[Effects of IL-1 and TNF-alpha on the proliferation and cytosolic Ca2+ of lens epithelial cell in vitro].

OBJECTIVE: To study the effects of interleukin 1 (IL-1) and tumor necrosis factor alpha (TNF-alpha) on the proliferation and cytosolic Ca(2+) of bovine lens epithelial cells (BLEC) in vitro. The effects of IL-1 and TNF-alpha on posterior capsular opacification (PCO) and the mechanism of effect on the proliferation were investigated. METHODS: The effects of IL-1 and TNF-alpha on the proliferation of BLEC were detected with MTT staining colorimetry, and cytosolic Ca(2+) was measured by fluorescence determination with Fura-2. RESULTS: IL-1 10(2) - 10(5) ng/ml, TNF-alpha 10(2) - 10(4) U/ml obviously could promote BLEC proliferation (P < 0.05 - 0.01), and increase cytosolic Ca(2+) (P < 0.05 - 0.01) at the same doses. CONCLUSIONS: IL-1, TNF-alpha contribute to the PCO. One of the mechanisms of IL-1 and TNF-alpha on proliferation is by cytosolic Ca(2+).

[A study on the prevention of selenite cataract with taurine].

OBJECTIVE: To evaluate the prevention of selenite cataract with taurine. METHODS: A prior injection of taurine was given intraperitoneally to Wistar rats 2 days before the selenite challenge and a following daily injection of taurine was needed until the experiment was finished. All of the rats were examined under a split-lamp microscope on selected days. RESULTS: 0.5% taurine showed no effect on lenses, 1%, 2% and 4% taurine could delay the development of nuclear opacity, and 2% and 4% taurine could also delay the development of cortical opacity. CONCLUSIONS: Taurine can prevent the development of selenite cataract. Its effect is dose-dependent.

[Sulfation of polysaccharides isolated from Indocalamus tesselatus and their anticytopathic effect on human immunodeficiency virus type I].

For preparation of sulfated polysaccharides isolated from bamboo leaves (Indocalamus tesselatus) with chlorosulfonic acid-pyridine, the experimental conditions as the molar ratio of reactants and temperature and time of reaction were discussed. A homogeneous sulfation method with good yield of more than 80% was obtained and the sufur content was found to be 21.9%. Conformation change in sulfated polysaccharide with changes of intrinsic viscosity, polarimetery and CD spectra were found to be related to the negative charge. The polysaccharide sulfates were shown to have better activity to inhibit cytopathic effect of human immunodeficiency virus(HIV) infection than native polysaccharides. The effective concentration was 10 micrograms.ml-1 while the cytotoxic concentration was 2.5 mg.ml-1.

[Field trials on the efficacy of albendazole composite against intestinal nematodiasis].

AIM: To study the anthelmintic effect of albendazole composite (containing 67 mg of albendazole and 83.3 mg base of pyrantel pamoate per tablet). METHODS: A randomized controlled study was carried out to compare the efficacy of a single dose of 3 or 2 tablets of albendazole composite versus a single dose of 400 mg of albendazole or 10 mg base/kg of pyrantel pamoate for treatment of intestinal nematodiasis including 1,864 cases infected with hookworm, 1,568 cases infected with Ascaris, 1,785 cases infected with Tricuris trichiura and 373 children infected with Enterobius vermicularis. RESULTS: In adults, the egg negative conversion rate of a single dose of 3 or 2 tablets of albendazole composite reached 65.0% and 52.7% for hookworm infection (P < 0.01), 100% and 100% for Ascaris infection, and 26.5% and 19.2% for Trichuris infection (P < 0.01), respectively. There were significantly better effect against hookworm with 3 tablets of albendazole composite than that with single albendazole or pyrantel pamoate (65.0% vs. 47.6% and 38.5%, P < 0.01). The effect of 2 tablets of albendazole composite against hookworm was also higher than that of single pyrantel pamoate (P < 0.01) and equal to single albendazole but the anthelmintic effect against Trichuris infection was lower than that of single albendazole (19.2% vs. 26.5%). In 2-6-year-old children, the effect of 1.5 tablets of albendazole composite against Enterobius vermicularis infection showed an egg negative conversion rate of 100% which was higher than that of single pyrantel pamoate (100% vs. 83.0%, P < 0.01). The worm collection data showed that the worm-expelling action of albendazole composite was much more rapid than that of albendazole. There were no adverse effects of albendazole composite on blood picture, liver or renal function and ECG. The side effect of both 3 and 2 tablets of albendazole composite was mild and transcient. CONCLUSION: Albendazole composite exhibits a synergistic effect of both albendazole and pyrantel pamoate.

[Antisense oligodeoxynucleotide targeted at the pre-s region of DHBV could inhibit virus replication in vivo].

To study the antiviral effect of antisense oligodeoxynucleotide (AS-ODN), AS-ODN phosphotioates targeted to DHBV Pre-S region (951-968nt) (5'-TATCTCCTCCATTGTTTG-3') was synthesized. Two day-old ducklings were infected i.p. with 150 microliters of DHBV (3 x 10(8) copies/ml DHBV). After 12 days, 6 DHBsAg and DHBV DNA positive ducks were divided into two groups. In treated group, each duck was injected i.v. AS-ODN 20 micrograms/gram weight/day for 10 dyas successively, while in the control group, each duck received the same volume of normal saline for 10 days. Sera from ducks prior to treatment and treated after 6, 10 days were collected, and after 10 days ducks were sacrificed and livers were examined. DHBsAg was significantly decreased in all three treated ducks after 10 days of AS-ODN treatment. One of these ducks showed decreased DHBsAg even 6 days after treatment. Two out of three treated ducks showed decreased serum DHBV DNA, while one remained unchanged. In contrast, none of the control group showed decreased DHBsAg and DHBV DNA did not change. By Southern blot, DHBV DNA in the liver significantly decreased in 2/3 of the treated ducks and decreased in the other duck. In all control ducks, DHBV DNA remained strongly positive. Data indicated that AS-ODN targeting at Pre-S region could partly inhibit the replication of DHBV.

[A new target position simulating system of X-knife].

This acticle describes a new target position simulator system of X-knife. It adopts a new method of simulating with target ball to ball hole as a substitute for the traditional way of simulating with target pointer to pointer. It has the advantages of precise localization, convenient operation and dependable repeatability. Clinical application has demonstrated that it is an ideal target position simulating wystem of X-knife.

[Cloning and expression in E. coli of 2,5-DKG reductase I from Corynebacterium].

Two different 2,5-DKG reductases was purified from Corynebacterium sp. SCB3058, then its genomic DNA was used as template, a segment containing 2,5-DKG reductase I was amplified by PCR and cloned into pGEM3zf(+) to obtain the recombinant plasmid pGEM813. The sequence analysis showed that the cloned segment was 1107 bp in length, contained a single open reading frame of 834 nucleotides, which encoded a 34 kD protein consisted of 278 amino acids. After the primary control sequence was deleted, the expression vector pBL4 was constructed by plasmid pBL. With induction of temperature, a 34 kD protein was specifically expressed in E. coli DH5 alpha in high level. The expressed recombinant protein accounted for 20% of the total cell protein and had high specific enzyme activity. In spite of 30 degrees C, 37 degrees C or 42 degrees C induction, the specific activity of enzyme was almost the same level. These results indicate that most of the recombinant protein induced at 42 degrees C was existed with inclusion bodies. This work was the bases of constructing a recombinant Erwinia which can produce 2-KLG directly from D-glucose by one-step fermentation.

[Absorbable self-reinforced biomaterials used for fracture fixation implants].

A series of absorbable polymers used for osteosynthesis implants are introduced in this review. To reduce the degradation rate and increase the strength of them, some methods, such as self-reinforced or coating with other polymers, have been presented. Some clinical and animal's experiments have been discussed.

[A method for measuring the isocentre positional accuracy of X knife system].

The mechanical isocentre positional accuracy of the X Knife system is a very important technological parameter in the course of the X Knife therpy. In this paper, a film-concentric circle method for measuring the mechanical isocentre positional accuracy is introduced. A simulation ball set at the isocentre of the X Knife mechanical positional system, as a substitute for target, was imaged on a film (the simulation ball's image is a circle and its diameter is 8 mm), and a collimator was imaged on the same film (the collimator's image is also a circle and its diameter is 30 mm). The distance between these two circles' centres is the error of the isocentric system. This method has been used in our work.

[Effects of hairy root huangqi on hemopoietic function].

The effects of hairy root Huangqi on hemopoietic system were studied in this paper. The oral administration of 5-20 g/kg hairy root Huangqi per day for 5-12 days could increase the count of RBC and reticulocytes in mice with hemorrhagic anemia, raise the hemoglobin, RBC and hematocrit in mice with hemolytic anemia, enhance the number of WBC and nucleared cells in bone marrow of mice treated with cyclophosphamidum or x-rays. The experiments showed that hairy root Huangqi promoted the hemopoietic function. The effects were similer to those of natural Huangqi.

[Study of photoreflectance in Cd1-xMnxTe/Cd1-yMnyTe superlattices of II - VI diluted magnetic semiconductors].

The observation of photoreflectance spectra of Cd1-xMnxTe/Cd1-yMnyTe superlattices was carried out at temperature T = 80K. The samples were grown by molecular beam epitaxy (MBE) technique. The heavy-and light-hole excitonic transition structures 11H, 22H, 33H and 11L were observed and the theoretical calculations, including the strain effects, were performed. By comparison, both agree well.

[Flotation spectrophotometric determination of platinum on isochromatic dye ion pair with tetrabromofluorescein and rhodamine 6G].

The flotation spectrophotometric method for the determination of platinum with isochromatic dye ion-pair is described. Rhodamine 6G (R6G) was produced and entered into aqueous phase when pH = 5.5 buffer solution was added to the Pt (SnCl3)5 (R6G)3 by isopropyl ether flotation. And then R6G. TBF was floated by tetrabromofluorescein(TBF)with toluene. The floatation was dissolved in acetone and absorbance of the solution was measured at 530nm. The sensitivity was raised because of the six dyes (Pt : R6G : TBF = 1 : 3 : 3) assistant effect. The molar absorptivity of this method is 5.92 x 10(5)L x mol(-1) x cm(-1). Beer's law was obeyed in the range of 0-2 microg/10mL with a relative standard deviation of 3.8%. The method was applied satisfactorily to the determination of platinum contained in catalysts without separation.

Modulation of intestinal estrogen receptor by ovariectomy, estrogen and growth hormone.

Ovarian hormone deficiency decreases and estrogen (E2) and growth hormone (GH) administrations increase intestinal absorption of calcium (Ca++). However, the underlying mechanisms are uncertain. To examine whether alterations in the binding characteristics of intestinal estrogen receptors (ERs) are involved, we developed and validated methods for simultaneous measurement of intestinal ERs in cytosolic and nuclear fractions and applied these techniques to four groups of female rats: sham-operated, ovariectomized (Ovx), Ovx + 5 micrograms E2/kg b.wt./day and Ovx + 8 mg GH/kg. b.wt./day. All animals were killed on day 21, and mucosal cells harvested from the duodenum for ER determination. The cytosolic and nuclear ERs were 117.2 +/- 2.7 fmol/mg protein and 64.9 +/- 1.2 fmol/mg DNA, respectively, in sham-operated rats and decreased by 16.1% and 17.0% to 98.4 +/- 1.7 fmol/mg protein and 53.8 +/- 1.3 fmol/mg DNA, respectively in Ovx rats (P < .001). E2 therapy prevented completely the decrease in cytosolic and nuclear ERs that occurred in Ovx rat (126.1 +/- 2.9 fmol/mg protein and 68.0 +/- 3.0 fmol/mg DNA, respectively, in the E2-treated group). Similarly, GH administration prevented the decrease in cytosolic and nuclear ERs that resulted from ovariectomy (119.2 +/- 3.2 fmol/mg protein and 63.4 +/- 1.3 fmol/mg DNA, respectively, in the GH-treated group). The Kd of nuclear ER-ligand complex was 2.0 +/- 0.03 nM in sham-operated rats and was slightly modulated by Ovx, E2 and GH (3.3 +/- 0.02, 2.33 +/- 0.09 and 2.23 +/- 0.04 nM, respectively, P < .001), but the Kd of cytosolic ER-ligand complex was not altered by Ovx, E2 or GH. Our findings indicate that E2 deficiency down-regulates, whereas E2 and GH administrations up-regulate intestinal ERs and prevent ovariectomy-induced decrease in receptor binding affinity. We conclude that E2 deficiency, E2 and GH may modulate intestinal Ca++ absorption, in part, by altering the abundance and binding characteristics of intestinal ERs.

Microsomal epoxide hydrolase polymorphism and risk of spontaneous abortion.

We conducted a nested case-control study to examine the association of microsomal epoxide hydrolase polymorphism with spontaneous abortion. The analysis included 127 cases and 107 controls from a rural community in China. The prevalence of the homozygous wild-type (AA), the heterozygous variant (Aa), and the homozygous variant (aa) in exon 3 of epoxide hydrolase was 13.4%, 34.6%, and 52.0% in cases and 27.1%, 30.8%, and 42.1% in controls, respectively. In contrast, the variant genotypes in exon 4 of epoxide hydrolase were less frequent in cases than controls. Using women with genotype AA as the referent, the adjusted odds ratio of spontaneous abortion was 2.69 [95% confidence interval (CI) = 1.33-5.42] for those with genotype Aa or aa in exon 3; but it was 0.45 (95% CI = 0.22-0.94) for those with genotype Aa or aa in exon 4, indicating that the two variants have opposite associations with spontaneous abortion. The findings persisted after adjustment for age, education, parity, smoking, alcohol use, occupation, and pesticide exposure, as well as in subgroup analysis. Moreover, for the variant genotypes Aa or aa in exon 3, the odds ratio was twice as great in those cases with three or more spontaneous abortions than in those with fewer spontaneous abortions.

Toxic methemoglobinemia: a rare but serious complication of transesophageal echocardiography.

Cyanosis occurring during a transesophageal echocardiogram (TEE) is usually related to the respiratory depressant effect of the administered sedative. However, acute toxic methemoglobinemia related to topical benzocaine is a rare and not well recognized but potentially life threatening cause of cyanosis during or after TEE. A case of this condition is presented and the literature on its frequency, diagnosis and management is briefly reviewed.

Ventricular tachycardia after oxytocin injection in patients with prolonged Q-T interval syndrome--report of two cases.

Oxytocin is widely used in obstetric settings to stimulate uterine contraction and prevent postpartum hemorrhage. Its adverse effects which include transient hypotension and increase heart rate could be life-threatening in patients with fixed cardiac output or hypotension resulting from hemorrhage. We reported two cases suspected to have preexisting prolonged Q-T interval syndrome (PQTS) who developed ventricular tachycardia immediately after intravenous injection of oxytocin. Anesthetic management of and use of oxytocic agents in patients with PQTS were discussed.
