Subpopulations of T-lymphocytes in multiple myeloma.

The percentage and absolute numbers of T lymphocytes bearing Fc receptor for IgG and IgM were evaluated in 13 patients with multiple myeloma and in a group of controls of the same age range. An increase in the percentage of the TG cells was found, whereas TM cell numbers were not different from those of the controls. In order to better define the properties of the TG lymphocytes, their ability to suppress the PWM-induced B cell differentiation was tested in an in vitro experimental assay. TG cells from multiple myeloma exert a suppressor activity as the TG of the controls in this system. The possible interpretation of suppressor T cell increases in these patients is discussed.

A bovine muscle extract with anti-cirrhotic and anti-inflammatory properties in the rat.

An extract from bovine muscle, prepared by proteolytic digestion, inhibits the development of CC1(4)-induced liver cirrhosis when administered either subcutaneously or orally to the rat. It has a significant effect on survival time, liver and body weight, and gross and microscopic features, collagen, water, protein and triglyceride content of liver. It also accelerates the regression of CC1(4)-induced cirrhosis. Most of these effects were also achieved with a heat-stable or lipid fraction of the extract. The heat-stable fraction also shows anti-inflammatory activity in carrageenan- and egg white-induced edemas and in cotton pellet granuloma.

Effects of creatine administration on experimental liver fibrosis and on creatine and phosphocreatine content of rat muscle.

Creatine administration has no effect on the induction of liver cirrhosis caused by CCl4 in the rat. The muscle creatine content decreases in rats given CCl4 and this change is not modified by creatine treatment. Administered on its own it depresses the level of muscle creatine in normal rats. Creatinine administration increases the uptake of radioactive creatine by the muscle.

B lymphocytes in newborns.

B lymphocyte markers in 24 full term newborn infants were evaluated within four days after birth. It was demonstrated that the percentage of cells with surface immunoglobulins (SmIg) in the blood of these subjects was normal when the lymphocytes were stained with polyvalent as well as monospecific antisera. The percentage of cells bearing Fc receptors were found to be slightly, but not significantly, reduced. The ability of newborn lymphocytes to form rosettes with mouse erythrocytes was significantly reduced. The hypothesis that the B dependent system reaches complete maturity already at the first days after birth is discussed.

Cytochemical study in human lymphocyte subpopulations as defined by Fc receptors.

Three subpopulations of human T lymphocytes (TG, TM, and Tnon-G/non-M cells) were evaluated by means of six different cytochemical reactions. Elevated staining activity was demonstrated in all the populations when stained for acid phosphatase (AcP), beta-glucuronidase (BG) and N-acetyl beta-glucosaminidase (NABG). While the majority of TM cells displayed an intense acid alpha-naphthyl acetate esterase (ANAE) positivity, only a smaller proportion of the Tnon-G/non-M exhibited a positive reaction and a much lower activity was found in the TG group. TM cells were also alpha-naphthyl acetate esterase (NAE)-and alpha-naphthyl butyrate esterase (NBE)-positive, showing a dot-like pattern. These reactions were present in a lower proportion in the Tnon-G/non-M population and nearly completely absent in the TG cells. The present study identifies different cytochemical phenotypes for the different T-lymphocyte subsets and the analysis of these phenotypes may indicate that the TM is the most cytochemically differentiated subpopulation.

5'-Nucleotidase activity in liver homogenates of rats treated with CCl4, colchicine, cycloheximide, emetine, ethanol, ethionine and 5-fluorotryptophan.

5'-Nucleotidase activity an enzyme marker of the plasma membranes, increases in female rat liver homogenates following ethionine administration, while homogenates from males show no changes. Treatment with CCl4, colchicine, cycloheximide, emetine, ethanol and 5-fluorotryptophan does not significantly modify the 5'-nucleotidase activity of liver homogenates of either female or male rats.

[Properties of human lymphocytes binding to mouse red blood cells].

Certain properties of the bond between lymphocytes and mouse red blood cells were studied. The turnover of the receptor on the lymphocytes of twelve healthy subjects was evaluated. After two hours of incubation in a medium devoid of additives the receptor is released and its resynthesis is completed in twenty-four hours. In experiments employing proteolytic enzymes inhibition of rosette formation is observed, whereas neuraminidase, under the same experimental conditions, significantly increases the number of rosette forming cells. The percentage of mouse rosette forming cells in nineteen patients affected with chronic lymphatic leukaemia was then evaluated (17 with type B and 2 with type T). On the basis of these cases and other recently published data, an ontogenetic explanation for the appearance of this receptor is proposed.

Adhesion of enteropathogenic bacteria to cells in tissue culture.

Adhesion to the intestinal mucosa is the primary determinant of virulence among bacteria causing intestinal disease. Most current test systems involve the use of animal or foetal tissue. This paper illustrates the ability of enteropathogenic bacteria to adhere to mammalian cells. The test described enables considerable differences in adherence to be demonstrated between bacterial strains.

Changes in serum proteins and in major liver constituents following carbon tetrachloride and a high-fat choline-free diet in rats.

Both carbon tetrachloride and a high-fat, low-protein choline-free diet cause hepatomegaly in rats due to an accumulation of both lipids and proteins with a concomitant increase in deoxyribonucleic acid content. CCl4 causes a decline in hepatic glycogen. Serum albumin level is decreased in CCl4-treated rats and increased in those fed the experimental diet.

The influence of cycloheximide and chloramphenicol on rat serum proteins.

The effects of cycloheximide (CHM) and chloramphenicol (CAP) on rat serum proteins and on their electrophoretic patterns have been investigated. It has been shown that total serum protein is decreased by both treatments. CHM causes a fall in transferrin and alpha2(3-8) globulin content, while CAP reduces only alpha2(3-8) globulins. Different microscopic features of hepatic steatosis are described with both drugs. The results are briefly discussed.
