[Chromatographic-mass spectrometric analysis of sialic acids of disialosyllactosylceramides from the calf thymus].

Gas-liquid chromatography/mass spectrometry of the compounds obtained on methanolysis and deuterioacetylation of the permethylated gangliosides has demonstrated that disialosyllactosyl ceramides of calf thymus contain three components - NeuAc alpha 2----8NeuAc alpha 2----LacCer, NeuAc alpha 2----8NeuGc alpha 2----LacCer and NeuGc alpha 2----8NeuGc alpha 2----LacCer.

Molecular species analysis of lipids by metastable ion mass spectrometry.

A convenient universal and fast mass spectrometrical method designed for the molecular species analysis of natural lipids is described. In contrast to the commonly employed procedures the method does not require chemical or enzymatic treatment and does not include chromatographic steps. The method relies on the recognition of ions characteristic of individual molecular species in the mass spectrum of a particular lipid fraction, that is accomplished on the basis of metastable ion spectra. The efficiency of this approach is demonstrated with a variety of natural lipids: triglycerides, glycerophospholipids, sphingomyelin and ornithinolipids. The advantages and limitations of the method as well as possible further developments are discussed.

[New anthracycline antibiotics taurimycins A and B produced by a Str. tauricus No. 7036-1 culture].

Taurimycins A and B are new anthracycline antibiotics belonging to the group of glycosides. They were isolated from the mycelium of culture 7036-1 classified with Str. tauricus. The empiral formula of taurimycin A is C14H51O16N,M+813. The empirical formula of taurimycin B is C41H49O16N,M+811. The absorption maxima in the UV spectra of the antibiotics (in alcohol solutions) were observed at 235, 257, 292, 466, 482, 494, 515 and 527 nm. The IR spectra showed absorption bands at 3450, 2945, 2870, 1740 and 1610 cm-1. Hydrolysis of the antibiotics with hydrochloric acid resulted in formation of an aglycone identified as epsilon-pyrromycinone.

[A new antibiotic, corallomycin, from an Act. longispororuber].

A new antibiotic named corralomycin (C34H28O9) was isolated from the mycelium of culture 6795-38 classified as belonging to Act. longispororuber. The maximum UV absorption spectra of the antibiotic in an alcohol solution were observed at the wavelengths of 228, 268 and 420 nm, the absorption bonds of 3400, 2980, 2865, 1710, 1625, 1595, 1165, 880 and 780 cm(-1) were recorded in the IR spectrum. Two acetyl derivatives of corallomycin, i.e. diacetate (C38H32O11) and tetraacetate (C42H36O13) were prepared.

[Gangliosides of mouse liver and ascites hepatoma 22a].

The ganglioside composition of the ascite hepatoma 22a and that of mouse liver was compared. GM2 was shown to be the main ganglioside in both tissues; however, the gangliosides of the hepatoma differed from liver gangliosides by a higher amount of disialoganglioside GD1a and by contents of monosialoganglioside GM1 and disialoganglioside GD1b which are absent in normal mouse liver. Trisialogangliosides which are characteristic for normal liver cells were not detected in the hepatoma. The liver and hepatoma gangliosides differed also by the type of sialic acids. In the liver the individual ganglioside fractions belong to either the N-glycolyl- or the N-acetylneuraminyl series, whereas the ganglioside fractions of the hepatoma are characterized by simultaneous presence of both sialic acid types.

[Structure of carminomycins II and III].

Carminomycins II and III, the main components of the carminomycin complex were isolated in pure state. Their crystalline exalates and acetate of cardminomycin II were prepared. The PMR spectra of both carminomycins and the 13C-NMR spectra of the oxalates were obtained. The molecular weights of the antibiotics were determined by mass-spectrometry. On the basis of the PMR spectra it was shown that carminomycins II and III had similar structures and differed in the stereoisomerism of the nitrogen-free fragment linked to the amino sugar. This was confirmed by the 13C-NMR spectra. The above fragment (C7H15O3) is analogous to the fragment of baumycins A1 and A2 described earlier.

[New amino acid from the antibiotic, ristomycin A].

A new amino acid E was isolated from a mixture of the products of the reductive hydrolysis of ristomycin A 57% HJ in the presence of red phosphorus. Its characterization was performed. The new amino acid was formed as a result of reductive dehydration of the respective beta-oxyamino acid present in the native antibiotic and being completely destroyed during general acid or alkaline hydrolysis.

Diol lipids. Acylated ethyleneglycol glycosides. A new type of natural glycolipid.

A new type of glycolipid has been detected in ripening corn seeds. The presence of ethyleneglycol, galactose, glucose and fatty acids was shown by degradation studies. The products of alkaline deacylation were identified as galactosyl- and glycosylethyleneglycol by thin-layer chromatography and combined gas-liquid chromatography-mass spectrometry. The native ethyleneglycol galactolipid was isolated by distribution of the total lipids between heptane and 95% methanol, following silica gel column chromatography of the methanol soluble fraction. Analysis of the alkaline deacylation products of the isolated ethyleneglycol lipid as well as examination of the mass-spectrum of its tetraacetate showed the new lipid to have the structure of 1-acyl-2-(O-beta-D-galacto-pyranosyl)ethyleneglycol with palmitic, oleic and stearic acids as the main fatty acid components. The fragmentation patterns under electron impact of trimethylsilyl ethers of synthetic ethyleneglycol glycopyranosides and of 1-palmitoyl-2-(O-beta-D-tetraacetylgalactopyranosyl)ethyleneglycol are described.
