[Synthesis and antibacterial activity of 7-substituted-1-ethyl(2-fluoro-ethyl)-6,8-difluoro-1,4-dihydro-4- oxoquinoline-3-carboxylic acid].

Eighteen 7-substituted-1-ethyl (2-fluoroethyl)-6, 8-difluoro-1, 4-dihydro-4-oxoquinolone-3-carboxylic acid were designed and synthesized. The results of preliminary antibacterial test showed that most of the compounds exhibited definite activities in test against pathogenic bacteria. Especially, compound 17 and 18 showed better activities and surpassed fleroxacin when compared in vitro.

[Chitosan pin fixation of rabbit proximal tibia osteotomy].

Degradatable chitosan pin, 4 millimeters in diameter and 30 to 40 millimeters in length, was studied radiographically and histologically (oxytetracycline-labeling) at 3, 6, 12, 24 and 48 weeks after the implantation for fixation of a transverse proximal tibia osteotomy in fourty rabbits. The results showed that new bone formation and oxytetracycline uptake were increased 3-6 weeks after operation and the fracture was completely healed 12 weeks after operation, but in two rabbits the chitosan pin broke, in 3 weeks. Histologically, there was a massive inflammation (non-specific foreign body reaction) 3-12 weeks after implantation, which gradually subsided 24 weeks aften operation. Chitosan pin was slowly degradated in bone and was not replaced by normal tissue 48 weeks after operation. The questions to be addressed in future include how to increase the mechanical strength of chitosan pin and how to improve the characteristics of its surface.

[Application of charge transfer reaction in the determination of aminosalicylate sodium].

A spectrophotometric method for the determination of aminosalicylate sodium has been developed based on the charge transfer reaction. The Beer's law is obeyed in the range of 40-160 microg/mL of aminosalicylate sodium. The apparent molar absorptivity of the complex at 660nm is 1.17 X 10(3)L x mol(-1). The average standard deviation of ten determination is 0.67%. The composition of aminosalicylate sodium 2, 6-Dichloroquinone chlorimide complex is 1:1. The proposed method has been applied to the determination of aminosalicylate sodium tablets with satisfactory results.

Low birth weight in China and Finland.

Although a developing country, China has a lower occurrence of low birth weight (LBW) than many developed countries. This study of two population-based one-year birth cohorts, from Finland in 1985-86 and China in 1992, shows the occurrence of low birth weight (LBW) (1000- < 2500 g) among singletons to be 2.6 percent in the Chinese cohort and 3.0 percent in the Finnish one, and that of preterm births (28- < 37 weeks) 2.7 percent and 4.5 percent, respectively. The main component of LBW is term LBW (57.4 percent) in the Chinese case and preterm LBW (64.7 percent) in the Finnish case. The perinatal mortality rate (PMR) was twice as high in the Chinese cohort (13.0 vs. 5.9 per thousand). The occurrence of LBW in the Finnish cohort decreased to 2.3 percent after crosstabulation of the Finnish mothers to conform in structure to the population of Chinese mothers in terms of maternal age, marital status and maternal smoking. The result suggests that the lower incidence of LBW in the Chinese cohort seems to be a reflection of the Chinese socio-cultural environment, which provides Chinese mothers with favourable characteristics. The Finnish excess LBW would have disappeared if the mothers had possessed those characteristics as well. The excess perinatal deaths in the Chinese series might be explained by the different levels of perinatal health care in the two countries.

Biochemical characterization of recombinant factor IX.

Mature human factor IX is a 55,000-d glycoprotein with a modular domain structure and numerous posttranslational modifications. A recombinant form of human factor IX (rFIX) has been produced from a Chinese hamster ovary cell line that was engineered for high-level protein processing and expression. To ensure that the recombinant molecule contains the requisite structural and functional features of the plasma-derived form, rFIX was subjected to detailed biochemical and biophysical characterization. The laboratory studies showed that the posttranslational modifications and primary, secondary, and tertiary structures of rFIX were similar to those of plasma-derived factor IX (pdFIX). In addition, rFIX displayed a high degree of purity and a product release specification for specific activity that is > or = 200 IU/mg.

Mortality and hospitalizations of 24-year-old members of the low-birthweight cohort in northern Finland.

We followed 11,355 children born in 1966 up to the age of 24 years to investigate the impact of birthweight on long-term outcomes of death and development. As expected, low-birthweight (LBW, 1500-2,499 gm) children showed a higher risk of death than normal-birthweight (NBW, > or =2,500 gm) children before the age of 15 years. Rate ratios for LBW children ranged from 25 for the first week of life to 2.5 for ages 5-14 years. Rate ratios of very-low-birthweight (VLBW, <1,500 gm) children for the first year of life were 10 times higher than those of LBW children. Similarly, for the first 14 years of life, surviving LBW children experienced a higher risk of being hospitalized as well as staying in hospitals for over 2 weeks if they were hospitalized. The LBW children still had a higher risk of receiving sick pensions at the age of 24 years.

Increased receptor sensitivity to neuropeptide Y in the hypothalamus may underlie transient hyperphagia and body weight gain.

Disruption of neural signaling by microinjection of a neurotoxin, colchicine (COL), in the ventromedial hypothalamus (VMH) of rats results in rapid and transient hyperphagia and body weight gain. Since neuropeptide Y (NPY) is a potent hypothalamic orexigenic signal and continuous NPY receptor activation by intracerebroventricular (icv) NPY infusion results in hyperphagia and obesity, we tested the hypothesis that altered NPYergic signaling may underlie the transient hyperphagia in COL-injected rats. Immediately following COL (4 microg) microinjections in the ventromedial nucleus (VMN) rats displayed hyperphagia both during the lights-on and lights-off periods. Concomitant with hyperphagia, preproNPY mRNA levels in the arcuate nucleus and NPY levels in the paraventricular nucleus decreased in a time-dependent manner. However, food intake in response to intracerebroventricular injections of NPY (29, 117 and 470 pmole) was significantly higher in COL-injected rats and the latency to initiation of feeding was markedly reduced as compared to controls. The smallest dose of NPY which was virtually ineffective in control rats, evoked near maximal intake in COL-injected rats. This enhanced response lasted for only 4 days paralleling the transient hyperphagia. The NPY Y1 receptor antagonist 1229U91 (5 or 30 microg/rat, icv) significantly suppressed feeding in COL-treated rats thereby indicating that hyperphagia in these rats was dependent upon endogenous NPY. Overall, these studies demonstrate that not only high levels, but low levels of NPY may also result in hyperphagia and increased body weight and this hyperphagia may be attributed to the rapid development of NPY Y1 receptor hypersensitivity.

Maternal menstrual history and small-for-gestational-age births in a population-based Chinese birth cohort.

The relationship between maternal menstrual history and small-for-gestational-age (SGA) births is examined in a population-based birth cohort of 5291 mothers with singleton births and 28 completed gestational weeks in Qingdao, China, in 1992. Multivariate analyses suggest that the length of the menstrual cycle and age at menarche had independent effects on SGA after controlling for sex of the infant, physical size of the mother, blood pressure at the first antenatal visit, parity, gestational week at the first antenatal visit, number of antenatal visits, number of previous spontaneous abortions and maternal educational attainment and occupation. The relative risk (RR) of SGA among thin mothers (BMI < = 21) is 1.63 (95% confidence interval (CI) 1.16-2.29) if the mother's menarche started after 14 years (reference to < = 14 years), 1.75 (95% CI 0.96-3.20) if the mother's menstrual cycle was 29-30 days and 2.92 (95% CI 1.42-6.03) if the menstrual cycle > = 31 days ( < = 28 as a reference category for both). Among normal weight mothers (BMI 21-24), only a menstrual cycle equal or longer than 31 days still had a significant effect on SGA (RR 2.17, 95% CI 1.18-4.08), and among fat mothers no significant effects were observed. The results encourage further evaluation of the association between maternal menstrual background and fetal development among other ethnic populations and exploration of the possible biological mechanisms behind this relationship.

Effects of direct current on dog liver: possible mechanisms for tumor electrochemical treatment.

Mechanisms of tumor electrochemical treatment (ECT) were studied using normal dog liver. Five physical and chemical methods were used. Two platinum electrodes were inserted into an anesthetized dog's liver at 3 cm separation. A voltage of 8.5 V direct current (DC) at an average current of 30 mA was applied for 69 min; total charge was 124 coulombs. Concentrations of selected ions near the anode and cathode were measured. The concentrations of Na+ and K+ ions were higher around the cathode, whereas the concentration of Cl- ions was higher around the anode. Water contents and pH were determined near the anode and the cathode at the midpoint between the two electrodes and in an untreated area away from the electrodes. Hydration occurred around the cathode, and dehydration occurred around the anode. The pH values were 2.1 near the anode and 12.9 near the cathode. Spectrophotometric scans of the liver sample extract were obtained, and the released gases were identified by gas chromatography as chlorine at the anode and hydrogen at the cathode. These results indicate that a series of electrochemical reactions take place during ECT. The cell metabolism and its environment are severely disturbed. Both normal and tumor cells are rapidly and completely destroyed in this altered environment. We believe that the above reactions are the ECT mechanisms for treating tumors.

Decreased beta2-adrenergic receptor density on peripheral blood mononuclear cells in myasthenia gravis.

Beta2-adrenergic receptors (beta2AR) are present on both lymphocytes and skeletal muscle cells. Antibodies and T cells that react with these receptors are present in patients with myasthenia gravis (MG). Immune reactivity against the beta2AR may thus modify both the immune and the muscle functions in MG. In this study, we analysed the density and affinity of beta2AR on peripheral blood mononuclear cells using a radioligand binding assay. The density (Bmax) of the receptor on cells from patients with MG was significantly lower than that on cells from patients with other neurological disorders and healthy individuals. The affinity (Kd) of the receptor and the concentration of the second messenger, cAMP, in the cells did not differ between the groups. Serum antibodies against beta2AR were demonstrated in 22% of 27 MG patients vs. 0% of 26 healthy controls. Incubation of cells with serum or purified IgG containing antibodies against the beta2AR resulted in a decline in ligand binding of the receptor in samples from three out of five patients. Thus, this study suggests that a downregulation of the beta2AR may occur in MG. This downregulation might be of importance in the patho-genesis of the disease and its symptoms.

Central ANG II receptor involved in carotid sinus reflex resetting in chronically stressed rats.

The performance of carotid sinus baroreceptor reflex (CSR) was characterized in chronically stressed rats by changing intracarotid sinus pressure (ISP) and constructing ISP-MAP (mean arterial pressure) relationship curve. The role of central angiotensin II (ANG II) receptors in the changes of CSR performance induced by chronic stress was determined. Rats were subjected to foot-shock stress for two weeks. The carotid sinus was isolated from the systemic circulation and the ISP changed in a stepwise manner. The results showed that in chronically stressed rats, ISP-MAP relationship curve shifted upward, the set point was significantly higher than that obtained from the unstressed group, and the reflex gain and the MAP range were significantly smaller than those in unstressed rats. After intracerebroventricular injection of saralasin (20 ng), MAP range was augmented and the set point decreased significantly. Injection of vehicle did not lead any significant differences between the parameters of the reflex measured before or after injection in either the stressed or the unstressed rats. Furthermore, administration of ANG II (10 microg) induced a significant increase in the set point and decrease in the reflex gain in the unstressed rats. The responses of CSR to ANG II were completely blocked by pretreatment of saralasin. These findings suggest that chronic stress could induce the decreased CSR function in the normotensives and central ANG II receptors involved in the resetting of CSR in the chronically stressed rats.

Polymorphism of mitochondrial DNAs of Yunnan domestic water buffaloes, Bubalus bubalis, in China, based on restriction endonuclease cleavage patterns.

Restriction endonuclease cleavage patterns of mitochondrial DNA(mtDNA) of three local types of Yunnan native water buffalo were analyzed using 18 enzymes which recognize six nucleotides. Among the 12 animals analyzed, 3 of 18 enzymes, BamHI, EcoRI, and Scal, revealed polymorphisms. Three mtDNA types were identified. The results indicate that a relatively low level of mtDNA variation exists in Yunnan domestic water buffaloes. The origin of Chinese buffalo derived from Yunnan province of China is discussed.

The interactive language of the hypothalamus for the gonadotropin releasing hormone (GNRH) system.

The enormous diversity in neurochemical signals employed within the network of afferents to GnRH neurons is well-documented. An examination of novel and accumulating knowledge on the operation of these messengers indicates the presence of an interactive language governing GnRH secretion. The basic operational structures identified to date to affirm this interactive form of communication summarized in this review are the following: (i) the demonstration of interconnections within various components of the afferent network; (ii) coexistence and possible co-release of excitatory and inhibitory neurotransmitters/neuromodulators; (iii) co-action of various messengers at synaptic targets, and (4) modulation by gonadal steroids of the synthesis and release of signals and their receptors, and induction of synaptic plasticity for the timely relay of signals for GnRH secretion. Unraveling the molecular sequelae that promote this interactive communication to elicit periodic GnRH secretion is now a new challenge.

Role of pyruvate carboxylase in facilitation of synthesis of glutamate and glutamine in cultured astrocytes.

CO2 fixation was measured in cultured astrocytes isolated from neonatal rat brain to test the hypothesis that the activity of pyruvate carboxylase influences the rate of de novo glutamate and glutamine synthesis in astrocytes. Astrocytes were incubated with 14CO2 and the incorporation of 14C into medium or cell extract products was determined. After chromatographic separation of 14C-labelled products, the fractions of 14C cycled back to pyruvate, incorporated into citric acid cycle intermediates, and converted to the amino acids glutamate and glutamine were determined as a function of increasing pyruvate carboxylase flux. The consequences of increasing pyruvate, bicarbonate, and ammonia were investigated. Increasing extracellular pyruvate from 0 to 5 mM increased pyruvate carboxylase flux as observed by increases in the 14C incorporated into pyruvate and citric acid cycle intermediates, but incorporation into glutamate and glutamine, although relatively high at low pyruvate levels, did not increase as pyruvate carboxylase flux increased. Increasing added bicarbonate from 15 to 25 mM almost doubled CO2 fixation. When 25 mM bicarbonate plus 0.5 mM pyruvate increased pyruvate carboxylase flux to approximately the same extent as 15 mM bicarbonate plus 5 mM pyruvate, the rate of appearance of [14C] glutamate and glutamine was higher with the lower level of pyruvate. The conclusion was drawn that, in addition to stimulating pyruvate carboxylase, added pyruvate (but not added bicarbonate) increases alanine aminotransferase flux in the direction of glutamate utilization, thereby decreasing glutamate as pyruvate + glutamate --> alpha-ketoglutarate + alanine. In contrast to previous in vivo studies, the addition of ammonia (0.1 and 5 mM) had no effect on net 14CO2 fixation, but did alter the distribution of 14C-labelled products by decreasing glutamate and increasing glutamine. Rather unexpectedly, ammonia did not increase the sum of glutamate plus glutamine (mass amounts or 14C incorporation). Low rates of conversion of alpha-[14C]ketoglutarate to [14C]glutamate, even in the presence of excess added ammonia, suggested that reductive amination of alpha-ketoglutarate is inactive under conditions studied in these cultured astrocytes. We conclude that pyruvate carboxylase is required for de novo synthesis of glutamate plus glutamine, but that conversion of alpha-ketoglutarate to glutamate may frequently be the rate-limiting step in this process of glutamate synthesis.

Effect of alum on free-living and copepod-associated Vibrio cholerae O1 and O139.

The effects of alum [KAl(SO4)2] on free-living and copepod-associated Vibrio cholerae O1 and O139 were investigated by using plate counts and immunofluorescence direct viable counting (DVC). Growth of alum-treated cells in 0.5/1000 Instant Ocean seawater was inhibited, i.e., no growth was obtained on Luria-Bertani (LB) agar or thiosulfate-citrate-bile salt-sucrose (TCBS) agar. However, a significant number of the inhibited cells maintained viability, as measured by DVC. In comparison, a significant number of V. cholerae organisms associated with zooplankton, most of which were crustacean copepods, were viable but nonculturable, with only a small number of cells retaining culturability on LB and TCBS agar. Both DVC and viable plate counts (CFU) were significantly greater for V. cholerae O1 and O139 associated with zooplankton than for V. cholerae in water alone, i.e., without copepods. It is concluded that alum is an effective coagulant but not an effective killing agent for V. cholerae and that association with copepods offers protection for V. cholerae O1 and O139 against alum and chlorine treatments.

Specificity of the high-mannose recognition site between Enterobacter cloacae pili adhesin and HT-29 cell membranes.

Enterobacter cloacae has been implicated as one of the causative agents in neonatal infection and causes a septicemia thought to be initiated via the gastrointestinal tract. The adhesion of radiolabeled E. cloacae to HT-29 cells was concentration and temperature dependent and was effectively blocked by unlabeled bacteria or by millimolar concentrations of alpha-mannosides and micromolar concentrations of high-mannose oligosaccharides. A variety of well-characterized mannose oligosaccharides were tested as inhibitors of adhesion. The best inhibitor was the Man9(GlcNAc)2-tyrosinamide, which was considerably better than other tyrosinamide-linked oligosaccharides such as Man7(GlcNAc)2, Man6(GlcNAc)2 or Man5(GlcNAc)2. Further evidence that the bacteria preferred Man9(GlcNAc)2 structures was obtained by growing HT-29 cells in the presence of glycoprotein processing inhibitors that block mannosidase I and increase the amount of protein-bound Man9(GlcNAc)2 at the cell surface. Such cells bound 1.5- to 2-fold more bacteria than did control cells. The adhesin involved in binding to high-mannose structures was purified from isolated pili. On sodium dodecyl sulfate-gels, a 35-kDa protein was identified by its specific binding to a mannose-containing biotinylated albumin. The amino acid sequences of several peptides from the 35-kDa subunit showed over 85% identity to FimH, the mannose-specific adhesin of Salmonella typhimurium. Pili were labeled with 125I and examined for the ability to bind to HT-29 cells. Binding showed saturation kinetics and was inhibited by the addition of Man9(GlcNAc)2-tyrosinamide but not by oligosaccharides with fewer mannose residues. Polyclonal antibody against this 35-kDa protein also effectively blocked adhesion of pili or E. cloacae, but no effect was observed with nonspecific antibody. These studies demonstrate that the 35-kDa pilus subunit is a lectin whose specificity is directed toward Man, (GlcNAc)2 oligosaccharides.

Growth factor-mediated K+ channel activity associated with human myeloblastic ML-1 cell proliferation.

ML-1 cell proliferation is dependent on the presence of serum growth factors. Removing serum from the culture medium results in growth arrest and promotes differentiation. In this study, we found that a 4-aminopyridine-sensitive K+ channel was highly expressed in proliferating ML-1 cells and significantly diminished in G1-arrested ML-1 cells induced by serum deprivation but was restored within 30 min in these cells with addition of 10% fetal bovine serum (FBS) or 5 ng/ml epidermal growth factor (EGF). Intracellular adenosine 3',5'-cyclic monophosphate (cAMP) levels, but not guanosine 3',5'-cyclic monophosphate, were significantly increased in serum-deprived cells stimulated by FBS or EGF, and the effects of FBS and EGF on the channel activation were mimicked by exogenous cAMP. In inside-out patches, K+ channel activity was significantly increased by the cAMP-dependent protein kinase catalytic subunit, whereas the effect of EGF on K+ channel activation was blocked by Rp-8-(4-chlorophenylthio)adenosine 3',5'-cyclic monophosphothioate. Together, our results demonstrate that serum growth factors stimulate K+ channel activity in proliferation of ML-1 cells through protein kinase-induced phosphorylation and suggest an important molecular mechanism for serum growth factor-stimulated mitogenesis in ML-1 cells.

Several new targets of antitumor agents.

Alpha-fetoprotein (AFP), as a hepatoma-promoting factor, has become a new target of anti-hepatoma agents. It is a new approach for the treatment of tumors to inhibit or block oncogene expression. Informational drugs are being developed for gene therapy applications as inhibitors of oncogene expression. The induction of tumor cell differentiation is another new strategy of drug therapy of tumors. Common action mode of many antitumor drugs is to induce apoptosis of tumor cells. Suicide genes, as targeting therapy of tumors, improve the present chemotherapy, exhibiting broad application prospects.
