were screened by G418. The growth rates of the transfected HeLa cells and their sensitivity to adriamycin(ADM) and cisplatin (CDDP) were examined by cell count and MTT uptake. Colony formation in soft agar was used to embody the malignant phenotype in vitro. RESULTS: There was little difference between HeLa cells transfected with sense RNA plasmids and pLXSN vector. The growth rate of HeLa cells transfected with antisense RNA declined, and their ability to form colony in soft agar was decreased. The sensitity of the antisense hsp90 beta transfected HeLa cells to ADM and CDDP increased 2.1-5.8 fold; the LD50 decreased to 1/2-1/5 that of the wild-type HeLa cells. CONCLUSION: Hsp90 beta is involved in the maintenance of malignant phenotype and drug resistance in HeLa cells.

[The relationship between prostate cancer and multi-estimating results of prostate-specific antigen].

OBJECTIVE: To analyze the relationship between prostate cancer and multi-estimating results of prostate-specific antigen (PSA). METHODS: In accordance with the results of 76 cases checked on PSA over 2 years, univariate and multivariate analysis were performed in prostate cancer connected with PSA, PSA density (PSAD), PSA velocity (PSAV), age-specific PSA (PSA-age), and free to total PSA ratio (fPSA/tPSA) by means of t test, Chi-square test, and logistic regression. RESULTS: Of the 76 cases observed, 16 were diagnosed with prostate cancer. By univariate analysis, all factors of PSA, PSAD, PSAV, age-PSA and fPSA/t PSA showed conspicuous correlations with prostate cancer. However, in multivariate analysis, only did fPSA/t PSA (P = 0.006) with regressive ratio being positive number and PSAD (P = 0.018) with regressive ratio being negative number reveal statistical differences with prostate cancer. CONCLUSION: In multi-estimating results of PSA, fPSA/t PSA as a risk factor and PSAD as a protective factor were more correspondant with the principle for screen test of prostate cancer.

[Long-term therapeutic effects of extended radical resection and radical resection of cancer of cardia and stomach fundus].

OBJECTIVE: To study the best range of radical resection in the treatment of cancer of the cardia and fundus of stomach. METHODS: 418 patients with cancer of the cardia and fundus of stomach underwent radical resection. Of them 192 were treated by extended radical resection (extended group), and 226 by radical resection (radical group). The 5-year and 10-year survival rates were followed up and compared in the two groups. RESULTS: Analysis failed to demonstrate significant difference between the two operations for TNM stage I and II (P > 0.05). For stage III, however, the 5-year and 10-year survival rates in the extended group increased by 14.2% and 15.9% as compared with those in the radical group (P < 0.05). The two groups survival rates were similar for stage IV (P > 0.05). CONCLUSIONS: To completely clean the lymph nodes of splenic hilus and artery and improve long-term survival rate, extended radical resection including spleen and body and tail of the pancreas should be recommended for stage III patients with cancer of the cardia and fundus of stomach when their serosa was involved or lymph node metastasis took place.

[Long-term effect on preoperative ulnaris arterial intubation chemotherapy in treatment of stage III breast cancer].

OBJECTIVE: To evaluate preoperative ulnaris arterial intubation chemotherapy as a step of multidisciplinary treatment of stage III breast cancer. METHODS: 109 patients with stage III breast cancer were studied. 64 patients (group A) were treated by preoperative ulnaris arterial intubation chemotherapy and others (group B) were not. The 5 and 10-year survival rates and disease-free survival rates were analyzed. RESULTS: In patients with stage III breast cancer, the overall response rate of group A was 76.6%, and the response rate to histological study was 89.1%. The 5-year and 10-year survival rates of group A were 64.0% and 47.9% respectively. The disease-free survival rate of group A was 59.4% at 5 years and 43.3% at 10 years. All were higher than the 5-year and 10-year survival rates (31.1%, 21.9%) and the 5-year and 10-year disease-free survival rates (22.2%, 12.5%) of group B (P < 0.05). CONCLUSION: Preoperative ulnaris arterial intubation chemotherapy is an effective measure for the treatment of stage III breast cancer. It can improve the long-term effects on stage III breast cancer and reduce local relapse and distant metastasis.

[Multivariate analysis of prognostic factors in renal cell carcinoma].

OBJECTIVE: To assess the effect of prognostic factors on renal cell carcinoma (RCC). METHODS: 316 cases of RCC were reviewed retrospectively. Their survival rates were calculated by Kaplan-Meier method and statistical differences were determined by Log-rank test. Significant prognostic factors were evaluated by Cox's multivariate proportional hazard model. RESULTS: After 40.3 +/- 18.5 month follow-up, the overall 5-year survival rate was 62.3%. By multivariate analysis, nine factors were included in Cox's multivariate proportional hazard model. M was the most important prognostic factor in RCC (P = 0.0013), and the others in turn were T (P = 0.0182), age (P = 0.0347), performance status (P = 0.0423), N (P = 0.0471), lymphadenectomy (P = 0.0542), grade (P = 0.0775), serum albumin (P = 0.1536), and serum creatinine (P = 0.4543). CONCLUSIONS: The significant prognostic factors in RCC were T, N, and M. Age and performance status showed the effect on prognosis of RCC. Lymph-node dissection also revealed a meaningful effect on relative lower stage of RCC.

[A cohort study on the relationship between vegetable intake and risks of lung cancer in the Tin Corporation (YTC) miners in Yunnan].

OBJECTIVE: To understand the relationship between vegetable intake and risk of lung cancer in the Tin Corporation (YTC) miners in Yunnan. METHODS: The reproducibility and validity of frequency of vegetable intake in Food Frequency Questionnaire (FFQ) were studied and the correlations between different vegetable intake and frequency and lung cancer relative risk among high risk population of lung cancer by Chi-square, single- and multi-Cox regression were analyzed. RESULTS: Results showed that the reproducibility and validity of wax gourd, cucumber, spinach, wild celery, fresh bush pepper, fresh soybean, sweet potatoes, asparagus, fresh bamboo shoot were good (r = 0.20 - 0.33, P < 0.05); There were significantly different incidences of lung cancer in different frequencies of wax gourd, cucumber, wild celery, fresh bush pepper out of 11 vegetables intake, and also in different total vegetable intakes (chi(2) = 8.83 - 30.64, P < 0.05). 11 vegetables and total vegetable intake were significantly negatively correlated with risks of lung cancer (RR < 1, P < 0.05). Only 2 vegetable frequencies were negatively correlated with risk of lung cancer by adjusting confounders such as age, occupational history, and smoking (P < 0.05). CONCLUSION: This suggested that vegetables intake may have a role in decreasing the incidence of lung cancer.

[Pharmacognostical studies on the stem and leaf of Mussaenda pubescens].

Morphological and microscopic chracteristics of Mussaenda pubescens Ait. f, a traditional Chinese medicine were described and illustrated with line drawings. TLC and UV analysis of the medicinal material were also undertaken. These studies provide referential information for identification and development of this medicinal material.

[Preparation of tea polyphenols sustained-release microcapsule].

With ethycellulose (EC) as matrix tea polyphenols (TP) microcapsule was prepared by using emulsion solvent evaporation technique. Effect of preparation condition on size and drug entrapment efficiency of microcapsule was investigated. We also investigated the drug entrapment effect of microcapsule. The result showed that the tawny microcapsule with highly porous surface was obtained by using polyvinyl alcohol (PVA) and sodium dodccyl sulate (SDS) as dispersant. The stirring rate, polymer concentration and drug payload affected the size and drug entrapment efficiency of microcapsule. The matrix and preparation method do not damage the ingredient of TP. The microcapsule can improve the stability of TP and sustain TP released.

[Influence of oxygen content on photoluminescence from a-SiOxNy thin films].

Amorphous SiOxNy thin films were deposited by PECVD technique. Strong photoluminescence bands centered at about 340 and 600 nm and peaks centered at 365, 375, 380, 735 and 745 nm from as prepared samples were observed at room temperature. Influence of partial pressure of H2 during deposition was investigated. The results show that strength of PL and oxygen content increase with H2 partial pressure decrease, the position of PL peaks does not change, and the central position of PL bands shifts. The possible origin of PL and mechanism of the variations are discussed according to experimental results.

[Determination of palladium by ion exchange resin phase spectrophotometry].

A new method for the determination of palladium by ion exchange resin phase spectrophotometry has been established. It is based on the fixation of palladium as 1-(5-bromo-2-phridylazo)-2-naphthol-6-sulphonic acid (5-Br-PAN-S) compelled on a anion-exchange resin. The resin phase absorbances at 690 and 800 nm are measured directly and the linear range of 0-20 micrograms for palladium. The method has been applied to the determination of palladium in water samples and catalysts with satisfactory results.

[Color reaction of cationic surfactants on bromphenol red and its analytical applications].

The suitable conditions, mechanism, characteristics and analytical applications of bromphenol red (BPR) reacting on cationic surfactants (CSAA) such as cetylpyridinium bromide (CPB) or cetyltrimetryl ammonium bromide (CTAB) were investigated. The results show that the single molecular CSAA reacts on BPR to form an ion-associate complex at pH 5.0-6.0, and its molar ratio (CSAA:BPR) is 3:2. Based on this color reaction, the critical micelle concentration (CMC) of CTAB and CPB was determined, and the possibility for the determination of trace CTAB and CPB were proposed.

[Determination of trace copper, zinc, iron, calcium and magnesium in lymph by flame atomic absorption spectrophotometry].

This paper reports the determination of trace Copper, Zinc, Iron, Calcium and Magnesium in lymph by flame atomic absorption spectrophotometry. The lymph samples were digested with HNO3-HClO4. The method is convenient and rapid. The rates of standard recovery of Copper, Zinc, Iron, Calcium and Magnesium were 99.2%, 103.0%, 102.0%, 104.0% and 98.5% respectively.
