Superoxide anions and other components of human renal adenocarcinoma.

The levels of superoxide anion production, cytochrome P450, ornithine decarboxylase(E.C.4.1.1.17), catalase(E.C.1.11.1.6), deoxyribonucleic acid, ribonucleic acid and protein have been studied in human kidney and renal clear-cell adenocarcinoma tissues. The levels of superoxide anion production, ornithine decarboxylase, catalase and ribonucleic acid in the tumor tissue are very different from those in the kidney.

Temporal changes in activities of enzymes reducing ring-A of progesterone in the fetus and placenta of the rat.

The development of delta4-5alpha-reductase and 3alpha- and 3beta-hydroxysteroid dehydrogenases in rat fetuses and placentas was detected by the production of specific metabolites during in vitro incubation with progesterone. The metabolites, 5alpha-pregnane-3,20-dione, 3alpha-hydroxy-5alpha-pregnan-20-one, and 3beta-hydroxy-5alpha-pregnan-20-one, were identified by isolation and purification by thin-layer chromatography and reverse isotope dilution, followed by recrystallization of the free compound and a derivative. The delta4-5alpha-reductase and the 3beta-hydroxysteroid dehydrogenase are associated with the particulate fraction. However, 3alpha-hydroxysteroid dehydrogenase is a soluble enzyme present in the supernatant fraction of homogenized placentas and fetuses. The activities of these enzymes were greater in the placenta than in the fetus. As pregnancy progresses, the activities increase in the fetus, but diminish in the placenta. Although the placenta secretes only marginal amounts of progesterone, it has a notable capacity to metabolize progesterone to 5alpha-reduced metabolites. The fetus has the ability to utilize progesterone as early as day 13, and its potential to convert progesterone into ring-A reduced products increases during gestation.

Steroid metabolism in rat submaxillary glands.

The ability of rat submaxillary gland to metabolize C21 steroids was studied by culturing explants with labeled progesterone, pregnenolone and 17 alpha-hydroxyprogesterone. The metabolites isolated were identified by paper chromatography, thin layer chromatography and combined gas liquid chromatography mass spectrometry. The radiochemical purity of certain metabolites was confirmed by recrystallization to constant specific activity. The capability of this gland to reduce C21 steroids was confirmed with the isolation of 3 alpha-hydroxy-5 alpha-pregnan-20-one and 5 alpha-pregnane-3 alpha, 20 beta-diol. The presence of delta 5, 3 beta hydroxy steroid dehydrogenase was shown by the conversion of pregnenolone to progesterone. Only in the incubations with 17 alpha-hydroxyprogesterone a compound with a molecular weight 290 was detected.

Receptor characteristics of specific estrogen binding in the renal adenocarcinoma of the golden hamster.

Linear sucrose gradient analyses reveal that all estrogen-induced and -dependent primary renal tumor cytosols examined contain an 8 S and variable amounts of 4 S receptor in low ionic buffer concentrations. Similar results were obtained with extracts of primary metastases of these tumors. Sucrose gradients containing high salt (0.4 M KCl) convert the 8 S receptor in both the hamster renal tumor and uterus to a 4 to 5 S complex. Scatchard plot analysis reveals that the renal tumor cytosol estradiol-receptor complex has a Ka of 1.7 X 10(9) M-1 and 9.2 X 10(-10) M binding sites. Competition for the tritiated 17beta-estradiol binding sites in the renal tumor was similar to that in the uterus with respect to estrogenic compounds. Nonestrogenic steroids exhibited minimal competition at the same concentrations or higher. Substitution in the ring structure, particularly in position 3 of the phenolic A-ring, resulted in a considerable loss in the ability of such compounds to compete for these receptors. Aniestrogens were effective competitors for these estrogen receptors only at higher concentrations relative to the tritiated estradiol.

Preparation of prostatic plasma membranes Distribution of (Na-+, K-+)-ATPase and Mg-2+-ATPase in the rat ventral prostate.

1. A method is described for the isolation of a plasma membrane fraction from rat ventral prostate. This fraction is greatly enriched in (Na-", K-+)-ATPase but also contains a small amount of Mg-2+-ATPase and 5'-nucleotidase activities. 2. The activity ratio (Mg-2+ plus Na-+ plus K-+/Mg-2+ or Mg-+ plus Na-+) of the plasma membrane (Na-+, K-+)-ATPase was 1.7. The enzyme system specifically requires ATP as substrate and is inhibited by Ca-2+ or ouabain.

The synthesis of prostaglandin F by human endometrium in organ culture.

Slices of human endometrium obtained from hysterectomy specimens were cultured for 48 hours in an organ culture medium supplemented with ethanol (control, vehicle), 17-beta-estradiol (.5 mug/ml), or progesterone (.5 mug/ml). Uncultured endometrial tissue, cultured tissues, and the media were assayed for prostaglandin F (PGF) by a radioimmunoassay technique. Hematoxylin and eosin and periodic acid-Schiff stain histologic controls were done on all tissues. The concentrations of PGF in picograms/milligram, corrected for percent recovery, in the differently treated tissues were: preculture 298; culture control 2210; estrogen-treated 2680; progesterone-treated 1260. All differences except those between estrogen and control (p greater than .10) and progesterone and control (p less than .10) are significant at the p = .02 level or better. Progesterone appears to inhibit PGF synthesis which occurs during in vitro culture of human endometrium; estrogen tended to increase PGF synthesis in this system.

Biochemical comparison of estrogen receptors of the hamster hypothalamus and uterus.

Comparison of the soluble estradiol receptor proteins in the hypothalamus and uterus of the golden Syrian hamster revealed that both have sedimentation coefficients of 8S in a low ionic strength buffer and migrate similarly on analytical disc-gel electrophoresis in both 5% and 7% acrylamide gels. The competition of [3H]-17beta-estradiol binding by unlabeled estrogenic as well as nonestrogenic compounds was similar with the receptors present in the two tissues. The affinity constants for the hypothalamic and uterine receptors were 4.3 X 10-9M-1 and 1.6 X 10-10M-1, respectively; the number of binding sites when expressed on an equivalent protein basis (i.e. 1 mg/ml) was 0.18 X 10-10M and 3.1 X 10-10M, respectively. Identical amounts of soluble receptors with similar sedimentation characteristics were found in hypothalami from male and female animals gonadectomized 64 h prior to use. A titration of binding sites in the uteri of intact and ovariectomized animals revealed decreased binding in the supernatant preparation from intact animals, probably due to the presence of bound endogenous unlabeled hormone.

Junctional specialization in estrogen-induced renal adenocarcinomas of the golden hamster.

Estrogen-dependent renal adenocarcinoma and normal proximal tubules of the hamster kidney exhibit junctional differences. Although both cell types possess gap junctions, the neoplastic cells have in addition a cytoplasmic configuration of gap-junctional membrane (annular nexuses) not found in the kidneys of untreated or estrogenized hamsters or in the nontumorous kidney adjacent to the adenocarcinoma. The presence of these unique structures in the renal tumor and its abdominal metastases was demonstrated by electron microscopy with the use of lanthanum impregnation. A possible correlation between these structures and the estrogen sensitivity of the kidney neoplasm is made.
