Stoichiometric and reversible phosphorylation of a 46-kDa protein in human platelets in response to cGMP- and cAMP-elevating vasodilators.

Recently, we reported the purification of a 46-kDa membrane-associated platelet protein which is phosphorylated in intact platelets and platelet membranes by cGMP- and cAMP-dependent protein kinases (Halbrügge, M., and Walter, U. (1989) Eur. J. Biochem. 185, 41-50). Here we demonstrate that both cGMP- and cAMP-dependent protein kinases catalyze the rapid incorporation of up to 1.4 mol of phosphate/mol of this purified vasodilator-stimulated phosphoprotein (VASP). A specific rabbit antiserum was prepared which recognized both the 46-kDa dephospho form and the 50-kDa phospho form of VASP in Western blots. In untreated washed platelets, VASP was found to be present primarily as a 46-kDa dephosphoprotein. Sodium nitroprusside (100 microM) raised the intracellular platelet cGMP concentration from approximately 0.44 to 4.1 microM, without a significant effect on the cAMP level, and converted up to 50% of VASP to the 50-kDa phospho form. Prostaglandin E1 (10 microM) raised the platelet cAMP concentration from approximately 4.4 to 28.4 microM, without a significant effect on the cGMP level, and shifted up to 67% of VASP to the 50-kDa phospho form. Removal of the vasodilators sodium nitroprusside and prostaglandin E1 from the platelet suspension was followed by a return of the cyclic nucleotide concentration to basal levels and subsequent conversion of the 50-kDa phospho form of VASP to the 46-kDa dephospho form. The results support the hypothesis that VASP phosphorylation is an important component of the intracellular mechanism of action of these vasodilators in human platelets.

[Research schools in pharmacy. 6. Gunther Wagner and his students].

The aim of this article is to trace the historical development of Günther Wagner and his research school. Wagner is an idealy teacher. The scientific programme is represented by 2 books, 502 papers and 54 dissertations. It has some main roots: synthesis of potential active substances and pharmaceutical analytic. Wagner had 991 students (pharmacists), 45 made their doctor graduate under Wagner and 6 are becoming professor. The research conditions in Leipzig were good and the research school of Wagner has an important public and social recognition in the GDR and in other countries.

[Research schools in pharmacy. 5: Alexander Tschirch (1856-1939) and his students].

Alexander Tschirch was one of the most important teachers of pharmacy. In 1890 he was appointed Professor of Pharmacy and Pharmacognosy at Bern and he was a good director of his institute. The scientific programme is represented by 21 books, 388 papers and 158 dissertations. The main topics were investigations of plants (phytochemical) and researchs for pharmacopoeias. Tschirch had 2300 Students, 158 made their doctor-graduate and 11 are becoming professor. The research conditions in the pharmaceutical institute Bern were good and the research school of Tschirch has an important public and social recognition, also Tschirchs work provided the bases of the modern phytochemical research of plants.

[The development of pharmacy in German universities in the 19th century].

This article analyses the development of pharmacy as an academic discipline in Greifswald, Göttingen, Ingolstadt-Landshut-München, Breslau and Marburg. The research shows that the establishing of independent institutes of pharmacy under competent leadership (for instance Ernst Schmidt in Marburg) played a decisive role in developing the discipline pharmacy.

[Research schools of pharmacy. 4. Heinrich A. Beckurts (1855-1929) and his students. The history of pharmaceutical science: 37].

In 1886 Heinrich A. Beckurts was appointed Professor of pharmaceutical Chemistry at Braunschweig. He was one of the most important teachers of pharmacy there and a good director of his institute. The scientific programme is represented by 146 papers an 52 dissertations; main roots were research of plants (phytochemical) and researchs for pharmacopoeias. Beckurts had 2000 students, 52 made their doctor-graduate under Beckurts and five are becoming professor. The research conditions in the pharmaceutical Institute Braunschweig were good and the research school of Bekkurts has an important public and social recognition, also Beckurts work provided the basis of the modern study for pharmacy.

[The contribution of German professors to pharmaceutical chemistry and phytochemistry in the 19th and 20th centuries. 40. The history of pharmaceutical science].

The aim of this article is to trace some features of the historical development of phytochemistry. In the period from 1870 to 1920 pharmacy at universities attempted a new phase, the emancipation as an independent science. The article analyses the contributions to phytochemistry by the professors E. Schmidt (1845-1921), H. Thomas (1859-1931), H. Beckurts (1855-1929) and their research schools as well as those of K. Polstorff (1846-1911) and m. Scholtz (1861-1919).

Low-grade soft tissue sarcomas of the extremities. Analysis of risk factors for metastasis.

Of 130 patients with low-grade localized soft tissue sarcomas of the extremities treated at Memorial Hospital between 1968 and 1978, 18 (14%) developed metastases. The risk of metastasis could not be predicted by the clinical or pathologic size, tumor depth, proximal location, recurrence before presentation or subsequent to treatment at our institution, the type of surgical and adjuvant therapies, and the adequacy of surgical margins. The median age of patients with metastases was 10 years older than the other patients (57.5 years and 47.5 years, respectively). Of the more common sarcomas in this study, malignant peripheral nerve tumors and malignant fibrous histiocytomas had the highest prevalence of metastasis (27% (three of 11) and 17% (three of 18), respectively). Although patients with low-grade extremity sarcomas treated during the first 5 years of this review had an increased rate of metastasis, no other patient, tumor, or treatment variable examined significantly differed between the two time periods. Although grade alone is a good predictor of the risk of metastasis from soft tissue sarcomas of the extremity, 14% of patients in this series with low-grade histology developed metastases. Further review of larger numbers of these tumors should be undertaken in an effort to more definitively identify risk factors predictive of metastatic potential in low-grade sarcomas.

Horizontal two-dimensional electrophoresis with immobilized pH gradients using PhastSystem.

Protocols for horizontal two-dimensional electrophoresis with immobilized pH gradients in the first dimension were modified for horizontal micro two-dimensional electrophoresis using PhastSystem. Different equilibration conditions of the first-dimensional immobilized pH gradient gel strip prior to second-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis were evaluated. Silver stained two-dimensional patterns were obtained within 3.5 h.

Soft-tissue sarcoma: initial characteristics and prognostic factors in patients with and without metastatic disease.

Five hundred and sixty-five patients with soft-tissue sarcoma were admitted to Memorial Sloan-Kettering Cancer Center from July 1, 1982 to July 1, 1985. Clinical and pathologic factors were analyzed to identify initial characteristics and to determine factors associated with prognosis and the development of metastasis. In decreasing order of frequency, the most common sites of primary tumor were the extremity (51.1%), retroperitoneum/viscera (28.8%), trunk (15.9%), head and neck (3.7%) and breast (0.4%). The most common histologic types were liposarcoma (21.4%), malignant fibrous histiocytoma (20.2%), leiomyosarcoma (20.0%), fibrosarcoma (11.0%), and tendosynovial sarcoma (9.6%). One hundred and twenty-eight patients (22.7%) had metastases in a median follow-up period of 19 months. Metastasis was more common in patients with retroperitoneal and visceral sarcomas than in patients with extremity sarcomas. Patients with leiomyosarcomas were more likely to develop metastases than those with other histologic types. Fibrosarcoma and liposarcomas were the least likely to develop metastases in this time frame. Tumors greater than 5 cm in diameter occurred more frequently in patients with metastases. Actuarial survival was related to the site of primary tumor and the histologic type and grade of malignancy. Multivariate analysis indicated that patients with low-grade sarcomas, extremity sarcomas, and fibrosarcomas and patients without metastatic disease demonstrated significantly improved survival. In patients with metastases, disease-free interval (less than 365 or greater than or equal to 365 days) was significantly related to overall survival.(ABSTRACT TRUNCATED AT 250 WORDS)

Prognostic factors for local recurrence and survival in patients with localized extremity soft-tissue sarcoma.

The charts of 423 patients with localized extremity soft-tissue sarcoma treated at our institution over a 10-year period (1968-1978) were reviewed. Data were subjected to both univariate and multivariate analysis, with independent variables in the multivariate analysis appearing in capital letters. Factors associated with an increased risk of local recurrence included the following: AGE greater than 53; PRESENTATION WITH RECURRENT DISEASE; HIGH TUMOR GRADE; positive regional nodes; TREATMENT BY LIMB-SPARING SURGERY (LSS); invasion of vital structures (LSS only); INADEQUATE MARGINS; and biopsy with delayed definitive resection. Survival was adversely affected by the following: AGE greater than 53; PAINFUL MASS; PROXIMAL SITE; SIZE greater than or equal to 10 cm; HIGH TUMOR GRADE; POSITIVE NODES; invasion of vital structures; TREATMENT BY AMPUTATION; INADEQUATE MARGINS; and local recurrence after treatment at our institution. Significant variations in both local recurrence and survival according to histopathology were also observed, with EMBRYONAL RHABDOMYOSARCOMA, ANGIOSARCOMA, and MALIGNANT PERIPHERAL NERVE TUMORS emerging as independent predictors of local recurrence. Using the Cox models for local recurrence and survival, patients were stratified into high-, intermediate-, and low-risk categories based on the presence or absence of each variable. Risk factor analysis should be part of the overall evaluation of each patient with extremity sarcoma.

[The university pharmacy and the university pharmacist in history].

The major purpose of the research is to show the influence of university pharmacy and university pharmacist on the development of pharmaceutical science. The lectures in materia medica were frequent realized in university pharmacy since middle age. In the 18th century the university pharmacy was in some cases the precursor to the institute of pharmaceutical chemistry. In the paper also a summary is given of the importance of extensive activity of the university pharmacists J.D. Leers, G.L.C. Rousseau, J.R. Spielmann, F.C. Lieblein, A.F. Duflos, E.F. Jahns, W. Mettenheimer, C. Brunnengräber, C.A. Laubinger, J. Stamm, O.H. Gerke and E. Weber.

[Research schools in pharmacy. 2. Ernst A. Schmidt (1845-1921) and high area of scholarship; the history of pharmaceutical science. 33].

The aim of this article is to trace the historical development of Ernst Schmidt and his research school. Schmidt was an ideally director and teacher. The scientific programme is represented by two books, 197 papers and 117 dissertations. The scientific programme has two main roots: research of plants (phytochemical), especially alkaloids (63% of the publications) and chemical researches. Schmidt had 1200 students, 109 made their doctor-graduate under Schmidt and 14 are becoming professor. The research conditions in the pharmaceutical Institute Marburg were very good and the research school of Schmidt has an important public and social recognition still today.
