Inactivated Saccharomyces cerevisiae as virulence-enhancing agent in the toxicity test of Salmonella typhi and the immunity assay of typhoid vaccine in mice.

Mucin has been used as virulence-enhancing (VE) agent in estimation of the toxicity of bacteria to mice and the potency of bacterial vaccines such as typhoid vaccine. Discontinuance of the acceptable brand of mucin made it necessary to search for an effective alternative. Five percent inactivated Saccharomyces cerevisiae (IY) showed a significant higher VE effect on S. typhi in mice (132 colony forming units/LD50) with lower viscosity when compared to 5% mucin (740 colony forming units/LD50), and either one could promote bacterial lethal toxicity in mice more than 10(4) times. The strong correlation (r2 = 0.9765) between the immune dose and the ratio of mouse death was found by using 5% IY as VE agent in the active mouse protection test for typhoid vaccine. The relative potencies of a typhoid vaccine to the reference vaccine obtained by using 5% mucin or 5% IY as VE agent were not significant difference. Therefore, IY was a suitable substitute for mucin as a VE agent in the toxicity test of bacteria and in the immunity assay of typhoid vaccines.

Tentative diagnoses facilitate the detection of diverse lesions in chest radiographs.

The influence of clinical history upon detection has been frequently studied. In a unique investigation, detection of diverse abnormalities commonly encountered in practice was tested, and the influence of clinical history was conceptualized as suggesting a tentative diagnosis. Categorical prompts that were correct for specific abnormalities resulted in detection superior to that with unprompted reading. Prompts that were plausible but incorrect for abnormal and normal cases led to very few false-positive responses, and subsequent reports of specific abnormalities were as accurate as in unprompted reading. It is concluded that appropriate clinical history results in improved perceptual performance rather than simply improved decision-making.

Modulators of pain in the spinal cord.

Intrathecal injection of subanalgesic doses of morphine (7.5 nmol) and dynorphin-A-(1-13) (1.25 nmol) in combination resulted in a marked analgesic effect as assessed by tail flick latency in the rat. The analgesic effect of the composite of dynorphin/morphine was dose-dependent in serial dilutions and not accompanied by any signs of motor dysfunction. Synergism between dynorphin and morphine can also be demonstrated without motor dysfunction, when dynorphin is injected together intrathecally with a serial increasing dosages of morphine. Metenkephalin showed an analogous synergistic effect with dynorphin and also morphine in the spinal cord, although dynorphin (and also metenkephalin) antagonized morphine analgesia by the intraventricular route. The underlying mechanism of the interactions among different classes of opioid ligands at different levels of the central nervous system deserves further study.

[Comparison of plaque reduction and active mouse protection tests for estimating the potency of Japanese encephalitis vaccine].

The plaque reduction test for assaying the potency of Japanese encephalitis vaccine was evaluated by comparing with the active mouse protection test. The relative potency of tested vaccine to the standard vaccine were 1.40 +/- 0.250 by the plaque reduction test with coefficient variation of 0.179 and 2.54 +/- 1.152 by the active mouse protection test with coefficient variation of 0.454. There was no significant difference between the plaque reduction and the active mouse protection test in term of relative potency, but the plaque reduction method had a better reproducibility. Furthermore, the plaque reduction method showed a higher sensitivity and time saving.

Intraluminal meconium calcification without distal obstruction.

Intraluminal meconium calcification of unknown mechanism is reported in three neonates who did not have distal intestinal obstruction. The condition was benign in all three cases and did not require surgical intervention. This new form of intra-abdominal calcification should be differentiated from other forms that invariably are due to surgical disorders.

Enteroclysis aided by an electric pump.

An electric pump is recommended that facilitates enteroclysis in patients undergoing radiographic examination of the small bowel. Because the pump produces a constant and rapid flow of barium, and thus increased distention, its use enhances the demonstration of small bowel lesions. The authors have performed more than 30 examinations using this pump with total success.

Biological properties of Trimeresurus mucrosquamatus venom and its fractions.

Trimeresurus mucrosquamatus venom was separated into two principle fractions by Sephadex G-100 gel filtration. Fraction I (FI) contained the main part of lethal and hemorrhagic activities and minor part of proteolytic activity on casein. The lethal dose of FI determined by the i. v. injection was significantly smaller than that of the i. p. injection, while the result of fraction II (FII) was reversed. The venom was susceptible to heat, pH and cysteine-HCl but not to albumin, sodium glutamate, sorbitol and glycine.
