Cell components required for deletion of an autoreactive T cell repertoire.

T cells become tolerant to self antigens during their development in the thymus. Clonal deletion of thymocytes bearing T cell receptor (TcR) which recognize self antigens is a major mechanism for generating tolerance. In the present study we have used allogeneic bone marrow (BM) chimeras, prepared with various combinations of mouse strains and focusing especially on expressions of I-E molecules and Mls-1a antigens on the cell surface, to investigate both immunohistochemically and by flow cytometry the cell components that contribute to the clonal deletion of T cells positive for V beta 6 TcR. The V beta 6 TcR expression is strongly associated with T cell recognition of both I-E and Mls-1a antigens. We found that I-E+ cells derived from donor BM (and thus not of recipient lineage) represented a primary requirement for deletion of Mls-1a-reactive thymocytes which bear V beta 6 TcR. Immunohistochemical analysis revealed that the donor-derived I-E+ cells were distributed mainly to the thymic medulla and that the V beta 6+ cells were eliminated from the thymic medulla between 2 and 3 weeks following BM transplantation. In contrast, Mls-1a+ cells of either donor or recipient origin might be responsible for the deletion, even though cortical epithelial cells appeared not to express Mls-1a antigens.

Central nervous system congenital malformations, especially neural tube defects in 29 provinces, metropolitan cities and autonomous regions of China: Chinese Birth Defects Monitoring Program.

Data from the Chinese Birth Defects Monitoring Program (CBDMP) over the period of October 1986 to September 1987 were analysed to study the descriptive epidemiology of congenital malformations of the central nervous system (CNS), especially neural tube defects (NTDs) in China. A total of 4628 CNS congenital malformations were recorded within seven days of delivery among 1,243,284 live and stillbirths of 28 or more weeks gestation in 945 hospitals from all 29 provinces, metropolitan cities and autonomous regions of China. Neural tube defects account for 73.55% of these cases, hydrocephaly for 24.63% and microcephaly for 1.82%. The prevalence rates at birth of NTDs and congenital malformations of the CNS in China were 27.37 and 37.22 per 10,000 respectively. More NTDs were observed in females (35.68 per 10,000 female births) as compared to males (19.23 per 10,000 male births). The prevalence of NTDs in rural areas (51.69 per 10,000 births) was higher than that in urban areas (15.45 per 10,000 births).

Sequential analysis of distributions of donor-derived thymocytes bearing T-cell antigen receptor (TCR) and donor-derived Ia+ cells in thymuses of fully allogeneic bone marrow chimera in mice.

Lethally irradiated SJL/J mice were reconstituted with B10 bone marrow cells, and the process of thymic reconstitution by donor-derived cells positive for I-A or V beta 8 molecules was investigated. The donor-derived Ia+ cells appeared in the medulla on day 7 after reconstitution. The Ia+ cells became confluent up to day 14, and the cellularity in the medulla on day 17 was almost the same as that in the normal thymus. Dull V beta 8+ thymocytes were first recognized in the cortex on day 10 and were identifiable in the medulla by day 14. The V beta 8+ cells seemed to be mainly CD4+8+ double-positive. Furthermore, most of the V beta 8+ cells in the medulla of chimeras given cyclosporin A for 3 weeks after reconstitution appeared to be CD4+8+ thymocytes which bear a low concentration of TCR exist in the thymic medulla at a relatively early stage when donor-derived Ia+ cells have already settled there. The coincidental appearance and coexistence of Ia+ cells and TCR+ thymocytes in the medulla suggest that these histological characteristics may be related to the selection of thymocytes in this area.

[The laboratory examination of Candida albicans].

This study compares the laboratory methods in detecting candidas causing oral candidiasis and suggests that smear method is the main one. PAS stain is the best for diagnosis, and the potassium hydroxide method is the easiest and quickest in handling. As to those uncertained by smear method, culture should be done. For those positive on Sabouraud's medium, the germ tube test should be performed and positive results can be regarded as candida albicans. If negative the chlamydospore production as well as the carbohydrate fermentation and assimilation should be done.

[Treatment of childhood anorexin with spleen deficiency by principle of invigorating spleen and benefiting qi].

UNLABELLED: 101 children of anorexia with Spleen deficiency were divided randomly into two groups, the invigorating Spleen(IS) group of 58 cases treated by modified Yi-Gong-San, and the replenishing zinc(RZ) group of 43 cases treated by 0.2% zinc sulphate syrup. One course of treatment was 3 months. RESULTS: the clinical effects of IS group were notable in promoting appetite, increasing body weight and height, eliminating polyhidrosis, preventing common cold and tracheitis etc. The total effective rate was 96.6% in IS group which was significantly better than 72.1% in the RZ group (P less than 0.01). The effect of examination showed that the levels of hair zinc after treatment were markedly increased in both groups (P less than 0.01). The average increased value of hair zinc was 62.2 +/- 9.1 (ppm) in IS group which was higher than 42.1 +/- 7.2 (ppm) in RZ group (P less than 0.05). Moreover excretion rate and Hb after treatment in the IS group were markedly higher those in the RZ group (P less than 0.01). Chemical analysis showed that the concentration of zinc in modified Yi-Gong-San was very slight (0.73 mg/pack). It indicated that arising hair zinc in IS group was not simply supply of zinc but also the treatment in appetite, digestion and absorption, with which the body was supplied enough zinc naturally by food taken.

An immunocytochemical study on the distribution of ferritin and other markers in 36 cases of malignant histiocytosis.

The distribution of ferritin in 36 autopsy cases of malignant histiocytosis was investigated by immunocytochemical staining, together with the detection of alpha 1-antichymotrypsin, alpha 1-antitrypsin, lysozyme, S-100 protein, and ricinus communis agglutinin in the consecutive sections. The results showed that ferritin-positive tumor cells were present in every case. The quantity of cellular ferritin in well-differentiated histiocytes was higher than that in atypical histiocytes. Double labeling showed that ferritin and alpha 1-antichymotrypsin might be located either in one tumor cell or in separate cells. Our data suggest that ferritin may be a tumor associated antigen in malignant histiocytosis, playing a regulatory role for tumor cell differentiation.

A liver-specific nuclear factor interacts with the promoter region of the large surface protein gene of human hepatitis B virus.

The outer envelope of the 42-nm virion of the human hepatitis B virus (HBV) is composed of the large, the middle, and the major surface proteins. Whereas the middle and the major surface proteins are transcribed from the SPII promoter of the pre-S/S gene, the large surface protein is transcribed from the SPI promoter located upstream of SPII. We have previously shown that transcription of SPI (comprising nucleotides [nt] -380 to +17) occurs preferentially in differentiated hepatoma cell lines (H.K. Chang and L.P. Ting, Virology 170:176-183, 1989). In this report, we further demonstrated that a sequence of 95 base pairs in the upstream region of SPI (nt -95 to +17) was necessary and sufficient for such preferential expression in differentiated hepatoma cells. By analysis of the expression of the chloramphenicol acetyltransferase gene in a series of mutants with deletions at the 5' end of SPI, we identified a positive transcriptional cis-acting element mapping at nt -95 to -72 which appears to play a key role in the regulation of the expression of the large surface protein. This region shared a high degree of sequence homology with regulatory sequences of several liver-specific genes from human, mouse, and rat, with a consensus sequence (G/A)GTTA(A/C)TNNT(C/T)NNC(A/C). We further identified a nuclear factor present in the nuclear extracts of differentiated human hepatoma cell lines which interacted specifically with this element of the SPI promoter. This nuclear factor was similar to the rat liver-specific factor HNF-1, since an oligonucleotide containing the recognition sequence of HNF-1 could efficiently compete for the human factor in a footprinting assay. The sequence at nt -93 to -68 which was bound by this factor in SPI was termed the HNF-1-binding element. Activation of the SPI promoter by human differentiated hepatocyte nuclear factor 1, described in this report, probably explains, first, the formation of the 42-nm virion specifically in liver but not in several other tissues despite the synthesis of the middle and the major surface proteins in those tissues, and second, why only differentiated hepatoma cell lines are able to produce 42-nm-like virion particles on transfection by HBV DNA.

[In vitro assessment of candidacidal activity of human leukocyte myeloperoxidase preparation].

Myeloperoxidase (MPO), present in the azurophilic granules of human polymorphonuclear neutrophils, is important in the oxygen-dependent microbicidal activity of neutrophils. The purpose of our study was to investigate the therapeutic potency of the MPO preparations. This paper is to present our primary work on MPO isolation and its microbicidal activity assay. White blood cells, isolated freshly from normal donors, were lysed with cetyltrimehylammonium bromide to liberate myeloperoxidase. The enzyme preparation was partially purified by 50% (NH4)2SO4 precipitation followed by 65% (NH4)2SO4 precipitation. A hundred million leukocytes yielded 1.03 mg protein of the MPO preparation with the RZ of 0.31. The specific activity of the MPO preparation was about 29.25 u/mg. When 0.672 units of the MPO preparation were incubated with about 10(7) clinical isolates of Candida albicans in the presence of 0.2 mmol/L H2O2 and 0.14 mol/L NaCl. It was detected that 95.58 +/- 0.64% of the organisms were killed in the methylene blue staining system.

[Catheter ablation in eight patients with Wolff-Parkinson-White syndrome].

After a follow-up study of 13-39 months, the clinical effect of catheter ablation in 8 patients with WPW syndrome and recurrence of SVT was reported. Five cases of left free-wall accessory pathway underwent catheter ablation attempts by direct current shocks delivered in coronary sinus. 3 of them were free from SVT without any medications after ablation, 1 case was improved. The other one was unsuccessful. Catheter ablation was attempted by delivery of shocks at coronary sinus orifice in 3 patients with posteroseptal accessory pathway. Arrhythmias were controlled without the need of drug therapy in 2 cases and the clinical symptoms were relieved in 1 case after shocks. We consider that the catheter ablation attempted to block the conduction of left free-wall and posteroseptal accessory pathway induces much less damages than surgical ablation and offers the advantages of shorter hospitalization, lower cost and a much shorter convalescence. The authors stated that it is a viable alternative to surgical ablation for those patients who are not responsive or allergic to drugs or those who are not willing to take a life-long period of medications. Some concerns for increasing the success rate and reducing the complications of ablation were also discussed.

Identification of a pterin derivative in Escherichia coli DNA photolyase.

DNA photolyase from Escherichia coli contains reduced flavin adenine dinucleotide plus a second chromophore, partially characterized in previous studies. Both chromophores function as sensitizers in catalysis. The second chromophore has been identified as a 6-substituted pterin derivative. The compound is oxidized with permanganate to yield 6-carboxypterin or reduced with sodium cyanoborohydride to yield a 5,6,7,8-tetrahydropterin derivative. The second chromophore exhibits spectral properties (lambda max = 360, 255 nm, pH 2) similar to that observed for 7,8-dihydropterin cations. The compound does not exhibit a spectrally detectable pKa around 4 but is converted to a dication (lambda max = 346, 255 nm) in strong acid (pKa approximately 1). Similar ionization behavior is observed with 7,8-dihydropterin derivatives that are alkylated at N(5). The instability of the second chromophore in weakly alkaline solution is due to a fully reversible conversion to a labile bleached form. As compared with other pterin derivatives, the hydrolytic instability is unusual but is very similar to that observed for 5,6-dialkyl-7,8-dihydropterinium salts. It is proposed that the second chromophore is a 7,8-dihydropterin with substituents at positions 5 and 6. The discovery that a pterin derivative functions as a photosensitizer in DNA repair is apparently the first example of a photobiological function for pterins.

Effect of higenamine on action potential of ventricular myocardial cells.

Action potentials of isolated ventricular myocardial fibers of pigs and the electrophysiological actions of dl-demethylcoclaurine (DMC) or higenamine on the porcine myocardial cells were studied by glass microelectrodes. The action potentials of porcine ventricular myocardial fibers were similar to those of other mammalian hearts. Amplitude of the action potential was 115 +/- 9 mV, duration of action potential 242 +/- 30 ms, resting membrane potential -85 +/- 5 mV and the maximum rise of depolarization of action potential 151 +/- 27 V/s. After perfusion with normal Tyrode's solution containing DMC 5 micrograms/ml, the amplitude of action potential was increased and the duration of action potential was prolonged, the amplitude and duration of the plateau of action potential were increased and the slope of phase 2 of action potential was reduced (p less than 0.05). The resting potential, the slope of phase 3 and the maximum depolarization rate of phase 0 of action potential did not change. In Tyrode's solution containing Mn++ 3 mM, these electrophysiological actions of DMC disappeared. DMC may abolish the conduction block induced by K+-rich solution. In Tyrode's solution containing K+ 32 mM, the upstroke of action potential showed 2 phases under influence of DMC. The second phase disappeared when Mn++ was added. All findings indicate that DMC can be considered as an activator of the slow channel. The electrophysiological mechanism and clinical significance of DMC were discussed.
