Human IgE and IgG antibodies to mosquito proteins detected by the immunoblot technique.

Immunoblot technique was used in this study to detect IgE and IgG antibodies in human sera against mosquito antigens. Mosquito proteins were separated on SDS-polyacrylamide gels and transferred electrophoretically to nitrocellulose papers. After incubation with sera from different individuals, the precipitated bands were analyzed with enzyme-labeled goat anti-human immunoglobulins. Distinct patterns of antigen were recognized by IgE and IgG antibodies.

Multiple effects of honokiol on catecholamine secretion from adrenal chromaffin cells.

The molecular mechanism of honokiol, extracted from the bark of Magnolia obovata, was studied using bovine adrenal chromaffin cells as a model system. Honokiol inhibits catecholamine secretion induced by carbachol and DMPP and that induced by exposure to high K+ and Ba2+ but to a lesser extent. The inhibitory effects of trifluoperazine and honokiol on carbachol-, high K(+)- and Ba2(+)- induced secretion were not additive. The results suggest that honokiol interferes with the interaction between the acetylcholine receptor and its agonists and that honokiol may also affect the steps in exocytosis after intracellular calcium has been raised, possibly at the site(s) where calmodulin acts.

[Microbial populations and periodontal condition--evaluation by differential phase contrast microscopy].

The purpose of this study was to determine correlations between various clinical assessments of inflammatory periodontal disease and the proportion of bacterial morphotypes in the subgingival flora of sites representing widely varying states of periodontal disease. Phase-contrast microscopy was used to determine the proportion of varying bacterial morphotypes at 154 sites. For each site the following clinical assessments were recorded: gingival index, probing depth, plaque index and calculus index. Statistically significant (p less than 0.01) positive correlations were found between gingival index and spirochetes or motile rods, and between probing depth and spirochetes. Significant negative correlations were found between gingival index and coccoid cells, and between probing depth and coccoid cells or rods. The significant increase in the proportion of spirochetes or motile bacteria occurred when bleeding upon probing was observed. The ratio of motile to non-motile cells at advanced disease sites was in the vicinity of 1:1. The data obtain in this study may be useful in monitoring the effects of various treatment modalities on the periodontal flora and may be helpful in determining the presence or absence of active disease.

Prenatal diagnosis and postnatal management of surgically correctable fetal malformations.

Twenty newborns, prenatally diagnosed to have congenital malformations, received surgical correction postnatally in recent 7 years. Using serial ultrasound examinations, the following lesions were found: multicystic dysplastic kidney (3 cases), ureteropelvic and ureterovesical junction stenosis (1 case), Mullerian duct cyst (1 case), sarococcygeal teratoma (3 cases), adenomatoid malformation of lung (1 case), omphalocele (1 case), gastroschisis (1 case), duodenal atresia (1 case), ileal atresia (1 case), meconium peritonitis (2 cases), imperforate anus (1 case), adrenal cyst (1 case), congenital diaphragmatic hernia (1 case) and a pair of xipho-omphalopagus twin. The results of surgical corrections were good in all except one baby with omphalocele who died of respiratory distress syndrome.

Characterization and comparison of epsilon-crystallin and lactate dehydrogenases in the lenses of vertebrates and invertebrates.

Screening of lens homogenates for the identification of lactate dehydrogenases was undertaken for the representative species from five major classes of vertebrates plus the cephalopod of invertebrates. The duck and caiman lenses appeared to contain the highest enzymatic activity of this glycolytic enzyme among all species examined. Biochemical isolation and characterization of epsilon-crystallins from the duck and caiman lenses revealed differences between these structural crystallins and the authentic lactate dehydrogenase of the avian heart regarding some of the kinetic properties. This is in contrast with the claim that duck epsilon-crystallin is identical to heart-type lactate dehydrogenase.

Two antiplatelet agents from Magnolia officinalis.

Magnolol and honokiol are two position isomers isolated from the bark of Magnolia officinalis. Both inhibited the aggregation and ATP release of rabbit platelet-rich plasma induced by collagen and arachidonic acid without affecting that induced by ADP, PAF or thrombin. Aggregation of washed platelets was more markedly inhibited than that of platelet-rich plasma, while the aggregation of whole blood was least affected by both inhibitors. Thromboxane B2 formation caused by collagen, arachidonic acid or thrombin was in each case inhibited by magnolol and honokiol. The rise of intracellular calcium caused by arachidonic acid or collagen was also suppressed by both agents. Collagen-induced intracellular calcium increase in the presence of indomethacin was suppressed by magnolol. It is concluded that the antiplatelet effect of magnolol and honokiol is due to an inhibitory effect on thromboxane formation and also an inhibition of intracellular calcium mobilization.

(Na+ -K+)ATPase activity in erythrocyte membranes of spontaneously, one kidney-one wrapped, and deoxycorticosterone acetate--NaCl hypertensive rats.

(Na+ -K+)ATPase activity in erythrocyte membranes of spontaneously (SHR), one kidney-one wrapped, and deoxycorticosterone acetate (DOCA)-NaCl hypertensive rats was studied. (Na+ -K+) ATPase activity decreased in both prehypertensive (6 weeks old) and hypertensive (14 weeks old) stages of SHR, suggesting that the alteration of this enzymic activity may be due to a pre-existing defect in the membrane rather than being a consequence of hypertension. By contrast, (Na+ -K+)ATPase activity remained unchanged in the one kidney-one wrapped hypertensive rats, whereas that of one kidney-one wrapped normotensive rats as well as that of DOCA-NaCl hypertensive rats was increased significantly (P less than 0.05). These changes were specific for (Na+ -K+) ATPase, since Mg2+-ATPase activity was not altered. The susceptibility of (Na+ -K+)ATPase to the inhibitory action of ouabain was not changed significantly. These findings indicate that (Na+ -K+)ATPase activities of erythrocyte membranes isolated from the different types of hypertensive rats were subject to different changes. Whether this phenomenon applies to the clinical distinctions among the various types of hypertension remains a subject for further investigation.

Multiple dosing increases the ocular bioavailability of topically administered flurbiprofen.

The ocular bioavailability of radiolabeled flurbiprofen was determined after single or multiple topical dosing of rabbits. Three doses at half-hour intervals of a 0.03% solution produced higher concentrations in the tissues than those after a single dose of either the 0.03% or the 0.1% solution. Three doses of 0.03% flurbiprofen gave intraocular concentrations in the range that inhibits cyclo-oxygenase activity. Administration of more than three doses did not significantly increase the concentration of flurbiprofen in most tissues. The drug distribution pattern in the ocular tissues after multiple dosing was the same as after single dosing. Although multiple dosing with the 0.03% concentration resulted in higher tissue levels of flurbiprofen, increasing the concentration to 0.1% did not result in greater ocular bioavailability of the drug.

Smooth muscle tumours of the digestive tract: report of 160 cases.

Of 160 patients seen in the period 1951-84 with smooth muscle tumours of the digestive tract, 71 proved to have leiomyomas, 87 had leiomyosarcomas and 2 had leiomyoblastomas. Tumour diameter was frequently greater in patients with leiomyosarcoma. The surgical mortality was 2.8 per cent after treatment of leiomyoma and 10.3 per cent after treatment of leiomyosarcoma. After resection of leiomyosarcoma the 2-year survival rate was 86 per cent and the 5-year survival rate was 43 per cent. The only hope of cure of these malignant lesions lies in surgical resection but, even when the lesion is incurable, resection may allow worthwhile palliation.

Evidence for expression of the facilitated glucose transporter in rat hepatocytes.

The eukaryotic facilitated glucose transporter (GT) is expressed by many cell types, with the notable exception of hepatocytes; however, GT is expressed by several hepatoma cell lines, including the well-differentiated lines Fao, Hep3B, and HepG2. We report on studies carried out to determine the aspect(s) of the transformed phenotype that might be responsible for activating GT expression. Using RNA blot analysis with probes derived from rat GT cDNA, we found that GT was expressed by rat hepatocytes under two conditions (i) in vitro, when isolated hepatocytes were placed in cell culture, and (ii) in vivo, when rats were subjected to starvation for greater than or equal to 2 days. However, GT expression was not an obligatory feature of hepatomas, since two primary hepatocellular carcinomas did not express any GT mRNA. GT expression in hepatocytes was reduced by addition of dimethyl sulfoxide or sodium butyrate to the culture medium. Since these reagents are known to promote differentiation in some cell culture systems, their effect on hepatocytes may be to maintain the GT repression normally observed in vivo. Inclusion or exclusion in the culture medium of several other agents that enhance hepatocyte viability (serum, insulin, corticosteroids, epidermal growth factor, or triiodothyronine) did not affect GT expression. It is unclear whether the two conditions that led to GT expression in hepatocytes are related by a common signaling mechanism. Possibly, both cases involve a "stress" response: in vivo, a normal physiological response to starvation; in vitro, a response to a major alteration in the cellular environment.

Exsheathment of microfilariae of Brugia pahangi in the susceptible and refractory strains of Aedes aegypti.

Exsheathment of microfilariae of Brugia pahangi was studied in susceptible (Liverpool) and refractory (Bora-Bora) strains of Aedes aegypti. It was found that the microfilariae tend to carry their sheaths into the haemocoel of both strains of Ae. aegypti within two hours after the engorgement of mosquitoes from a rat parasitized by filariae. The percentage of sheathed microfilariae in the haemocoel then progressively decreased to 0% at eight hours and to 1% at 24 hours post-ingestion in the Bora-Bora and Liverpool strains, respectively. Those microfilariae that remained in the midgut more than two hours after ingestion were most likely to cast off their sheaths there. The percentage of microfilariae exsheathed in the midgut progressively increased to about 91 and 78% at 24 hours post-ingestion in the Bora-Bora and Liverpool strains, respectively. These results suggested that the exsheathment of microfilariae occurs both in the haemocoel and in the midgut of two strains of Ae. aegypti.

The ocular distribution of bunolol in the eyes of albino and pigmented rabbits.

The ocular administration of a 50 microL instillation of bunolol hydrochloride, a beta 1- and beta 2- adrenoceptor blocking agent, resulted in significantly higher drug levels in the choroid/retina, iris, and ciliary body of pigmented rabbits compared with albino rabbits following topical administration. The concentrations in these tissues also persisted longer in the pigmented rabbits' eyes. However, no statistically significant differences in tissue levels were observed in the cornea or conjunctiva. The results of this study support the previously reported finding with timolol which showed longer retention of the drug in the iris, ciliary, choroid, and retina of pigmented rabbits than albinos.

A comparative study on schizophrenia diagnosed by ICD-9 and DSM-III: course, family history and stability of diagnosis.

Data from the Taipei Center of the International Pilot Study of Schizophrenia were reanalyzed using the ICD-9 and DSM-III diagnoses at 7-year follow-up. Patients diagnosed as schizophrenic according to DSM-III were shown to be a more homogeneous group in terms of their clinical manifestations, social functions and family psychiatric history than those defined as schizophrenic by ICD-9. The discordant cases of ICD-9 schizophrenia and DSM-III affective disorders were found to be different from the concordant schizophrenic group, but similar to the concordant group of affective disorders diagnosed by ICD-9 and DSM-III. Thirty-five per cent of mood-incongruent psychotic major depressive disorders defined by DSM-III at initial evaluation were diagnosed as schizophrenia at 7-year follow-up.
