Turnover of palmitic and arachidonic acids in the phospholipids from different brain areas of adult and aged rats.

[3H]Palmitic acid and [14C]arachidonic acid were injected together into the cerebral ventricle of 4-month and 24-month-old rats. At different time intervals from the injection, the distribution of these fatty acids in the lipids from different brain areas was examined. The fatty acids were rapidly incorporated into the lipids through different mechanisms. The time-specific activity relationship indicate that the utilization of the fatty acid differs according to the different areas and aging decreases the utilization of both the fatty acids. The decline of arachidonic acid incorporation into phospholipids is particularly evident, indicating that aging affects mainly the utilization of polyunsaturated fatty acids.

Effect of chronic treatment with phosphatidyl serine on phospholipase A1 and A2 activities in different brain areas of 4 month and 24 month old rats.

The activities of phospholipase A1 and A2 were tested in mitochondria and microsomes from different cerebral areas of both 4 month and 24 month old rats, after 30 days i.p. treatment with ox Brain Cortex Phosphatidyl Serine (BC-PS). In the brain areas from control animals a rather great decline in its ability to hydrolize the fatty acids of the main phosphoglycerides was found. Taking into account only the effect of treatment with BC-PS on the modification of phospholipase activities caused by aging, we found that the treatment was able to balance the enzymatic functions altered by aging in several of the area examined. The importance of this result on the lipidic metabolism of the aging brain is also discussed.

[Strain gauges for measuring the tension at the site of sutures. Use of the original instrument with a personal technic].

The Authors have developed an instrumentation suitable for suture line tension measuring during plastic operations for large incisional hernias. A full description is given of the measuring technique and of the original instrumentation, including two steel bars called "fachiri" and an electric measuring system with strain gauges. With the employment of this technique, relaxing incisions of the muscle aponeurosis can be reduced to the minimum. Being the suture line tension one of the most important causes of relapsing, the Authors believe that this technique provides a sure advantage for the long term results of plastic operations.

[Enteral nutritional support in the treatment of the high-risk surgical patient].

The Authors report about their experience of 27 cases of Total Enteral Feeding. The role of the nitrogen balance inversion, from negative to positive, as an expression of protein anabolism, is specially emphasized. This parameter proves more suitable than the serum albumin level increase, of low statistical significance.

Turnover of ethanolamine phosphoglycerides in different brain areas of adult and aged rats.

The turnover of ethanolamine glycerophospholipids (EGP) has been determined in six different cerebral areas of 4-month and 22-month-old rats, by injecting [3H]glycerol together with [14C]ethanolamine into the lateral ventricle of the brain. The areas examined behave quite differently in respect to their utilization of the most simple precursors of phosphoglyceride biosynthesis. The incorporation of both glycerol and ethanolamine is already complete as early as 2-4 hours and then reutilization begins, at least for the so called fast pools of phosphoglycerides. The different slopes of the specific activity of the two precursors in EGP suggest a high degree of variance among catabolic rates in the different brain regions. In aged rats the utilization of the water-soluble precursors of EGP synthesis decreases in all brain areas and these data suggest that aging may have a different effect on the catabolic activities as well as phospholipid biosynthesis.

The influence of polyunsaturated phosphatidylcholine on brain lipid synthesis during aging.

The synthesis of choline phosphoglycerides and ethanolamine phosphoglycerides is noticeably decreased in brain microsomal membranes of aged rats as compared to that of two-month-old rats. Experimental evidence is presented that shows that the addition of a diglyceride preparation obtained from a polyunsaturated phosphatidylcholine to microsomes of aged rats practically restores the decreased synthesizing activity of both choline and ethanolamine phosphoglycerides. It is proposed that adding diglycerides to aged membranes affects the properties of the membranes and probably enzyme-lipid interactions.

Interaction of ethidium bromide with ribosomes. Absorption, fluorescence, circular dichroism and sedimentation studies.

It is shown in this work that the binding of ethidium bromide to yeast ribosomes occurs through intercalation in the double-stranded rRNA regions and produces changes in the ribosomes structure, yielding unfolded subparticles, and even partial separation of proteins from rRNA at very high binding ratios. The addition of Mg2+ prevents these structural changes, probably by partial inhibition of the dye binding.

Binding of ethidium bromide to ribosomal RNA. Absorption, fluorescence, circular and electric dichroism study.

The interaction between ethidium bromide and ribosomal RNA has been studied by means of absorption, fluorescence, circular and electric dichroism measurements in the near ultraviolet and visible regions at low ionic strength (1 . 10(-3) and 6 . 10(-3). The results have been interpreted on the basis of a model of interaction involving the intercalation of the phenanthridinium ring of the dye in the double-stranded regions of the RNA molecule, resulting in an increase of the dye-dye interactions as compared to DNA, and a stiffening of the intercalation regions.

Serine incorporation into phospholipids in vivo and serine phosphoglyceride metabolic transformations in cerebral areas from adult and aged rats.

Serine phosphoglycerides (SPG) in the nervous tissue are synthesized by base-exchange reaction. SPG may be decarboxylated and converted into ethanolamine phosphoglycerides (EPG). This latter reaction may be followed in turn by several enzymatic reactions leading to choline phosphoglycerides (CPG) via three successive methylations. These metabolic pathways are important for several reasons and their contribution to membrane phospholipid renewal has been clearly demonstrated. Base-exchange is the only metabolic route to synthesize SPG; it was therefore possible to test this pathway in vivo by injecting labeled serine. Serine incorporation into phosphoglycerides was reduced in cortex and hippocampus from aged rat brain as compared to adult, while no change in the SPG successive metabolic transformation was found. Based on both the results presented herein and previous ones obtained in vitro the authors suggest that the reduced serine incorporation may be due to reduced uptake into cells from aged animals. This finding leads to the conclusion that the above mechanisms for partial renewal of the phospholipid molecule may play an important role in the brain structure of aged animals.

TAF test in primary esophageal carcinoma: comparison with other tumor markers.

An ELISA method for the determination of circulating specific HSV-TAA antibodies has recently become available (TAF test). The presence of TAF was tested in serum of 154 patients with primary esophageal carcinoma, collected in three institutions. The overall TAF-test positivity rate was 57.1%, being significantly lower in stage IV than in stage III patients. The concordance rate between TAF and CEA, ferritin, TPA, SCC and TATI was low, suggesting that TAF is probably independent of the other tumor markers evaluated. The clinical role of TAF-test determination in patients with esophageal carcinoma is currently under evaluation.

Combination chemotherapy using vincristine, adriamycin, cyclophosphamide (VAC) alternating with ifosfamide and etoposide (IE) for advanced soft tissue sarcomas: a phase II study.

Chemotherapy options in patients with advanced soft tissue sarcomas (STS) remain presently inadequate. In this phase II study the activity and toxicity of a combined alternating VAC/IE regimen in advanced and/or metastatic STS was evaluated. VAC (vincristine, total dose of 2 mg, adriamycin 70 mg/m2 and cyclophosphamide 600 mg/m2) was given as intravenous bolus infusion on day 1, followed after 21 days by IE (ifosfamide, 1.8 g/m2 infused over 1 h, daily for 5 consecutive days, with mesna uroprotection, plus etoposide at 500 mg/m2 infused over 2 h on the first day). Courses were repeated every 3 weeks. Twenty patients were treated, 12 of whom had previously been given chemotherapy for advanced and/or metastatic disease. Treatment was feasible in ambulatory setting, with good patient compliance. Myelosuppression, overall acceptable, was the major dose-limiting toxicity, while non-hematological side effects were minimal. An overall response rate of 45% was observed (95% CI, 26%-66%), with 2 complete and 7 partial remissions, achieving 75% in the subset of untreated patients. Median survival time was 10 months in the whole group (range, 4-26+ months) and 14 months in responder patients (range, 9-26+ months).

Clinical relevance of vascular endothelial growth factor and thymidine phosphorylase in patients with node-positive breast cancer treated with either adjuvant chemotherapy or hormone therapy.

PURPOSE: To determine the role of the two angiogenic peptides, vascular endothelial growth factor (VEGF) and thymidine phosphorylase (TP) (the latter also being a target enzyme for cytotoxicity of 5-fluorouracil and methotrexate), and conventional prognostic factors in predicting relapse-free survival (RFS) and overall survival (OS) probabilities in two cohorts of patients with node-positive breast cancer (NPBC) treated with either adjuvant chemotherapy (CMF [cyclophosphamide, methotrexate, 5-fluorouracil] schedule) or hormone therapy (tamoxifen). PATIENTS AND METHODS: We studied two groups of 137 and 164 patients with NPBC, median follow-up of 72 months for both, treated with adjuvant chemotherapy or hormone therapy, respectively. The cytosolic levels of VEGF and TP were determined in the primary tumor by original immunometric methods. The association between VEGF and TP and of these angiogenic peptides with other prognostic indicators were tested by using the Spearman correlation coefficient (for continuous variables) or the Kolmogorov-Smirnov test (for dichotomous variables). Results of the clinical outcome were analyzed by both univariate and multivariate (for RFS only) Cox regression models in which VEGF and TP were treated as continuous variables. RESULTS: In the CMF group, the concentrations of VEGF and TP ranged from 5.8 to 7798 pg/mg of protein (median, 87.5 pg/mg) and from 1.2 to 904 U/mg (median, 138.2 U/mg), respectively. There was no significant association between the two angiogenic peptides. VEGF was not associated with any other variable, whereas TP showed a positive association with age and an inverse association with the number of involved nodes. In the tamoxifen group, the concentrations of VEGF (5.9-2482; median, 79.3 pg/mg protein) and TP (6.1-1542; median, 146.5 U/mg) were similar to those of the CMF group, and the two angiogenic peptides were not correlated. VEGF was positively associated with age and was inversely associated with estrogen receptor and progesterone receptor, whereas TP was not associated with any other variable. Univariate analysis in the CMF group showed that VEGF and TP were significantly predictive of both RFS and OS. Likewise, the number of involved axillary nodes was significantly associated with both RFS and OS. Univariate analysis in the tamoxifen group showed that TP did not significantly influence either RFS or OS. On the contrary, VEGF levels were significantly predictive of both RFS and OS, as were the number of involved nodes, estrogen receptor concentrations, and progesterone receptor concentration. In the multivariate analysis on RFS in the CMF group, VEGF, TP, their first-order interaction term, and age were significant and independent predictive factors. In the tamoxifen group, only VEGF and the number of involved nodes were significant and independent predictive factors. DISCUSSION: The results of our study suggest that high levels of TP and low levels of VEGF characterize the patients with NPBC treated with adjuvant CMF who have the highest likelihood of favorable outcome. Low levels of VEGF and the presence of less than three involved axillary nodes characterize the patients with NPBC treated with adjuvant tamoxifen who have the highest likelihood of favorable outcome. This information may be useful to plan future studies to better select the patients with NPBC for conventional adjuvant treatments as well as to monitor the efficacy of novel therapeutic strategies of adjuvant therapy based on inhibition of angiogenesis.
