Assay of long-chain acyl-CoAs in a complex reaction mixture.

A method has been developed which allows the quantitative analysis of labeled or unlabeled acyl-CoAs in complex reaction mixtures. The method is based on (a) a quantitative solubilization of acyl-CoAs and lipids, directly in the reaction vessel, by 0.05 M Tris-HCl, pH 7.5/CHCl3/CH3OH (1/3/3, v/v/v); (b) monodimensional TLC of aliquots of the whole reaction mixture, resolving malonyl-CoA, acetyl-CoA, long-chain acyl-CoAs, polar lipids and neutral lipids plus free fatty acids; and (c) quantitation by TLC densitometry and/or TLC radiochromatography. All fractions--and particularly long chain-acyl-CoAs--can then be analyzed for distribution and label of fatty acyl moieties.

Assay of stearoyl-CoA synthesis in microsomes from normal and Trembler mouse sciatic nerves.

Using a new one-step technique which allows the characterization and a quantitative assay of long-chain acyl coenzyme A (acyl-CoA), the activity of the stearoyl-CoA synthetase was studied in the membrane fraction from sciatic nerves of normal and Trembler mice. A synthesis of acyl-CoAs was observed in both cases, but, whatever the experimental conditions, there was a two-fold increase of the specific activity of the ligase in the PNS of the Trembler mouse.

Alteration of sulfatide synthesis in control and Trembler mice during Wallerian degeneration and remyelination.

Sulfatide synthesis from sulfate is much greater in the peripheral nerves of the Trembler mouse. After nerve transection, during Wallerian degeneration, this synthesis rate drops down very rapidly in both normal and Trembler mice. Twenty-four hours after permanent transection, the rate of synthesis is reduced by 80% in the mutant and 50% in the normal mouse. Four days after transection, the synthesis rate in the Trembler is only 9% of that observed in intact nerves, and 21% of that in the intact nerves of normal animals. After 5 d the synthesis remains constant. Thus, enhanced synthesis of sulfatides in the Trembler mouse is probably not caused by Wallerian degeneration. After crush of the sciatic nerve, the synthesis rate decreases very rapidly in the normal mouse as it does after permanent transection. But during regeneration, from the 7th day, it rises dramatically and 14 d after crush, a 2.5-fold increase in the synthesis rate is observed, compared to that in the contralateral control nerve. This synthesis rate returns to normal 1 mo after crush. In the Trembler, the synthesis decreases for 2 d after crush and increases from then on, eventually reaching the value of the contralateral control Trembler nerve within 2 mo. In the mutant there is no prominent peak of sulfatide synthesis during regeneration.

[Fractionation of a sciatic nerve homogenate from normal, trembler and quaking mice on a continuous sucrose gradient].

Two fractions, characterized by biochemical criteria [quantity and density of particulate material, localization of myelin specific protein--Po, P1 and Pr--and of 2',3'-cyclic nucleotide 3'-phosphohydrolase (CNP) and of 5'-nucleotidase], were isolated from sciatic nerve homogenate of normal, Trembler and quaking, young and adult Mice, on a continuous sucrose gradient. The fraction, at 0.58-0.6 M, the richest in membrane particles contains the myelin. The fraction at 0.7-0.75 M should contain the plasma membrane of the Schwann cell which synthesizes myelin.

[Biochemical aspects of myelinogenesis in the peripheral nervous system].

The excised sciatic nerve of the trembler mouse synthesizes minute amounts of C20 and C22 saturated fatty acids as compared to the normal PNS. No lignoceric acid synthesis is observed. The microsomal fraction contains however an elongase able to synthesize with nearly normal levels the saturated very long chain fatty acids.

[Pathology of myelin; a biochemical approach].

In the sciatic nerves of the Trembler mouse, considered as a good model for the study of the hereditary hypertrophic neuropathies, the levels of the different classes of lipids, of alkanes and of very long chain fatty acids are reduced. Those of cholesterol esters and lysophosphatidylcholine are increased.

Lipid composition of sciatic nerve from dysmyelinating trembler mouse.

The amounts and the distribution of the various lipids were studied in the sciatic nerves from normal and trembler mice. When compared to the normal, the total lipidic amount was reduced by 66% in the trembler mouse, and each class of lipid was decreased nearly the same way, except for the cholesterol esters the value of which increased five times. The level of each individual phospholipid was decreased, phosphatidylcholine being the least affected and phosphatidylserine the most altered.

[Biosynthesis of long chain fatty acids and alkanes in Saccharomyces cerevisiae].

Cell-free extracts of Saccharomyces cerevisiae catalyse the formation of very long chain fatty acids with an even number of carbon atoms. These acids may be alpha-oxidated leadint to odd-chain acids. Both even and odd chain acids may be subjected to reductive decarboxylation giving rise to alkanes with respectively odd or even aliphatic chains.
