Antigenic analysis of Chlamydiae by two-dimensional immunoelectrophoresis. I. Antigenic heterogeneity between C. trachomatis and C. psittaci.

Tw0-dimensional immunoelectrophoresis was used for the antigenic analysis of Chlamydia. Antigens of chlamydiae were found to be successfully solubilized with the nonionic detergent Triton X-100. Nineteen and 16 distinct components were observed for Chlamydia trachomatis strain L2/434/Bu and C. psittaci strain Meningopneumonitis/Cal-10, respectively. The triton-solubilized antigens of chlamydiae differ strikingly between species since only one strong reciprocal cross-reacting antigen was observed between 434 and Cal-10. However, the antigens of Trachoma strain K/UW-31/Cx and LGV strain 434 were found to be very similar.

Antigenic analysis of Chlamydiae by two-dimensional immunoelectrophoresis. II. A trachoma-LGV-specific antigen.

Two-dimensional immunoelectrophoresis was utilized to study precipitins in hyperimmune rabbit serum made against chlamydiae and from patients with chlamydial infections. An antigen of Triton X-100-solubilized L2/434/Bu organisms with an electrophoretic mobility of 0.65 relative to bovine serum albumin at pH 8.6 was excised from the agarose gel of electrophorograms as antigen-antibody complexes and used to immunize rabbits. A monospecific antiserum to antigen 0.65 was obtained that reacted with Trachoma-LGV strains L2/434/Bu, B/TW-5/OT, and K/UW-31/Cx, but not with the mouse pneumonitis (Nigg) strain or the psittacosis strain meningopneumonitis (Cal-10). The Trachoma-LGV specificity of antigen 0.65 was further shown by indirect immunofluorescence straining with the monospecific antiserum of chlamydial inclusions in infected HeLa cells. Precipitins with a specificity for antigen 0.65 were indentified in 15 of 18 sera from patients with diagnosed Chlamydia trachomatis infections LGV, trachoma, nongonococcal urethritis, and nongonococcal cervicitis by using monospecific antiserum to antigen 0.65 in the peak suppression test. Thus, antigen 0.65 appears to be a Trachoma-LGV-specific antigen that has considerable promise for serodiagnosis.

A simplified method for immunological typing of trachoma-inclusion conjunctivitis-lymphogranuloma venereum organisms.

Responding to the pressure of large numbers of trachoma-inclusion conjunctivitis (TRIC)-lymphogranuloma venereum (LGV) isolates from field studies requiring serotyping, we have developed a simplified, less-precise method that utilizes cell culture-grown organisms to produce mouse antisera which is tested agaist prototype TRIC-LGV antigens in the micro-immunofluorescence test. Cell cultures with as few as 5 to 15% of cells showing inclusions produced adequate antibody in mice 4 days after single injection. Knowledge of the reaction of prototype antisera with the antigens has allowed typing of most isolates tested from the pattern of cross-reaction of their antiserum.

Effect of polycations, polyanions and neuraminidase on the infectivity of trachoma-inclusin conjunctivitis and lymphogranuloma venereum organisms HeLa cells: sialic acid residues as possible receptors for trachoma-inclusion conjunction.

The infectivity of trachoma-inclusion conjunctivitis (TRIC) organisms (TW-5) was enhanced by pretreatment of HeLa cell monolayers before inoculation with diethylaminoethyl (DEAE)-dextran (30 mug/ml) and poly-l-lysine (10 mug/ml) and inhibited by dextran sulphate (250 mug/ml), fetuin (4%), ovomucoid (5%), N-acetyl neuraminic acid (0.5%), and Cholera vibrio neuraminidase (100 U/ml). The infectivity of lymphogranuloma venereum organisms (434) was not affected by DEAE-dextran, fetuin, and neuraminidase, was slightly inhibited by poly-l-lysine, and was inhibited by dextran-sulphate, ovomucoid, and N-acetyl neuraminic acid. The study suggested that sialic acid residues on the cell surface may be specific receptors for TRIC organisms. The receptors for TRIC organisms (TW-5 and TW-3) could be specifically blocked with inactivated (56 C for 30 min) TRIC organisms at the ratio of one live to 100 inactivated TRIC organisms, but not by inactivated lymphogranuloma venereum (434) or influenza virus (A(2)/Jap 305).

Interfacial dehydration by alcohols: hydrogen bonding of alcohols to phospholipids.

The interaction between alcohols (ethanol and n-butanol) and dipalmitoylphosphatidylcholine (DPPC) in carbon tetrachloride was studied by Fourier transform infrared spectroscopy (FTIR). Upon addition of the alcohols, the P = O stretching band of DPPC at 1260 cm-1 shifted to lower frequency (red-shift). The red-shift indicates that the P = O vibration became slower possibly because the heavier alcohol molecules replaced the water molecules hydrogen bonded to the PO2 moiety. The formation constants between the PO2 group and ethanol or n-butanol (n-butanol data in parenthesis) were 19.0 M-1 (7.1 M-1) when estimated from the spectral change. A new absorbance peak appeared at 3265 cm-1 (3275 cm-1) representing the DPPC-alcohol complex. The formation constant of this complex was also 19.0 M-1 (7.1 M-1). The identical formation constant suggests that the DPPC-alcohol complex was formed at the PO2 moiety of DPPC with hydrogen bonding to the alcohol OH. At higher alcohol concentrations, the absorbance peak of DPPC-alcohol complex shifted to 3225 cm-1 (3235 cm-1). Apparently, the lower frequency shift at higher alcohol concentration occurred by the formation of alcohol multimers (dimer, trimer, and tetramer) interacting with DPPC.

Importance of reinfection in the pathogenesis of trachoma.

The authors' epidemiologic studies of trachoma on Taiwan and experimental monkey eye infections with and without Chlamydia trachomatis immunization are reviewed for the data they provide on the pathogenesis of trachoma. These studies indicate that trachoma is an immunopathologic disease in which the more severe progressive trachoma infections with pannus and scar formation occur only after reinfection. This hypothesis is supported by a 10-year study of 32 family households that were followed with repeated clinical and laboratory observations. Although most cases of active trachoma healed spontaneously, there were 26 persons in nine families who developed clinical and laboratory evidence of 29 episodes of new trachoma eye infection. On the basis of the clinical disease, its persistence, and the laboratory findings, these 29 new infections could be divided into 10 primary, nine secondary, and 10 tertiary infections. Details of the different stages of infection leading to chronic trachoma are presented.

Comparison of ophthalmic silver nitrate solution and erythromycin ointment for prevention of natally acquired Chlamydia trachomatis.

During prospective studies of infants born vaginally to women with cervical Chlamydia trachomatis infection, we evaluated 27 infants given 0.5% erythromycin ointment and 93 given 1% silver nitrate solution as eye prophylaxis, according to the preference of the parents or delivery room personnel. The cumulative proportion of infants developing chlamydial conjunctivitis was 25% for both groups (P = 0.37, Mantel-Cox test). The cumulative proportion of infants developing chlamydial infection at any anatomic site was 74% for those given erythromycin and 70% for those given silver nitrate (P = 0.93). The two groups did not differ significantly in cumulative proportions developing nonchlamydial conjunctivitis. These results indicate that, as it is routinely used in our hospital, erythromycin ointment was not more effective than silver nitrate as prophylaxis against chlamydial conjunctivitis. The influence of delayed administration on efficacy of prophylaxis requires further evaluation.

Oral contraceptives do not alter the course of experimentally induced chlamydial salpingitis in monkeys.

BACKGROUND AND OBJECTIVES: The effects of oral contraceptive use on chlamydial infections and ensuing PID are not well understood. In this study, the effects of oral contraceptives on the clinical course of acute chlamydial salpingitis were investigated. STUDY DESIGN: Monkeys (n = 4) in which salpingeal auto-transplants had been established were given oral contraceptive pills consisting of a combination of estrogen and progesterone (Norlestrin 1/50, Parke-Davis, Ann Arbor, MI). After one complete cycle of hormone treatment, monkeys were inoculated with C. trachomatis serovar E in the subcutaneous salpingeal pockets while under continuous hormone treatment. Inoculated pockets were biopsied serially for isolation of organisms and histopathological evaluation. Two monkeys not given oral contraceptives were studied in parallel. RESULTS: The duration of infection as evidenced by recovery of organisms was not appreciably different between the treated and untreated monkeys. Chlamydia could be recovered from pocket tissues through day 10 post-infection in control and treated animals, and on day 14 post-infection in one of two control animals. Thereafter all isolation became negative. The inflammatory infiltrate consisted primarily of mononuclear cells (50-100 cells per 40 x field). CONCLUSIONS: Our results indicate that administration of oral contraceptives did not affect the duration of shedding of organisms nor the histopathology of acute chlamydial infection of the reproductive tract.

Acute Disease↗

Chlamydia pneumoniae and cardiovascular disease.

Chlamydia pneumoniae is a ubiquitous pathogen that causes acute respiratory disease. The spectrum of C. pneumoniae infection has been extended to atherosclerosis and its clinical manifestations. Seroepidemiologic studies have associated C. pneumoniae antibody with coronary artery disease, myocardial infarction, carotid artery disease, and cerebrovascular disease. The association of C. pneumoniae with atherosclerosis is corroborated by the presence of the organism in atherosclerotic lesions throughout the arterial tree and the near absence of the organism in healthy arterial tissue. C. pneumoniae has also been isolated from coronary and carotid atheromatous plaques. To determine whether chronic infection plays a role in initiation or progression of disease, intervention studies in humans have been initiated, and animal models of C. pneumoniae infection have been developed. This review summarizes the evidence for the association and potential role of C. pneumoniae in cardiovascular disease.

Japanese encephalitis surveillance in China (Province of Taiwan) during 1968-1971. I. Geographical and seasonal features of case outbreaks.

During 1968-1971 Japanese encephalitis (JE) surveillance was conducted with WHO-assisted programs in Taiwan area. Emphasis was placed on: (1) active case-finding by hospital visits; (2) blood collection from every patient at the suitable time; and (3) the enforcement of standard diagnostic criteria on the results of hemagglutination-inhibition tests. Each year, approximately 90% of reported patients were etiologically examined. JE virus etiology was established in 277 (1968), 279 (1969), 269 (1970) and 158 (1971) cases. Despite the apparent concentration of cases in several cities, practically all the cultivated plains and basins were established as potentially endemic for JE. JE outbreak is found to have been clearly associated with season, not only in subtropical but also in tropical Taiwan. Each year it occurred consistently in mid-July in the southernmost county, 2-3 weeks after the peak of rice transplantation and its duration was a few weeks. On the western side of the island, although less consistently, JE outbreak seems to occur one to three weeks later than in the south. Outbreak in eastern Taiwan was a mid-September phenomenon for 3 years. Observed seasonal lag, which was significant between two neighboring localities on same latitude, suggests that paddy water management is more directly related to date of outbreak than local climate. Seasonal characteristics further suggests that the area may be separated into eight ecologic subdivisions. JE-virus transmission to a man during April at temperatures below the average of 20 C was discussed.

Japanese encephalitis surveillance in China (Province of Taiwan) during 1968-1971. II. Age-secific incidence in connection with Japanese encephalitis vaccination program.

A baseline age/morbidity pattern for Japanese encephalitis (JE) epidemic in China (Province of Taiwan) in the 1960s was sought in JE surveillance. The pattern is characterized by: (1) high incidence in age groups 2-4 years, (2) moderately high incidence in age groups 5-7 years, (3) negligible incidence in children under 1 year of age as well as in adults, and (4) the variability of relative height of peak incidence co-incident with the JE-epidemic size in situ. Concurrently with surveillance, a JE-vaccination program was implemented by the Government. The children of target age groups received two doses of vaccine each during April. Vigilant supervision was given to the campaign to achieve a high coverage rate with two-dose vaccination and meticulous record-keeping. During 1969-1972, 2-year olds were the exclusive target; 52.2% (1969), 71.5% (1970), 74.5% (1971), and 83.0% (1972) of the target age group were covered. A considerable change from the baseline age/morbidity pattern seen in 1968 occurred during the above period. The comparison of epidemicity-adjusted incidence by year indications that incidence reduction in the 2- and 3-year old children in 1971 were substantial and suggests the possible impact of vaccination. The total JE incidence for the period 1968-1971 appears to be much lower in vaccinated population (23 reported and 8 confirmed/0.9 million) than in unvaccinated population (1,330 reported and 679 confirmed/2.2 million).
