[The family approach to a case of a battered child].

In the case of a battered child, the authors propose that the treatment be made on a family basis in order to secure a therapeutic alliance with the parents. Symptomatic treatment, based on legal and medical measures, does not allow this type of therapeutic approach. The use of a day center for the child and the parents, and a multidisciplinarian team work (pediatrician, child psychiatrist, social worker) facilitate this alliance and diminish negative counter attitudes.

[Differentiation and tumorigenicity of human malignant melanocytes. Comparative study of two "non pigmented" and pigmented lines from the same fibroblastoid cells].

A morphological, caryological and biochemical study of two established cell lines of human malignant melanocytes derived from the same original tumour, when compared with heterotransplantation results in nude Mice, showed that "non-pigmented" cells are more tumorigenic than pigmented cells. Differentiation and tumorigenicity, although concomitant, here seem to be two independent phenomena.

A new approach to the differential diagnosis of human malignant melanomas.

In an attempt to improve the diagnosis of melanotic tumors, we have compared the diagnosis obtained by histological examination of 216 skin tumors and their metastases with that obtained by using a conjunction of four techniques: tissue culture, cytoenzymology, in situ electron microscopy and 5-S-cysteinyldopa (5-S-CD) assay. In 46 cases the final diagnosis as determined by one or more of these tests differed from the initial histological diagnosis, but was confirmed by repeat histological examination. We conclude that this method presents a valuable new approach to the differential diagnosis of human malignant melanoma.

Determination of 5-fluorouracil in plasma by GC/MS using an internal standard. Applications to pharmacokinetics.

An analytic method, sensitive to 5 ng/ml and specific, has been set up for the determination of unchanged 5-fluorouracil in human plasma, using gas chromatography--mass spectrometry. Its applications to the pharmacokinetic study of unchanged 5-Fluorouracil in man following an intravenous bolus, 8 hours infusion and oral administration confirmed the non-linearity of the kinetic model. (The clearances obtained with an infusion--from 5 to 60 1/min--are 10 to 60 times higher than the ones observed with a bolus--from 0.5 to 1.4 1/min.) The bioavailability of the oral form seems to be about 60 per cent and the absorption appears to be very fast. A attempt to search a mathematical non-linear model of the 5-Fluorouracil kinetics, using computer simulation, is presented.

[Recognition of onomatopoeias and animal cries during focalized lesions of the cerebral cortex (author's transl)].

The recognition of animal cries and their onomatopoeias was compared during the course of right or left unilateral cortical lesions. A double dissociation is noted; in left-sided affections there is a more severe deficiency to onomatopoeia than to cries, whereas in right-sided lesions the quality of the performances is reversed. These results suggest that the right hemisphere is dominant in the auditive recognition of familiar sounds, and supply information as to the linguistic value of onomatopoeias.

Ultrastructure of spontaneously differentiated human malignant melanocytes cultured from primary tumors.

Ultrastructural study of early subcultures and established lines of human malignant melanocytes derived from primary tumors showed that melanocytes passed through a phase of dedifferentiation during which they took on a fibroblast-like appearance; then they had a phase of spontaneous redifferentiation. The fibroblast-like cells were characterized by the presence of network-like organelles in their cytoplasm, and the melanocytes by melanosomes.

Cellular localization of tyrosinase in human malignant melanoma cell lines.

Human malignant melanocytes show characteristic morphologic modifications which are particularly evident in their specific organelles: melanosomes. These modifications are conserved in cell culture. The ultrastructural localization of tyrosinase, the enzyme which converts tyrosine and dopa into melanin, was determined in 13 human melanoma cell lines. The different cell lines possess 4 distribution patterns of melanin synthesis based on dopa oxidase activity. The two first pathways, which involve the Golgi apparatus, seem to differ by the amount of enzyme within this organelle. The third pathway mainly involves the smooth endoplasmic reticulum, whereas tyrosinase is visible only in vesicles in the fourth. Some cells synthesize the enzyme in the manner observed in very early embryos.

Determination of flunitrazepam, desmethylflunitrazepam and clonazepam in plasma by gas liquid chromatography with an internal standard.

A method is developed for gas-chromatographic determination of flunitrazepam, desmethylflunitrazepam and clonazepam in plasma, using methylclonazepam as internal standard. Following extraction of the benzodiazepines and their hydrolysis, the benzophenones are analyzed by gas-chromatography, with a glass column filled with 3% OV-225 on Gas Chrom Q and a 63Ni electron capture detector. The technique has a good selectivity. Its limit of sensitivity is about 0.5 ng/ml of plasma. It is suitable for investigations on pharmacokinetics and bioavailability as well as for monitoring the correlation between plasma level and therapeutic efficacy and toxicology.

Ultrastructural comparison between cultured and tumor cells of human malignant melanoma.

Tumor cells from 28 human malignant melanomas were compared by electron microscopy with cultured cells of 17 established human melanoma cell lines derived from the same types of tumors. Both tumor and cultured cells were classified into five types according to their different fine-structural characteristics. There was a correlation between the cell types observed in the tumor fragments and in the cell lines. This evidence suggests that cultured human melanoma cells retained in vitro the characteristics of the melanoma cells in vivo, from which they were derived.

[Effect of the administration of 9,12 dimethyl-1,2 benzanthracene on the plasma and pituitary levels of prolactin in rats during the estrous cycle].

Plasma determination of prolactin in the Rat after administration of DMBA has shown that the hormonal peak was practically suppressed whereas in the pituitary the concentration of prolactin 4 was much higher than the one found in the controls whatever the oestral state. The experiments reported concerning plasma and pituitary, permit us to propose several possible mechanisms for the carcinogenic action of DMBA.
