OBJECTIVE: To study the patterns of gene expression of main collagen, type I and type II in lumbar discs. METHODS: The level of collagen gene mRNA expression of type I and type II was investigated by in situ hybridization on fetal, adults and pathologic specimens with cDNA probe. Collagen in various specimens was observed by staining of sirius scarlet. RESULTS: Positive mRNA hybridization signals of collagen type I were concentrated around outlayer of annulus fibrosus. Type II collagen was highly expressed in nucleus pulposus. In adult and pathologic specimens, no hybridization signal of type II collagen was observed, and the expression level of type II collagen mRNA decreased. Positive signals appeared in nucleus pulposus. CONCLUSION: Obvious changes took place in the regulation of collagen gene expression with aging. Expression of collagen in adult and pathological disc was decreased. Abnormal appearance could be seen between collagen gene type I and type II. Expression of collagen type I mRNA appeared in nucleus pulposus.
OBJECTIVE: To explore the correlation between nm23-H1 gene and malignant and semi-malignant bone tumors. METHOD: Thirty surgical specimens were collected, including 8 osteosarcomas, 7 giant cell tumors, 4 chondrosarcomas, 2 Ewing's sarcomas, 2 malignant fibrohistocytomas, 1 neurofibrosarcoma, 1 schwannoma, and 5 metastatic cancers. The expression of nm23-H1 in these specimens was observed by in situ hybridization and immunohistochemical staining. RESULT: The expression of nm23-H1 in 4 cases of primary giant cell tumor (GCT) was high, but was low or absent in 3 recurrent ones. In 4 cases of chondrosarcomas, the expression was positive. Its expression was basically negative in Ewing's tumor, malignant fibrous histiocytoma and metastatic sarcomas. CONCLUSION: There is no association of nm23-H1 with osteosarcoma.
OBJECTIVE: To study the effective ways to restore sight and control the postoperative corneal astigmatism in eccentric penetrating keratoplasty. METHODS: Marks were made on the meridian where the center of recipient bed was located, and at that meridian the distance between the external edge of recipient bed and the limbus (two point marking technique) was also marked. Afterward, the donor graft was cut down from a donor eye at the corresponding position and sutured on the recipient bed. This technique was performed on 23 cases with eccentric corneal ulcer and perforation, and they were followed up for 3 months to 2 years. RESULTS: The grafts of 18 eyes remained transparent. The visual acuities were obtained > 0.1 in 16 cases, > 0.4 in 6 cases. Among them, visual acuity of 1.0 was obtained in 2 cases. CONCLUSIONS: Two point marking technique can reduce the postoperative corneal astigmatism and obtain satisfactory visual acuity in eccentric penetrating keratoplasty. This technique is simple and easy to perform.
OBJECTIVE: To investigate the clinical characteristics and to compare the effects of several therapies of Mooren's corneal ulcer. METHODS: 550 consecutive cases of Mooren's corneal ulcer inpatients were analysed, including the age, sex, laterality of the eye, ulcer location, perforative rate, cure rate of surgeries, recurrent rate, and the effects of conjunctiva excision, lamellar keratoplasty (LKP), LKP and plus cyclosporin A eye drop. RESULTS: The average age of onset of the corneal ulcer was 48.4 years old, the ratio of the male to the female patients was 1:0.74, the bilateral disease was 30% of the total cases, 31.5% of the bilateral disease occurred in the younger group, and 68.5% of the bilateral ulcer occurred in the older group, ulcers located at the limbus of the palpebral fissure were 70% of the total cases, perforative rate was 13.3%, 43.2% of the perforation occurred in the younger group, and 56.8% of the perforation occurred in the older group, recurrent rate of the post-operation was 25.6%, the cure rate of the first procedure of LKP and plus cyclosporin A eye drop was 73.7%, the final cure rate was 95.6%, and the preservative rate of the corneal anatomical structure of the surgeries was 99.7%. CONCLUSION: The study primarily provided the clinical characteristics of Mooren's corneal ulcer patients of China. The available data do not support the present concept of the classification of the two clinical types. The bilateral ulcer may be the malignant type of Mooren's ulcer. LKP and plus cyclosporin A eye drop was an effective treatment of Mooren's corneal ulcer.
Recent work on operant visual learning and memory in Drosophila has suggested at least three distinct memory phases. Trying to disrupt memory pharmacologically, we fed flies with ouabain or the depolarizing drugs potassium chloride (KCl), lithium chloride (LiCl) and monosodium glutamate for some specific time before training. The depolarizing drugs abolished memory very soon after training. Ouabain exerted no effect on memory within the first 20 min but abolished it more than 30 min after training. These drugs had no diminishing effects on the visual discrimination and behavioral performance of the flies during training. This result suggests that memory disruption may not be induced by nonspecific effects of the drugs. In addition, reversal training of the KCl-fed flies indicates that KCl appears not to impair the retrieval mechanism of flies. These results suggest that the specific disruptive effects of the drugs on memory formation and the existence of a short-term memory phase, are susceptible to disruption of the depolarizing drugs but unaffected by ouabain.
1. To investigate the effect of dietary fat source on host resistance to intracellular pathogens, weanling female C3H/Hen mice were fed one of three experimental diets containing, 20% by weight, lard, soybean oil or 17% menhaden fish oil plus 3% corn oil. After 4 weeks, survival of mice (n = 12/treatment group) injected intraperitoneally with 2 x 10(6) colony forming units of live Listeria monocytogenes was determined. In a second study, bacterial clearance from the liver and spleen at 2, 4 and 7 days post-challenge was determined (n = 8/treatment group). 2. We found that the survival of mice fed the diets with soybean oil or menhaden fish oil was significantly lower than those fed lard (P < 0.05). Survival rates were 58% (7/12), 33% (4/12) and 100% (12/12), respectively, for mice fed soybean oil, menhaden fish oil and lard. In the second study, mice fed menhaden fish oil had approximately 1 log10 greater bacteria in their spleens at day 4 than mice fed lard or soybean oil (P < 0.001). There were no significant treatment differences in the number of bacteria recovered from liver samples. 3. In summary, dietary fat source significantly affects murine resistance to Listeria, with diets rich in n-3 polyunsaturated fatty acids, such as from fish oil, having the most detrimental effect.
The objective of this study was to investigate the impact of feeding mice a diet rich in n-3 polyunsaturated fatty acids (PUFA) from fish oil on the interferon-gamma (IFN-gamma) response during an active infection with Listeria monocytogenes. Weanling female C3H/Hen mice were fed experimental diets containing 20% by weight one of the following fats: soybean oil, lard, or a mixture of menhaden fish oil and corn oil (17:3, w/w). After 4 weeks, mice were injected with 10(5) live L. monocytogenes, and the concentration of IFN-gamma in serum and spleen was determined 0, 2, 4, and 7 days postinfection by enzyme-linked immunosorbent assay (ELISA). Fish oil-fed mice showed significantly higher IFN-gamma in their blood at 2 and 4 days postchallenge compared with mice fed the soybean oil-containing or lard-containing diets (p < 0.001). A higher concentration of IFN-gamma was also found in the spleen homogenate of fish oil-fed mice on day 4 postchallenge (p < 0.005). To examine in vitro IFN-gamma production, splenocytes were isolated from fish oil-fed and soybean oil-fed mice on day 4 postchallenge and cultured with concanavalin A (1 microgram/ml and 10 micrograms/ml) for 24 and 48 h. There were no significant differences in the IFN-gamma concentration in cell culture supernatants between these diet treatments. This study demonstrated that the elevation in the concentration of IFN-gamma in blood and spleen during murine listeriosis is accentuated and prolonged by dietary n-3 PUFA, and these effects may not be due to changes in IFN-gamma production.
We studied the mRNA expression of p53 and MDM2 gene in osteosarcoma. Twenty osteosarcomas were examined with biotin-labelled oligodeoxynucleotide probe in situ. Of 20 tumors, 8 (40%) and 6(30%) showed positive expression of p53 and MDM2 genes, respectively. Among these, 2 tumors revealed co-expression of both p53 and MDM2 genes. This study confirmed the existence of aberrant mRNA expression of p53 and MDM2 genes in osteosarcoma.
Studying the denaturation of oxidative hemoglobin of blood preserved at room temperature, it is helpful to overall understand erythrocyte injurious during blood preserved at room temperature. Using electron spin resonance technique, we have researched the denaturation of oxidative hemoglobin of blood preserved at 35 degrees C. We discovered that blood preserved at 35 degrees C, appears ESR absorption of high spin methemoglobin (g = 6) after preservation time is 36 hours. The high spin methemoglobin ESF absorption will increase when preservation time increases. When blood is preserved at 4 degrees C, it dose not emerge ESR absorption during 60 hours preservation. SOD activity will gradually decline in erythrocyte of blood preserved at 35 degrees C when preservation time increases. Oxidative denaturation of hemoglobin is probably caused with gradual declining of antioxidation system effect in erythrocyte.
PURPOSE: To study the role of epikeratophakia in treating pediatric unilateral aphakia after traumatic cataract extraction. METHODS: We performed epikeratophakia in 43 children to treat aphakia after traumatic cataract extraction, using the corneal lenses made by ourselves. The rehabilitation of the the postoperative vision and the prevention of amblyopia were observed with a mean follow-up period of 20 months. RESULTS: All the lenses remained transparent. Postoperatively, 31 cases (72.1%) achieved uncorrected visual acuities (V.A) over 0.2, 32 cases (74.4%) achieved corrected V.A over 0.4. Most of the cases achieved the best preoperative corrected V.A with spectacles of less than 3 diopters. No severe complication occurred. CONCLUSION: Epikeratophakia is predictable with quality lens and correct surgical technique. The result suggests that the epikeratophakia is one of the best treatment for the pediatric aphakia especially for those who are not optimal for IOL implantation.
PURPOSE: To evaluate the efficacy of epikeratophakia for the treatment of severe Terrien's degeneration. METHODS: Four eyes with severe Terrien's corneal degeneration are studied. The lesions were located at the peripheral cornea and the corneal thickness was less then 1/3 of normal. Plano keratoplasty was performed. A lamellar lenticule with a 2 mm scleral rim was grafted on the cornea of which the epithelium had been removed. Keratometry and cycloplegic refraction were performed pre- and post-operatively. RESULTS: Follow-up was between 3 and 13 months (mean, 10 months). All grafts remain clear. The properative mean cylinder was 11.5 diopters and the postoperative mean cylinder was 3.9 diopters. Best corrected visual acuity was improved in 3 eyes. Interface opacification is the main complication, like other forms of epikeratoplasty. Epithelial ingrowth occurred in one eye and the lenticule was replaced. CONCLUSIONS: Epikeratophakia is a simple and effective way for the treatment of severe Terrien's degeneration. This procedure not only increases the corneal thickness, but also hepls the cornea to preserve its transparency, since the Bowman's layer remains intact.
Evaluating of 216 first time patients in our department,we divided two groups of complained teeth and non complained teeth,analyzed the location of numbers of focus,and the clinical data.The results showed the most important key procedure was to do best of full teeth examination and systematic treatment.
Four peptides of human immunodeficiency virus (HIV) located in the regions of gp41, gp120, p24 of HIV-1 and gp36 of HIV-2 have been synthesized according to the published amino acid sequences and the positions of antigenetic determinants of viruses by a solid-phase method. An indirect ELISA for detecting antibody against HIV 1/2 was established using these synthetic peptides as the coating antigens. We detected 41 control sera provided by National Institute for the Control of Pharmaceutical and Biological Products. The results indicated that the general coincident rate were 100%, the variant coefficient was less than 10%. A comparison of our reagent with Huayi, Pasteur and Jinhao reagents in detection of 90 positive sera and 140 normal sera for HIV-1/2 showed that the coincident rate was 100% with Pasteur and Jinhao, 99.57% with Huayi. All of 186 sera from patients with other diseases were negative. The reagent are stable at 37 degrees C for 4 days. The results indicated that our reagent is highly specific, sensitive and stable, it can be used for detection of HIV-1/2 infections.

The role of a transmembrane Ca2+ gradient in anion transport by Band 3 of human resealed erythrocyte ghosts has been studied. The results show that a transmembrane Ca2+ gradient is essential for the conformation of erythrocyte Band 3 with higher anion transport activity. The dissipation of the transmembrane Ca2+ gradient by the ionophore A23187 inhibits the anion transport activity. The extent of this inhibition approaches 90% as the Ca2+ concentration on both sides of the ghost membrane is increased to 1.0 mM and half-maximum inhibitions is observed at 0.25 mM Ca2+. Addition of ATP (0.4 mM) to the resealing medium can partly reestablish the transmembrane Ca2+ gradient by activation of Ca(2+)-ATPase and alleviate the inhibition to some extent. N-ethylmaleimide, an inhibitor of erythrocyte Ca(2+)-ATPase, prevents such restoration. Electron micrographs reveal that numerous larger intramembranous particle can be observed on the P-faces of freeze-fractured resealed ghosts in the absence of a transmembrane Ca2+ gradient.
OBJECTIVE: To examine the relationship between ankylosing spondylitis and heterogeneity of HLA-B27 in Chinese. PATIENTS AND METHODS: We subtyped HLA-B27 in 76 patients with ankylosing spondylitis and 1 200 healthy Chinese of Han nationality. Polymerase chain reaction was designed to amplify specific regions of class I (exon 2 and 3). Amplified sequences were tested with eight sequence-specific oligonucleotides to distinguish all HLA-B27 subtypes. RESULTS: The HLA-B27 was closely related to ankylosing spondylitis and at least four subtypes presented in Chinese patients. B2704 was the most common subtype, occurring in 54.8% and 50.85% respectively in HLA-B27 positive ankylosing spondylitis patients and healthy controls. The main HLA-B27 subtypes in the HLA-B27 positive patients distributed in similar proportion to those of HLA-B27 positive healthy controls (P < 0.1). CONCLUSIONS: The prevalences of various subtypes of HLA-B27 vary in different races. HLA-B2704 is the main subtype in Chinese Han nationality. The HLA-B27 specific residues that are shared by the subtypes might be involved in the pathogenesis of ankylosing spondylitis.
OBJECTIVE: Giant cell tumor of bone (GCT) is a comparatively common bone tumor in Chinese. Though GCT is generally considered as a benign tumor, it can pursue an aggressive course. The present study attempts to clarify whether the flow cytometric study can be an useful tool for prognosis of GCT patients. METHODS: DNA content and cell cycle studies on 96 pathologically confirmed GCT specimens and anti-GCT monoclonal antibody (GCF-5 McAb) labeling study on 30 GCTs were made by flow cytometry. RESULTS: 29 GCTs (30.2%) exhibited aneuploid and 67 (69.8%) diploid. Of 71 primary tumors, 16 (22.5%) exhibited aneuploid. The DNA index was 0.97+0.15, and the average S, G2/M phase cells 15.23+6.15%. Of 25 recurrent tumors, 13 (52%) were aneuploid, with an average DI of 1.08+0.17. The average S, G2/M phase cells was 19.56+7.12%. Between the primary and recurrent tumor groups, there was significant statistical difference both in S, G2/M phase cell percentage (P < 0.05) and DNA content (P < 0.01). There was no significant difference between Jaffe's grade I and grade II groups in DNA content or in S, G2/M phase cell percentage. GCF-5 McAb could combine with almost all cells of S, G2/M phase and part of the cells in G0/G1 phase. Significant relationship existed (r=0.468, P < 0.05) between the percentages of S, G2/M phase cells and GCF-5 positive cells. Between the primary and the recurrent tumor groups, there was a significant difference in the percentage of both S, G2/M phase cells and GCF-5 positive cells. CONCLUSION: Apparently flow cytometric study is of value in the evaluation of the biological behavior of GCT as well as the prognosis of the patients.
