Ammonia removal from compost leachate using zeolite. III. Regeneration of zeolite columns.

The effects of hydraulic reteneration time (HRT), and the strength of the regenerating solution on the ammonia adsorption capacity and the zeolite regeneration were studied using bench-scale packed zeolite columns. A 0.6 M NaCl solution fed at a HRT of I h was preferred for the regeneration process, and more than 95% of adsorbed ammonium ions were recovered after using 7-8 bed volumes (BV) of the regenerating solution. The adsorption-regeneration time ratio was approximately 5:1. High concentration of potassium ions in the composting leachate competed with NH4+ ions for the exchange sites, resulting in a reduction in the efficiencies of ammonia removal and zeolite column regeneration. However, Zeolite still proved to have a great potential as a medium for ammonia removal in treating composting leachate.

Ammonia removal from compost leachate using zeolite. II. A study using continuous flow packed columns.

Bench-scale packed zeolite columns were set up and operated to investigate the continuous removal of ammonium ions from compost leachate. The effects of hydraulic retention time (HRT), and particle size of the zeolite on the ammonia adsorption capacity were studied. For both the coarse particle and the powdered zeolite columns, higher ammonia removal efficiencies were achieved with longer HRT (i.e., lower influent flow rate) tests. At the same HRT, ammonia removal efficiencies from tests with powdered zeolite were generally 20% higher than tests with the coarse particle zeolite. A HRT of 6 hours was found appropriate for efficient ammonia removal, and an operating capacity of 1.31 mg N/g zeolite was obtained. Over 98% of the ammonia input from the influent was consistently removed for over 5 bed volumes (BV) of compost leachate flowing through the zeolite column. Zeolite proved to have a great potential as a medium for ammonia removal in treating composting leachate.

Hydatidosis in a Chapman's zebra (Equus burchelli antiquorum).

A 14-year-old male Chapman's zebra had been treated for a cervical granulomatous cellulitis for 3 months prior to sudden death associated with myocardial fibrosis. Incidental findings at necropsy included multiple 3-8-cm unilocular cysts in the liver and lungs. Cysts contained either a clear fluid with or without hydatid sand or a gelatinous substance with foci of mineralization. Light and scanning electron microscopic examination of the hydatid sand revealed free protoscolices as well as intact and ruptured brood capsules with protoscolices attached to the germinal membrane. The protoscolices had 2 rows of 36-38 rostellar hooks with a length of 25-30 microm. The cyst wall consisted of inner germinal, intermediate laminated, and outer adventitial layers. Hydatidosis caused by Echinococcus granulosus was diagnosed based on the unilocular cysts, multiple protoscolices formed in a brood capsule, typical trilayered cyst wall, and herbivorous intermediate host. This is the first reported case of animal hydatidosis in Taiwan. The infection is thought to have been established 12 years ago in South Africa prior to importation.

The clinical features and surgical results of malignant eyelid tumors.

BACKGROUND: To determine the clinical characteristics and surgical results of patients with malignant eyelid tumors. METHODS: A retrospective study was used to review patients who had malignant eyelid tumors and underwent surgery at our hospital from 1984 to 1999. The therapies for these tumors were surgical excision with immediate peripheral tissue examination of frozen section and Mohs' micrographic surgery. RESULTS: We reviewed 77 patients with malignant eyelid tumors. Their ages ranged from 17 to 98 years with mean age of 63.4 years. The three most common tumors, in order of frequency, were basal cell carcinoma (45 cases, 58.4%), sebaceous adenocarcinoma (14 cases, 18.2%), and squamous cell carcinoma (13 cases, 16.9%). Tumor developed most commonly on the lower eyelid. Six (7.8%) patients had local recurrence after an average period of 3 years and 9 months. Meanwhile, regional metastases were found in 7 patients (9.1%) and distant metastases were found in 4 patients (5.2%). CONCLUSION: In our study, wide excision with frozen section control for malignant eyelid tumor was a relatively useful method. Cosmetically and functionally acceptable results can be achieved after eyelid reconstructive surgery.

A peritoneal mesothelioma in a captive aardwolf (Proteles cristatus).

A 10-year-old male aardwolf (Proteles cristatus) was presented abdominal distention and emaciation for 3 months. Physical examination revealed firm abdominal masses with effusions. Cytologic assessment of the effusion showed uniform round tumor cells with a surface brush border. Necropsy showed white velvety masses covering the peritoneal surface of the liver, gall bladder, stomach, omentum, mesentery, spleen, intestine, abdominal wall and diaphragm. Histologic examination demonstrated papillary projections, lined with cuboidal tumor cells supported by fibrous connective tissue cores, arising from the serosa of visceral organs. Cytoplasmic vacuolation and a surface brush border were evident on some cells under light microscopy. Tumor cells stained positive for both cytokeratin (AE1/AE3) and vimentin. Electron microscopy showed prominent surface microvilli, rough endoplasmic reticulum, mitochondria and desmosomes in tumor cells. This may be the first reported case of peritoneal mesothelioma in a captive wild aardwolf.

[Development of donor cells in chicken-duck chimeric embryos].

Chicken W chromosome-specific DNA fragments were obtained after transformation, extraction, purification and digestion with EcoR I of pUGD1201. The W chromosome-specific DNA probe was a 1.2 kb insert, which was labelled by the random-technique using a Digoxigenin Labeling Kit. Chicken PGCs from a line of White Leghorns were microinjected into Maya duck blastoderms at stage X. The embryos at 6,8,9,12,16,18 days of incubation were used to study the development of chicken PGCs in the chimeric embryos. Used chicken W chromosome-specific DNA probes by in situ hybridization, development of donor cells in chicken-duck chimeric embryos were studied. In 54 chimeric embryos, there were different degrees of chimerism in the organs. The chimeric rate of the livers was the highest, which of the gonads was the lowest. It is demonstrated that somatic and germline chimeras can be yielded successfully by transferring stage X chicken blastodermal cells.

Selective termination of aneuploidy utilizing rapid fluorescence in situ hybridization detection techniques.

Twin pregnancy following assisted reproductive technology with a euploid fetus and a coexisting aneuploid co-twin constitutes a conflicting situation; therefore, it is important for the genetic constitution of each co-twin to be diagnosed accurately and promptly for parental genetic counseling and subsequent aggressive management. A 35-year-old woman, gravida 1, with a 2-year history of infertility, presented bilateral fallopian tubal obstruction at her infertility workups, for which she received in vitro fertilization; subsequently she conceived a twin pregnancy. She underwent genetic amniocentesis at 16 weeks' gestation, as indicated by an advanced maternal age. Presented with the diagnosis of twin pregnancy with discordancy for trisomy 21, a rapid fluorescence in situ hybridization (FISH) technique for aneuploidy mapping was applied for subsequent abdominal selective fetal reduction. The FISH technique facilitates the rapid analysis of uncultured amniocytes. Normal (disomic) and trisomic samples can be distinguished clearly and rapidly for subsequent selective fetocide. The FISH technique is an important tool in prenatal diagnosis and clinical applications.

[Genetic studies of 13 loci in Guangdong Han population by two multiplex PCR systems and its forensic application].

OBJECTIVE: The genetic studies of 13 short tandem repeats(STRs) loci in two multiplex amplification systems were carried out on Chinese Han population in Guangdong. METHODS: DNA samples from 328 unrelated individuals were screened. The 13 loci were D3S1358, vWA, FGA, D8S1179, D21S11, D18S51, D5S818, D13S317, D7S820, D16S539, TH01, TPOX and CSF1PO. The PCR products were analyzed and genotyped by ABI 377-96 Sequencer. RESULTS: The combined power of discrimination (DP) was 0.999999999999993 and the combined paternity of exclusion(PE) was 99.999%. These 13 loci met the Hardy-Weinberg expectations. CONCLUSION: The two multiplex amplification systems were very useful in forensic case investigation.

Serum withdrawal-induced post-transcriptional stabilization of cyclooxygenase-2 mRNA in MDA-MB-231 mammary carcinoma cells requires the activity of the p38 stress-activated protein kinase.

Overexpression of the cyclooxygenase-2 (COX-2) gene is observed in several neoplastic diseases. However, molecular mechanisms involved in the regulation of expression of COX-2 are not well understood. In this report, we describe a unique post-transcriptional regulatory mechanism of COX-2 mRNA stabilization in MDA-MB-231 cells, a highly metastatic cell line derived from a human mammary tumor. High levels of COX-2 mRNA, protein, and enzyme activity were induced by serum withdrawal, which were potently inhibited by the addition of serum or >100-kDa serum factor. Nuclear run-on analysis and actinomycin D chase experiments indicate that regulation is primarily at the level of post-transcriptional mRNA stability. Interestingly, SB203580, an inhibitor of the p38 stress-activated protein kinase (SAPK), and overexpression of the dominant-negative p38alpha construct potently inhibited the serum withdrawal-induced COX-2 mRNA levels. Indeed, the half-life of COX-2 mRNA decreased from 9 to 4.5 h after SB203580 treatment, suggesting that signal transduction by the p38 SAPK pathway is required for COX-2 mRNA stability.

Analysis of chlorophenoxy acid herbicides in water by large-volume on-line derivatization and gas chromatography-mass spectrometry.

This work presents a modified method to analyze chlorophenoxy acid herbicides in water samples. The herbicides 2,4-D (2,4-dichlorophenoxyacetic acid). Silvex (2,4,5-trichlorophenoxypropionic acid) and 2,4,5-T (2,4,5-trichlorophenoxyacetic acid) were used to evaluate the method. The method involves extraction of samples by a graphitized carbon black cartridge, and on-line derivatization in the GC injection port using a large-volume (10-20 microl) direct sample introduction (DSI) device with tetraalkylammonium salts. The analytes were then identified and quantitated by ion-trap gas chromatography-mass spectrometry. The large-volume DSI injection-port derivatization technique provides sensitivity, fast and reproducible results for chlorophenoxy acid herbicides residues, to quantitation at 0.1 to 0.2 microg/l in 500-ml water samples. An enhanced characteristic mass chromatogram of molecular ions of butylated chlorophenoxy acid herbicides with a significant chlorine isotope pattern by electron impact ionization MS allows us to determine herbicides residues at trace levels in aqueous samples. Recovery of the herbicide residues in spiked various water samples ranged from 70 to 99% while RSDs ranged from 1 to 13%.

Amperometric immunosensors based on protein A coupled polyaniline-perfluorosulfonated ionomer composite electrodes.

A very sensitive immunosensor based on polyaniline/ Nafion/protein A (PA/NF/PrA) composite electrodes has been developed for the amperometric immunoanalysis with urease-labeled immunoreagents. The use of urease conjugated goat anti-RIgG (GaRIgG-Ur) as the labeled antibody and urea as the substrate with an amperometric detection at -200 mV (vs Ag/AgCl) resulted in a dynamic range of 50-2000 ng mL-1 and a low detection limit of 10 ng/mL (64 pM) for the immunoanalysis of rabbit immunoglobulin G (RIgG). Because of the special affinity between protein A and RIgG, the PA/NF/PrA electrode can be regenerated repetitively by changing the pH of the buffer solutions. Characteristics of the PA/NF/PrA/RIgG immunosensor and optimal conditions for the competitive immunoanalysis of RIgG with FIA were studied.

The effects of spatial frequency overlap on face recognition.

The effects of spatial frequency overlap between pairs of low-pass versus high-pass images on face recognition and matching were examined in 6 experiments. Overlap was defined as the range of spatial frequencies shared by a pair of filtered images. This factor was manipulated by processing image pairs with high-pass/low-pass filter pairs whose 50% cutoff points varied in their separation from one another. The effects of the center frequency of filter pairs were also investigated. In general, performance improved with greater overlap and higher center frequency. In control conditions, the image pairs were processed with identical filters and thus had complete overlap. Even severely filtered low-pass or high-pass images in these conditions produced superior performance. These results suggest that face recognition is more strongly affected by spatial frequency overlap than by the frequency content of the images.

Induction of integrin-associated protein (IAP) mRNA expression during memory consolidation in rat hippocampus.

We have previously demonstrated that hippocampal integrin-associated protein (IAP) gene expression is associated with memory formation in a one-way inhibitory avoidance learning in rats. In the present study, we further investigated the role and mechanism of IAP involved in memory consolidation in rats. Because of the minute amount of IAP present in the brain, we have adopted the quantitative reverse transcription-polymerase chain reaction (RT-PCR) method. Our results revealed that hippocampal IAP mRNA expression is approximately fourfold higher in rats showing good memory retention (GM, retention score of 600 s) at 3 h, but not at other time points, after training when compared with the poor memory rats (retention score < 80 s). On the other hand, integrin alphav mRNA level was markedly increased ( approximately twofold), while integrin beta3 mRNA level was decreased ( approximately 50%) at 1 h post-training. Further, separate sets of RT-PCR analysis revealed that IAP5 and IAP6 mRNA expressions, but not that of IAP7, were markedly increased in GM rats 3 h post-training. Moreover, regional distribution studies revealed that different isoforms of the IAP gene are similarly distributed in different brain areas, while IAP7 has been the predominant form present in astrocyte cells. These results together suggest that IAP mRNA expression is indeed induced upon training, rather than that the GM rats have constitutively higher levels of IAP. The unparallel change of IAP and integrin mRNA expressions as far as time-course is concerned suggests that they are possibly involved in different forms and stages of memory processing. Further, IAP5 and IAP6 are more closely associated with memory consolidation, while IAP7 may constitute the major isotype for signal transduction in astrocyte cells.

Does face recognition rely on encoding of 3-D surface? Examining the role of shape-from-shading and shape-from-stereo.

It is now well known that processing of shading information in face recognition is susceptible to bottom lighting and contrast reversal, an effect that may be due to a disruption of 3-D shape processing. The question then is whether the disruption can be rectified by other sources of 3-D information, such as shape-from-stereo. We examined this issue by comparing identification performance either with or without stereo information using top-lit and bottom-lit face stimuli in both photographic positive and negative conditions. The results show that none of the shading effects was reduced by the presence of stereo information. This finding supports the notion that shape-from-shading overrides shape-from-stereo in face perception. Although shape-from-stereo did produce some signs of facilitation for face identification, this effect was negligible. Together, our results support the view that 3-D shape processing plays only a minor role in face recognition. Our data are best accounted for by a weighted function of 2-D processing of shading pattern and 3-D processing of shapes, with a much greater weight assigned to 2-D pattern processing.

Characteristics of pale, soft, exudative broiler breast meat.

The objective of the study was to characterize pale breast meat, compare it with normal colored breast meat, and determine whether it should be considered pale, soft, and exudative (PSE). Characteristics of 20 normal and 20 pale broiler breasts, obtained at a commercial slaughter plant, were evaluated. Compared with normal meat, the pale breast meat had a significantly (P < 0.05) lower pH (5.7 vs. 5.9), higher color L* value (60.0 vs. 55.1), higher drip loss (1.34 vs. 0.87%), lower marinade uptake (31.2 vs. 44.3%), and lower cooking yields (95.2 vs. 105.8%). Protein solubility in pale samples was slightly (P < 0.05) lower than in normal samples, which suggests increased protein denaturation in the pale breasts. Correlations between pH and L* value (r = -0.76), pH and marinade uptake (r = 0.64), sarcoplasmic protein solubility and L* value (r = -0.71), and sarcoplasmic protein solubility and moisture uptake (r = 0.66) and cooking yield (r = 0.66) were significant (P < 0.05). Correlations between total protein solubility and moisture uptake or cooking yields were not significant. The low ultimate pH of pale breast muscle appears to be the main determinant of its low water-holding capacity (WHC). This lower pH was unrelated to a higher lactate concentration or glycolytic potential of the pale muscle. Further research is needed to determine the causes of the low pH and possible measures to increase the pH (and functionality) of pale broiler breast muscle. Because the pale breast muscle has a low WHC, it can be considered PSE.

Nitric oxide synthase inhibitor ameliorates oral total parenteral nutrition-induced barrier dysfunction.

The expression of inducible nitric oxide synthase (iNOS) is increased in the intestine and results in mucosal damage after endotoxin challenge. Although the oral administration of total parenteral nutrition (TPN) solution promotes bacterial translocation (BT) and increases the intestinal permeability, the role of NO in the nutrition-induced loss of mucosal barrier function remains unclear. The distribution of fluorescein isothiocyanate-dextran (FITC-dextran, 4400) across the lumen of small intestine in rat was examined to investigate the role of NOS activity on the intestinal permeability under oral TPN feeding. Fifty-one rats were randomly divided into 4 groups. Group I (control group) was fed with rat chow, group II received TPN solution orally. Groups III and IV received TPN solution supplemented with NOS inhibitors. On day 9, FITC-dextran was injected into the intestinal lumen. After 30 min, blood samples were taken from portal vein and analyzed for plasma FITC-dextran level by fluorescence spectrophotometry. Samples of small intestine were frozen and sectioned in a cryostat for morphological and NOS histochemical studies. Homogenates of small intestine were used for NOS activity measurement. The plasma level of FITC-dextran showed a significant increase (P < 0.05) in rats fed with oral TPN compared with the control ones. Supplement with NOS inhibitors significantly decreased the intestinal permeability in groups III and IV compared with group II. Similarly, the total NOS activities showed a significant 2-fold increase (P< 0.05) in group II, and NOS inhibitors decreased the elevated NOS activity. These data suggest that oral TPN feeding for 9 days leads to an increase in permeability to dextran and the total NOS activity of small intestine, and both induction of the intestinal permeability and NOS activity were inhibited by treatment with NOS inhibitors. Addition of S-methylisothiourea (SMT), an iNOS selective inhibitor, profoundly inhibited 66% of the induced iNOS activity (P < 0.05) and reduced 74% of the diet-induced increase in intestinal permeability (P < 0.05) in group II. The induced permeability change in rats receiving oral TPN is mainly due to the activity of intestinal mucosal iNOS. The induction of iNOS is an important mediator for intestinal barrier dysfunction. Administration of SMT, which specifically decreases iNOS activity, is useful in the prevention of diet-induced barrier failure.

Sphingosine-1-phosphate signaling via the EDG-1 family of G-protein-coupled receptors.

The bioactive lipid sphingosine-1-phosphate (SPP) is abundantly formed and released during the activation of platelets by thrombotic stimuli. Once exported, SPP interacts with the G-protein-coupled receptors (GPCR) of the EDG-1 family. SPP binds to EDG-1 with the dissociation constant of approximately 8 nM and induces signal transduction events such as mitogen-activated protein kinase (MAP kinase) activation, decrease of cAMP levels, remodeling of the actin cytoskeleton, among others. EDG-1 is a prototypical member of a large family of GPCRs that interact with glycero- and sphingolysolipid phosphates, namely, SPP and lysophosphatidic acid (LPA). Three other GPCRs, trivially termed EDG-3, EDG-5, and EDG-8, are also high-affinity receptors for SPP. The four SPP receptor subtypes regulate different intracellular signal transduction pathways. In vascular endothelial cells, cooperative signaling between EDG-1 and EDG-3 subtypes of SPP receptors results in adherens junction assembly, cell survival, morphogenesis into capillary-like networks, and angiogenesis. SPP acts distinctly, albeit cooperatively, with polypeptide angiogenic factors, resulting in the formation of mature neovessels. Thus SPP signaling as an extracellular mediator via the EDG-1 family of GPCRs may be a heretofore unrecognized mechanism for the regulation of angiogenesis and vascular endothelial cell function.
