Purification, solution properties and crystallization of SIV integrase containing a continuous core and C-terminal domain.

The C-terminal two-thirds segment of integrase derived from the simian immunodeficiency virus has been cloned, expressed in Escherichia coli, and purified to greater than 95% homogeneity. The protein encompasses amino-acid residues 50-293 and contains a F185H substitution to enhance solubility. In dilute solutions at concentrations below 1 mg ml(-1), the enzyme is predominantly dimeric. At the higher concentrations (>10 mg ml(-1)) required to enable crystallization, the enzyme self-associates to form species with molecular weights greater than 200 kDa. Despite the apparent high aggregation in solution, the enzyme crystallizes from a 8%(v/v) polyethylene glycol (molecular weight 6000) solution in a form suitable for X-ray diffraction studies. The resulting single crystals belong to the space group P2(1)2(1)2(1), with unit-cell parameters a = 79.76, b = 99.98, c = 150.2 A, alpha = beta = gamma = 90 degrees and Z = 4. Under X-ray irradiation generated with a rotating-anode generator, the crystals diffract to 2.8 A resolution and allow collection of a native 3 A resolution diffraction data set.

A new model of cancer cachexia: contribution of the ubiquitin-proteasome pathway.

A new model of cachexia is described in which muscle protein metabolism related to the ubiquitin-proteasome pathway was investigated. Cloning of the colon-26 tumor produced a cell line, termed R-1, which induced cytokine (noninterleukin-1beta, interleukin-6 and tumor necrosis factor-alpha)-independent cachexia. Implantation of R-1 cells in mice elicited significant (20-30%) weight loss and decreased blood glucose by 70%, and adipose tissue levels declined by 95% and muscle weights decreased by 20-25%. Food intake was unaffected. The decrease in muscle weight reflected a decline in insoluble, but not soluble, muscle protein that was associated with a significant increase in net protein degradation. The rate of ubiquitin conjugation of proteins was significantly elevated in muscles of cachectic mice. Furthermore, the proteasome inhibitor lactacystin blocked the increase in protein breakdown but had no significant effect on proteolysis. Several markers of the ubiquitin-proteasome pathway, E2(14k) mRNA and E2(14k) protein and ubiquitin-protein conjugates, were not elevated. Future investigations with this new model should gain further insights into the mechanisms of cachexia and provide a background to evaluate novel and more efficacious therapies.

Interacting appetite-regulating pathways in the hypothalamic regulation of body weight.

Various aspects of the complex spatio-temporal patterning of hypothalamic signaling that leads to the development of synchronized nocturnal feeding in the rat are critically examined. Undoubtedly, as depicted in Fig. 7, a distinct ARN in the hypothalamus is involved in the control of nocturnal appetite. At least four basic elements operate within this ARN. These are: 1) A discrete appetite-driving or orexigenic network of NPY, NE, GABA, GAL, EOP, and orexin transduces and releases appetite-stimulating signals. 2) Similarly, anorexigenic signal-producing pathways (e.g., CRH, GLP-1, alpha MSH, and CART) orchestrate neural events for dissipation of appetite and to terminate feeding, possibly by interrupting NPY efflux and action at a postsynaptic level within the hypothalamus. It is possible that some of these may represent the physiologically relevant "off" switches under the influence of GABA alone, or AgrP alone, or in combination with NPY released from the NPY-, GABA-, and AgrP-coproducing neurons. 3) Recent evidence shows that neural elements in the VMN-DMN complex tonically restrain the orexigenic signals during the intermeal interval; the restraint is greatly aided by leptin's action via diminution of orexigenic (NPY) and augmentation of anorexigenic (GLP-1, alpha MSH, and CART) signals. Since interruption of neurotransmission in the VMN resulted in hyperphagia and development of leptin resistance, it seems likely that the VMN is an effector site for the restraint exercised by leptin. The daily rhythms in leptin synthesis and release are temporally dissociable because the onset of daily rise in leptin gene expression in adipocytes precedes that in leptin secretion. Nevertheless, these rhythms are in phase with daily ingestive behavior because the peak in circulating leptin levels occurs during the middle of the feeding period. These observations, coupled with the fact that circulating levels of leptin are directly related to adiposity, pose a new challenge for elucidating the precise role of leptin in daily patterning of feeding in the rat. 4) A neural timing mechanism also operates upstream from the ARN in the daily management of energy homeostasis. Although the precise anatomical boundaries are not clearly defined, this device is likely to be composed of a group of neurons that integrate incoming internal and external information for the timely onset of the drive to eat. Evidently, this network operates independently in primates, but it is entrained to the circadian time keeper in the SCN of rodents. Apart from its role in the onset of drive to eat, the circadian patterns of gene expression of NPY, GAL, and POMC denote independent control of the timing device on the synthesis and availability for release of orexigenic signals. The VMN-DMN-PVN complex is apparently an integrated constituent of the timing mechanism in this context, because lesions in each of these sites result in loss of regulated feeding. The accumulated evidence points to the PVN and surrounding neural sites within this framework as the primary sites of release and action of various orexigenic and anorexigenic signals. A novel finding is the identification of the interconnected wiring of the DMN-mPVN axis that may mediate leptin restraint on NPY-induced feeding. The chemical phenotypes of leptin and NPY target neurons in this axis remain to be identified. These multiple orexigenic and anorexigenic pathways in the hypothalamic ARN appear to represent redundancy, a characteristic of regulated biological systems to provide a "fail-safe" neural mechanism to meet an organism's constant energy needs for growth and maintenance. Within this formulation, the coexisting orexigenic signals (NPY, NE, GAL, GABA, and AgrP) represent either another level of redundancy or it is possible that these signals operate within the ARN as reinforcing agents to varying degrees under different circumstances. Daily changes in hypothalamic gene expression of neuropeptide Y, galanin, proopiomelanocortin, and adipocyte leptin gene expression and secretion: effects of food restriction.

The participation of hypothalamic neuropeptide Y (NPY)-, galanin (GAL)-, and opioid-producing neurons in the restraint on food intake exerted by adipocyte leptin has recently been recognized. To further understand the interplay between the central appetite-stimulating- and peripheral appetite-inhibiting signals in the management of daily food intake, we have examined the daily patterns in expression of the hypothalamic neuropeptides and leptin receptor (R) and adipocyte leptin gene expression and secretion in freely feeding (FF) rats. These analyses were extended to determine the impact of food restriction (FR) to 4 h daily for 4 weeks. Groups of FF and FR rats were killed at 4-h intervals during a 24-h period, and hypothalamic NPY, GAL, POMC, and leptin-R gene expression and leptin gene expression were evaluated by RNase protection assays and serum leptin and corticosterone (CORT) levels were estimated by RIA. The following new findings emerged: 1) In FF rats, hypothalamic NPY messenger RNA (mRNA) levels fluctuated during the course of 24 h with high levels at 0700 h and 1100 h followed by a decrease at 1500 h during the lights-on phase that was sustained throughout the dark phase (1900 h-0500 h) of the light-dark cycle. Hypothalamic GAL and POMC mRNA also displayed daily patterns but with a different time course; GAL and POMC gene expression were elevated 4 h later than NPY mRNA at 1100 h and 1500 h. 2) Although FR to 4 h between 1100 h and 1500 h resulted in maintenance of body weight compared with a steady weight gain in FF rats, the daily patterns of fluctuations in hypothalamic neuropeptide gene expression were abolished. 3) In FF rats, hypothalamic leptin-R and adipocyte leptin gene expression and serum leptin levels displayed a daily pattern temporally different from that of hypothalamic neuropeptide gene expression. Adipocyte leptin mRNA remained low during the lights-on phase but increased at the onset of the lights-off phase (1900 h) and remained elevated through the dark phase. 4) Hypothalamic leptin-R gene expression, like that of adipocyte leptin gene expression, rose abruptly at the onset of nocturnal feeding behavior but receded progressively to low range thereafter. 5) On the other hand, a dichotomy in the daily rise in adipocyte leptin gene expression and leptin secretion was observed in FF rats. Unlike adipocyte leptin mRNA, serum leptin increased at 2300 h, 4 h after initiation of ingestive behavior. 6) In FR rats, adipocyte leptin gene expression fluctuated little over the 24-h period but, as in FF rats, leptin hypersecretion peaked 4 h after initiation of food intake. 7) In both FF and FR rats, increased serum CORT levels preceded serum leptin rise. Overall, these results show that in FF rats, gene expression of hypothalamic appetite stimulating peptides first rise and then fall to nadir during the lights-on phase when leptin levels are in low range; adipocyte leptin mRNA rises before impending ingestive behavior and increased leptin secretion reaching peak manifests itself during nocturnal feeding. The FR regimen, which curtailed the normal body weight gain, abolished these daily fluctuations in gene expression of hypothalamic orexigenic peptides and adipocyte leptin but permitted feeding-associated increased leptin secretion. Thus, it may be important to consider the daily patterns of gene expression and availability of hypothalamic orexigenic peptides in investigations aimed at elucidating the central mechanisms underlying the feedback action of the normal and altered leptin secretion patterns.

Nonstrategic subjective threshold effects in phonemic masking.

Three backward-masking experiments demonstrated that the magnitude of the phonemic mask reduction effect (MRE) is a function of subjective threshold and that the magnitude is also independent of stimulus-based response strategies. In all three experiments, a target word (e.g., bake) was backward masked by a graphemically similar nonword (e.g., BAWK), a phonemically similar nonword (e.g., BAIK), or an unrelated control (e.g., CRUG). Experiments 1 and 2 had a low percentage (9%) of trials with phonemic masks and differed only in baseline identification rate. Experiment 3 controlled baseline identification rate at below and above subjective threshold levels, with 9% phonemic trials. The results were that identification rates were higher with phonemic masks than with graphemic masks, irrespective of the low percentage of phonemic trials. However, the magnitude of the phonemic MRE became large only when the baseline identification rate was below subjective threshold. The pattern of the phonemic MRE was interpreted as a result of rapid automatic phonological activation, independent of stimulus-based processing strategies.

[Changes of serum biochemical parameters during hypothermia and hypoxia in rats].

OBJECTIVE: To study the effects of hypothermia and hypoxia on serum biochemical parameters. Method Acute hypobaric hypoxia experiment in cold environment was carried out in 48 healthy Wistar rats to observe changes of hepatic, cardiac and renal functions. RESULT: Hepatic, cardiac and renal functions changed non-prominently after acute hypoxia exposure under cold condition. Under hypoxic exposure of the same degree, serum lactic dehydrogenase(LDH), alanine transaminase(ALT), blood urea nitrogen (BUN) and creatinine (Cr) increased more significantly at 10 degrees C than those at 20 degrees C (P < 0.01) while creatine kinase (CK) decreased significantly at 10 degrees C than that at 20 degrees C. CONCLUSION: After acute hypoxia in cold environment, the changes in cardiac function did not simply equal to the changes by cold environment plus changes by acute hypoxia.

[A feasibility study on the highly-efficient electronic fluorescent lamp used as a lighting source for cultivation of higher plant in space].

OBJECTIVE: To testify that the Highly-efficient Electronic Fluorescent Lamp (HEFL) can be used as a sole lighting source for the cultivation of higher plants in space. METHOD: The HEFL was utilized as the lighting source for the culture of three varieties of Luctuca sativa L in the lately-constructed Space Higher Plant Cultivation Ground-based Experimental Facility(SHPCGEF). Other culturing conditions were: temperature 20 +/- 0.2 degrees C, relative humidity (75 +/- 1)%, average photosynthetic active radiation (PAR) 70 micromol m-2 s-1 PPF, average wind velocity 0.45 m/s, photoperiod 24 h light/0 h dark, peat + vermiculite substrate culture, useful growing area 1.2 m2, growing period 28 d, one variety was cultured at every batch. Following plant maturing and being harvested, observations of external morphology of above-ground parts of the plants, measurements and calculations of edible biomass output and photosynthetic efficiency, analysis of nutrient compositions such as proteins, fats, carbohydrates, amino acids, vitamins and elemental compositions of lettuce leaves, and comparisons with formerly-related experiments were made. RESULT: All of the above-mentioned targets met our demands, some of them were superior to the results of other similar experiments. CONCLUSION: The HEFL can completely meet the needs for the growth and development of some higher plants planned to be grown in space, its physical characters basically accord with the demands, so it can be utilized as the only lighting source for higher plant growth in space environmental conditions.

[Endarterectomy for carotid stenosis].

OBJECTIVE: To discuss the indications, anesthesia and surgical techniques of carotid endarterectomy (CEA). METHODS: Twenty-two sides of carotid stenosis were revealed in 20 patients. CEA was performed under general anesthesia in 21 sides and local anesthesia in one. The time of carotid occlusion ranged from 27 to 52 minutes (average 35.5 min) in 20 operations, and intraoperative shunt was used in the other two. Fifteen operations were monitored with EEG. RESULTS: The postoperative course was uneventful in 17 patients after 18 operations, while transient muscle strength decrease on contralateral side and ipsilateral lower facial paresis occurred after three operations in the other three patients. Nineteen patients were followed-up for 38 months on the average. Among nine patients with TIA, ischemic attacks ceased in eight and decreased in one. Improvement was achieved in six patients with cerebral infarction. One patient without symptoms of carotid ischemia was still normal after operation. CONCLUSION: Carotid endarterectomy is a safe and effective way for treating carotid stenosis.

Comparison of some antineoplastic drugs on inhibiting thrombin catalizing fibrinogen clotting in vitro.

OBJECTIVE: To classify the effect of thrombin, the key enzyme which enables fibrinogen to form fibrin (fibrinogen clotting) on the formation of metastasis by comparing the inhibition of some antineoplastic drugs on fibrinogen clotting in vitro. METHODS: Time intervals of different drugs to reach a maximum OD (340 nm) data in fibrinogen solution added with thrombin were used in this work. RESULTS: It was found that L-4-oxalysine (8-40 micrograms/ml) and arabinosyl cytosine (10-50 micrograms/ml) could inhibit the effect of thrombin by extending the fibrinogen clotting time to 100%-150% (P < 0.001) and 61%-100% (P < 0.001) while the other antimetastatic drugs razoxane, probimane, adriamycin, harringtonine homoharringtonine and alpha-anordrin at the treatment concentrations showed no such activity. The positive rate of drugs to thrombin activity was approximately 25%. CONCLUSION: It suggests that L-4-oxalysine and arabinosyl cytosine may even exhibit antimetastatic effect through thrombin-fibrinogen pathway, and thrombin might operate in tumor metastasis for only limited step but crucial to fibrin formation in tumor nodules.

[Analysis of mandibular movement and functional classification for malocclusion patients].

OBJECTIVE: To classify the paths of mandibular movements of patients with malocclusion. METHODS: 223 patients were examined clinically and mandibular Kinesiograph(MKG) was applied. RESULTS: Each kind of malocclusion has its corresponding characteristic paths of mandibular movement. The paths of mandibular movements of patients with the same class of malocclusion are similar. It was also found that the mandibular movement was related not only with occlusion but also with jaws, muscles, and joints. Meanwhile, they affect each other. CONCLUSION: Functional malocclusion classification includes functional class I, II, III and IV malocclusion.

[Clinical study of anastrozole in the treatment of postmenopausal women with advanced breast cancer].

OBJECTIVE: To study the effect and adverse reactions of anastrozole in the treatment of postmenopausal women with advanced breast cancer. METHODS: A multicenter, open, non-randomized and crossover clinical trial on the effect of anastrozole was conducted. Sixty-one postmenopausal women with advanced breast cancer were treated by oral anastrozole, 1 mg once every day. Each patient received 4-6 weeks of treatment. Plasma estradiol concentrations were determined 2 weeks before treatment and within 3 days after treatment discontinued. Effect and adverse reaction were evaluated. RESULTS: The response rate in 60 evaluable cases treated with anastrozole was 21.7% (CR = 0, PR = 13, MR = 2). The effect was correlated with postmenopausal time, previous endocrine therapy and ER status. Plasma level of estradiol was suppressed by 58.3%. The main adverse reactions, including nausea, fatigue, facial flush, were generally tolerable. CONCLUSION: As an aromatase inhibitor capable of decreasing plasma estradiol level, anastrozole is therapeutically effective for advanced breast cancer in postmenopausal women. Side effects are mild and tolerable.

[Adenocarcinoma of the urinary bladder: a report of 27 cases].

OBJECTIVE: To analyze the therapeutic results of urinary bladder adenocarcinoma. METHODS: Twenty-seven urinary bladder adenocarcinoma patients were treated from 1970 through 1997. There were 12 cases of urachal adenocarcinoma and 15 cases of bladder adenocarcinoma. Hematuria with or without urinary tract irritation symptoms was the most common clinical manifestation. RESULTS: The overall 5-year survival rate was 25.9%. Among patients with urachal adenocarcinoma, 7 received extended partial bladder resection. None of them had local recurrence. Patients with bladder adenocarcinoma mainly received partial bladder resection, with which 4 developed local recurrence at the primary site. The 5-year survival rate of urachal adenocarcinoma and bladder adenocarcinoma was 33.3% and 20.0%, respectively. One patient of bladder adenocarcinoma with skin and inguinal lymph node metastases and 2 patients of urachal adenocarcinoma with lung metastasis treated with chemotherapy and radiotherapy survived 24, 28 and 60 months, respectively. CONCLUSION: Extended partial bladder resection is the treatment of choice for urachal adenocarcinoma while radical cystectomy for bladder adenocarcinoma. Comprehensive therapy should be given to patients with metastatic or recurrent disease to improve survival.

[The effect of Xuezhikang on oxidation of low-density lipoproteins in vitro].

OBJECTIVE: To study the effect of Xuezhikang on oxidation of human low-density lipoprotein. METHODS: In vitro experiment was used to study the effect of Xuezhikang on the change of human low-density lipoprotein. Relative electrophoresis movement rate after oxidation of low-density lipoprotein mediated by addition of copper ion was observed. RESULTS: Oxidation of low-density lipoprotein mediated by copper ion was accelerated as the time of action prolonged and the process was accompanied by increased relative electrophoresis movement rate. Oxidation of low-density lipoprotein by copper ion was inhibited significantly in the presence of Xue zhikang as compared with absence of XuezhiKang at all time points. A greater reduction of reactive electrophoresis movement rate up to 33.5%, (P < 0.05) was noted. With increase of Xuezhikang concentration, the extent of reduction increased. CONCLUSION: It is shown that human low-density lipoprotein can be oxidized by mediation of copper ion and XuezhiKang may inhibit the extent of oxidation of low-density lipoprotein by copper ion.

[The detection of T-cell receptor gene rearrangement in myelodysplastic syndromes].

OBJECTIVE: To analyse the T cell receptor (TCR) gene rearrangement in myelodysplastic syndromes (MDS). METHODS: Polymerase chain reaction (PCR) was used to detect the TCRVgammaI-Jgamma gene rearrangement in 36 cases of MDS. RESULTS: 8 cases (22.2%) were found to have TCRVgammaI-Jgamma gene rearrangement. The TCR gene rearrangement positive rate was higher in RAEB, CMML and RAEBT subtypes than that in RA and RAS subtypes (P < 0.05). The average time MDS evolving to acute leukemia was shorter in the MDS patients with TCRVgammaI-Jgamma for gene rearrangement than that in patients without (P < 0.01). CONCLUSION: TCR gamma gene rearrangement can also be found in some MDS patients and the detection of TCRgamma gene rearrangement might be helpful to identify the prognosis and the development of MDS.

[Treatment of severe acute renal failure with continuous renal replacement therapy].

OBJECTIVE: To present the experience in 101 severe acute renal failure (ARF) patients treated with continuous renal replacement therapy (CRRT) in our hospital from May 1986 to January 1999. METHODS: Patients'age, sex, previous health status and characteristics of renal failure were recorded. The acute physiology and chronic health evaluation II (APACHE II) score were calculated before CRRT treatment. RESULTS: 60 (59.4%) patients survived the acute phase of their illness, and 41 (40.6%) patients died. In the survival group, there were significantly less patients with pre-renal causes of renal failure and refractory course of uremia, low APACHE II score before treatment and less need of mechanical ventilation or vasoactive supporting. No difference was found in the percentage of oliguria between the survival and total group. CONCLUSION: CRRT is the treatment of first choice in ARF, especially in those complicated cases with severe parachymal renal diseases. The patients' age, previous health status, APACHE II score before CRRT, delayed occurrence of acute renal failure, need of mechanical ventilation and vasoactive drugs were predictors of a worse prognosis in severe ARF patient. The prognosis of renal type ARF patients improved with CRRT.

[Affect of insulin-like growth factor I and estradiol on the growth of uterine leiomyoma].

To study roles of insulun-like growth factor 1(IGF-I) and estradiol(E2) in the growth of leiomyoma. Protein expressions of IGF-I receptor(IGF-I-R), estrogen receptor(ER), and cell proliferation associated nuclear antigen(ki-67) were analysed by streptavidin-peroxide method in 40 cases. Serum level and local concentration of IGF-I were determined by immunoradiometric assay(IRMA) in 20 cases. Serum level of estradiol was determined by radioimmunoassay in 20 cases. Twenty normal fertile women were studied as control. Protein expressions of IGF-I-R and ER in uterine leiomyoma were significantly higher than in myometrium(P < 0.05); and local concentration of IGF-I positively correlated with ki-67 (r = 0.6513, P < 0.05); and expression of estrogen receptor. The serum levels of IGF-I and E2 had no significant difference between leiomyoma patients and normal women. IGF-I and E2 promote the growth of leiomyoma and IGF-I may act as a mediator of estrogen.

[Effects of Squama manitis on cardiac systolic function and platelet aggregation].

OBJECTIVE: To study the effect of Squama Manitis(S. M.) on cardiac function and platelet aggregation in vitro. METHOD: Isolated working rat hearts were prepared, and cardiac performance was measured before and after adding S. M. in the Krebs-Hensleit(K-H) perfusion buffer. Controls with K-H buffer were also investigated. Rich platelet plasma and Poor platelet plasma were centrifuged from the blood of 15 patients with coronary heart disease. Each sample was divided into three parts for use by three groups: control, group of S. M. and group of aspirin. Platelet aggregation was stimulated by adenosine diphosphate. RESULT: LVSP, +dP/dtmax and CO of isolated working rat hearts were significantly increased after treating with S. M. liquor. S. M. had no obvious effect on -dP/dtmax and LVEDP. These parameters did not change significantly in the control. Percentages of platelet aggregation were significantly lower in the group of S. M. and the group of aspirin as compared with the control. CONCLUSION: S. M. increases cardiac systolic function, has positive inotropic effect on myocardium and inhibits platelet aggregation.
