Engineering the monomer composition of polyhydroxyalkanoates synthesized in Saccharomyces cerevisiae.

Polyhydroxyalkanoates (PHAs) have received considerable interest as renewable-resource-based, biodegradable, and biocompatible plastics with a wide range of potential applications. We have engineered the synthesis of PHA polymers composed of monomers ranging from 4 to 14 carbon atoms in either the cytosol or the peroxisome of Saccharomyces cerevisiae by harnessing intermediates of fatty acid metabolism. Cytosolic PHA production was supported by establishing in the cytosol critical beta-oxidation chemistries which are found natively in peroxisomes. This platform was utilized to supply medium-chain (C6 to C14) PHA precursors from both fatty acid degradation and synthesis to a cytosolically expressed medium-chain-length (mcl) polymerase from Pseudomonas oleovorans. Synthesis of short-chain-length PHAs (scl-PHAs) was established in the peroxisome of a wild-type yeast strain by targeting the Ralstonia eutropha scl polymerase to the peroxisome. This strain, harboring a peroxisomally targeted scl-PHA synthase, accumulated PHA up to approximately 7% of its cell dry weight. These results indicate (i) that S. cerevisiae expressing a cytosolic mcl-PHA polymerase or a peroxisomal scl-PHA synthase can use the 3-hydroxyacyl coenzyme A intermediates from fatty acid metabolism to synthesize PHAs and (ii) that fatty acid degradation is also possible in the cytosol as beta-oxidation might not be confined only to the peroxisomes. Polymers of even-numbered, odd-numbered, or a combination of even- and odd-numbered monomers can be controlled by feeding the appropriate substrates. This ability should permit the rational design and synthesis of polymers with desired material properties.

Clinical significance of the coronary calcification score by multidetector row computed tomography for the evaluation of coronary stenosis in Japanese patients.

BACKGROUND: The coronary artery calcification (CAC) score measured by multidetector row computed tomography (MDCT) has emerged as a marker for predicting coronary artery disease (CAD). To evaluate the clinical significance of the CAC score, coronary artery stenosis as assessed by coronary angiography (CAG) was compared with the CAC score determined by MDCT, risk factors and medications. METHODS AND RESULTS: Subjects included 374 consecutive patients who underwent ECG-gate CT angiography using MDCT. The accuracy in patients with a CAC score >or=400 was 84%, and significantly lower than that in patients with a CAC score =0. In addition 92 patients (68 males, 24 females; mean age, 63+/-11 years) who underwent both MDCT and CAG within a 1-month period were selected for further investigation. Patients with significant coronary stenosis had a significantly higher CAC score than those without stenosis. In addition, a higher number of stenosed vessels was associated with a higher CAC score. The subjects were divided into 3 groups according to the CAC score: low (0-12), intermediate (13-444) and high (>or=445). The CAC score was significantly associated with age, and plasma levels of total cholesterol and hemoglobinA1c, and logistic regression analysis revealed that significant coronary stenosis as assessed by CAG was most closely correlated with the CAC score (p=0.03). CONCLUSIONS: The CAC score determined by MDCT can predict CAD independent of other factors, such as age, metabolic diseases and medications, when coronary stenosis can not be diagnosed because of severe calcification.

New approach to pulmonary vein isolation for atrial fibrillation using a multielectrode basket catheter.

BACKGROUND: Pulmonary vein (PV) isolation using a circular catheter creates an entrance block from the left atrium (LA) to the PV, which eliminates paroxysmal atrial fibrillation (PAF). A new approach to PV isolation during distal PV pacing is to use a basket catheter. METHODS AND RESULTS: Fifty consecutive patients with PAF underwent basket-catheter-guided PV isolation. PV pacing was performed from the distal electrode pair of the basket catheter. The exit breakthrough point was targeted for segmental PV isolation. The endpoint was the elimination of bidirectional PV - LA conduction. A repeat ablation procedure was performed in 12 of 14 patients who had recurrence of AF. The recovery of PV -LA conduction was noted in 24 of the 48 PVs, and 5 PVs (21%) had unidirectional block. At 12 months, 80% of patients were free of AF without antiarrhythmic drugs. No PV stenosis >50% was detected at 12 months after the procedure. CONCLUSIONS: This new approach for PV isolation during distal PV pacing using a basket catheter is useful for confirming bidirectional PV - LA conduction block. PV isolation that creates not only an entrance block but also an exit block at the PV - LA junction may be required to cure paroxysmal AF.

Angiotensin II type 1 receptor blockers do not promote coronary collateral circulation in patients with coronary artery disease.

We previously reported that angiotensin-converting enzyme inhibitors (ACE-Is) promote collateral circulation in patients with coronary artery disease (CAD). There have been many reports on the beneficial effects of angiotensin II type 1 receptor blockers (ARBs) on the cardiac microvasculature. Therefore, the following studies were performed to evaluate the association between treatment with an ARB and the enhancement of coronary collateral circulation as assessed by the Rentrop Score (RS) (Study 1) and to compare these results to those obtained with an ACE-I (Study 2). The subjects were 456 patients with angina who underwent coronary angiography. Study 1: Those who had one (1-V), two (2-V) or three significantly stenosed vessels (3-V) and who received only an ARB without any other anti-hypertensive medication were defined as the ARB group (n=81), and age-, sex- and body mass index-matched subjects (n=146) were selected as a comparative group. There were no significant differences in the percentage of patients with RS>or=1 between the two groups. Study 2: Those who received an ACE-I as the only anti-hypertensive treatment were defined as the ACE-I group (n=67), which was matched to the ARB group in Study 1. The percentage of patients with RS>or=1 in the ACE-I group was significantly higher than that in the ARB group as assessed by a Cochran-Mantel-Haenszel analysis. In addition, patients with 3-V disease who were treated with an ACE-I, but not an ARB, were most likely (odds ratio [confidence Interval]): 27.7 [4.8-161.0]) to show enhanced collateral circulation, as assessed by a multiple logistic regression analysis. These results suggest that treatment with an ACE-I, but not treatment with an ARB, was associated with the enhancement of collateral circulation in patients with CAD.

[Different expression and significance of VEGF and PCNA in aorta and pulmonary artery smooth muscle cells of rats with hypoxia pulmonary hypertension].

AIM: To study the expression of vascular endothelial growth factor (VEGF) and proliferating cell nuclear antigen(PCNA) in aorta and pulmonary artery smooth muscle cells of rats with hypoxia pulmonary hypertension(HPH). METHODS: The rat chronically HPH models were set up in the hypobaric hypoxia cabin. The experimental rats were divided into 3 groups: control group (rearing in a normoxia environment), two hypoxia groups (oxygen-deficient time being 2 weeks and 3 weeks, respectively). Expression of VEGF and PCNA were detected by immunohistochemical staining and image analysis. RESULTS: There was VEGF expression in vascular smooth muscle cells of aorta, pulmonary artery trunk and pulmonary arteriole in control group. In two hypoxia groups VEGF expression in vascular smooth muscle cells of pulmonary artery, trunk and pulmonary arteriole increased significantly, while no difference was found in smooth muscle cells of aorta. Expression of PCNA was very little in vascular smooth muscle cells of aorta, pulmonary artery trunk and pulmonary arteriole in control group. In two hypoxia groups, the PCNA expression increased only in vascular smooth muscle cells of pulmonary arteriole; there was no difference of PCNA expression in vascular smooth muscle cells of aorta and pulmonary artery trunk between hypoxia groups and control group. CONCLUSION: There is difference of VEGF expression in pulmonary artery trunk and aorta smooth muscle cells during the formation of chronic HPH, suggesting that VEGF expression may play very important role in the formation of chronic HPH.

Study on the genetic mutations of 17 alpha-hydroxylase/17,20-lyase deficiency in Chinese patients.

OBJECTIVE: To investigate the CYP17A1 gene mutations in Chinese patients with 17 alpha-hydroxylase/17, 20-lyase deficiency. METHODS: Clinical data were retrospectively analyzed. The CYP17A1 gene mutations were detected in 5 cases with 17 alpha-hydroxylase/17, 20-lyase deficiency and their relatives. The genomic DNA of the patients was isolated from whole blood. Seven pairs of primers were used to amplify eight exons and exon-intron boundaries of the CYP17A1 gene. The amplified PCR products were purified by agarose gel and then directly sequenced. In order to confirm the DNA sequences of different alleles, some fragments were inserted into pMD 18-T vector and then sequenced. Sequencing results were compared to the established human CYP17A1 sequence. RESULTS: Briefly, we found 2 kinds of compound mutations, of which were: (1) 6436-6438(TAC-->AA), causing amino acid Y329K, 418X; (2) 6531-6532(GC-->A), causing amino acid L361F, 418X. Among the five cases, four were homozygous for 6436-6438(TAC-->AA), whereas one was compound heterozygous for 6436-6438(TAC-->AA)/6531-6532(GC-->A). The clinical characteristics of 5 cases were all completely combined defects of 17 alpha-hydroxylase/17, 20-lyase, and they all carried two alleles of CYP17A1 gene mutations that all shifted the reading frame and resulted in truncated protein which lack of the activity center site of P450C17, of which corresponding with their clinical feature. CONCLUSION: Nine alleles have the mutation of 6436-6438(TAC-->AA), accounting for 90% of total alleles (9/10). That suggests this kind of mutation may have racial specificity. More study should be done to have better understanding of the function of the truncated P450C17 enzymes.

Enhanced expression of halp gene confers cellular resistance to H2O2 induced senescence.

OBJECTIVE: To investigate the H2O2-induced expression of human histone acetyltransferase-like protein (hALP), a telomerase regulation-associated gene, and its effects on the stress-triggered cellular senescence. METHODS: The induced expression of hALP was measured by semi-quantitative RT-PCR and immunofluorescent histochemistry after treatment of HeLa cells by H2O2. The effects of hALP expression on cellular responses to H2O2 were analyzed by MTT, flowcytometry, and SA-beta-gal staining, respectively. RESULTS: hALP mRNA could be dose-dependently induced by treatments of 0.2-1.6 mmol/L H2O2, and the induction could be observed after 6 hours and kept for 36 hours in the presence of 0.4 mmol/L H2O2. Meanwhile, the immunofluorescent staining showed marked stronger nuclear intensity of hALP protein in H2O2-treated HeLa cells. In the treatment of H2O2, the ectopic expression of hALP enhanced continuous growth and overcame G2/M arrest as well as decreased senescence-associated beta-gal staining. On the contrary, the transfected clones with antisense or blank vector and original He-La cells presented growth suppression, G2/M delay and higher percentage of SA-beta-gal activities in the presence of H2O2. CONCLUSIONS: The expression of hALP could be up-regulated by treatment of H2O2, and elevated expression could enhance cellular resistance to H2O2-induced cellular senescence. The data might be of references to elucidation of basic biological function of hALP gene and its associated telomerase activity.

[Inhibitory effect of p53 gene on telomerase activity and proliferative activity in salivary adenoid cystic carcinoma cells].

OBJECTIVE: To evaluate the inhibitory effect of p53 gene on salivary adenoid cystic carcinoma (SACC) cells. METHODS: Adenoviral vector pDeltaE1-p53 was constructed and transfected into SACC-83 cells. The enhanced p53 expression was measured by reverse transcription polymerase chain reaction (RT-PCR), and the effects of transfected p53 on SACC-83 cells were analyzed by TRAP-PCR-ELISA, luciferase reporter, flow cytometry (FCM), soft agar assay and tumorigenicity test. RESULTS: The expression of p53 gene in SACC-83 cells was increased after introduction of pDeltaE1-p53. The telomerase activity and the transcriptional activity of hTERT promoter were inhibited. The cells cycles of transfected SACC-83 were arrested in G(1) phase and the rate of colony-formation was decreased, and similarly the tumorigenicity in nude mice was also decreased. CONCLUSIONS: The introduction of wild-type p53 by adenoviral vector could suppress the telomerase activity and malignant phenotypes of salivary adenoid cystic carcinoma cells.

[Effect of Rho kinase on hypoxia stimulated proliferation of pulmonary arterial smooth muscle cells].

OBJECTIVE: To study whether Rho kinase is involved in the proliferation of pulmonary arterial smooth muscle cells (PASMCs) induced by hypoxia. METHODS: Rat's PASMCs were isolated and cultured. Methyl thiazolyl tetrazolium (MTT) was used to determine the growth of cells. The cell cycle was analyzed by flow cytometry. Western blot was used to assess the expression of Rho kinase. RESULTS: The results of MTT showed that the number of PASMCs was increased after exposure to low oxygen tension for 12 hours compared with PASMCs in normoxia. After exposure of PASMCs to hypoxia for 24 hours, the cellular proliferation peaked, and was higher than that of PASMCs exposed to normal oxygen tension, hypoxia and Y27632 (Rho kinase inhibitor) groups, then it declined when exposed for 48 hours. Analysis of cell cycle indicated that the ability of cell proliferation increased significantly in PASMCs exposed to hypoxia compared with normoxia. Rho kinase level was higher in PASMCs exposed to hypoxia compared with normoxia. CONCLUSION: Hypoxia stimulates proliferation of PASMCs and promotes PASMCs into the phase of mitosis. The expression of Rho kinase is increased in PASMCs that are exposed to hypoxia, and it shows that Rho kinase is activated by hypoxia in PASMCs. Rho kinase maybe involve in the proliferation of PASMCs induced by hypoxia.

Protein expression analysis of Halobacillus dabanensis D-8T subjected to salt shock.

To investigate the mechanism of salt tolerance of gram-positive moderately halophilic bacteria, two-dimensional gel electrophoresis (2-D PAGE) was employed to achieve high resolution maps of proteins of Halobacillus dabanensis D-8T. Approximately 700 spots of proteins were identified from these 2-D PAGE maps. The majority of these proteins had molecular weights between 17.5 and 66 kDa, and most of them were distributed between the isoelectric points (pI) 4.0 and 5.9. Some protein spots were distributed in the more acidic region of the 2-D gel (pI <4.0). This pattern indicated that a number of proteins in the strain D-8T are acidic. To understand the adaptation mechanisms of moderately halophilic bacteria in response to sudden environmental changes, differential protein profiles of this strain were investigated by 2-D PAGE and Imagemaster 2D Platinum software after the cells were subjected to salt shock of 1 to 25% salinity for 5 and 50 min. Analysis showed 59 proteins with an altered level of expression as the result of the exposure to salt shock. Eighteen proteins had increased expression, 8 proteins were induced, and the expression of 33 proteins was down-regulated. Eight of the up-regulated proteins were identified using MALDI-TOF/MS and MASCOT, and were similar to proteins involved in signal transduction, proteins participating in energy metabolism pathways and proteins involved in stress.

[Experimental studies of carcino-embryonic antigen immunosensor based on piezoelectric resonate technology of quartz crystal].

In order to construct a new type of piezoelectric quartz immunosensor for the determination of carcino-embryonic antigen (CEA), the sensor detection pools were consisted of plastic loops and crystals that were 10 MHz quartz AT-cut with gold coated electrodes and the immobilization of the monoclonal antibody against CEA onto gold electrode surface of the quartz crystal was accomplished via Thiol method. Then the immunosensors were used in clinical laboratory. The experimental results showed that the piezoelectric immunosensor had good response to CEA, the frequency shifts were linearly dependent on CEA concentration in the range of 1.56 to approximately 50.00 ng/ml, other antigens such as alpha fetoprotein (AFP), prostate specific antigen (PSA), human chorionic gonadotropin (hCG) did not interfere with the sensor's response. The results obtained from this method were in satisfactory accordance with those obtained by radio immunoassay(P>0.05), The correlation coefficient was 0.90. Piezoelectric immunosensor for determination of CEA has the advantage of high sensitivity, high specificity, unnecessary labeling, simple performing, rapid analysis, low cost, real-time detection and repeated use, etc. It can be used for detecting serum CEA in clinical laboratory.

[Effects of dusts on workers' health in enzyme production plants].

OBJECTIVE: To assess the health effects of dusts in enzyme production plants. METHODS: The concentration of enzyme-containing dusts, the enzyme and the wheat-containing dusts and their health effects on workers were investigated in three enzyme production plants. Air samples were collected by high volume sampler and personal sampler. Total dust was weighed and its content of enzyme was analyzed by enzyme activity method. Health effects were assessed by the questionnaire, the pulmonary function and the skin prick test. RESULTS: It was found that the geometric mean of enzyme-containing dust was 8.91 mg/m(3), the industrial enzyme was 1.68 mg/m(3), and the wheat-containing dust was 6.93 mg/m(3). The enzyme-containing dust higher than 20 mg/m(3) caused eye symptoms. The wheat-containing dust at 6.93 mg/m(3) might result in skin and nose symptoms. The sensitization could be observed in enzyme and wheat exposed workers. CONCLUSION: Adverse effects may arise for the health of the workers if the concentration of enzyme-containing dusts and the wheat-containing dusts is up to a certain limit and it is necessary to make the limit of these indexes.

Experimental study on plasticity of proliferated neural stem cells in adult rats after cerebral infarction.

OBJECTIVE: To investigate whether there is endogenous neural stem cell proliferation and whether these proliferated neural stem cells represent neural plasticity in the adult rats after cerebral infarction. METHODS: Cerebral infarction models of rats were established and the dynamic expression of bromodeoxyuridine (BrdU), BrdU/polysialylated neural cell adhesion molecule (PSA-NCAM) were determined by immunohistochemistry and immunofluorescence staining. BrdU was used to mark dividing neural stem cells. PSA-NCAM was used to mark the plasticity of neural stem cells. RESULTS: Compared with controls, the number of BrdU-positive cells in the subventricular zone (SVZ) and hippocampus increased significantly at 1st day after cerebral infarction (P < 0.05), reached maximum at 7th day, decreased markedly at 14th day, but it was still elevated compared with that of the controls (P < 0.05). The number of BrdU-labeled with PSA-NCAM-positive cells increased significantly at 7th day (P < 0.05), reached maximum at 14th day, markedly decreased at 28th day, but it was still elevated compared with that of the controls (P < 0.05). It was equal to 60% of the number of BrdU-positive cells in the same period. CONCLUSION: Cerebral infarction may stimulate the proliferation of endogenous neural stem cells in situ and most proliferated neural stem cells represent neural plasticity.

[Influence of temperature on hydrolysis and acidogenesis of kitchen wastes in two-phase anaerobic digestion].

The experiment evaluated the solubilization rate and VFA production rate of kitchen wastes with controlled-temperature at 25 degrees C, 37 degrees C, 40 degrees C and 50 degrees C, and subsequently investigated the lactic acid production and constituents of individual organic acids at various temperature levels. Results show that the increase of temperature when below 37 degrees C may improve both hydrolysis and acidogenesis rates and acidogenesis rate decreases while hydrolysis rate increases above 37 degrees C. The maximum VFA concentration of 34.4 g x L(-1) was achieved at 37 degrees C and a higher hydrolysis rate of 82% was obtained at 50 degrees C. Formic and acetic acid were predominant VFA, while propionic and butyric acid took a relatively little part. And lactic acid concentration remained relatively high. Accroding to the experiment an optimum temperature to optimize both hydrolysis and acidification is 37 degrees C.

[Microbiochemical analyzer based on continuous spectrum and its test for clinic use].

A microbiochemical analyzer based on continuous spectrum and its test for clinic use are introduced. The principle of splitting behind was adopted to design the micro biochemical analyzer, which has the characteristics of little disorder light, real time collection of continuous spectrum signal (340-770 nm), small volume, light weight, small wastage of sample and reagent and rapidness and direct view of check. It has wide application in the emergency treatment, middling and small hospitals, rescue of battleground, and medicinal research. Representative items for clinic test were selected, such as uric acid, total cholesterin and albumin in our experiments. Comparative test and analysis of the daily check-up samples were made respectively by our microbiochemical analyzer and by the standard one of the hospital (Olympus AU2700, RT-1904C, Beckman LX-20). The test results show that our micro biochemical analyzer can meet the requirement of clinic use.

Low-density lipoprotein oxidized to various degrees activates ERK1/2 through Lox-1.

Although the standard procedure for preparing extensively oxidized low-density lipoprotein (Ox-LDL) is to incubate it with 10muM CuSO(4) at 37 degrees C for 24h, it is not well known how important the degree of oxidation of LDL is for inducing cell signaling. Since Lox-1 (an Ox-LDL receptor) contributes to cell proliferation through extracellular-signal-regulated kinase (ERK)1/2 activation and subsequently induces plaque growth, we analyzed ERK activity using LDL with various degrees of oxidation, from minimally Ox-LDL, which is mainly in human plasma, to extensively Ox-LDL using capillary electrophoresis (cITP). The cITP was a suitable tool for evaluating the degree of oxidation of LDL for analyzing the optimal conditions for the oxidation of LDL by CuSO(4) to obtain LDL that was oxidized to a degree comparable to that in human plasma. In addition, both minimally and extensively Ox-LDL induced similar levels of ERK1/2 activation through Lox-1 in human coronary artery smooth muscle cells. These results indicate that both minimally and extensively Ox-LDL may be important for the progression of plaque growth through Lox-1. Since most previous reports have provided data only using extensively Ox-LDL, a re-evaluation is needed to analyze several signals that use LDL which has been oxidized to various degrees.

Rate of iron transfer through the horse spleen ferritin shell determined by the rate of formation of Prussian Blue and Fe-desferrioxamine within the ferritin cavity.

Iron (2+ and 3+) is believed to transfer through the three-fold channels in the ferritin shell during iron deposition and release in animal ferritins. However, the rate of iron transit in and out through these channels has not been reported. The recent synthesis of [Fe(CN)6]3-, Prussian Blue (PB) and desferrioxamine (DES) all trapped within the horse spleen ferritin (HoSF) interior makes these measurements feasible. We report the rate of Fe2+ penetrating into the ferritin interior by adding external Fe2+ to [Fe(CN)6]3- encapsulated in the HoSF interior and measuring the rate of formation of the resulting encapsulated PB. The rate at which Fe2+ reacts with [Fe(CN)6]3- in the HoSF interior is much slower than the formation of free PB in solution and is proceeded by a lag period. We assume this lag period and the difference in rate represent the transfer of Fe2+ through the HoSF protein shell. The calculated diffusion coefficient, D approximately 5.8x10(-20) m2/s corresponds to the measured lag time of 10-20 s before PB forms within the HoSF interior. The activation energy for Fe2+ transfer from the outside solution through the protein shell was determined to be 52.9 kJ/mol by conducting the reactions at 10 approximately 40 degrees C. The reaction of Fe3+ with encapsulated [Fe(CN)6]4- also readily forms PB in the HoSF interior, but the rate is faster than the corresponding Fe2+ reaction. The rate for Fe3+ transfer through the ferritin shell was confirmed by measuring the rate of the formation of Fe-DES inside HoSF and an activation energy of 58.4 kJ/mol was determined. An attempt was made to determine the rate of iron (2+ and 3+) transit out from the ferritin interior by adding excess bipyridine or DES to PB trapped within the HoSF interior. However, the reactions are slow and occur at almost identical rates for free and HoSF-encapsulated PB, indicating that the transfer of iron from the interior through the protein shell is faster than the rate-limiting step of PB dissociation. The method described in this work presents a novel way of determining the rate of transfer of iron and possibly other small molecules through the ferritin shell.

A critical role for calponin 2 in vascular development.

Calponin 2 (h2 calponin, CNN2) is an actin-binding protein implicated in cytoskeletal organization. We have found that the expression of calponin 2 is relatively restricted to vasculature from 16 to 30 h post-fertilization during zebrafish (Danio rerio) development. Forty-eight hours after injecting antisense morpholino oligos against calponin 2 into embryos at the 1-4-cell stage, zebrafish demonstrated various cardiovascular defects, including sluggish axial and head circulation, absence of circulation in intersegmental vessels and in the dorsal longitudinal anastomotic vessel, enlarged cerebral ventricles, and pericardial edema, in addition to an excess bending, spiraling tail and twisting of the caudal fin. Knockdown of calponin 2 in the Tg(fli1:EGFP)(y1) zebrafish line (in which a fli1 promoter drives vascular-specific enhanced green fluorescent protein expression) indicated that diminished calponin 2 expression blocked the proper migration of endothelial cells during formation of intersegmental vessels. In vitro studies showed that basic fibroblast growth factor-induced human umbilical vein endothelial cell migration was down-regulated by knockdown of calponin 2 expression using an antisense adenovirus, and overexpression of calponin 2 enhanced migration and hastened wound healing. These events were correlated with activation of mitogen-activated protein kinase; moreover, inhibition of this pathway blocked the promigratory effect of calponin 2. Collectively, these data suggest that calponin 2 plays an important role in the migration of endothelial cells both in vivo and in vitro and that its expression is critical for proper vascular development.
