Mössbauer spectra of photosystem-I-reaction centres from the blue-green alga Chlorogloea fritschii.

Substantial amounts of iron have been shown by Mössbauer spectroscopy to be present in Photosystem-I preparations from the blue-green alga Chlorogloeta fritschii. Changes in the spectra on chemical reduction provide evidence that some of this iron is very similar to that found in the 4Fe-4S centres of ferredoxins. Such reduced samples also show e.p.r. signals consistent with maximum reduction of iron-sulphur centres A and B of Photosystem I. An unchanged component in the spectra indicates, assuming all centres A and B are reduced, the presence of another iron-containing species.

Sézary syndrome. Tartrate-resistant acid phosphatase in the neoplastic cells.

Tartrate-resistant acid phosphatase has been known to be of diagnostic value in hairy cell leukemia. However, occasionally neoplastic cells of other varieties of lymphoproliferative disorders may contain tartrate-resistant acid phosphatase. The authors have studied four patients with Sézary syndrome who had typical cutaneous lesions with extensive lymphoid infiltrates and circulating atypical E-rosetting lymphoid cells. The abnormal Sézary cells accounted for 23-69% of the peripheral mononuclear cells and often showed convoluted or folded nuclei. These cells in all four patients were strongly positive for acid phosphatase resistant to tartaric acid inhibition. Enzymatic cytochemical studies for acid phosphatase with and without tartrate may be helpful in the differential diagnosis of cutaneous T-cell lymphomas from variants of chronic dermatitis.

Reliability of theophylline clearance in determining chronic oral dosage regimens.

Predicted and approximated mean serum theophylline concentrations were compared to assess the feasibility of using theophylline clearance, calculated from the intravenous steady-state serum theophylline concentration, to determine appropriate oral dosage requirements. Ten patients with reversible airway disease (age 7 to 64 years) received theophylline (as Aminophylline USP) by constant i.v. infusion. Theophylline clearance was calculated for each patient from steady-state serum theophylline concentrations determined from blood samples obtained after a least 48 hours of constant theophylline infusion. Aminophylline USP tablets were then adminstered orally. Blood levels measured at peak and trough times following oral theophylline administration were used to calculate an approximate mean serum theophylline concentration. The relationship between predicted and approximated mean serum theophylline concentrations was calculated and analyzed by orthogonal least square regression line. Serum theophylline concentrations ranged from 5.1 microgram/ml to 20.1 microgram/ml, and clearances ranged from 0.03 liter/kg/hr to 0.09 liter/kg/hr. During oral therapy, the maximum concentration ranged from 10.4 microgram/ml to 23.8 microgram/ml, while the minimum ranged from 6.5 microgram/ml to 15.2 microgram/ml. The difference between predicted and approximated mean serum concentrations was within +/-1.6 micrograms/ml in all cases. The study indicates that the theophylline clearance calculated from i.v. infusion data can be used to determine appropriate oral dosage requirements.

Interpretation of the Mössbauer spectra of the four-iron ferredoxin from Bacillus stearothermophilus.

The Mössbauer spectra of both oxidized and reduced ferredoxin from Bacillus stearothermophilus have been analysed using computer fits to theoretical spectra obtained from a spin Hamiltonian. A consistent set of parameters was obtained from fits to spectra obtained over a wide range of temperature and magnetic field. These results are interpreted in terms of a model for the active centre which is consistent with its electronic and magnetic properties in both redox states. In the model for the oxidized centre all four iron atoms have essentially the same valence, intermediate between ferric and ferrous, with one pair spin-up and the other pair spin-down. On reduction the extra electron goes predominantly to one pair of iron atoms which become ferrous with the other pair remaining substantially unchanged. Using this model it is possible to obtain relationships between the spin Hamiltonian parameters for individual iron atoms and those for the coupled centre. This can give further insight into the relation between the observed electron paramagnetic resonance and Mössbauer spectra.

Be, Li, and Na redistribution near a porcelain/Ni alloy interface shown by ion microprobe mass analysis.

During porcelainization the non-preoxidized alloy develops a Be-rich reaction zone in the interface which separates a Be-depleted alloy from a Li and (to a lesser extent Na) depleted region of the ceramic. Thus, Be diffuses to the interface through the alloy Be degrees leads to Be++ + 2 e- occurs at the interface; some of the electrons are electronically conducted to the porcelain-air interface where they are consumed as 2 e- + 1/2 O degrees 2 leads to O=, and the resulting negatively charged porcelain surface attracts Li+ (and Na+).

Aluminum hydroxide: evaluation of two dosage forms and two dosing schedules in reducing intestinal phosphate absorption.

A crossover, randomized, eight-week trial using eight adult volunteers was undertaken to (1) determine the efficacy of aluminum hydroxide in decreasing gastrointestinal phosphate absorption, (2) compare the effectiveness of "with meal" and "between meal" dosing schedules, and (3) compare the effectiveness of the capsule and liquid dosage forms. Four treatments, each with a daily dose of approximately 6 g, were taken on a three-times-a-day regimen. The effectiveness of the treatment regimens was measured by using two variables: the decrease in total urinary phosphorus excretion, and the increase in the percent tubular reabsorption of phosphorus (TRP). Aluminum hydroxide treatment produced a significant change in urinary phosphorus excretion (p less than 0.0005) and in percent TRP (p less than 0.0005). For the study population, total urinary phosphorus excretion was the determining factor in changing the percent TRP. The increase in TRP and the decrease in total urinary phosphorus excretion was of the same degree regardless of dosage form or dosage schedule. Aluminum hydroxide was effective in decreasing phosphorus absorption as measured in this study. It appears that aluminum hydroxide capsules could be used with equal efficacy as the liquid dosage form in controlling hyperphosphatemia in patients with chronic renal failure who will not comply with a treatment program using the liquid.

Effect of theophylline on allergen skin tests.

The effect of theophylline on allergen-induced wheal and flare responses of 15 adults with atopy was studied in a double-blind, crossover study. After intradermal skin tests to determine baseline wheal and flare responses, subjects were assigned randomly to consecutive seven-day courses of theophylline 16 mg/kg/day (as Aminophylline USP) then of placebo, or the reverse. Subjects were tested with mixed weeds and mixed grasses allergens and with histamine diphosphate, codeine phosphate and a buffered saline control. Skin tests were repeated two hours after the last dose of theophylline or placebo in each treatment course, and serum theophylline concentrations were measured. The surface areas of the wheal and flare responses were measured by planimetry. No statistically significant difference (p greater than 0.05) was found between theophylline and placebo in the suppression of the allergen-induced wheal and flare response. Serum concentrations of theophylline were within the accepted therapeutic range. In therapeutic steady-state serum concentrations, theophylline does not significantly suppress allergen-induced wheal and flare response of atopic patients, and withdrawal of the drug before skin testing is unnecessary.

Identification of a non-magnetic iron centre and an iron-storage or -transport material in blue--green algal membranes by Mössbauer spectroscopy.

At least two species of iron were identified in crude membrane preparations of the blue-green algae Chlorogloea fritschii and Anacystis nidulans by Mössbauer spectroscopy. One species was non-magnetic with chemical shift more characteristic of a ferredoxin-like centre than of any known low-spin Fe(II) biological compounds. Studies on cells grown under depleted-iron conditions show the other iron species to be part of the system for iron metabolic transport and/or storage.

Pseudomonas aeruginosa bacteremia in a dialysis unit. 1. Recognition of cases, epidemiologic studies and attempts at control.

Infections commonly occur in patients undergoing dialysis and have been related to diminished host resistance of uremic patients, the arteriovenous fistulas and bacteriologic contamination of dialysis fluids. The occurrence of four cases of bacteremia due to Pseudomonas, three of which were type 7, and the presence of this serotype in the dialysis fluids suggested an important association between infection and growth of bacteria in the fluids. Attempts to reduce levels of bacteria in the dialysis fluid were unsuccessful using dialysate free of glucose in clinical trial, despite in vitro studies demonstrating poor growth of Pseudomonas in this medium. A filter placed with the recirculating system was only partially successful. The second paper of this series traces the portal of entry of bacteria from dialysate to the blood through reutilized coils.

Combined growth-inhibitory responses and ultrastructural alterations produced by 1,3-bis(2-chloroethyl)-1-nitrosourea and dexamethasone in rat glioma cell cultures.

The effect of 0.0001 to 10 muM 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) and 1 muM dexamethasone on cell proliferation was studied by measuring cell densities in control and drug-treated rat glioma (strain C6) monolayer cultures. When C6 cultures were exposed to 0.01 to 10 muM BCNU, the growth rates decreased for 2 days as control cell populations continued to proliferate at log phase rates. These growth-inhibitory responses were dose dependent and ranged from 20 to 80%, relative to control growth. Subsequently, the growth rates increased and the inhibitory responses ranged from 0 to 12% 4 days later. Cell densities in C6 cultures exposed to 1 muM dexamethasone for 1 day did not differ significantly from controls. Then cell proliferation ceased and the inhibitory response remained at 50% relative to controls in stationary phase. When 0.03 muM BCNU and 1 muM dexamethasone were supplied simultaneously to C6 cultures, a 35% inhibitory response occurred after 1 day. This response did not differ significantly from that observed with 0.03 muM BCNU alone. After 4 days, the inhibitory response did not decrease in cultures containing both drugs, but did decrease to 13% in the 0.03 muM BCNU-treated cultures. In 1 muM BCNU-treated cultures, the response was 66% after 1 day, which decreased to 21% 5 days later. When 1 muM BCNU was supplied to C6 cultures that were pretreated for 1 day with 1 muM dexamethasone, the response was 91% the following day, and this decreased to only 54% 5 days later. Dose-response curves showed that the inhibitory responses after 1 day in these pretreated cultures exposed to 0.001 to 10 muM BCNU increased up to 22% relative to the responses produced by either drug alone. After 5 days, the responses in the pretreated cultures exposed to 0.001 to 1 muM BCNU was 50%, which was similar to the response produced by 1 muM dexamethasone alone. Ultrastructural studies revealed that control and 1 muM BCNU-treated C6 cells contained 18 mitochondria, but the treated cells were 10% smaller after 1 day. Cells exposed to 1 muM dexamethasone for 1 day conount of granular endoplasmic reticulum increased greater than 80% in cells treated with BCNU for 1 day or dexamethasone for 2 days. C6 cells pretreated with dexamethasone and exposed to BCNU for an additional day (a) contained 23 mitochondria, (b) did not decrease in size, and (c) exhibited a greater than 250% increase in the amount of granular endoplasmic reticulum. These results demonstrate that combined growth-inhibitory responses and ultrastructural alterations occur when C6 cells are treated sequentially with 1 muM dexamethasone and BCNU.

The mechanism of action of dicyclomine hydrochloride on rabbit detrusor muscle and vas deferens.

Dicyclomine inhibited responses of rabbit detrusor muscle to transmural stimulation more than atropine or lidocaine, and also inhibited responses to acetylcholine and to KCl. Responses of rabbit vas deferens to noradrenaline and transmural stimulation were inhibited by dicyclomine. Dicyclomine was as potent a local anesthetic as lidocaine, as determined by the guinea-pig wheal test. 45Ca++ exchange in the rabbit detrusor, measured by the lanthanum methods, was not altered by atropine, lidocaine or dicyclomine. It is concluded that dicyclomine not only acts as an anticholinergic agent but also impairs excitation-contraction coupling and release of transmitter from autonomic nerves.

Organizing patient data for use in clinical pharmacy practice and education.

A simple, low-cost manual data retrieval system, which provides selective access to all patient cases monitored by pharmacists, is described. The system uses notched cards which are coded by clinical descriptors. The codes are based on a modification of the International Calssification of Diseases, Adapted, disease-diagnosis coding system. This multiple entry system is flexible and can be easily converted to a computer-based system.

Ovarian surgery in pregnancy.

A series of 57 cases of ovarian disease during pregnancy is presented; 10 of the patients were referred to the Mayo Clinic for treatment. During the 17 year period under investigation (1955 through 1971), there were 25,189 deliveries in Rochester, Minnesota. Thus, the corrected incidence of operable ovarian disease in pregnancy was one case in every 536 deliveries. Simple ovarian cysts were the most common lesions found (38 of 57 cases). The over-all abortion rate was about 25 per cent (14 of 57 patients); six of the patients were threatening to abort prior to surgery. Therefore, the incidence of spontaneous abortion after surgery was 16 per cent (eight of 51 patients). Progestational agents given before and after operation did not affect the abortion rate.
