Xenotransplantation of microencapsulated canine islets into diabetic rats.

Islets of Langerhans were isolated in high yields from canine pancreata. In the procedure, the pancreata were perfused and digested with collagenase, and the islets were then purified on histopaque density gradients. As many as 60,000 islets were isolated from a single pancreas. Islets were encapsulated in alginate-polylysine-alginate membranes with the aid of an air-jet droplet generator. In vitro studies demonstrated that the isolated and encapsulated islets secreted insulin in response to glucose and IBMX challenge for at least 9 weeks. In in vivo studies 6 diabetic Wistar rats were transplanted with 5,000 to 8,000 encapsulated islets each. The diabetic condition was reversed in all recipients for up to 112 days. In control animals, which received free, unencapsulated islets, the xenografts remained functional for fewer than 21 days. Microcapsules retrieved from normoglycemic transplant recipients 1 and 2 months posttransplantation were shown to contain viable islet tissue, and no cellular overgrowth was observed on capsular surfaces. The results of the study indicate a considerable clinical potential of microencapsulated canine islet xenografts.

Astragalus membranaceus and Polygonum multiflorum protect rat heart mitochondria against lipid peroxidation.

We isolated rat heart mitochondria and induced lipid peroxidation with ADP and FeSO4. Oxygen consumption and MDA formation were measured for quantitating the amount of lipid peroxidation. Using these methods, we screened the water extracts of 14 Chinese medicinal herbs for their effect on lipid peroxidation. It was found that Astragalus membranaceus inhibited 42.1 +/- 3.4% of oxygen consumption and 39.8 +/- 3.2% of MDA production at concentration of 2 mg dried herb/ml mitochondrial suspension. At the same concentration, Polygonum multiflorum inhibited 52.1 +/- 7.3% of oxygen consumption and 50.9 +/- 5.3% of MDA production. Other herbs did not inhibit lipid peroxidation to 50% of control at concentration up to 6 mg dried herb/ml mitochondrial suspension. Purification and identification of the active component(s) in Astragalus membranaceus and Polygonum multiflorum as well as their clinical application await further studies.

Growth inhibitors against tumor cells in Cordyceps sinensis other than cordycepin and polysaccharides.

Cordyceps sinensis is a parasitic fungus that has been used as a Chinese medicine for a long time. In the present study, inhibitory effects of crude methanolic extracts of C. sinensis fruiting bodies on various tumor cell lines were demonstrated. The crude methanolic extracts were fractionated into 15 fractions by silica gel column chromatography. Two of the 15 fractions (CS-36-39 and CS-48-51) significantly inhibited the growth of K562, Vero, Wish, Calu-1, and Raji tumor cell lines. The inhibitory activities were not due to the polysaccharides, which have been removed in the extracting process. The polarities of these two fractions indicated that they were different from that of cordycepin. Therefore, it is suggested that tumor cell growth inhibitors, other than cordycepin and polysaccharides, are contained in C. sinensis.

Ischemic renal failure in chronic hypoxic rats.

The after effects of renal ischemia were studied in hypoxia-adapted rats. It was felt that chronic hypoxia animals which had already adapted to a low oxygen level might be more tolerant of renal ischemic insult; however, chronic hypoxia is always accompanied by polycythemia, which may cause severe RBC trapping and consequently might enhance renal damage after renal ischemia. Chronic hypoxic rats were prepared by exposure in an altitude chamber 15 h per day for 4 weeks. The plasma sodium, potassium, urea, and creatinine levels were determined to compare the changes in these parameters between the baseline and 3 h after a 45-min occlusion of both renal arteries in 12 sea-level (SLB) controls and in 12 chronic hypoxic (CHB) and 11 chronic hypoxic plus RBC pheresis (to reduce hematocrit: CH + P) rats. From the parameters measured, the CHB rats were found to be more tolerant of renal ischemia. However, this was not the case in the rats with pheresis. It is concluded that after chronic hypoxia, some humoral factors in the plasma may play an important role in reducing the renal damage after ischemic insult.

Experimental podophyllotoxin (bajiaolian) poisoning: III. Biochemical bases for toxic effects.

The biochemical mechanism underlying the toxicity of podophyllotoxin is investigated. Previous studies from our laboratories suggested that hepatocytes were extremely sensitive to the toxicity of podophyllotoxin and a disruption of protein synthesis was suspected. Dose-response and time-course studies on the effects of podophyllotoxin on protein. RNA, and DNA syntheses on hepatocellular cultures were made. Inhibitions of protein, RNA, and DNA syntheses were demonstrated, and a direct correlated dose-response relationship on such effects was also evident. Inhibition of protein synthesis appeared to be a direct toxic effect of podophyllotoxin and occurred independently from that of RNA. The reduction of DNA synthesis was reflected by a reduction of H3-thymidine incorporation, which is believed to be the consequence of inhibition on the mitotic activity by podophyllotoxin.

[Factors related to the choice of clinic between Chinese traditional medicine and Western medicine].

The study applied Andersen's health-service utilization model to analyze the basic demographic, enabling and need factors related to the choice of traditional Chinese medicine clinic or modern Western medicine clinic by single-method-treatment (i.e. traditional Chinese medicine or modern Western medicine only) patients. During the period from August 1989 to October 1989, systemic sampling was done and a structured questionnaire survey was carried out among patients from the Out-patient Departments of 13 teaching hospitals accepting reimbursement by Labor Medical Insurance in Taiwan. The total number of valid respondents was 579: 378 (65.3%) were visiting modern Western medicine clinics and 201 (34.7%), traditional Chinese medicine clinics. There were 339 (58.6%) males and 240 (41.4%) females, aged from 15 to 85 years old, with a mean of 40.7 years. Under univariate analysis, the significant variables (p < 0.05) related to visiting the two types of clinics were: nativity, religious belief, career, general health condition, severity of illness of this episode, types of disorder as neuromusculoskeletal, digestive, circulatory, endocrine-metabolic and sense-and-skin. By logistic regression analysis, the significant variables (p < 0.05) relating to visiting two types of clinics were religion, career, and two kinds of disorders. Folk-religion believers, farmers and businessmen favored traditional Chinese medicine; and patients who suffered from musculoskeletal, sense organs or skin disorders were also likely to visit traditional Chinese medicine clinics.

Studies of the synthesis, immunology, and cytotoxicity of a cyclic octapeptide corresponding to TNF-alpha-(59-66).

In an attempt to investigate possible binding domains of the tumor necrosis factors (TNF), we have previously synthesized a cyclic hexapeptide corresponding to murine TNF-(127-132) (cTNF-1). In this report, we describe the synthesis and biological activity of another cyclic octapeptide corresponding to human TNF-alpha-(59-66) (cTNF-2). The design of these cyclic peptides is based on their high sequence homology with corresponding fragments of human TNF-alpha or TNF-beta. Similar to cTNF-1, the cyclic octapeptide cTNF-2 displayed low in vitro cytotoxicity against human HeLa and HEP-2 cell lines. The cyclic peptides cTNF-2 and cTNF-1 were then tested for the induction of interleukin-1 (IL-1) production from human peripheral blood mononuclear cells and monocytes in vitro. At low concentrations, the IL-1 levels induced by these cyclic peptides were similar to that of recombinant TNF-alpha. However, the IL-1 production by cTNF-2 stimulation was dose-dependently increased and reached that of a lipopolysaccharide (LPS; 0.1 micrograms/mL) level. These findings suggest that the fragments corresponding to human TNF-alpha-(59-66) and murine TNF-(127-132) may represent certain binding domains of the tumor necrosis factors that elicit IL-1 production.

High-performance liquid chromatography of the quinazolinocarboline alkaloid dehydroevodiamine.

A high-performance liquid chromatographic method has been developed for the determination of dehydroevodiamine (DeHE), an active principle from Evodia fruit. Plasma was denatured with acetonitrile and centrifuged, the supernatant was separated and blown dry, and the residue was redissolved in water. Bile was acidified with perchloric acid and centrifuged to yield the supernatant. Aliquots were used for analysis. Elution was isocratic on a reversed-phase column with acetonitrile-water-phosphoric acid (64:35:0.8, v/v) adjusted to pH 3.5 as the mobile phase. Ultraviolet detection was at a wavelength of 367 nm. The detection limits were 2 ng/ml for plasma and 10 ng/ml for bile. The intra-day and inter-day variations were mostly below 10%.

Stimulation of inositol phosphate production and GTPase activity by compound 48/80 in rat peritoneal mast cells.

Phospholipase C activity, GTPase activity and cytosolic-free calcium concentration in mast cells were stimulated by compound 48/80. Accumulation of inositol phosphates in rat mast cells was stimulated by guanosine 5'-[gamma-thio]-triphosphate. Guanosine 5'-[gamma-thio]triphosphate, however, exhibited no effect upon the purified phospholipase C activity and upon phospholipase C in the mast cell homogenate. The stimulatory effect of compound 48/80 upon phospholipase C activity of intact mast cells was observed to have been well correlated with that on GTPase activity of mast cell homogenate. Compound 48/80 exhibited no effect upon the binding of radioactive guanosine 5'-[gamma-thio]triphosphate to mast cell homogenate. Phospholipase C activity was verified by the above results to become affected by compound 48/80 through guanine nucleotide-binding regulatory protein.

Renal functional response to ischaemic renal failure in chronic hypoxic rats.

1. Chronic hypoxic rats are always polycythaemic. It is possible that an increase in packed cell volume may enhance erythrocyte trapping with a consequent increase in renal damage after renal ischaemia. These experiments were designed to assess renal functional changes after renal arterial occlusion in chronic hypoxic rats. 2. Chronic hypoxic rats were prepared by exposure (15h/day) to an altitude chamber (5486m) for 4 weeks. 3. After 45 min of left renal arterial occlusion, there were significant decreases in the excretion of potassium, p-aminohippurate and inulin and in the p-aminohippurate extraction ratio in 12 sea level ischaemic insulted kidneys. In 12 chronic hypoxic rats, the same parameters were changed after left renal ischaemia but only the p-aminohippurate ratio was significantly altered. 4. Administrations of 1 or 5 mg/kg phosphoramidon did not cause any significant improvement in the measured renal parameters in both kidneys and in both groups of rats after ischaemia. 5. In the second experiment, the rats were challenged by rapid infusion of 10 ml of saline intravenously, and urine was collected for 90 min from each ureter. Four hours after left renal arterial occlusion, the insulted kidney showed increased water and sodium excretion in both sea level and chronic hypoxic rats. However, 24 h after left renal ischaemia, the responses of sea level and chronic hypoxic rats were different. Urinary excretion was significantly reduced in sea level rats, but was almost normal in chronic hypoxic rats. 6. This report suggests that some beneficial factors after chronic hypoxia might play important roles in reducing the damage after renal ischaemia.

Antihepatotoxic activity of phenolic flavan-3-ols and their derivatives.

The protective activity against carbon tetrachloride induced hepatotoxicity of several phenolic flavan-3-ols and their derivatives has been assessed. Our research showed that monomers possessing a pyrogallol moiety as the B-ring had greater activity and this was not directly related to the stereo-chemistry of the hydroxyl group at C-3 in the flavan unit. However, when a galloyl group was linked to the hydroxyl group to form a gallate, this product exhibited markedly more activity than other analogs. These results suggest that the antihepatotoxic activity of phenolic flavan-3-ols and their derivatives seem to be related to the galloylation at the C-3 hydroxyl group in the flavan skeleton rather than the structure of another moiety or the degree of condensation.

Effects of heat therapy on renal hemodynamics, compensatory hypertrophy and glomerulonephritis in rats.

To investigate whether back heating (BH) affects renal function, we applied an electric heating pad on the back skin overlying the kidneys in rats. In the acute experiment on saline-expanded, normal conscious rats, BH up to 42 +/- 1 degrees C for 15 min induced diuresis, natriuresis, kalliuresis, increased urinary kallikrein excretion as well as an increase in renal blood and plasma flow and glomerular filtration rate (GFR). When the rats were uninephrectomized and subjected to either BH twice a day for 1 week or a sham procedure, the study group showed a higher GFR per gram kidney weight and a higher increase in kidney/body weight ratio. When the rats were injected daily with cationic bovine serum albumin for 30 days and then subjected to either BH twice a day for 26 days or a sham procedure, the BH group showed a higher degree of proteinuria during the recovery period and a higher kidney/body weight ratio on the 27th day. Acute BH for 15 min in anesthetized rats increased body temperature, perirenal temperature, heart rate, stroke volume, cardiac output and cardiac index. Taken together, these results suggest that BH may affect the kidneys in normal and pathologic states. These effects may be at least partly related to a change in systemic hemodynamics.

Effects of palmatine on motor activity and the concentration of central monoamines and its metabolites in rats.

We used behavioral and biochemical methods to investigate the sedative effect of palmatine on locomotor activity and the concentration of monoamine in rats. It was found that palmatine enhanced the hypomotility induced by alpha-methyl-p-tyrosine, reserpine and 5-hydroxytryptophan, but reduced the hypermotility produced by L-dopa plus benserazide and p-chlorophenylalanine. Furthermore, palmatine significantly decreased the concentration of dopamine and homovanillic acid in the cortex and the concentration of serotonin in the brain stem, and it increased the concentration of 5-HT in the cortex and 5-hydroxyindole acetic acid in the brain stem. These results suggest that the sedative mechanism of palmatine may be related to the decrease in the concentration of catecholamine in the cortex and serotonin in brain stem and the increase in the concentration of 5-HT in the cortex.

Characterization of agonist-induced internalization and down-regulation of beta-adrenergic receptors in isolated rat lung and heart preparations.

Using isolated lung and heart preparations from male Sprague-Dawley rats, this study characterizes whether the beta-adrenergic receptor (BAR) agonist isoproterenol (ISO) can induce internalization and down-regulation of BAR in these tissues. The results showed that ISO-induced BAR internalization was time- and concentration-dependent in the lung preparations. When the lung and heart preparations were compared, however, the same ISO (10 microM, 30 min) treatment only induced BAR internalization in the lung but not in the heart preparations. Pretreatment of hypertonic sucrose medium (0.45-0.6 M) for 30 min inhibited about 50% of ISO-induced BAR internalization in the lung preparations. Treatment of either ISO (10 microM) or terbutaline (10 microM) for 3 hr or 12 hr did not significantly decrease the number of BAR in both tissue preparations. However, ISO (10 microM, 12 hr) increased the KD value of the radioligand [125I]-iodocyanopindolol for BAR in the heart homogenates. This ISO-induced KD change was not observed in the presence of cycloheximide (20 micrograms/ml), a protein synthesis inhibitor. In the presence of cycloheximide (20 micrograms/ml), ISO also did not decrease the BAR number in either tissue preparation.

Mass isolation of highly purified canine islets using an automated method and Histopaque Ficoll gradients.

Eighteen pancreata from adult mongrel dogs were used for the study of islet isolation. The pancreas was distended with collagenase in Hanks' solution. The automated screen method and Histopaque Ficoll gradients were used to isolate and purify the canine islets. In vitro, the viability of isolated islets was assessed by both histology and perifusion studies. In vivo, the islet function was evaluated by using a nude mice xenograft model. Fair to good isolation and purification was found in 12 experiments. Before and after purification, the isolated islet count was 4767.1 +/- 560.1 and 3637.7 +/- 333.4 islet equivalence (I.E.)/gm pancreatic tissue. The purity was above 90%. Aldehyde Fuchsin stain disclosed islets with copious beta granules. The stimulation index of islets responding to 16.7 mM glucose plus 1 mM 3-isobutyl-1-methylxanthine (IBMX) versus 1.67 mM glucose was 12.93 +/- 4.75. Normoglycemia was restored and maintained for up to 2 weeks in 7 of 10 and up to 3 weeks in 5 of 10 diabetic nude mice transplanted with canine islets. In conclusion, the automated screen method and Histopaque Ficoll gradients afford a good yield of highly purified canine islets, and functional viability was verified both in vitro and in vivo. This will be an ideal model for isolation of human islets.

Synthetic and cytotoxic studies of alpha-methylene-gamma-butyrolactone bearing pyrimidines.

A total of ten pyrimidine alpha-methylene-gamma-butyrolactones were synthesized as potential antitumor agents on the basis of their possible action as Michael acceptors for DNA/RNA and cellular enzymes. The synthesis of these heterocycles involved a convenient Reformatsky-type reaction of pyrimidinyl ketones with ethyl alpha-(bromomethyl)acrylate. The preliminary in vitro cytotoxic assay indicated that these synthetic compounds were essentially active against the growth of KB, Hep-2, HeLa and Colo-205 cells. Among them, 5'-biphenyl-5'-(uracil-1-ylmethyl)-2'-oxo-3'-methylenetetrahydr ofuran (5f) demonstrated to be the most potent antileukemic agent.

[Factors related to the choice of different hospitals providing Chinese traditional medicine].

A structured questionnaire was answered by patients who used the Outpatient Departments of two types of Chinese traditional medicine clinics: clinics associated with western-style teaching hospitals, and regular Chinese medicine clinics which were area-matched with, but not located in the teaching hospitals. The study applied the Andersen behavior model and discussed the factors related to the choice of one type or the other clinic by one-method-treatment (i.e. traditional only, not mixed with western, medicine) patients. The total number of valid questionnaires was 362. Under the analysis of logistic regression, the variables which were significantly related to the choice of the types of the hospitals were age, insurance status, first visit or follow-up, general health condition, severity of illness, musculoskeletal and neurological disorders. The older the patients, the more likely they were to visit teaching hospitals. First-visit or uninsured patients also favored teaching hospitals. Patients with perceived stable conditions or more serious illness were likely to use teaching hospitals. Patients with musculoskeletal or neurological disorders favored regular clinics.

A new method of tissue processing that causes no shrinkage or distortion.

BACKGROUND: The routine method of tissue processing or dehydration usually causes shrinkage and/or distortion. More noticeable changes are seen in large specimens rich on hydrated tissues, such as embryos. Most likely this is due to the step-wise concentration change of the solvents. EXPERIMENTAL DESIGN: A new method of gradual solvent or medium exchange for tissue processing was developed. By using a peristaltic pump, pure solvent or embedding medium was added slowly to the processing chamber and the overflowing processed solution was slowly drained off. A computer spreadsheet program was used to calculate the concentration changes. After adding four to five times the processing volume of the solvent, the concentration of the adding solvent in the processing chamber could reach 98.15 to 99.32%. After reaching a desired endpoint, pure solvent could be used to replace the processing solution. RESULTS: We compared microencapsulated pancreatic islets and liver tissue processed traditionally and with the new method. By using this design, a smooth linear and gradual change of the concentration of the processing solutions could be obtained. No shrinkage or distortion of tissues due to the dehydration artifact was noticed, and the tissue seemed to be ideally suited for accurate quantitative histologic measurements. CONCLUSIONS: A very simple but efficient method for gradual solvent and medium exchange had been designed. This could efficiently prevent the shrinkage and distortion commonly produced by traditional tissue processing methods and prepare the tissue for quantitative histologic and pathologic studies.
