Glutamic acid decarboxylase in tubules and glomeruli isolated from rat kidney cortex.

4-Aminobutyric acid (GABA) synthesis was examined in purified glomeruli and tubules of rat kidney cortex that were incubated in the presence of [2,3-3H2]glutamate. The GABA that was formed was separated from glutamate using anion-exchange resin, and identified by means of an automatic amino acid analyser. In the renal cortex only the tubules were able to form GABA (35.0 nmol mg-1 h-1); the remaining GABA synthesis found in the glomerular preparations can most probably be attributed to a contamination by cortical tubules (9%), as shown by determination of a known tubular marker enzyme (L-gamma-glutamyltransferase). Hydroxylamine (1 mM) and ethanolamine-O-sulfate (10 mM), well-known inhibitors of cerebral GABA formation and GABA catabolism respectively, inhibited renal tubular GABA formation at 100% and 44% respectively.

Activity of phosphatases and cellular contents in glomeruli and tubules isolated from growing rat renal cortex.

The activity of acid and alkaline phosphatases was determined in glomeruli and tubules isolated from 14-day old to adult rat renal cortex. Within a nephron fraction, each phosphatase activity produced a specific pattern which was not similar to that in other fractions. In the glomeruli the acid phosphatase activity increased between 14 and 21 days of life and was always higher than in the tubules where it did not significantly change with age. By using tartrate sodium and formaldehyde it was shown that the acid phosphatase isoenzymes are distributed according to a specific pattern which does not change during maturation. The alkaline phosphatase activity in glomeruli and in tubules increases with age but is always lower in the former fraction than in the latter. Furthermore, in young as well as in adult rats, glomeruli were characterized by a higher DNA content and lower RNA and protein/DNA ratios than the corresponding tubular fragments.

Isoproterenol-sensitive adenylate cyclase in glomeruli isolated from young and adult rat renal cortex.

The stimulation of the adenylate cyclase activity of isolated rat renal glomeruli by isoproterenol decreased during kidney maturation. A potentiation of the isoproterenol response by GTP was demonstrated in young and adult animals. Both KF and GPP(NH)P enhanced the enzyme activity under basal conditions in young and adult glomeruli. GTP had no significant effect upon the basal activity of the adenylate cyclase in glomeruli of 14-day old rat kidney while it had one in 21-day old and adult kidneys.

Quantitative bone scan and bone metastases in prostatic cancer.

Bone scan is an essential method of investigation for the detection of metastases; it is also used to follow the evolution of the disease and the response to treatment in prostatic cancer. The authors tried a bone scan quantification method to better evaluate the efficiency of hormonal treatment. 28 patients were followed up by this method. Taken as a whole, quantitative scans show results very similar to those of standard scans. However, there are some differences between the two methods of interpretation. The quantitative method seems to be more effective.

[Thyroid metastases from a nephro-epithelioma].

The authors report a rare case of thyroid metastasis from a nephro-epithelioma, which appeared four years after nephrectomy, and was revealed by a Basedow-type goiter. It was treated by total thyroidectomy. A cervical recurrence was diagnosed in the course of a routine checkup and arteriography. Death occurred 22 years after thyroidectomy and 26 years after nephrectomy. The diagnosis is often only made at necropsy. The slow growth of these metastases does not preclude extended survival after surgical excision, and requires active therapy.

Reactivation of the secretion from the prostatic gland in cases of reduced fertility. Biological study of seminal fluid modifications.

The efficiency of Pygenum africanum treatment was tested on a population where the acid phosphatase activity was shown to be reduced at two successive examinations. The treatment caused an increase in acid phosphatase activity and total protein secretion. However, the fact that the men who had IgA present in their ejaculate reacted more favorably to the treatment shows that Pygenum africanum is more efficient in cases where there is no prostate gland inflammation.

[Transurethral resection of the prostate. Our experience in a series of 244 cases. Mid-term results].

Two hundred and forty four patients were treated by transurethral resection of the prostate (68% for adenomas and 32% for epitheliomas) between June 1980 and August 1982. The indications for surgery are based on obstruction of the urinary outflow tract, and the weight of prostatic tissue, which can best be assessed by ultrasound. TURP is the technique of choice for small and medium-size prostates. Hemorrhage is the most frequent complication at operation. Resorption TURP syndrome is extremely rare if an isotonic liquid is used. Urinary tract infections are best treated preoperatively. 2.6% of the patients suffered from urethral stricture despite the use of a 24 French resectoscope and early removal of the catheter. The greater the surgeon's experience, the lower the incidence of definitive urinary incontinence.

[Distribution of various sites of hormonal action between microvessels, glomeruli and tubules isolated from the renal cortex of rabbits].

Collagenase dispersion of rabbit kidney cortex followed by centrifugation on discontinuous sucrose gradient, allowed the simultaneous obtention of microvascular, glomerular and tubular fractions. Microscopic studies and the measurements of cellular renin activity and the muscle-specific-enzyme creatine kinase showed that these vessels were arteriolar in nature and that they contained the preglomerular arterioles. The glomerular or tubular contamination rates were assessed by means of enzymatic markers. AVP stimulated only the tubular fraction whether GTP was present or not. In the absence of GTP, beta-isoproterenol stimulated only the tubular enzyme while in the presence of GTP, the microvascular and especially the glomerular AC were also stimulated. GTP enhanced considerably the microvascular response to bPTH-(1-34) while, in the glomeruli and tubules the effects of GTP and hormones were only additive. The possible physiological significance of these results are discussed.

[The upper urinary tract in pregnant women. Physiopathologic, ultrasonic and urographic study].

This study of the upper urinary tract with the voluntary co-operation of expectant mothers, and based on 316 ultrasound examinations and 138 urographies, confirms the high incidence of uretero-pyelocalyceal dilatations is pregnancy. Compression by the gravidic uterus and the hormonal environment of pregnancy are the best-known etiologies. The frequency of associated urinary infections and the slow regression of the dilatation in the post-partum period (three months) will be noted.

[Our experience with the treatment of urethral stenosis in a series of 90 cases at the Strasbourg Hospital Center].

Ninety patients presenting with urethral stenosis were treated surgically at the Strasbourg Hospital, involving 100 internal urethrotomies and sixteen urethroplasties. The authors consider that short bulbous stenosis, with no previous treatment, should be treated endoscopically; that, if the stenosis recurs after two internal urethrotomies, or if the stenosis is infected or located in the pendulous or membranous urethra, a urethroplasty is indicated, as in cases of long bulbous stenosis; and that, even though the rate of complications is high following Johansson's urethroplasty, this procedure is indicated in cases of infection. The use of free full-thickness grafts led to a 100% success rate.

[Epidemiology of renal epithelioma in adults].

The frequency of renal epithelioma is very high in the Bas-Rhin department (France) (9,92 per 100 000 among the male population). The various risk factors are considered: the ethnic group, genetic factors, tobacco, alcohol; all these factors are not very important. But the intake of animal lipids and proteins could be an important factor to induce renal epithelioma.
