The role of selenium-dependent and selenium-independent glutathione peroxidases in the formation of prostaglandin F2 alpha.

In recent years, growing evidence suggests that glutathione peroxidases (GSH-Pxs), both selenium-dependent GSH-Px (Se-GSH-Px) and selenium-independent GSH-Px (non-Se-GSH-Px) play an important role in the biosynthesis of prostaglandins and leukotrienes and in the regulation of key enzymes associated with the arachidonic acid cascade. The precise nature of their involvement in eicosanoid metabolism, however, is not yet completely understood. In the study reported here, we have systematically determined the catalytic efficiencies of Se-GSH-Px and non-Se-GSH-Px toward prostaglandin (PG) G2 (PGG2) and PGH2. Se-GSH-Px exhibited high catalytic activity for the reduction of PGG2 as indicated by Km and Vmax values of 12 microM and 78 mumol/min/mg, respectively, whereas PGH2 was found to be a poor substrate, an indication that Se-GSH-Px reduces the hydroperoxide moiety but not the endoperoxide moiety of PGG2. The kinetic constants of Se-GSH-Px toward PGG2 were comparable to those determined for such classical substrates as H2O2 and cumene hydroperoxide. In contrast to Se-GSH-Px, non-Se-GSH-Px associated with cationic isozyme II of glutathione S-transferases (GSTs) from sheep lung cytosol was very active in the conversion of PGH2 to PGF2 alpha with a Vmax of 960 nmol/min/mg and a Km of 77 microM. This study shows that PGF2 alpha formation by non-Se-GSH-Px occurred in a GSH-dependent reduction of either PGG2 or PGH2. When PGG2 was used as the substrate for non-Se-GSH-Px, a novel intermediate compound appeared and was later identified by several methods of structural analysis as 15-hydroperoxy PGF2 alpha. Thus, the reductive cleavage of the endoperoxide occurs faster than the 15-hydroperoxide reduction allowing 15-hydroperoxy PGF2 alpha to accumulate briefly. A study of GSTs from several different tissues and species indicated that the transformation of PG endoperoxides to PGF2 alpha is catalyzed specifically by GST isozymes, which contain Ya size subunits. This specificity of GST isozymes in PG biosynthesis, coupled with their tissue-specific expression, may be a mechanism by which the body modulates the type of PGs produced in these tissues. Also, these results suggest a possible interaction of Se-GSH-Px and non-Se-GSH-Px in the biosynthesis of PGF2 alpha.

Primary structure of elephant pituitary prolactin.

Tryptic digests of elephant pituitary prolactin (elePRL) were separated by reverse phase high performance liquid chromatography (HPLC) and paper electrophoresis. From the amino acid composition, the amino acid sequencing of selected peptides, and from their alignment with expected tryptic peptides from ovine prolactin (oPRL), the primary sequence of elePRL is proposed.

Primary structure of fox pituitary growth hormone.

Tryptic digests of fox growth hormone (fGH) were separated by reverse phase high performance liquid chromatography (HPLC) and by paper electrophoresis. From the amino acid composition of these tryptic peptides and from their alignment with the expected tryptic peptides from bovine growth hormone (bGH), the primary structure of fGH is proposed. There are only 17 amino acid residues which are different in these two growth hormone molecules.

Effect of centrally and peripherally administered beta-endorphin on food intake in rats.

A role for beta-EP in the regulation of food intake has been suggested as a contributory factor in the obesity of some genetically obese animal models. Studies undertaken to determine whether continuous administration of beta-EP could alter food intake in normal rats are described. The present studies demonstrated that continuous subcutaneous infusion with beta-EP was ineffective in modulating food intake, but that acute intraperitoneal or intracerebroventricular administration stimulated food intake in previously food deprived or satiated animals, respectively. These results suggest that beta-EP is not involved in the long-term regulation of food intake, but under certain conditions it may play some role in the regulation of individual meals. It is speculated that the latter activity may result from the action of other appetitive regulatory hormones.

Haemorrhagic shock encephalopathy.

Three children suffered from acute onset of convulsions and progressive coma associated with hyperpyrexia, profound shock and generalized bleeding tendency. No causative agent could be identified. Despite aggressive resuscitation they all died. Post-mortem examination revealed cerebral oedema, petechial haemorrhages of the gut, lungs and kidneys, and a generalized depletion of lymphocytes in the lymphoid organs. Features were compatible with haemorrhagic shock encephalopathy, which is a highly fatal disease. The possible role of hyperpyrexia in its pathophysiology is discussed. More careful case identification and research in various possible aetiological factors may help elucidate its pathogenesis.

Lack of mixed agonist-antagonist properties of [Gln8-Gly31]-beta h-EP-Gly-Gly-NH2 and [Arg9,19,24,28,29]-beta h-EP in the rat vas deferens neuroeffector junction: studies with naloxone, beta-funaltrexamine and ICI 174,864.

The 1-27 truncated fragment of beta h-endorphin (beta h-EP) as well as [Gln8,Gly31]-beta h-EP-Gly-Gly-NH2 or [Arg9,19,24,28,29]-beta h-EP exhibited opiate agonist activity in the rat vas deferens bioassay; the potency of these peptides was 3 to 6 times less than that of beta h-EP. None of these compounds exhibited any degree of antagonism towards the inhibitory action of beta h-EP. Naloxone antagonized and reversed the inhibitory action of beta h-EP and its analogues though with varying potencies. The apparent naloxone-pA2 value for beta h-EP was 8.94; that for [Gln8-Gly31]-beta h-EP-Gly-Gly-NH2 was 8.08 and that for [Arg9,19,24,28,29]-beta h-EP was 8.38. beta-Funaltrexamine (beta-FNA) potently antagonized the inhibitory action of beta h-EP following non-equilibrium kinetics. Tissue preincubation with 10 nM beta-FNA for 60 min followed by extensive washing caused a 10-fold increase in the beta h-EP IC50. However, 10 nM beta-FNA caused only a 1.2 increase in the IC50 of [Gln8,Gly31]-beta h-EP-Gly-Gly-NH2 and a 4.1-fold increase in the IC50 of [Arg9,19,24,28,29]-beta h-EP. In contrast, preincubation of the tissue with 3 microM ICI 174,864 did not modify the potency of beta h-EP or its structural analogues. However, a 60 min pretreatment with 10 microM beta-FNA followed by the addition of 3 microM ICI 174,864 revealed a further decrease in the potency of the opiopeptins compared with tissues exposed to beta-FNA alone or ICI 174,864 alone. In conclusion, the inhibitory action of these peptides is remarkably sensitive to beta-FNA antagonism; in addition the peptides act as pure opiate agonists in marked contrast with the agonist-antagonist properties described in the CNS.

Insulin-like growth factor-II (IGF-II): a potential autocrine/paracrine growth factor for human breast cancer acting via the IGF-I receptor.

Insulin-like growth factor-II (IGF-II) is a potent mitogen for several types of cultured cells and tissues. We have studied the interaction of IGF-II with a panel of cultured human breast cancer cell lines, examining the possibility that these cells synthesize and secrete IGF-II activity which could have autocrine/paracrine functions. Synthetic IGF-II was mitogenic in five of seven cell lines tested, including the estrogen receptor-positive lines MCF-7L, ZR75-1, and T47D and the estrogen receptor (ER)-negative lines Hs578T and MDA-231. IGF-II was slightly less potent than IGF-I in stimulating DNA synthesis in MCF-71 cells, an effect that paralleled its ability to compete for [125I]IGF-I binding in these cells. Affinity labeling studies revealed that IGF-II could also compete for binding to the 130,000 mol wt alpha-subunit of the IGF-I receptor. A monoclonal antibody to the IGF-I receptor inhibited the mitogenic effects of IGF-II in MCF-7L and MDA-231 cells, suggesting that this receptor mediates the growth effects of IGF-II in these breast cancer cells. Using a RIA and a RRA, IGF-II-like activity was detected in conditioned medium extracts processed to remove IGF-binding proteins from several breast cancer cell lines, with the highest levels found in conditioned medium from MCF-7L and T47D cell lines. IGF-II mRNA transcripts in MCF-7L and T47D cells were identified by Northern blot analysis and were confirmed by RNase protection assay. IGF-II mRNA was increased by estrogen in MCF-7L cells.(ABSTRACT TRUNCATED AT 250 WORDS)

[Protective effects of an ethanolic extract and essential oil of Curcuma kwangsinensis S. against experimental liver lesions in mice].

The ethanolic extract and the essential oil of Curcuma kwangsinensis were found to be effective in lowering the elevated SGPT induced by CCl4, TAA and BSP retention. Histological examination showed that hepatic damages of the extract-and oil-pretreated mice were less than those of the CCl4 control group. The ethanolic extract helped to shorten sodium pentobarbital sleeping time in both normal and CCl4 intoxicated mice. In addition, the essential oil exhibited an inhibitory effect on triglyceride accumulation in mice. The toxicity of the extract and the oil was found to be low: oral LD50 was 86.8 +/- 12g crude drug/kg (ethanolic extract) and 1.10 +/- 0.08g/kg (essential oil).

[The increase of protein synthesis by zinc can resist the damage induced by streptozotocin on islet cells].

The beneficial effect of ZnCl2 on islet cells injured by streptozotocin was observed directly on the cultured cells and its possible mechanism was analysed. The results were as follows: (1) The number of viable cells in culture was reduced from 70 x 10(5)/ml to 43.93 +/- 1.16 x 10(5)/ml 12 hours after adding normal saline plus STZ (3.0 mmol/L), and this reduction could be alleviated by adding ZnCl2 (0.25, 0.5, 1.0 mmol/L) in varying amounts and a dose-response relationship was found. The number of viable cells was returned to 47.39 +/- 0.88 x 10(5)/ml, 58.06 +/- 2.29 x 10(5)/ml, and 67.72 +/- 1.48 x 10(5)/ml respectively in the culture with ZnCl2 of different levels. (2) The effect of ZnCl2 was reversed when cycloheximide (100 micrograms/ml), a protein synthesis inhibitor, was added into the cultural medium, and the number of viable cells was again decreased from 63.17 +/- 2.15 x 10(5)/ml to 45.77 +/- 0.76 x 10(5) ml. No obvious effect was observed when cycloheximide was given alone. (3) The experiment of 3H-leucine incorporation into islet cells showed that protein synthesis was increased slightly and insignificantly after adding ZnCl2 (1.0 mmol/L). However, there was a significant increase of protein synthesis in cells when ZnCl2 (1.0 mmol/L) was added with STZ (3.0 mmol/L). The results suggest that ZnCl2 has a protective effect on the damaged islet cells induced by STZ. The increase of protein synthesis may be one of the mechanisms involved in the action of ZnCl2, which strengthens the repairing ability of islet cells after injury.

Changes in cardiac and respiratory function of Antarctic Research Expedition members--preliminary report.

Cardiac and respiratory functions were estimated in 16 Chinese Antarctic Research Expedition members (Third expedition) before and after spending three months in the Antarctic. No significant changes were found in ECG examination. In cardiac function test, prolonged LVET and abbreviated PEP were found. PEP/LVET decreased significantly. Respiratory ventilation function examination showed an increase in FEV1 and FEV25 and a decrease in ERC. Results showed that cardiac and respiratory functions remained almost unchanged in the Expedition members. It is also suggested that a physiological compensation may have occurred in these members' cardiac functions either in the Antarctic or during the return sea voyage.

Utilization of mevalonate by aorta for the synthesis of medium-chain n-fatty acids (C8, C10) and acylglycerols.

[2-14C]Mevalonate was incorporated into acylglycerols by bovine aorta. The majority of the radioactivity was found in diacylglycerols. Hydrolysis, followed by analysis by thin-layer chromatography, paper chromatography, gas chromatography, and gas chromatography-mass spectrometry showed that the radioactivity was greatest in the medium-chain n-fatty acids, caprylic (C8) and capric (C10) acids. Triacylglycerols and diacylglycerols have important functions in the maintenance of the metabolic integrity of organs and their constituent cells. Therefore, medium-chain fatty acids may play significant roles, such as solubilization and transport of cholesterol and regulation of metabolism, as constituents of diacylglycerols. The results suggest that the trans-methylglutaconate shunt of mevalonate metabolism functions in the aorta.

High molecular weight forms of insulin-like growth factor II and its binding protein identified by protein immunoblotting.

Insulin-like growth factor (IGF)II is a mitogenic polypeptide which circulates in association with a binding protein(s). Immunoblotting studies were performed in human serum and indicate that:(1)a approximately 200 kDa covalently-linked IGF-II/binding protein complex is antigenically related to the 30 kDa binding protein, (2)IGF-II prohormone is associated with this complex, and (3)a major portion of the IGF-II prohormone immunoreactivity in human serum is present in fractions which would not be detected by standard radioimmunoassay methods. Our data provide insight regarding the inter-relationships of IGF-II and its binding protein, and direct evidence for the presence of IGF-II prohormone in human serum.

Structural characterization of Ba2YCu3O7 by high resolution transmission electron microscopy.

We show that high-resolution transmission electron microscopy can be used directly to reveal the structure of the metal framework and its defects in Ba2YCu3O7. The use of dynamical effects under carefully selected and controlled conditions allows the detection of long-range order in the disposition of the oxygen vacancies, but inevitable misalignments make the quantification of the order parameter difficult. Specimen preparation methods are compared, and a wide-angle convergent-beam pattern is demonstrated.

Beta-endorphin-(1-27) antagonizes beta-endorphin- but not morphine-, D-Pen2-D-Pen5-enkephalin- and U50, 488H-induced analgesia in mice.

beta-Endorphin-(1-27), administered intraventricularly has been previously reported to block the analgesia induced by beta-endorphin injected intraventricularly. The present study was to determine if the blocking effect of beta-endorphin-(1-27) was specific to beta-endorphin which stimulates epsilon receptors, but not to other opioids with activity at different opioid receptors. The antagonistic effects of beta-endorphin-(1-27) on the analgesia induced by beta-endorphin (epsilon-opioid receptor agonist), D-Ala2-NMePhe4-Gly-ol-enkephalin(DAGO) and morphine, (mu-opioid receptor agonists), D-Pen2-D-Pen5-enkephalin(DPDPE) and D-Ala2-D-Leu5-enkephalin(DADLE) (delta-opioid receptor agonists) and U-50, 488H (kappa-opioid receptor agonist) were studied. beta-Endorphin-(1-27) injected intraventricularly, at doses which, when injected alone did not produce analgesia, antagonized the analgesia induced by beta-endorphin given intraventricularly. However, the analgesia induced by DAGO, morphine, DPDPE, DADLE and U-50, 488H given intraventricularly was not antagonized by beta-endorphin-(1-27). The data suggest that beta-endorphin-(1-27) selectively blocks the analgesia induced by the stimulation of epsilon receptors but not by the stimulation of mu, delta, and kappa receptors. The results support the previously proposed hypothesis that beta-endorphin produces its analgesia by stimulating specific epsilon receptors.

Sister chromatid exchange and cell cycle patterns of normal human bone marrow cells after in vitro exposure to cytostatic drugs.

The effect of different cytostatic chemotherapeutic drugs on the frequency of sister chromatid exchanges and growth kinetics of the bone marrow cells in normal adults was studied by means of an in vitro bromodeoxyuridine chromosome labeling method. Cytosine arabinoside, harringtonine, methotrexate, vincristine, and 6-mercaptopurine were tested. Three of these agents induced significantly high frequency of sister chromatid exchange (p less than 0.01 or p less than 0.05). The sister chromatid exchange frequency induced by cytosine arabinoside and that induced by 6-mercaptopurine were higher than those of the control group. The increased frequency of sister chromatid exchange predominated in C and A group chromosomes. At the concentration adopted by the authors, the growth kinetics were little influenced by these drugs.

Synthetic human seminal alpha-inhibin-92 selectively suppresses follicle-stimulating hormone release in vivo.

A 92-amino acid polypeptide, alpha-inhibin-92 (alpha-IB-92), has been isolated and characterized from human seminal plasma and found to be active in suppressing follicle-stimulating hormone (FSH) release in vitro. In the present in vivo study, intravenous injection of synthetic alpha-IB-92 (4 and 20 micrograms) significantly suppressed FSH release (P less than 0.001), whereas this peptide had no effect on luteinizing hormone (LH) release in 1-day orchidectomized male rats. In contrast, third ventricular injection of alpha-IB-92 (0.02, 0.4, 4, or 20 micrograms) had no effect on FSH and LH release in 1- or 2-day orchidectomized rats. These results indicate that alpha-IB-92 exerts a FSH suppressing activity by direct action on the pituitary gland.

Isolation and characterization of growth hormone from blue fox pituitary glands.

Growth hormone has been purified to homogeneity from blue fox pituitary glands. It has 191 amino acids with two disulfide bridges and a single tryptophan residue. The somatotropin activity is only 8% when compared with the bovine hormone in the receptor-binding assay. From radioimmunoassay data using baboon antisera to porcine or bovine growth hormone, the fox hormone has 14-17% immunoreactivity of bovine or porcine hormone.

New segment synthesis of alpha-inhibin-92 by the acyl disulfide method.

The thiocarboxyl group reacts with diaryl disulfides to give an unsymmetrical acyl disulfide in dimethylformamide (DMF) and a symmetrical diacyl disulfide in aqueous DMF. Both acyl disulfides react with the alpha-amino group to form the peptide bond. The method was used in a new segment synthesis of alpha-inhibin-92 (alpha-IB-92) with use of 2,2'-dipyridyl disulfide as activator. Thiocarboxyl peptides were synthesized by the solid-phase method on 4-[alpha-(Boc-Gly-S)benzyl]phenoxyacetamidomethyl-resin. The segments alpha-IB-92-(1-34)SH (I), Msc-alpha-IB-92-(35-65)SH (II), Msc-alpha-IB-92(66-92)OH (III), and Msc-alpha-IB-92-(35-92)OH (VI) were prepared in yields of 33, 36, 41, and 25%, respectively, with use of crystalline symmetrical anhydrides in double and triple coupling protocols. Segments I, II, and III were used in a 3-segment synthesis of alpha-IB-92 with an overall yield based on starting resin of about 8% while a 2-segment synthesis using I and IV gave 11%. An all stepwise synthesis of alpha-IB-92 gave 4.5%.
