Cell proliferation in a malignant angioendothelioma during sequential chemotherapy.

The rate of cellular proliferation was studied in the neoplastic cells of a malignant angioendothelioma and in its inflammatory infiltrate by means of optical radioautography after incorporation ex vivo of tritiated thymidine. One third of the normal looking, as well as the atypical, endothelial cells were engaged in the cell cycle of proliferation. This large pool has been effectively suppressed during subsequent chemotherapy which resulted in partial regression of the neoplasm. The proliferative activity of the infiltrate was higher inside the neoplasm than at its periphery.

Double test of spontaneous rosettes with sheep and mouse erythrocytes. Statistical studies and usefulness in malignant evolutive and nonevolutive lymphoproliferative diseases.

We have demonstrated the log normality of the distribution of sheep rosette-forming cells and mouse rosette-forming cells' values obtained with lymphocytes isolated from the peripheral blood of 135 healthy human beings and 57 patients suffering from chronic lymphocytic leukemia and well differentiated lymphocytic lymphosarcoma with bone marrow infiltration either in evolution or in remission. In evolutive cases, the absolute numbers of mouse rosette-forming cells rose as well as the lymphocytosis, whereas the absolute numbers of sheep rosette-forming cells were normal or even increased in spite of an impressive drop of their percentage. In nonevolutive cases, the absolute numbers of sheep rosette-forming cells and mouse rosette-forming cells were lowered to half of the normal values as a consequence of the lymphopenia induced by chemotherapeutic agents.

[Logarithmo-normal distributions of relative and absolute values of human lymphocytes forming spontaneous rosettes in presence of sheep and mouse erythrocytes in normal adult].

The percentages and absolute numbers of peripheral blood lymphocytes forming rosettes with sheep and mouse red blood cells were established in 135 normal human beings. We found SRFC values almost similar to the proportion of so called "active" rosettes, whereas the values of MRFC were low. SRFC and MRFC values had a logarithmico-normal distribution. The absolute number of the two types of rosettes was decreased in the subjects more than 70 years old.

[Hepatic lesions and coagulation disorders in pulmonary embolism: a clinico-pathological case].

The authors described the case of a 49-patient suffering from obstructive broncho-pneumopathy and ischemic myocardiopathy complicated by multiple pulmonary embolism. The clinics were dominated by signs of major hepatic deficiency and of intravascular disseminated coagulation with fibrinolysis. The clinical and biological parameters evoking pulmonary embolism in that intricated chart were discussed.

[Bronchospasm, a sign revealing pulmonary embolism].

As a clinical feature, and sometimes as a feature of the special mechanical tests of ventilation, bronchospasm has been found by the authors to be associated with pulmonary embolus. Bronchospasm was found to be the commonest feature in the 45 patients studied. The sudden onset of bronchospasm must be regarded as strong evidence for pulmonary embolus, especially in patients with a past history of bronchitis. The mechanism of these asthma-like features is discussed. The authors impugn the adrenergic system both to explain the bronchial overactivity of the patients with pulmonary embolus, and to understand how the attack commences.

[Sequential chemotherapy based on the hypothesis of a circadian rhythm of tumor proliferation].

The authors treated 41 cases of advanced solid tumors by a combination chemotherapy with methotrexate, vinblastine and cyclophosphamide administered in a sequential manner according to the hypothesis of a circadiam rhythm of proliferation of human neoplasia. A tumoral regression was observed in 66. 6 p. cent of adenocarcinomas and 86.2 p.cent of non glandular carcinomas.

Use of synchronization induced by cyclophosphamide in a methylcholanthrene sarcoma with circadian proliferation to rational sequential chemotherapy.

Owing to the circadian activity of tumoral proliferation, cyclophosphamide (CPA) morning injection exerts, on a methylcholanthrene induced sarcoma, a preferential oncostatic effect. This morning injection is followed by a temporary inhibition of mitoses preceding a high mitotic peak, explained by chemo-induced synchronization of tumor cells. Whereas single vinblastine (VLB) administration has no antitumoral effect, an injection of VLB during the described peak is able to improve oncostatic effect of cyclophosphamide. Moreover, the VLB administration at the same moment as CPA suppresses antineoplastic activity of the latter.

Histological modifications of the lymph nodes in mice bearing syngeneic grafts of tumours induced by methylcholanthrene.

Light and electron microscopical studies demonstrate that regional but also non-regional lymph nodes of C57BL mice grafted by tumours, induced by methylcholanthrene, are able to elicit a complex immunological response of the cellular, as well as of the humoral types. No significant difference was found according to the growth rate and the immunogenicity of the six tumours investigated. The cellular immunization is predominant during the two first weeks. It is mainly characterized by the appearance in the paracortical area of numerous immunoblasts which are in close connexions with'dendritic' macrophages showing multiple long cytoplasmic expansions. During the same period of time, the reticulum cells become hyperplasic in the sinus. From the 15th day to the death of the animals, the cellular immunizaiton progressively regresses while a humoral immunity develops. It shows itself as an hypertrophy of the lymph follicles and an increase in the number of medullary cord plasma cells.
