The analysis of alkaline phosphatase isoenzyme using 4-methylumbelliferyl phosphate as substrate on a cellulose acetate membrane.

A new procedure is established for the analysis of alkaline phosphatase isoenzymes. The electrophoretic separation on cellulose acetate membrane coupled with the detection of alkaline phosphatase activity with 4-methyl-umbelliferyl phosphate as a substrate is described. The proposed method would be useful for the analysis of sample of micro-scale quantities and low activities.

Discussion of Tanaka's paper.

The theory of quantification is a method of statistical data analysis of categorical data. In other words, this is a kind of data theory and is closely related to optimum scaling method. This method has been mainly developed by Guttman in Israel and Hayashi in Japan. The multidimensional scaling method, which has been recently developed, is considered to be a continuation of the theory of quantification. Tanaka discussed mathematically some of Hayashi's methods of quantification. The present paper, gives an overview of the methods developed by him and other closely related methods and gives the orientation of those methods introduced by Tanaka. Then, as an illustration of exploratory categorical data analysis, the experimental data of Grizzle are analyzed by using the second method of quantification. The data structure is shown heuristically as a spatial configuration of factors in two-dimensional Euclidean space.

The estimation of heart weight from routine chest X-ray films.

A formula to estimate heart weight was derived using a theory of quantification which employed the measurements of cardiac silhouette taken from routine postero-anterior projection chest X-ray film. The films obtained in 73 necropsied cases of essential hypertension, renal hypertension, mitral and aortic valvular diseases, and 62 necropsied cases with other diseases were used for derivation of the formula. The measurements of the horizontal distances from the midsternal line to the intersecting point of the lower margin of each rib along the left cardiac border, and of the greatest distance along the right cardiac border were employed; body height, age and sex were also included. The heart weight estimated by this formula coincided with the heart weight measured at necropsy with a high correlation coefficient of 0.81. The correlation rate in 34 external samples was also significantly high, being 0.72. These results indicate the practical usefulness of this formula.

Anti-testosterone antisera produced in mice of different strains and sexes.

Specific anti-testosterone antiserum was obtained from mice of the strain C57B1/6. Testosterone 3(O-carboxymethyl)oxime-keyhole limpet hemocyanin conjugate was administered to male and female mice of the strains C57B1/6 (H-2b), DBA/2 (H-2d), (C57B1/6 x DBA/2)F1, (H-2b/d), and AKR (H-2k). Antisera obtained from male and female C57B1/6 mice were more specific for testosterone than those from the other strains, and the cross-reaction with 5alpha-dihydrotestosterone was 25.0 and 29.2%, respectively. The titer of antiserum from C57B1/6 mice was the highest and that of antiserum from DBA/2 mice was the lowest. On the other hand, the titer of antiserum from the female was higher than that of antiserum from the male in each strain. These results indicate that the mouse strain and sex differences are important factors for antibody formation from challenged antigen.

Simple determination of high urinary excretion of 5-hydroxyindole-3-acetic acid with ferric chloride.

We describe a method for determination of urinary 5-hydroxyindole-3-acetic acid, a metabolite of serotonin and tryptophan and a useful indicator of metastasizing carcinoid tumor. The analysis is based on the color development with ferric chloride added to the residue obtained on evaporating a diethyl ether extract of the sample that has been acidified with sulfuric acid and saturated with sodium chloride. The color is measured by 510 nm. The reaction was made highly specific by the acidification so that nonspecific reactions with catecholamines and with other phenolic compounds such as salicylic acid could be eliminated.

Changes of serum anti-thyroid antibodies during and after pregnancy in autoimmune thyroid diseases.

Changes of serum anti-thyroglobulin haemagglutination antibodies (TGHA) and anti-thyroid microsomal haemagglutination antibodies (MCHA) were observed during pregnancy and after delivery in Graves' disease and autoimmune thyroiditis. Both TGHA and MCHA decreased as pregnancy progressed, and sometimes they became negative in late pregnancy. Transient increases of TGHA and MCHA were observed after delivery and the antibody titres reached peaks about 3-4 months post-partum in more than halft the patients. In some patients, antibodies developed after delivery. Similar transient increases of antibodies were observed after spontaneous and therapeutic abortion. These changes seem to be induced by physiological and immunological changes occurring during pregnancy and after delivery.

Changes of serum immunoglobulins IgG, IgA, IgM, and IgE during pregnancy.

The serum levels of immunoglobulins at various times throughout pregnancy were measured in 11 healthy women. The concentrations of IgG, IgA, and IgM decreased significantly in the second and third trimesters, the mean decreases at the second trimester being 18, 13, and 9%, respectively. When the decreases were expressed on the basis of serum total protein, the decreases in IgG and IgA were significant but the decrease in IgM was not. The level of IgE either decreased or increased during pregnancy. Maternal age, emesis, ABO-incompatibility, and the sex and weight of the baby at birth were not related to the initial concentration or to the extent of decrease of immunoglobulins during pregnancy. In a case of Rh incompatibility, increase of immunoglobulins was observed concomitantly with the transient appearance of anti-Rh(D) antibody. Immunoglobulin depletion in pregnancy seems to result from both immune suppression and hemodilution.

Enzyme immunoassay of serum testosterone.

An enzyme immunoassay of serum testosterone using the testosterone-glucoamylase complex is described. Testosterone was estimated by the enzyme immunoassay after extraction with hexan: ether (4:1) for serum from men and additional thin layer chromatographic step for serum from women. The within and between assay errors, measured as the coefficient of variation were 11.1 percent (n=8) and 12.0 percent (n=12). The sensitivity of this assay was 0.25 ng. The mean testosterone concentration (+/- SD) in 19 normal men and 4 normal cycling women were 5.3 +/- 1.8 and 0.52 +/- 0.12 ng/ml, respectively. The level of testosterone found by the present assay compared favorably with those obtained by other methods.

Determination of urinary total phenolic compounds with use of 4-aminoantipyrine: suggested screening test for hyperthyroidism and for catecholamine-producing tumor.

We describe a method for determining those urinary total phenolic compounds that are tyrosine analogs or metabolites, such as thyroxine and catecholamines. The urine sample, 4-aminoantipyrine in carbonate-bicarbonate buffer, and potassium ferricyanide solution are mixed and the quinoneimine dye that forms is measured at 500 nm. Some cases of hyperthyroidism, diabetes mellitus, nephrosis, obesity, hypertension, or catecholamine-producing tumor showed above-normal values, so that this determination seems useful as a screening test for these disorders.

Abnormal serum lactate dehydrogenase isoenzyme in a case of laryngeal carcinoma and thyrotoxicosis.

An abnormal lactate dehydrogenase (LDH) isoenzyme was found in the serum of a patient with laryngeal carcinoma and hyperthyroidism. On electrophoresis it migrated as an additional band between LDH-1 and LDH-2. Follow-up studies suggested that this higher molecular weight, rather thermostable LDH isoenzyme, might have originated from the cancer tissue, though a possible relationship with the thyrotoxic state cannot be excluded.

Enzyme immunoassay of testosterone using the testosterone-glucoamylase complex.

A highly sensitive, reproducible method was established for enzyme-coupled immunoassay of testosterone, involving coupling of testosterone to glucoamylase [EC 3.2.1.3] with a water-soluble coupling reagen carbodimide. No enzyme activity was lost during this coupling procedure. The sensitivity of the method was comparable to that of competitive protein binding assay.

Simple determination of 3alpha-hydroxy fraction and 3beta-hydroxy fraction of urinary 17-ketosteroids.

We describe a method for determination of the 3alpha-hydroxysteroid fraction and 3beta-hydroxysteroid fraction of 17-ketosteroids in urine. The method includes enzymatic hydrolysis with beta-glucuronidase and extraction with ethyl acetate and color development of 3beta-hydroxysteroids by 3beta-hydroxysteroid oxidase and of total 17-ketosteroids by the Zimmermann reaction. The value for 3alpha-hydroxysteroids is calculated by subtracting the value for 3beta-hydroxysteroids from the value for total 17-ketosteroids. Recovery was assessed and results compared with those by an accepted method. Results were satisfactory for measuring these fractions of 17-ketosteroids.
