The effects of Candida albicans on turpentine-induced hyperplasia of hamster cheek pouch epithelium.

This investigation aimed to determine the effect of Candida albicans on hyperplastic epithelium. Epithelial hyperplasia was induced in hamster cheek pouches by painting the mucosa with 50% (v/v) turpentine and liquid paraffin (TLP50) for 10 weeks. The pouches were then tied off with sutures after inoculation with C. albicans. Many of the pouches lost their sutures and were histologically normal after periods up to 8 weeks. Those which remained tied off showed epithelial dysplasia resembling changes seen in human candida leukoplakia. It is suggested from these preliminary experiments that C. albicans may be capable, in certain conditions, of causing further changes to hyperplastic epithelium resembling those seen in Candida leukoplakia.

Candida glabrata (syn. Torulopsis glabrata) associated with a chronic hyperplastic lesion of the palate.

A case initially presenting as chronic hyperplastic candidosis is described. After treatment with nystatin on three separate occasions over a seven-year period, a lesion similar to that diagnosed originally occurred in the same site. Further examination of this later occurrence showed it to be associated with the imperfect fungus Candida glabrata which had a high minimum inhibitory concentration for nystatin (greater than 30 micrograms/ml). The lesion eventually resolved following treatment with miconazole gel and surgical excision of the hyperplastic tissue. The patient has remained symptomless for a two-year follow-up period.

Sources of error in the registration of oral cancer.

The names of 102 patients with histologically confirmed squamous cell carcinoma of the oral cavity were checked against Cancer Registry data for the Trent Region. The study was carried out to determine the accuracy of site recording and the failure rate of registration. Although 94 of the patients were registered, 38 per cent of these were incorrectly coded with respect to site. The way in which such errors may occur is discussed and the procedure for registration of oral cancer is explained.

An investigation into the origin and nature of 'amyloid' in a calcifying epithelial odontogenic tumour.

Fresh and fixed tissue from a resection specimen of a calcifying epithelial odontogenic tumour (CEOT) was prepared for histological, histochemical, immunological and biochemical investigation in order to study the nature of the amyloid-like material. The fixed tissue gave positive reactions with Congo-red, Thioflavin T and the dimethylamino benzene (DMAB)-method for tryptophan. The diazotization-coupling (DC) method for tyrosine was negative. The major protein purified from the unfixed tissue by sequential gel filtration had a molecular weight of 9,800. The amino acid analysis of this protein had similarities with tuft enamel protein, immune amyloid and the variable light chain component (VK). From the data obtained in this study, it is not possible to determine the precise nature of the amyloid-like material in this CEOT. However, the results do support the concept that 'amyloid' should be considered as a term describing a broad group of related proteins.

Stereological analysis of histological parameters in experimental premalignant hamster cheek pouch epithelium.

Hamster cheek pouch mucosa was treated with the carcinogen DMBA in order to provide samples of epithelium in the various pathological stages of carcinogenesis. Stereological techniques were applied to histological sections of normal and carcinogen-treated epithelium in order to quantify cytological and tissue parameters that may be related to the features of epithelial atypia. The results were compared with values previously obtained for benign epithelial hyperplasia induced by a solution of oil of turpentine. Of the parameters investigated, the nuclear-cytoplasmic ratio, nuclear numerical density and volume-to-interface area ratios were best able to distinguish between benign and premalignant epithelium. The ability to ascribe actual numerical values to histological features which are usually only subjectively evaluated may be a useful diagnostic aid.

Stereological analysis of hamster cheek pouch epithelium after one application of DMBA or turpentine.

Both cheek pouches of two groups of five male hamsters were treated with one application of either 50% turpentine in liquid paraffin (TLP50) or a 0.5% solution of dimethylbenz(alpha)anthracene in liquid paraffin (DMBA); animals were killed 48 h later. Five untreated animals served as controls. Mucosal specimens were removed from each pouch, fixed in normal-acetic-alcohol and processed for light microscopy. Sections were cut at 4 micrometer and stained with haematoxylin and eosin. A systematic random sampling method was used to obtain sections for sterological analysis. Grids for point and intersect counting were superimposed on the projection screen of an automatic sampling microscope. Results showed that the epithelial nuclear-cytoplasmic ratio, nuclear density, nuclear diameter and surface area interface ratios were similar for the experimental groups and, with the exception of the last parameter, differed significantly from controls. The volume proportion of TLP50-treated epithelium was significantly larger than that of DMBA-treated epithelium. Derived values for the average nuclear volume in both experimental groups were almost twice that of controls; the cytoplasmic volumes were more than doubled. At 48 h the epithelial response to a single application of TLP50 or DMBA was essentially similar except that TLP50 probably induced greater hyperplasia.

Quantitative analysis of histological parameters in giant cell lesions of the jaws and long bones.

Stereological techniques have been used to compare histological parameters of the giant cell component in 10 giant cell tumours of long bones, 10 central giant cell granulomas of the jaws and 10 peripheral giant cell granulomas of the jaws. The giant cell parameters investigated were: mean profile diameter, mean diameter, mean profile axial ratio, volume-to-surface ratios, nuclear numerical density, profile area and cell volume. Serial sections were cut at 4 micron from routinely fixed, paraffin-embedded specimens of each case and two sections, separated by 100 micron, were stained with haematoxylin and eosin. Random fields were photographed until at least 25 giant cell profiles had been recorded from each of the two sections of each lesion. The processed film was projected for point and intersect counting, the entire procedure ensuring double-blind assessment of the cases under investigation. In general there was no significant difference for any of the parameters between the giant cells of central and peripheral jaw lesions. There was a significant difference, however, between the giant cells of central jaw lesions and long bone tumours in respect of both nuclear numerical density and mean absolute cell volume.

The effect of turpentine on hamster cheek pouch mucosa: a model of epithelial hyperplasia and hyperkeratosis.

Hamster cheek pouches were painted thrice weekly with 50% v/v turpentine in liquid paraffin over periods ranging from 1 day to 16 weeks. Pouch mucosa was examined histologically for changes and measurements of epithelial thickness were made with a calibrated eyepiece micrometer. An increase in epithelial thickness was first observed 48 hrs. after a single painting and was accompanied by inflammatory changes in both epithelium and connective tissue. Maximal epithelial thickening occurred after 9 weeks of thrice weekly painting. Cheek pouches of animals left without further treatment for up to 1 year following 9 weeks painting were indistinguishable from controls. The epithelial response to treatment with turpentine is that of a reversible benign epithelial hyperplasia with hyperkeratosis.

The calcifying epithelial odontogenic tumour-- report of four cases; two with long-term follow-up.

Four cases of calcifying epithelial odontogenic tumour are reported, all with pain as a presenting symptom. Two of the cases have long-term follow-up, one with a recurrence after 16 years and the other with no recurrence after 24 years. Only 17 per cent of cases reported in the literature have follow-up of 10 years or more, and yet the possibility of recurrence after this length of time must be considered. Recommendations for treatment are made based on the biological behaviour and recurrence rate following various types of treatment reported in the literature. It is recommended that patients in whom the diagnosis of calcifying epithelial odontogenic tumour has been made should be reviewed continuously.

The calcifying epithelial odontogenic tumor. A review and analysis of 113 cases.

A review of the world literature has revealed 113 cases of the calcifying epithelial odontogenic tumor. The clinical, radiographic, and histopathologic features are analyzed. Various treatments that have been carried out are related to the frequency of recurrence. The theories of histogenesis are discussed and an attempt is made to explain the nature and origin of the amyloid-like substance.

The treatment of Sjögren's disease in NZB/NZW F1 hybrid mice with azathioprine: a two-stage study.

NZB/NZW mice exhibit changes in their salivary and lacrimal glands similar to those occurring in Sjögren's syndrome (SS) in humans. The aim of this study was to determine the efficacy of azathioprine in the treatment of SS in NZB/NZW mice. The experimental animals comprised three groups; the first received the drug daily from 14 weeks of age, the second started therapy at 26 weeks of age and the third group remained untreated. The animals, in groups of five, were killed at four-weekly intervals, up to 42 weeks, using ether. The submandibular and lacrimal glands were assessed for the extent of lymphocytic infiltration using a modified focus scoring system, with light microscopy. During the progression of the disease, azathioprine produced a reduction in the number of lymphocytic foci. This reduction occurred when azathioprine was used both early and late in the condition.

Assessment of sulphasalazine as a treatment modality in Sjögren's disease in NZB/NZW F1 hybrid mice.

OBJECTIVE: Sulphasalazine is a recognised second-line agent in the treatment of rheumatoid arthritis. The aim of this study was to determine if it might prove useful in the treatment of Sjögren's syndrome. METHODS: The trial was performed in NZB/NZW F1 hybrid mice and efficacy was assessed on the basis of a reduction in the lymphocytic infiltration of the submandibular salivary and lacrimal glands. The animals were divided into three groups; the control group remained untreated, the second group received the drug from 14-42 weeks and the third group received sulphasalazine from 26-42 weeks. At four-weekly intervals throughout the study, five animals in each group were killed using ether and the glands assessed for lymphocytic infiltration using a modified focus scoring technique with light microscopy. RESULTS: No significant reduction in the lymphocytic infiltration occurred consistently throughout the trials with sulphasalazine. CONCLUSION: Sulphasalazine did not significantly alter the development of Sjögren's disease in NZB/NZW F1 mice.
