Pregnancy-associated uterine granulated metrial gland cells in mutant and transgenic mice.

PROBLEM: Granulated metrial gland (GMG) cells are pregnancy-specific uterine lymphocytes found in rodents. The lineage relationships of GMG cells are incompletely defined, although genetic and immunophenotyping studies suggest membership in the natural killer (NK) cell lineage. Pregnancy-specific functions have been postulated for GMG cells, but no successful depletion of these cells has been achieved that would permit assessment of any critical roles that might influence gestational outcome. METHOD: Routine histological methods for light microscopy were used to assess implantation sites from wild-type mice and mice of the following genotypes: tgE26, mi/mi, and p53 knockout. RESULTS: GMG cells are transient, histamine-negative cells found in the pregnant uteri of most mice. Pregnancies in the NK and T-cell-deficient tgE26 were characterized by infrequent GMG cells, very small placentae, and an elevated rate of fetal and perinatal mortality. In term placentae of mi/mi with dystocia. GMG cells were found in a new location along the plane of placental separation, and they appeared degranulated. A normal life-history was observed for GMG cells in p53 knockout mice. CONCLUSION: Pregnancies in mutant and transgenic mice indicate that GMG cells are natural killer cells and have critical roles in placental development and pregnancy success. The disappearance of GMG cells at term is independent of p53 gene expression.

cis Elements and trans factors are both important in strain-specific regulation of the leukotoxin gene in Actinobacillus actinomycetemcomitans.

Actinobacillus actinomycetemcomitans, the etiologic agent of localized juvenile periodontitis, produces a potent leukotoxin that kills human neutrophils. The production of leukotoxin RNA can vary more than 50-fold among isolates of A. actinomycetemcomitans, and strains expressing high levels of leukotoxin RNA are most often found at sites of periodontal disease. To assess the relative contributions of transcription factors and promoter sequences in setting the disparate levels of leukotoxin RNA found, we have undertaken classical cis/trans analyses. First, the leukotoxin promoter regions from moderately leukotoxic (Y4) and minimally leukotoxic (ATCC 33384) strains of A. actinomycetemcomitans were cloned, sequenced, and compared with the previously sequences leukotoxin promoter region of the high-producer strain JP2. The Y4 and ATCC 33384 promoter regions each contain a 528-bp segment that is absent from JP2. Interestingly, the analysis of various deletion constructs in A. actinomycetemcomitans indicated that Y4, despite the large insertion, initiates leukotoxin RNA synthesis at the same promoter as JP2 does. To perform cis/trans analyses, these three leukotoxin promoter regions were cloned into a plasmid upstream of the reporter gene beta-galactosidase. Each plasmid was transformed into JP2, Y4, and ATCC 33384, and the beta-galactosidase levels were determined. The results indicated that the sequences responsible for down-regulating leukotoxin RNA levels in Y4 relative to JP2 are found within the transcribed region of the Y4 leukotoxin operon. Importantly, in ATCC 33384, strain-specific trans factors and promoter sequence differences are equally significant in determining the lower levels of leukotoxin RNA. We hypothesize that either strain ATCC 33384 has a negative regulatory protein (which is missing or mutated in JP2/Y4) or that JP2 and Y4 carry an activator that is missing or mutated in ATCC 33384.

Human enhancer of filamentation 1, a novel p130cas-like docking protein, associates with focal adhesion kinase and induces pseudohyphal growth in Saccharomyces cerevisiae.

Budding in Saccharomyces cerevisiae follows a genetically programmed pattern of cell division which can be regulated by external signals. On the basis of the known functional conservation between a number of mammalian oncogenes and antioncogenes with genes in the yeast budding pathway, we used enhancement of pseudohyphal budding in S. cerevisiae by human proteins expressed from a HeLa cDNA library as a morphological screen to identify candidate genes that coordinate cellular signaling and morphology. In this report, we describe the isolation and characterization of human enhancer of filamentation 1 (HEF1), an SH3-domain-containing protein that is similar in structure to pl30cas, a recently identified docking protein that is a substrate for phosphorylation by a number of oncogenic tyrosine kinases. In contrast to p130cas, the expression of HEF1 appears to be tissue specific. Further, whereas p130cas is localized predominantly at focal adhesions, immunofluorescence indicates that HEF1 localizes to both the cell periphery and the cell nucleus and is differently localized in fibroblasts and epithelial cells, suggesting a more complex role in cell signalling. Through immunoprecipitation and two-hybrid analysis, we demonstrate a direct physical interaction between HEF1 and p130cas, as well as an interaction of the SH3 domain of HEF1 with two discrete proline-rich regions of focal adhesion kinase. Finally, we demonstrate that as with p130cas, transformation with the oncogene v-abl results in an increase in tyrosine phosphorylation on HEF1, mediated by a direct association between HEF1 and v-Abl. We anticipate that HEF1 may prove to be an important linking element between extracellular signalling and regulation of the cytoskeleton.

Synchrony in binary-oscillator networks with local couplings.

Since synchronous oscillations in the visual cortex may be responsible for some features of visual scene, many works have been done to study the dynamics of oscillatory neural networks. Most of the oscillator network models rely on a global connection to reach synchronization. Here we propose a class of neural network models based on a simplified binary-oscillator, and find that the neurons of networks with local couplings will be in global synchrony under some criteria.

Measurement of ABH blood group substances in human saliva by immunoassay using artificial antigens as standard substances.

Introducing artificial antigens (trisaccharide-albumin complexes for A, B and H activities) as reference substances for immunoassay, blood group substances, A, B, and H in human saliva were measured. A good correlation between hemagglutination inhibition titers and concentrations of A, B, and H in saliva was demonstrated. Since the anti-H 1E3 used showed the specificity for both H type 1 and H type 2 chains and anti-H 224 for H type 2 chain, we measured H type (1 and 2) and H type 2 separately, and found 10- to 20-fold higher concentration of H type (1 and 2) compared with that of H type 2 in human saliva. The H type 2 was not found in all the saliva samples from nonsecretors irrespective of ABO subtypes. The results suggest that about 90% of A and B blood group substances in human saliva is also type 1 chain.

[Activation of proto-oncogenes induced by MNNG on primary culture of human gastric epithelium and immortalized human gastric epithelial cell line].

Epidemiologic study has shown the association of nitrosamide compounds with the high incidence of stomach cancer in south China. To study the mechanism of gastric carcinogenesis, we have established an immortalized human gastric epithelial cell line GES-1. GES-1 cells and the normal gastric tissues were treated with different concentrations of MNNG for 24 hours. Point mutation at codon 12 of c-Ha-ras gene was found in cells and tissues (43%) as demonstrated by PCR-RFLP. Rearrangement of c-met gene and amplification of c-erbB2 gene were detected by Southern blot assay on the MNNG treated GES-1 cells. The results indicate that MNNG treatment was intimately associated with the activation of certain oncogenes. H-ras and c-met genes, serving as early targets of carcinogens may play important role in the carcinogenesis of human gastric epithelial cells.

Subtype B isolates of human immunodeficiency virus type 1 detected in Australia.

The human immunodeficiency virus type 1 (HIV-1) can be subtyped on the basis of nucleotide sequence variability. Knowledge of circulating HIV-1 genotypes or subtypes allows understanding of the origin and spread of HIV-1 in different geographical regions, and is required for rational vaccine development. A study was undertaken to determine the predominant HIV-1 subtype in Australia. Part of the HIV-1 envelope gene (including the variable domain, V3) was sequenced directly from DNA extracted from peripheral blood mononuclear cells of 17 HIV-1 seropositive people in Sydney, Australia. Phylogenetic analysis based on nucleotide sequence suggested that all patients (including individual cases acquired in New Zealand, Papua New Guinea and Thailand) were infected with HIV-1 subtype B. Octapeptides from the HIV-1 envelope V3 loop tip indicated variation but included a predominance of the most common subtype B octapeptides HIGPGRAF (4 cases), NIGPGRAF (3 cases) and PIGPGRAF (1 case). These data suggest that subtype B is the major HIV-1 strain in Australia (and probably in New Zealand and Papua New Guinea), although the importation of HIV-1 acquired overseas is likely to lead to the detection and dissemination of other subtypes in Australia.

An HIV-1 infected long-term non-progressor (LTNP): molecular analysis of HIV-1 strains in the vpr and nef genes.

We describe a long-term non-progressive injecting drug user (IDU) who was infected with human immunodeficiency virus type-1 (HIV-1) in 1984, and has survived with stable CD4+ T-cell counts (> 800/microliters blood) without any acquired immune deficiency syndrome (AIDS) related illness. With a goal to investigate the molecular nature of HIV-1 strains infecting this patient, we amplified the nef and vpr genes directly from the fresh uncultured peripheral blood mononuclear cells (PBMCs), and carried out co-culture studies. Sequence analysis of the nef gene (from 1994 samples) showed no deletions (as has been previously reported) expected for a 7 base pair duplication at the C-terminus which prematurely terminated the nef reading frame, whereas even after repeated attempts the nef gene could not be amplified from the 1992 PBMC samples. In contrast, the vpr gene (from 1992 and 1994 samples) revealed two distinct quasispecies with no apparent defects. We observed five amino acid substitutions, between residues 83-90, at the C-terminus which has been recently implicated in G2 cell cycle arrest as an early step to HIV-1 infection. In the light of recent evidence on the role of nef gene defects/attenuations in long-term survival of HIV-1 infected patients, it may be that the nef gene defect created by gene duplication, which eliminated the cysteine-206 crucial in disulfide bond formation, may play a role in chronic HIV-1 infection in this patient. These data further suggest that deletions in the nef gene may not be the only reason for long-term non-progression of HIV-1 infection in some individuals, but the gene defects like duplication and subtle mutations in the functional motifs of both nef and vpr genes may confer similar protection in HIV-1 infected patients surviving for longer periods of time with stable CD4 counts.

Experimental study on spontaneous DNA synthesis of tonsillar lymphocytes in chronic recurrent tonsillitis and focal tonsillitis.

The immune function of tonsils and the immunological mechanisms of focal tonsillitis are still controversial. We do have some knowledge about the structure of tonsillar cells, but their functional status is still not clear. In 40 cases of chronic recurrent tonsillitis and 20 cases of focal tonsillitis with glomerular nephritis, cell culture of tonsillar lymphocytes was removed in vitro and 3H-tymidin incorporation applied to investigate the spontaneous DNA synthesis of tonsillar lymphocytes. The results demonstrated that: i) In chronic tonsillitis, spontaneous DNA synthesis of tonsillar lymphocytes in children is significantly stronger than that in adults (19,946 +/- 5,612 vs 9,216 +/- 5,702 cpm) (p <0.01). ii) Spontaneous DNA synthesis of tonsillar lymphocytes in focal tonsillitis is significantly stronger than that in the chronic recurrent tonsillitis in patients of the same age (25,307 +/- 12,231 cpm vs 12,455 +/- 7,914 cpm) (p <0.01). This indicates that the tonsillar lymphocytes are active in focal tonsillitis, can produce large amounts of memory B-cell clones, which reach other parts of the body through the blood and cause autoimmune reaction which can be blocked by tonsillectomy, thus curing the systemic disease.

Influence of lipopolysaccharide and interleukin-2 on proliferation and synthesis of sulfated macromolecules in cultured rat glomerular epithelial cells.

OBJECTIVE: To investigate the effects of lipopolysaccharide (LPS) and human recombinant interleukin-2 (hrIL-2) on the synthesis of sulfated macromolecules in cultured rat GECs. METHODS: With 3H-thymidine (3H-TdR) and 35S-Na2SO4 dual isotope labelling technique, the proliferation and synthesis of sulfated macromolecules in cultured rat GECs were estimated according to the 3H-TdR uptake and 35SO4(2-)-contents in cell layers respectively. RESULTS: Both LPS (6.25-50.0 micrograms/L) and hrIL-2 (2.5-10.0 kU/L) decreased the uptake of 3H-TdR by GECs (P < 0.05). As for incorporation of 35SO4(2-) into cell layers, the results that both means and adjusted means in hrIL-2 groups were lower than those in controls (P < 0.05) implied that hrIL-2 could inhibit the synthesis of basement membrane HSPG while LPS hindered incorporation through its inhibitory effect on cell proliferation. CONCLUSIONS: We may infer from these results that LPS-like proinflammatory mediators could increase the subpopulation of large pores in capillary wall through the reduction of cell-associated HSPG, and IL-2-like cytokines could contribute to the development of proteinuria through its inhibitory effect on the production of basement membrane HSPG in immune-mediated glomerular disorders.

[Image morphological quantitative analysis of pathological changes of intra-acinar arteries in experimental pulmonary artery hypertension and outcomes of treatment].

The relative ratios of vessel lumen area/total area (EA/TA), vessel wall area/total area (MA/TA) and vessel wall thickness/vessel external diameter (WD/TD) of intra-acinar arteries were studied with image analysor in 35 rats of experimental pulmonary artery hypertension and outcomes of treatment. All the rats were divided into control group (R0), hypertension group (R1) and treated groups (R2), (R3) and (R4). The results showed relative ratios of EA/TA decreased significantly in the palmonary artery hypertension group in comparison with the control and treated groups, while the values of WA/TA and WD/TD increased significantly. The results may be valuable for morphological quantitative study, diagnosis of pulmonary artery hypertension and prediction of the disease prognosis.

[Labeling DNA probe by polymerase chain reaction].

The use of polymerase chain reaction (PCR) for labeling probe has been demonstrated to offer various advantages including efficient labeling of DNA fragments as small as 72 bp, direct labeling of genomic DNA, and labeling with subnanogram amounts of input DNA. Therefore, a procedure for the nonradioactive labeling of chromasomal DNA 203 bp fragments of Helicobacter pylori with the hapten digoxigenin (Dig) by PCR has been developed. The results showed that the concentration of probe labeled by PCR was much higher than that by random primer labeled. PCR has the advantage of rapidity and economy. It is a very effective technique for synthesis of Dig-labeled DNA probe.

[Antiproliferative effects on Ph+ leukemic cells transfected with antisense bcr/abl fusion gene].

To investigate the possible roles that bcr/abl fusion gene plays in Ph-chromosome positive leukemia cells, in this study the recombinant plasmids expressing antisense transcripts to bcr/abl fusion gene were transfected into K562 and BV173 cell lines by lipofectin reagent. Southern and Northern bilot hybridizations proved the integration and expression of exogenous DNA in target cells. Antisense transcripts to bcr/abl inhibited proliferation and colony-forming ability of target cells. Through flow cytometric cell cycle analysis it was found that during the early period of transfection decrease of cells in G0/G1 and S phases and increase in G2 phase were observed, suggesting block of G2 progression. Later, cells in G0/G1 phase decreased further and retardation of S and G2 progressions was obvious. The results also indicated that proliferative inhibition of target cells was accompanied by genomic DNA degradation.

[Use of an anorectal driving device in the sphincter-saving operation for low rectal carcinoma].

UNLABELLED: The anastomosis with tubular anastomotic instrument for the surgical resection of the low rectal carcinoma with preserved anus is difficult to perform because of the deep location of the tumor and retraction of the paraproctium to the pelvic floor. From 1986 a self-made anorectal driving device was applied to facilitate anastomosis with the tubular anastomotic instrument. Ninety-six cases of low rectal carcinoma were so treated in 1986-1990. Eighty-five cases in 1980-1985 treated by tubular anastomosis without the use of the driving device served as the control. The success rate of anastomosis increased from 45.9% to 77.1% (P < 0.01), incidence of anastomotic fistula decreased from 11.6% to 3.1% (P < 0.01), the average operation time was reduced from 211 to 183 minutes (P < 0.01). CONCLUSION: The pelvic and anoretal tissue can be pushed upwards by this device, which facillitates complete dissection of the tumor and anastamosis by tubular anastomotic instrument.

[Effect of electroacupuncture on cholecystokinin gene expression in rat hippocampus during penicillin-induced epileptic seizure].

Northern Blot and hybrization in situ techniques were used to investigate the effect of electroacupuncture (EA) on the changes of cholecystokinin (CCK) mRAN levels of the hippocampus in rat penicillin-induced epilepsy model. Epilepsy can significantly increase CCK mRNA levels in dentate gyrus and CA3 areas of hippocampus in diencephalic sections after penicillin-induced seizure, whereas EA not only can attenuate the seizure behaviors and EEG changes, but also can decrease the increase of CCK mRNA contents induced by the seizure. However, in the subiculum, dentate gyrus and CA3 areas of mesencephalic sections of rat hippocampus, EA can further increase the enhancement of CCK mRNA concentration induced by penicillin-induced seizure. The results suggest that EA inhibitory effects on the seizure's behaviors and epileptiform activities may be related to the alteration of CCK gene expression in the different area of hippocampus.

[Extraovarian peritoneal serous papillary carcinoma: a clinicopathologic analysis of 11 cases].

OBJECTIVE: To investigate the clinical features, diagnostic criteria, treatment methods and prognosis of extraovarian peritoneal serous papillary carcinoma (EPSPC). METHOD: Eleven cases histologically confirmed as EPSPC and treated in our hospital from Jan. 1989 to May, 1994 were retrospectively analysed. RESULT: All patients with EPSPC were given cytoreductive surgery followed by systemic chemotherapy of PAC (carboplatin, Adriamycin, cyclophosphamide) regimen. The interval between chemotherapeutic treatments was one month, and the whole course of chemotherapy lasted at least 2-3 years. All patients except 3 were followed up, and the mean survival time reached 17.1 months. CONCLUSION: Due attention should be paid to differential diagnosis of EPSPC. The prognosis may be improved by cytoreductive surgery and chemotherapy of the PAC regimen.

[Synthesis and distribution of endothelin in rats with pulmonary hypertension induced by chronic hypoxia].

OBJECTIVE: To study the synthesis and distribution of endothelin (ET) in the rat model of pulmonary hypertension (PH). METHOD: By chronic hypobaric hypoxia (10%-10.5% O2, 4-18 d), a stable rat model of PH was established. The plasma ET level in the rat was measured by radioimmunoassay and the ET-like immunoreactivity in the rat's lung tissue was observed morphologically by immunohistochemical staining. RESULTS: The concentrations of plasma ET were significantly higher in the hypoxic rat model than in control animals (P < 0.01), but there were no significant differences in different time groups of the hypoxic rats (P > 0.05). ET's synthesis was mostly localized within pulmonary vascular vessels sharply contrasted with normal rats in which there were little or no ET staining. CONCLUSION: The results suggested that hypoxia might be the main reason for promoting ET's synthesis and release.
