Partitioning apoptosis: a novel form of the execution phase of apoptosis.

Apoptosis is characterized by chromatin condensation, DNA cleavage, redistribution of phosphatidylserine, and apoptotic body formation via an actin-dependent process. We describe a novel form of the execution phase of apoptosis in human multiple myeloma cells that is morphologically and mechanistically distinct from classical apoptosis, but is caspase-dependent and inhibited by IL-6 and overexpression of Bcl-2. Electron microscopic analysis of these cells demonstrated chromatin condensation without nuclear fragmentation, and 'partitioning' of cell constituents into two components: a single, large bleb containing soluble protein and free ribosomes, and a region containing the nucleus, organelles, and RER. In some cases, the bleb separated, becoming a free vesicle exhibiting random kinetic motion. These morphologic features occurred despite inhibition of the actin and tubulin cytoskeletal systems. This novel form of apoptosis, called partitioning apoptosis, was observed in a variety of tumor cell types and in primary cells. The execution phase of apoptosis can occur in a manner that is morphologically and mechanistically distinct from classical apoptosis.

Antitumor effect of antisense ODC adenovirus on human prostate cancer cells.

Ornithine decarboxylase (ODC), the first enzyme of polyamine biosynthesis, was found to increase in cancer cells, especially prostate cancers. Some chemotherapeutic agents aimed to decrease ODC expression showed inhibitory effects on cancer cells. In this study, we examined the effect of adenoviral-transduced antisense ODC on prostate cancer cells. An adenovirus carrying antisense ODC (rAd-ODC/Ex3as) was infected to prostate cancer cells PC-3 and LNCap. Expression of ODC and concentration of polyamines in cells were determined by Western blotting and HPLC. MTT (3-(4,5-methylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide) assay was used to analyze the effect on cell growth. Cell cycle was evaluated by FCM and cellular invasion by Matrigel invasion assay. A nude mouse xenograft model was used to examine tumorigenicity. Expression of ODC in PC-3 and LNCap cells were reduced to 45 and 59%, and three polyamines were also decreased by the rAd-ODC/Ex3as treatment. Consequently, cell growth was substantially inhibited and cell cycle arrested at G1 phase. Matrigel invasion assay showed relatively low invasion. Marked suppression of tumor formation was observed in the xenograft model. This study suggests that rAd-ODC/Ex3as has the antitumor effect on the human prostate cancer cells.

The formation, role and removal of NO3-N during corona discharge in air for phenol removal.

Pulsed corona discharge carried out in the gas phase has been shown to be a low cost solution for wastewater treatment. In this study, the results show that the nitrogen dissociated during corona discharge in the gas phase formed nitric acid in solution, which resulted in a drop in solution pH. The phenol removal could be described in accordance with a first-order reaction in a basic buffer solution, but not in initial neutral and basic non-buffered solutions. This indicates that basic conditions facilitate phenol removal, whilst the formation of nitric acid in solution resulted in decrease in the phenol removal rate. This study also shows that most of the oxidants used for phenol removal came mainly from the dissociation of water molecules at the surface of the liquid phase and/or the gas phase very close to the liquid surface in discharging conditions, not from the gas dissociation in the bulk gas phase, and therefore, it is more effective to keep the solution basic than to change the component of gas in the reactor when removing phenol from water using corona discharge.

The influence of pH on hydrolysis and acidogenesis of kitchen wastes in two-phase anaerobic digestion.

Batch and semi-continuous experiments were carried out to determine the effect of pH on hydrolysis and acidogenesis of kitchen wastes in the two-phase anaerobic digestion. The results of the batch experiment showed pH adjustment could improve both hydrolysis and acidogenesis rates of kitchen wastes. Compared with pH at 5, 9 and 11, pH 7 provided an optimum working condition for anaerobic digestion of kitchen wastes. At pH 7, about 86% of the total organic carbon (TOC) and 82% of chemical oxygen demand (COD) were solubilized and the maximum volatile fatty acid (VFA) concentration of 36 g l(-1) was achieved on the fourth day. The total VFA yield was 0.27 g (g total solid (TS))(-1), which almost doubled the yield without pH adjustment (0.15 g (g TS)(-1). The acidified products with lower concentration of lactic acid were more favorable to the subsequent methanogenesis. At pH 7, most of the protein was degraded into ammonia nitrogen (NH4(+)-N), resulting in additional buffer of acidified liquid. According to this, a novel method of pH adjustment applying NH4(+)-N buffer was established. The semi-continuous experiment indicated that adjusting pH to 7 in the acidogenic stage in the two-phase anaerobic digestion process would increase both TS loading rate (16g l(-1) d(-1)) and recovery of biological energy (520 ml methane (CH4) (g TS)(-1)).

Application of aerobic granular sludge in polishing the UASB effluent.

Stable aerobic granules were successfully developed in a sequencing batch reactor (SBR) fed with glucose and sodium acetate as the main carbon sources, and then they were acclimatized to polish the effluent from an up-flow anaerobic sludge blanket (UASB) reactor. It was found that more than 95% chemical oxygen demand (COD) and 85% suspended solids (SS) in the UASB effluent could be removed by an aerobic granular SBR as a post treatment unit. The performance of the SBR was stable over the whole experimental period. At an organic loading rate (OLR) of 4.5 to 5.4 g l(-1) d(-1), the acclimatized aerobic granules had an average diameter of 3.5 to 6.0 mm and a minimal settling velocity of 72 m h(-1). The biomass concentration in the SBR was as high as 8.4 g (dry weight) l(-1). This study showed for the first time that aerobic granular sludge in SBR would be an effective means to polish the effluent from UASB reactor.

Immunological development of preterm infants in early infancy.

To evaluate the immunological development of preterm infants, especially in early infancy, we examined the serum cytokine levels and the expression of Th2 and Th1 chemokine receptors, CCR4 and CCR5, on days 0, 14 and 28 in 16 low birth weight infants (1720.38 +/- 502.80 g) born at less than 37 (33.63 +/- 3.29) weeks of gestation. Using an enzyme-linked immunosorbent assay (ELISA), serum interleukin (IL)-4 levels exhibited an increase on day 14, but decreased to the initial level on day 28 (P < 0.05). The significant elevation of serum transforming growth factor (TGF)-beta levels was confirmed on day 14 (P < 0.05) but decreased to the initial level on day 28 (P < 0.05). The expression of CCR4 and CCR5 were examined using reverse transcription-polymerase chain reaction (RT-PCR) and flow cytometric analysis. The RT-PCR confirmed the expression of CCR5-mRNA soon after birth, while there was no expression of CCR4-mRNA. Thereafter, the expression of CCR4-mRNA increased significantly and reached the level of CCR5-mRNA expression on day 28 (P < 0.05). Flow cytometric analysis, however, revealed that the expression levels of both CCR4 and CCR5 were low at birth. Thus, CCR4(+) CD4(+) cells were significantly increased from days 0-28 (P < 0.05), while CCR5(+) CD4(+) cells were not. Increased IL-4 and TGF-beta synthesis as well as increased CCR4(+) CD4(+) cells suggest that, under extra-maternal circumstances, there is a shift in bias toward Th2 responses even in preterm infants soon after delivery, while they may be capable of developing Th1 mediated responses soon after birth.

Combined factor V and factor VIII deficiency in a Thai patient: a case report of genotype and phenotype characteristics.

A Thai woman, with no family history of bleeding disorders, presented with excessive bleeding after minor trauma and tooth extraction. The screening coagulogram revealed prolonged activated partial thromboplastin time and prothrombin time. The specific-factor assay confirmed the diagnosis of combined factor V and factor VIII deficiency (F5F8D). Her plasma levels of factor V and factor VIII were 10% and 12.5% respectively. The medications and blood product treatment to prevent bleeding from invasive procedure included 1-deamino-8-d-arginine vasopressin, cryoprecipitate, factor VIII concentrate, fresh frozen plasma and antifibrinolytic agent. Gene analysis of the proband identified two LMAN1 gene mutations; one of which is 823-1 G --> C, a novel splice acceptor site mutation that is inherited from her father, the other is 1366 C --> T, a nonsense mutation that is inherited from her mother. Thus, the compound heterozygote of these two mutations in LMAN1 cause combined F5F8D.

Optimizing the acquisition time profile for a planar integral measurement system with a spinning slat collimator.

This article considers a hypothetical imaging device with a spinning slat collimator that measures parallel-planar-integral data from an object. This device rotates around the object 180 degrees and stops at N positions uniformly distributed over this 180 degrees. At each stop, the device spins on its own axis 180 degrees and acquires measurements at M positions uniformly distributed over this 180 degrees. For a fixed total imaging time, an optimal distribution of the scanning time among the data measurement locations is searched by a nonlinear programming method: Nelder-Mead's simplex method. The optimal dwell time is approximately proportional to the weighting factor in the backprojector of the reconstruction algorithm. By using an optimal dwell-time profile, the reconstruction signal-to-noise ratio has a gain of 23%-24% for the filtered backprojection algorithm and a gain of 10%-18% for the iterative algorithms, compared with the situation when a constant dwell-time profile is used.

Formation of sp(3) bonding in nanoindented carbon nanotubes and graphite.

Nanoindentation-induced interlayer bond switching and phase transformation in carbon nanotubes (CNTs) and graphite are simulated by molecular dynamics. Both graphite and CNTs experience a soft-to-hard phase transformation at room temperature at compressive stresses of 12 and 16 GPa, respectively. Further penetration leads to the formation of interlayer sp(3) bonds, which are reversible upon unloading if the compressive stress is under about 70 GPa, beyond which permanent interlayer sp(3) bonds form. During nanoindentation, the maximum nanohardness of graphite can reach 109 GPa, and CNTs 120 GPa, which is comparable to that of diamond.

Ion acceleration from the shock front induced by hole boring in ultraintense laser-plasma interactions.

Ion-acceleration processes have been studied in ultraintense laser plasma interactions for normal incidence irradiation of solid deuterated targets via neutron spectroscopy. The experimental neutron spectra strongly suggest that the ions are preferentially accelerated radially, rather than into the bulk of the material from three-dimensional Monte Carlo fitting of the neutron spectra. Although the laser system has a 10(-7) contrast ratio, a two-dimensional magnetic hydrodynamics simulation shows that the laser pedestal generates a 10 mum scale length in the coronal plasma with a 3 mum scale-length plasma near the critical density. Two-dimensional particle-in-cell simulations, incorporating this realistic density profile, indicate that the acceleration of the ions is caused by a collisionless shock formation. This has implications for modeling energy transport in solid density plasmas as well as cone-focused fast ignition using the next generation PW lasers currently under construction.

Precision measurement of the neutron spin asymmetryA(n)(1) and spin-flavor decomposition in the valence quark region.

We have measured the neutron spin asymmetry A(n)(1) with high precision at three kinematics in the deep inelastic region at x=0.33, 0.47, and 0.60, and Q(2)=2.7, 3.5, and 4.8 (GeV/c)(2), respectively. Our results unambiguously show, for the first time, that A(n)(1) crosses zero around x=0.47 and becomes significantly positive at x=0.60. Combined with the world proton data, polarized quark distributions were extracted. Our results, in general, agree with relativistic constituent quark models and with perturbative quantum chromodynamics (PQCD) analyses based on the earlier data. However they deviate from PQCD predictions based on hadron helicity conservation.

Alteration of heme-oxygenase-carbon monoxide pathway in calcified rat vascular smooth muscle cells.

OBJECTIVE: The aim of the present study was to investigate the changes in heme-oxygenase (HO)-carbon monoxide (CO)-cyclic guanosine monophosphate (cGMP) pathway in clacified rat vascular smooth muscle cells (VSMCs). METHODS: Calcification of cultured rat VSMCs was induced by incubation of VSMCs with beta-glycerophosphate. Cellular calcium content, ALP activities and (45)Ca uptake were measured. HO activity, HbCO formation and content of cGMP in VSMCs were determined. Immunocytochemistry for HO-1 expression was observed. RESULTS: In comparison of control VSMCs, the cellular calcium content, ALP activity and (45)Ca uptake in calcified VSMCs were obviously increased. Immunocytochemistry showed that HO-1 expression was weak and not well distributed in calcified cells as compared to non-calcified VSMCs, but interestingly, there was stronger staining in calcified nodules than in VSMCs. Compared with VSMCs, HO-1 activity in calcified cells decreased by 42.7% [36.4 +/- 2.8 pmol (mg Pr x h)(-1) vs 63.5 x 5.3 pmol (mg Pr x h)(-1), p < 0.01], and HbCO formation decreased by 39.2% (3.38 x 0.69 micromol/mg Pr vs 5.56 +/- 0.48 micromol/mg Pr, p < 0.05). The cGMP content in calcified VSMCs was 78.1% lower than that of non-calcified VSMCs (4.3 +/- 0.51 vs 19.6 +/- 1.2 pmol/mg, p < 0.01). CONCLUSION: The results showed that HO-CO-cGMP pathway in calcified vascular cells obviously changed, which might contribute to disturbance of vascular function.

Representation of a human head with bi-cubic B-splines technique based on the laser scanning technique in 3D surface anthropometry.

Three-dimensional (3D) anthropometry based on the laser scanning technique not only provides one-dimensional measurements calculated in accordance with the landmarks which are pre-located on the human body surface manually, but also the 3D shape information between the landmarks. This new technique used in recent ergonomic research has brought new challenges to resolving the application problem that was generally avoided by anthropometric experts in their researches. The current research problem is concentrating on how to shift and develop one-dimensional measurements (1D landmarks) into three-dimensional measurements (3D land-surfaces). The main purpose of this paper is to test whether the function of B-splines can be used to fit 3D scanned human heads, and to for further study to develop a computer aided ergonomic design tool (CAED). The result shows that B-splines surfaces can effectively reconstruct 3D human heads based on the laser scanning technique.

Knockdown of spinal cord postsynaptic density protein-95 prevents the development of morphine tolerance in rats.

The activation of spinal cord N-methyl-D-aspartate (NMDA) receptors and subsequent intracellular cascades play a pivotal role in the development of opioid tolerance. Postsynaptic density protein-95 (PSD-95), a molecular scaffolding protein, assembles a specific set of signaling proteins around NMDA receptors at neuronal synapses. The current study investigated the possible involvement of PSD-95 in the development of opioid tolerance. Opioid tolerance was induced by intrathecal injection of morphine sulfate (20 microg/10 microl) twice a day for 4 consecutive days. Co-administration of morphine twice daily and PSD-95 antisense oligodeoxynucleotide (50 microg/10 microl) once daily for 4 days not only markedly reduced the PSD-95 expression and its binding to NMDA receptors in spinal cord but also significantly prevented the development of morphine tolerance. In contrast, co-administration of morphine twice daily and PSD-95 missense oligodeoxynucleotide (50 microg/10 microl) once daily for 4 days did not produce these effects. The PSD-95 antisense oligodeoxynucleotide at the doses we used did not affect baseline response to noxious thermal stimulation or locomotor function. The present study indicates that the deficiency of spinal cord PSD-95 attenuates the development of opioid tolerance. These results suggest that PSD-95 might be involved in the central mechanisms of opioid tolerance and provide a possible new target for prevention of development of opioid tolerance.

Evidence of neuronal excitatory amino acid carrier 1 expression in rat dorsal root ganglion neurons and their central terminals.

The expression and distribution of the neuronal glutamate transporter, excitatory amino acid carrier-1 (EAAC1), are demonstrated in the dorsal root ganglion neurons and their central terminals. Reverse transcriptase-polymerase chain reaction shows expression of EAAC1 mRNA in the dorsal root ganglion. Immunoblotting analysis further confirms existence of EAAC1 protein in this region. Immunocytochemistry reveals that approximately 46.6% of the dorsal root ganglion neurons are EAAC1-positive. Most EAAC1-positive neurons are small and around 250-750 microm2 in surface area, and some co-label with calcitonin gene-related peptide (CGRP) or isolectin IB4. In the spinal cord, EAAC-1 immunoreactive small dot- or patch-like structures are mainly localized in the superficial dorsal horn, and some are positive for CGRP or labeled by isolectin IB4. Unilateral dorsal rhizotomy experiments further show that EAAC1 immunoreactivity is less intense in superficial dorsal horn on the side ipsilateral to the dorsal rhizotomy than on the contralateral side. The results indicate the presence of EAAC1 in the dorsal root ganglion neurons and their central terminals. Our findings suggest that EAAC1 might play an important role in transmission and modulation of nociceptive information via the regulation of pre-synaptically released glutamate.
