A large retinol-binding protein, interphotoreceptor retinol-binding protein, is found only in the interphotoreceptor matrix of the eye, and may function in vitamin A transport for the visual cycle. Interphotoreceptor retinol-binding protein is the major glycoprotein of this matrix, and can be isolated rapidly by affinity-adsorption onto concanavalin A-Sepharose. The yield is approx. 0.25 mg per bovine eye. Its apparent Mr is 250000 by gel-filtration chromatography, and 225000 by native polyacrylamide-gradient gel electrophoresis; this protein band displays endogenous retinol fluorescence on such gels. As measured by SDS-polyacrylamide gel electrophoresis, the apparent Mr is 140000. In the interphotoreceptor matrix most vitamin A-binding sites on this retinol-binding protein are unoccupied; however, addition of exogenous all-trans-retinol can saturate these sites. The apparent dissociation constant for retinol is 10(-6) M, as measured by fluorimetric titration.

Newly weaned guinea pigs weighing approximately 300 g were fed normal laboratory diets with drinking water containing 5.5% ethanol as the sole source of liquid for periods of 8-11 weeks. Growth was continuous with this diet. After this period, hearts were removed from anesthetized animals and perfused for 3 hr in a perfusion system in which pressure may be induced in the right ventricle in the face of constant coronary flow. Protein synthesis, assayed from the incorporation of labeled lysine and phenylalanine, was compared to that in hearts from identically treated weight-matched control animals who had been drinking water without ethanol. Protein synthesis in hearts from ethanol-drinking animals was decreased in the right ventricles exposed to normal pulmonary pressure, but was unchanged in the contracting but not working left ventricles. The data suggest that prolonged exposure even to low levels of ethanol in the growing animal may interfere with the cardiac protein synthetic response to the normal work stress.
The relative molar synthesis of cardiac contractile proteins has been measured in the perfused heart under control haemodynamic conditions. This synthesis, of myosin heavy chains, individual light chains (1 and 2), actin and tropomyosin, was determined from isolated guinea-pig hearts perfused for 3h simultaneously with constant specific radioactivities and concentrations of [3H]lysine and [3H]phenylalanine. The data strongly suggest that all of the proteins studied were synthesized from the same precursor pools of lysine and phenylalanine, since the ratio of the specific activities of the two labels was the same in all of the proteins. Measurement of molar synthesis of each contractile protein was the same with either labelled amino acid. Under control haemodynamic-perfusion conditions, the relative molar synthesis of the contractile proteins was actin greater than heavy chains greater than light chain 2 greater than light chain 1 greater than tropomyosin.

The human vasoconstrictor bioassay was used to assess the potency of open applications of Synemol cream (0.025%), Diprosone cream (0.05%), Aristocort-A cream (0.5%), and Valisone cream (0.1%). Intensity of vasoconstriction was determined eight, twenty-four, and thirty-two hours after application. Results obtained from the average intensity scores of the three determinations indicated that Synemol cream (0.025%) is actually a more active compound than are Diprosone cream (0.05%), Aristocort-A cream (0.5%), and Valisone cream (0.1%), and that its activity is longer acting.

This paper described the detailed examination of 121 patients who had suffered from traumatic brain damage. An important feature of the study was that all patients were examined by methods devised by the therapists in the department concerned, and the results subsequently integrated. The patients are described and their ability to return to independent existence is assessed in relation to the duration of unconsciousness and of post-traumatic amnesia (PTA). One method in particular, devised in the physiotherapy department, seems to suggest scope for developing recovery curves that might prove valuable indicators of the effectiveness of long-term therapy.

This report describes a double-blind between-group comparison of Tofranil and placebo in fifty-nine patients admitted to the Joint Service Medical Rehabilitation Unit suffering from low back pain. Patients were given 25 mg of either Tofranil or placebo three times a day for four weeks. Fifteen patients were not included in the analysis, nine dropping out for non-trial related reasons, two dropping out complaining of lost libido and four not taking the tablets, or taking additional drugs. One of the patients complaining of lost libido was taking Tofranil and the other was taking placebo. Over the whole sample there was no significant benefit for Tofranil over placebo as regards physical measurements. Both Tofranil and placebo groups showed a significant improvement during the trial on straight leg raise and backward flexion. For lateral flexion the Tofranil group was significantly worse than the placebo group on entering the trial, and during the trial the Tofranil group improved to match the placebo group. The clinician's pain and stiffness assessments and the patients' pain and stiffness assessments show a significant improvement for both the Tofranil and placebo groups during the trial. However, no difference is shown between Tofranil and placebo. The psychological tests show no difference between Tofranil and placebo and only a marinal improvenmet over initial condition. Further analysis according to initial diagnosis showed nothing conclusive. Numerically, the use of Tofranil produced a marked improvement in pain and stiffness in patients with 'disc lesion only' diagnoses, whereas placebo did not produce an improvement. However, this observation was far from reaching statistical significance. Side-effects were not severe for either drug. Additional medication was rarely used.
