Enzymatic methyl esterification of specific glutamyl residue in corticotropin.

Ovine corticotropin (alpha s-ACTH) was enzymatically methylated with purified calf brain protein methylase II (protein O-methyltransferase; S-adenosyl-L-methionine: protein-carboxyl O-methyltransferase, EC 2.1.1.24) and S-adenosyl-L-[methyl-14C]methionine. After incubation for 60 min at 37 degrees C, 30 mol % of the hormone was methylated on the basis of the [14C]methyl incorporation. In order to assess the location of methylation, the modified peptide was digested with pepsin. Analytical results derived from studies on the peptic digest led to the suggestion that the alpha s-ACTH-(6--28) peptide fragment was esterified. Because there is only one available methylation site at Glu-28, these results indicate that Glu-28 of alpha s-ACTH was specifically methyl esterified to yield [Glu(OMe)28]-alpha s-ACTH.

Secondary structure prediction of anterior pituitary hormones. Lack of correlation between predicted values and circular dichroism data.

The secondary structures of human somatotropin, human choriomammotropin, ovine and porcine prolactin, human, ovine and porcine beta-lipotropin, human and ovine lutropin, human thyrotropin, human corticotropin, alpha-melanotropin and human beta-melanotropin have been predicted by the method of Chou & Fasman. Predicted contents of alpha-helix and beta-sheet do not correspond well with values estimated from circular dichroism spectra.

Human beta-endorphin. Immunoreactivity of synthetic analogs with various chain lengths.

Immunoreactivity of synthetic human beta-endorphin analogs with various chain lengths has been examined using a specific radioimmunoassay. It was found that beta-endorphin-(1--21) and analogs of shortened chain exhibit no immunoreactivity, whereas beta-endorphin-(1--15) possesses significant in vitro opiate activity. It appears that immunoreactivity of beta-endorphin resides in the COOH-terminal segment of residues (22--31). The data also show the lack of correlation between opiate and immunological activities of beta-endorphin.

Enzymatic methyl esterification of pituitary polypeptides.

Methyl esterification of pituitary polypeptides by protein methylase II (S-adenosylmethionine:protein-carboxyl O-methyltransferase, EC. 2.1.1.24) has been investigated. Ovine lutropin and adrenocorticotropin (alpha1-39-ACTH) were found to be good methyl acceptor substrates, followed by beta-lipotropin. While the alpha-subunit of lutropin had nearly equal the methyl accepting activity of lutropin, the beta-subunit was devoid of accepting activity. The maximum amount of esterification occurred between 15 and 30 min at 37 degrees C) depending on the methyl acceptor molecule. The rate of the methyl esterification of adrenocorticotropin fragments was also studied. While alpha7-38-ACTH had less than half of alpha1-37-ACTH methyl accepting capacity, alpha1-17-ACTH did not serve as methyl acceptor. However, when a mixture of the two fragments was preincubated, the resulting mixture had full alpha1-39-ACTH activity.

Adrenocorticotropin. 52 Synthesis and biological activity of adrenocorticotropic peptides with cystine bridges.

Three analogs of the amino terminal nonadecopeptide of adrenocorticotropin which incorporate cystine cystine bridges between positions (5, 10), (3, 10), or (2, 10) have been synthesized. All of the peptide analogs showed reduced biological activity; however, the peptide with the 5, 10 cystine bridge was shown to possess significantly higher lipolytic activity in rat fat cells than the peptides with a 3, 10 or 2, 10 cystine bridge.

Synthesis of biological activity of human beta-endorphin analogs with disulfide bridges.

Three analogs of human beta-endorphin containing internal cystine bridges have been synthesized and their biological activities have been determined. It was found that the analogs with disulfide bridges between positions 17 and 26 or 11 and 26 retained full opiate activity as estimated by guinea ileum assay, whereas the analog with a cystine bridge between positions 7 and 26 showed reduced opiate activity. None of the analogs retained significant immunoactivity as revealed by radioimmunoassay.

Characterization of turkey myelin basic protein isolated by a simple procedure.

Highly purified basic proteins have been isolated from bovine and turkey brains by a novel method employing acid-acetone extraction. The final product gave a single band on polyacrylamide gel electrophoresis at pH 4.3 and in the presence of sodium dodecyl sulfate. Both proteins have arginine at the COOH-terminus while the NH2-terminal residue cannot be detected and is probably blocked. A higher ratio of histidine to lysine and a greater proportion of serine and valine was found for the turkey compared with the bovine protein. Both proteins contain one tryptophan and two methionine residues. However, it was found from cyanogen bromide treatment that there is a marked difference in the location of one of the methionine residues, while the tryptophan-containing peptides liberated after trypsin digestion have different mobilities on peptide maps. When dissolved in water these proteins give a typical random coil curve from circular dichroism (CD), whereas in 80% methyl alcohol they assume a 25% alpha-helix.

Demonstration by radioligand-binding assay of the structural similarity of a partially synthetic growth hormone recombinant molecule to its natural analog and to native human growth hormone.

The recombinant resulting from the noncovalent interaction of a natural amino-terminal fragment of reduced and carbamoylmethylated human GH (residues 1-134) with a synthetic carboxyl-terminal fragment of reduced and carbamoyl-methylated human GH (residues 141-191) reacts in seven immunoassay systems and two receptor assay systems in a similar manner and with comparable potency to its naturally occurring analog, but with less avidity overall than does the native hormone. This similarity of reactivity is strong evidence for similarity of conformation among these molecules. The demonstration that a synthetic fragment can restore radioligand-binding properties to a proteolytic fragment of human GH implies that partially synthetic variant GH molecules have biological activity.

Specific radioimmunoassay of human beta-endorphin in unextracted plasma.

With an antiserum against human beta-endorphin (beta-EP) crossreacting less than 2% with human beta-lipotropin (beta-LPH) by weight we have developed a radioimmunoassay that can detect 1 pg beta-EP in diluted raw plasma. In a.m. fasting plasma of 14 normal subjects beta-EP ranged from less than 5 to 45 pg/ml. beta-EP was elevated in untreated, but normal in successfully treated Cushing's disease; undetectable in a patient with adrenal adenoma; extremely high in Nelson's syndrome; and elevated in a patient with bronchogenic carcinoma before, but undetectable after tumor resection. In subjects with intact hypothalamic-pituitary-adrenal axis, beta-EP was undetectable after dexamethasone and increased after metyrapone administration and insulin-induced hypoglycemia. beta-EP concentration was considerably lower in serum than in simultaneously collected plasma, but increased in serum left unfrozen for several hours after clot removal. Thus, beta-EP behaves like a hormone responding to the same stimuli as ACTH and beta-LPH and blood appears to contain enzymes both generating and destroying immunoreactive beta-EP.

Studies on pituitary follitropin. V. Isolation of the subunits of the human hormone and reaction of the amino groups with acylating agents.

The alpha and beta subunits of human follitropin were isolated in a high state of purity. The tryptophan fluorescence of the native hormone and the isolated beta subunit are different. The N-terminus of the alpha and beta subunits was identified as valine and aspartic acid respectively. While recombination of the isolated alpha and beta subunits restores the electrophoretic mobility of the intact hormone, its receptor binding activity cannot be fully regenerated. Substitution of the human follitropin alpha by an ovine lutropin alpha subunit, to form a recombinant with the follitropin beta subunit, generates a complex with 2-3 receptor binding activity of the native human follitropin and the same activity as ovine follitropin. Acylation of the intact hormone does not disrupt the quaternary structure but leads to complete inactivation. Acylation studies with the subunits suggests the crucial role of the epsilon-amino groups of the alpha subunit in determining biological activity.

Adrenal steroids indirectly modulate morphine and beta-endorphin effects.

The pharmacological effects of morphine or the postulated physiological opiate, beta-endorphin, were compared in adrenalectomized and sham control animals. Pretreatment of adrenalectomized rats and mice with the synthetic glucocorticoid dexamethasone 2 hr before opiate injections abolished the adrenalectomy-induced sensitization to parenteral opiates. This effect of dexamethasone was completely blocked by cycloheximide, a protein-synthesis inhibitor. After parenteral injection ot tritiated morphine, total tritium counts in the blood and brains of adrenalectomized mice were greater than in sham controls; this effect was also blocked by dexamethasone pretreatment. Administration of SKF 525-A before morphine produced results which suggest that this putative inhibitor of the mixed-function oxidase metabolizing enzyme system also alters opiate potency. Collectively, these studies provide evidence that adrenalectomy causes a decrease in morphine metabolism which enhances the parenteral potency of this opiate through increased bioavailability. Furthermore, the dexamethasone reversal of this effect appears to depend upon its de novo induction of protein components of the opiate-metabolizing system. The possibility that a physiological interplay between endorphins and corticosteroids exists is suggested by the dexamethasone blockade of both the pharmacological effects and the toxicity of intravenous beta-endorphin in adrenalectomized mice.
