Evidence for a local action of growth hormone in embryonic tooth development in the rat.

Studies in non-dental embryonic tissues have suggested that an interaction between growth hormone and its receptor may play a role in growth and development before the foetal pituitary gland is competent. This study reports the distribution of growth hormone, its receptor and binding protein in developing rat tooth germs from embryonic day 17 to 21 and postnatal day 0 using antibodies specific for each of these proteins. Four foetal rats were processed at each time point (E17, E18, E20/21 and postnatal day 0). Following routine fixation and paraffin embedding, sections were treated with antisera to rat growth hormone, rat growth hormone binding protein and growth hormone receptor. Localization of antibody/antigen complexes was subsequently visualized by addition of biotinylated IgG and reaction with streptavidin peroxidase and diaminobenzidine. Assessment of the level of staining was qualitative and based on a subjective rankings ranging from equivocal to very strong staining. Overall, growth hormone and its binding protein were located both in the cellular elements and throughout the extracellular matrix, whereas the growth hormone receptor showed an exclusively intra-cellular location. All three proteins were detectable in cells of the dental epithelium and mesenchyme at the primordial bud stage (E17) which occurs prior to expression of pituitary growth hormone. At the cap stage of odontogenesis (E18-19), numerous cells in both the dental epithelium and mesenchyme were intensely immunoreactive for growth hormone, its binding protein and receptor. In the succeeding early bell stage (E20-21), most of the mesenchymal cells in the dental pulp were mildly positive for these proteins, while the dental epithelium and adjacent mesenchyme were more immunoreactive. At the late bell stage (postnatal day 0), all three proteins were localized in dental epithelium, differentiating mesenchymal cells the cuspal surface facing the epithelial-mesenchymal interface, preodontoblasts, and odontoblasts forming dentine. From these observations, immunoreactive growth hormone, its receptor and binding protein appear to be expressed in odontogenic cells undergoing histodifferentiation, morphodifferentiation and dentinogenesis in a cell-type and stage-specific pattern throughout embryonic tooth development. This suggests the possibility that growth hormone, or a growth hormone-like protein, plays a paracrine/autocrine role in tooth development in utero.

[Geographic distribution and exposure population of drinking water with high concentration of arsenic in China].

According to the data obtained from the "National Survey on Drinking Water Quality and Waterborne Diseases", the geographic distribution and exposure population of high arsenic drinking water were reported. From the data of more than 28,800 water samples, we found 9.02 million people drinking the water with As concentration of 0.030-0.049 mg/L, 3.34 million people having their water of 0.050-0.099 mg/L and 2.29 million people having water of > 0.1 mg/L. A total of 14.6 million people, about 1.5% of the surveyed population was exposed to As (> 0.030 mg/L) from drinking water. 80% of high-As-drinking water was groundwater. The situation of As in drinking water in provinces, autonomous regions and municipalities were listed. The locations of sampling site where water As exceeded the national standard for drinking water were illustrated.

[Pathogenesis and combined treatment of sterility in men with varicocele].

To investigate the pathogenesis of sterility in men with varicocele and summarize the effects of combined treatment, we anallyed ultrastructure of testicular specimens in 21 cases. With combined surgical and medicinal treatment, 16 men (76.2%) made their wives pregnant during a period of 2 years after the operation. Electron microscopy disclosed glycogen infiltration of sertoli cells, and decreased spermatogenesis especially involving the middle and late stages. Defective carbohydrate and energy metabolism in the testis is suggested as the key point in the pathogenesis of sterility, and a combined surgical and medicinal treatment is beneficial to the increase of pregnant rate.

[Extracorporeal shock wave lithotripsy for uric acid stones in upper urinary tract].

Since 1987, 121 patients with uric acid stones in upper urinary tract have been treated by an EDAP LT-01 ultrasound-guided piezoelectric lithotriptor. One hundred and twenty-one patients had 167 renal stones 0.5-3.5 cm in diameter (mean 0.86 cm), 9 had ureteral stones 0.6-1.3 cm (mean 0.84 cm). Auxiliary treatment were given to 2 patients with upper ureteral stones, the others were treated by ESWL in situ. A success rate of 80.99% (98 patients) was achieved in one session, 12.40% (15) in two sessions, 4.96% (6) in three sessions, and 1.65% (2) in four sessions. No serious complications were encounterd. Therefore, ESWL with the ultrasound-guided piezoelectric lithotriptor is considered to be a satisfactory procedure for the treatment of uric acid stones. The ultrasound localization of uric acid stones, the shocking methods during ESWL and the application of alkaline medicine after ESWL were discussed.

[Variation of female migraine after menopause and its prophylaxis and treatment].

320 cases of female migraine after menopause according to the diagnostic criteria were studied by using 1:1 matched analysis. It was found that the additional symptoms increased after menopause. Eighty cases available for follow-up were divided into two groups, on which the treatment tests were done. Each patient in group I took Nilestrioli 2 mg twice a month and Perphenazine 12 mg and Doxepin 75 mg per day, while each patient in group II took Tolfenamic acid 300 mg a day. Two months after treatment, the cure rates were 57.58% in group I and 27.78% in group II. Two monthes after the cessation of therapy, 2 cases had relapses in group I. These data indicate that the decrease of estrin disorder and the additional symptoms after monopause play important roles in the change of migraine physiology, and the corresponding treatment methods are rational.

Clinical observation of Chinese drugs in prevention of neonatal hyperbilirubinemia.

Observation of the preventive and curative effects of Herba Artemisiae Scopariae [symbol: see text], Fructus Gardeniae [symbol: see text], Radix Scutellariae [symbol: see text] and Radix et Rhizoma Rhei Preparata [symbol: see text] in 30 cases of neonatal hyperbilirubinemia showed that both the peak value of serum total bilirubin and the incidence of hyperbilirubinemia were obviously lower than that in the control group.

[FTRaman and FTIR spectroscopy in lens with senile cataract].

OBJECTIVE: In order to search for the main composition and conformational changes of lens proteins in senile cataract. METHODS: We firstly used Fourier transform Raman and infrared (FTRaman and FTIR) spectroscopy and semi-quantitative method in the integral calculus of related peaks to determine the conformational changes of lens protein during opacification of varying grade. RESULTS: The spectral features of lenses with grade III cataract were intermediate between human clear lenses (grade 0) and grade IV or grade V cataractous lenses. The spectral characteristics of lenses with grade IV were very similar to that with grade V lenses, and some peaks of them were not seen in human clear lenses. The two spectral analyses showed that some findings were identical and some findings were complementary. CONCLUSION: The development of lens opacification is correlated not only with the relative contents of alpha-, beta-, gamma-crystallines in lens proteins but also with the various damaged SH, trp, phe, -CH2 in lens protein of senile cataract.

[Research of daunomycin as an agent for preventing posterior capsule opacification].

OBJECTIVE: To investigate the inhibitory activity and the effective concentration of daunomycin against the proliferation of lens epithelial cells in vitro, and make progress in pharmacological prevention of posterior capsule opacification. METHODS: Cultured bovine, rabbit and human lens epithelial cells were exposed to 0.5, 2.5, 5.0, 7.5 and 10.0 micrograms/ml daunomycin solution for ten minutes. Growth of cells was observed, after 48 hours of exposure, absorbency of the cells was tested by using Giemsa staining and colorimetry, and the median lethal dose (LD50) was calculated respectively. RESULTS: Daunomycin significantly suppressed the proliferation of bovine, rabbit and human lens epithelial cells in vitro, showing dose-dependent manner. Daunomycin can effectively inhibit the proliferation of human lens epithelial cells at the concentration of 0.5 microgram/ml, almost reaching the greatest effect at the concentration of 7.5 micrograms/ml. The calculated LD50 for bovine, rabbit and human lens epithelial cells was 0.49, 4.30 and 4.06 micrograms/ml respectively. CONCLUSION: Short time exposure of daunomycin at low concentration can effectively inhibit the proliferation of lens epithelial cells in vitro, after further research in vivo, daunomycin may possibly become a valuable agent for the prevention of posterior capsule opacification.

[Histopathologic and immunochemical changes in Sjögren's syndrome].

To research into the causes of sjögren's syndrome, we investigated the lacrimal gland tissues of 6 cases of sjogren's syndriome by using electronmicroscopy and immunochemical methods. The results revealed that at the early stage the lacrimal gland cells showed degeneration, irregular arrangement and many intracellular vacuoles. At the middle stage, some lymphocytes infliltrated into the gland tissues. And at the advanced stage, lymphocytes and collagenous fibers were there in substitution of gland cells.

[Studies on the essential oil constituents of the stem bark of Magnolia obovata].

For the extensive studies of the chemical constituents of Magnollis obovata, the essential oil from Magnolia obovata was obtained by steam distillation. The chemical components of the oil were examined by means of capillary gas chromatography and gas chromatography-mass spectrometry. 29 of the 75 separated constituents were identified. The content of each component identified was determined by area normalization method, which showed the content of 9 compounds was higher than 1%.

[Minor Bupleurum decoction in the treatment of viral myocarditis models].

A myocarditis model of Balb/c mice established by means of coxsackie virus type B3 was treated with the Minor Bupleurum Decoction. The results indicate that the decoction can regulate T lymphocyte subsets, influence anti-cardiac antibody production, help the organism to get clear of viruses, and thus plays an important role in treating viral myocarditis.

[Supercritical-CO2 fluid extraction of the fatty oil in Terminalia chebula and GC-MS analysis].

The fatty oils in Terminulia chebula were extracted by supercritical-CO2 fluid extraetion, and their fatty acids and its relative contents were determined by GC-MS. 12 fatty acids were identified. Palmitic acid, linoleic acid and oleic acid of them are main constituents. Compared with petroleum ether extraction method, the extraction rate from SFE-CO2 is higher and extraction time shorter.

[Detection of flaviviruses by reverse-transcription polymerase chain reaction with universal primer set].

For the purpose of early and rapid diagnosis of flavivirus infection, the universal primer set was selected on the NS1 gene. Another five different internal primers were selected on the NS1 genes of DEN1, 2, 3, 4 and JEV. With the universal primer set designed, the NS1 fragment about the size of 413 bp and with the five internal primers, the NS1 fragments about the sizes of 262 bp (DEN1), 189 bp (DEN2), 392 bp (DEN3), 97 bp (DEN4) and 323 bp (JEV) were amplified respectively. The reverse transcription-polymerase chain reaction (RT-PCR) was used to amplify gene fragments of flaviviruses. cDNA of DEN1, 2, 3, 4 and JEV were successfully amplified with universal primer set and internal primers. Sera from 78 patients with dengue fever were assayed by nested PCR, DEN1 was detected from 18 of the 40 patients' sera and DEN2 was detected from 48 patients' sera. Sera from 42 patients with Japanese encephalitis were also assayed by nested PCR, JEV was detected from 35 of the 42 patients' sera. By nested PCR, we completed identification of flaviviruses within 2 days. The results showed that this method has the potential value in rapid clinical diagnosis of flavivirus infection.

[Analysis on current status of drinking water quality in rural areas of China].

An investigation on drinking water quality in rural areas of 180 counties in 26 provinces, municipalities and autonomous regions of China was carried out. The population surveyed was 89.39 million. 69.6% of which was supplied with ground water. Central water supply systems served 47.1% of population. Quality of drinking water was graded according to the "Guidelines for Implementation of the 'Sanitary Standard for Drinking Water' in Rural Areas". The rate of population supplied with unqualified drinking water was 42.7%. The bacteriological indices of drinking water exceeded the standard seriously. Organic pollution occurred extensively. Some regions supplied with water of high concentration of fluoride.

[Studies of magnetic circular dichroism and absorption spectra in alpha-NisO4 x 6H2O crystal].

Alpha-NiSO4 x 6H2O crystals were studied by magnetic circular dichroism (MCD) and absorption spectra measurements in the range lambda = 185-3300nm. The results show that there are three aborption bands, of which the peaks are related to the d-d transitions from the ground state to the excited states. The weak peaks of a progression of the three members were observed between the principal structures lambda = 654nm and lambda = 713nm due to spin-orbit coupling interacion. The weak peaks of a progression of four members were also observed around the new absorption peak at lambda = 585nm in MCD spectra.

[Strain effects in Zn1-xMn(x)Se superlattices of II -VI diluted magnetic semiconductors].

We report the strain effects of Zn1-xMn(x)Se superlattices of II -VI diluted magnetic semiconductors by the photoluminescence measurements with compostions x = 0.2, 0.3, 0.4 in the temperature range T = 11K to 300K. The results show that the exciton energy tends to a red shift with the increasing of x due to the strain effects. The distribution of strain in wells and barriers presents a considerable change when their thickness ratio is altered. Photoluminescence spectra peaks as a function of temperature are mainly dominated by the band gap of ZnSe. The temperature coefficient of exciton energy is obtained.

Characterization of radiation-induced apoptosis in rodent cell lines.

For REC:myc(ch1), Rat1 and Rat1:myc(b) cells, we determined the events in the development of radiation-induced apoptosis to be in the following order: cell division followed by chromatin condensation, membrane blebbing, loss of adhesion and the uptake of vital dye. Experimental data which were obtained using 4He ions of well-defined energies and which compared the dependence of apoptosis and clonogenic survival on 4He range strongly suggested that in our cells both apoptosis and loss of clonogenic survival resulted from radiation damage to the cell nucleus. Corroboratory evidence was that BrdU incorporation sensitized these cells to radiation-induced apoptosis. Comparing the dose response for apoptosis and the clonogenic survival curves for Rat1 and Rat1:myc(b) cells, we concluded that radiation-induced apoptosis contributed to the overall radiation-induced cell inactivation as assayed by clonogenic survival, and that a modified linear-quadratic model, proposed previously, modeled such a contribution effectively. In the same context, the selective increase in radiation-induced apoptosis during late S and G2 phases reduced the relative radioresistance observed for clonogenic survival during late S and G2 phases.
