Oxidative electrochemical switching in dithienylcyclopentenes, part 1: effect of electronic perturbation on the efficiency and direction of molecular switching.

The electro- and spectroelectrochemical properties of dithienylhexahydro- and dithienyhexafluorocyclopentenes are reported. The large effect of variation in the central cyclopentene moieties on the redox properties of the dithienylcyclopentenes is in striking contrast to the minor effect on their photochemical properties. The electronic properties of the oxidised compounds in the +1 and +2 oxidation state are reported, and the possibility of electrochemical cyclisation and cycloreversion were explored by UV/Vis spectroelectrochemistry. The efficiency of electrochemical switching is found to be dependent both on the central cyclopentene unit and on the nature of the substituents at C5 of the thienyl rings. For the hexahydrocyclopentene-based compounds oxidative ring closure of the ring-open form is observed, while for the hexafluorocyclopentene-based compounds oxidative ring opening of the ring-closed form is observed. However, the introduction of electroactive groups such as methoxyphenyl allows oxidative ring closure to occur in the hexafluoro compounds. The effect of electrolyte, solvent and temperature on the spectroelectrochemical properties were examined, and the switching process was found to be sensitive to the donor properties of the solvent/electrolyte system employed. In addition, thermally activated reversible isomerisation of the dicationic closed form was observed. The driving force for electrochemical ring opening and closure appears to be dependent on the relative stabilisation of the dicationic ring-open and ring-closed states. This study provides insight into the factors which determine the direction of cyclisation.

Oxidative electrochemical switching in dithienylcyclopentenes, Part 2: effect of substitution and asymmetry on the efficiency and direction of molecular switching and redox stability.

The electrochemical and spectroelectrochemical properties of a series of C5-substituted dithienylhexahydro- and dithienylhexafluorocyclopentenes are reported. The effect of substitution at C5 of the thienyl moiety on the redox properties is quite dramatic, in contrast to the effect on their photochemical properties. The efficiency of electrochemical switching is dependent both on the central cyclopentene unit and on the nature of the substituents, whereby electron-donating moieties favour oxidative electrochemical ring-closure and vice versa. Asymmetrically substituted dithienylcyclopentenes were investigated to explore the ring-closure process in more detail. The results indicate that electrochemically induced ring-closure occurs via the monocation of the open form. In the presence of electroactive groups at C5 of the thienyl ring (e.g., methoxyphenyl) initial oxidation of these groups is followed by intermolecular electron transfer, which drives ring-closure of the open forms.

Dynamic chiral selection and amplification using photoresponsive organogelators.

The ability to select, amplify, and lock dynamic equilibria is of great interest into understanding and applying chiral systems in Nature. The dynamic equilibrium between P and M helicity of a nonchiral diarylethene switch 3 could selectively be coaggregated in the gel state by complementary chiral switches 1 and 2 (that itself is also subjected to the same equilibrium between P and M helicity). Enantiomeric excess as high as 94% was observed during this dual task for 1 and 2 (arranging itself and 3 in only one conformation during aggregation). Interestingly, opposite chiral induction was observed, although the conformation of both 1 and 2 is R.

Cyclohexane-based low molecular weight hydrogelators: a chirality investigation.

Seven new 1,3,5-cyclohexyltricarboxamide-phenylalanine derivatives were synthesized in order to investigate the effect of the amino acid chirality on the gelating properties of these small molecules in water. Gelation tests have shown that enantiomerically pure homochiral 1,3,5-cyclohexyltricarboxamide-L-phenylalanine is a non-hydrogelator as it crystallizes from water, whereas the heterochiral derivatives with either two L-phenylalanine moieties and one D-phenylalanine (LLD), or vice versa (DDL), are very good hydrogelators. Concentration-dependent gel-to-sol transition-temperature (T(gs)) curves for LLD or DDL gels show a sigmoidal behaviour, which is in contrast to the logarithmic curves generally observed for gels derived from low molecular weight gelators (LMWGs). Such sigmoidal behaviour can be related to interactions between fibre bundles, which give rise to intertwined bundles of fibres. Transmission electron microscopy (TEM) images of LLD and DDL gels show a network of thin, unbranched, fibre bundles with diameters of 20 nm. Right-handed twisted fibre bundles are present in the LLD gel, whereas left-handed structures can be found in the DDL gel. Each bundle of fibres consists of a finite number of primary fibres. Gels consisting of mixtures of gelators, LLD and DDL, and nongelators (LLL or DDD) were investigated by means of T(gs) measurements, CD spectroscopy and TEM. Results show that the incorporation of nongelator molecules into gel fibres occurs; this leads to higher T(gs) values and to changes in the helicity of the fibre bundles. Furthermore, it was found that peripheral functionalization of the homochiral derivatives LLL or DDD by means of a second amino acid or a hydrophilic moiety can overcome the effect of chirality; this process in turn leads to good hydrogelators.

A light-actuated nanovalve derived from a channel protein.

Toward the realization of nanoscale device control, we report a molecular valve embedded in a membrane that can be opened by illumination with long-wavelength ultraviolet (366 nanometers) light and then resealed by visible irradiation. The valve consists of a channel protein, the mechanosensitive channel of large conductance (MscL) from Escherichia coli, modified by attachment of synthetic compounds that undergo light-induced charge separation to reversibly open and close a 3-nanometer pore. The system is compatible with a classical encapsulation system, the liposome, and external photochemical control over transport through the channel is achieved.

An iterative catalytic route to enantiopure deoxypropionate subunits: asymmetric conjugate addition of grignard reagents to alpha,beta-unsaturated thioesters.

A highly enantioselective (up to 96% ee) conjugate addition of Grignard reagents, in particular, MeMgBr, to alpha,beta-unsaturated thioesters is provided as well as its application to a diastereo- and enantioselective iterative route to syn- and anti-1,3-dimethyl arrays and deoxypropionate subunits. The versatility of the method is illustrated in the synthesis of (-)-lardolure, a multimethyl-branched insect natural pheromone.

Two-stage enzyme mediated drug release from LMWG hydrogels.

An enzymatically cleavable low molecular weight gelator-(model) drug conjugate system can be employed to effect a two-step enzyme mediated drug release, demonstrating the potential of LMWG systems for the development of drug delivery devices.

New approaches towards the synthesis of the side-chain of mycolactones A and B.

New approaches towards the synthesis of the C1'-C16' side-chain of mycolactones A and B from Mycobacterium ulcerans are reported. Chiral building block 4 (Fig. 2) with the correct stereochemistry was obtained starting from naturally occurring monosaccharides, i.e. D-glucose or L-rhamnose. The polyunsaturated moiety 3 was synthesized in only 3 steps from 2,4-dimethylfuran. The building blocks were connected through a Sonogashira coupling resulting in the fast and convergent assembly of an 8,9-dehydro analogue 2 of the side-chain of mycolactones A and B. The synthesis of 1 is at this stage hampered by the lack of a selective partial hydrogenation protocol for alkynes embedded in a conjugated system. Alternative strategies involving palladium catalyzed sp2-sp2 coupling between C7' and C8' or C9' and C10' (Fig. 1) were also explored.

Enantioselective synthesis of 2-aryl-4-piperidones via rhodium/phosphoramidite-catalyzed conjugate addition of arylboroxines.

[reaction: see text] The highly enantioselective synthesis of 2-aryl-4-piperidones by rhodium/phosphoramidite-catalyzed conjugate addition of arylboroxines to 2,3-dihydro-4-pyridones is described. Both enantiomers of a variety of products with sterically and electronically different R substituents were obtained in high isolated yield and with excellent enantioselectivity up to 99%.

cis-Dihydroxylation and epoxidation of alkenes by [Mn(2)O(RCO(2))(2)(tmtacn)(2)]: tailoring the selectivity of a highly H(2)O(2)-efficient catalyst.

The carboxylic acid promoted cis-dihydroxylation and epoxidation of alkenes catalyzed by [MnIV2O3(tmtacn)2]2+ 1 employing H2O2 as oxidant is described. The use of carboxylic acids at cocatalytic levels not only is effective in suppressing the inherent catalase activity of 1, but also enables the tuning of the catalyst's selectivity. Spectroscopic studies and X-ray analysis confirm that the control arises from the in situ formation of carboxylate-bridged dinuclear complexes, for example, 2 {[MnIII2O(CCl3CO2)2(tmtacn)2]2+} and 3 {[MnII2(OH)(CCl3CO2)2(tmtacn)2]+}, during catalysis. For the first time, the possibility to tune, through the carboxylate ligands employed, both the selectivity and activity of dinuclear Mn-based catalysts is demonstrated. To our knowledge, the system 1/2,6-dichlorobenzoic acid (up to 2000 turnover numbers for cis-cyclooctanediol) is the most active Os-free cis-dihydroxylation catalyst reported to date.

Cyclohexane bis-urea compounds for the gelation of water and aqueous solutions.

A new class of efficient hydrogelators has been developed by a simple modification of the peripheral substituents of cyclohexane bis-urea organogelators with hydrophilic hydroxy or amino functionalities. These bis-urea hydrogelators were synthesised in two or three steps using an alternative procedure to the common isocyanate method. Gelation was obtained with organic solvents, water and strongly basic aqueous solutions like 25% ammonia. Hydrogelation was found to depend on a delicate balance between the hydrophobicity of the alkyl chains, hydrophilicity of the terminal substituents and the enantiomeric purity of the compound. The hydrogels consisted of a network of fibers, in which all urea groups are involved in intermolecular hydrogen bonding. Most likely, gelation is driven by hydrophobic interactions of the methylene units, whereas hydrogen bond formation between the urea groups provides the necessary anisotropy of the aggregation and the high thermal stability of the gels.
