p37mos-associated serine/threonine protein kinase activity correlates with the cellular transformation function of v-mos.

A serine/threonine-specific protein kinase activity is closely associated with v-mos-encoded proteins. Experiments were conducted with several mutant forms of p37mos to determine whether or not the kinase function correlates with the biological activity of the mutant v-mos genes. Two mutants lacking cell transformation activity, one an arginine substitution for lysine-121 in the putative ATP-binding site and the other a 23-amino acid deletion from the C-terminal end of p37mos, had no kinase activity associated with their mutant proteins. However, a third mutant with reduced biological activity had drastically less kinase activity than the wild-type protein. The latter mutant was able to phosphorylate the kinase-inactive p37mos(Arg-121) protein in vitro. These results indicate that even though p37mos(Arg-121) can be phosphorylated in trans by other kinase molecules, it lacks the ability to phosphorylate itself in vitro. This provides a compelling argument that the protein kinase function of p37mos is an intrinsic property of the protein. Moreover, since the kinase function correlates with the cellular transformation activity of the v-mos gene, we predict that it is required for the biological activity of the v-mos gene.

Ankylosing spondylitis, HLA-B27, and Klebsiella: a study of lymphocyte reactivity of anti-Klebsiella sera.

Twenty three anti-Klebsiella antisera were tested for their cytotoxic activity and four for their binding capacity for peripheral blood lymphocytes (PBL) from patients with HLA-B27 positive ankylosing spondylitis (AS+B27+) and from B27 positive (AS-B27+) and B27 negative (AS-B27-) healthy individuals. None of the antisera showed specific activity against PBL from any particular group. The antisera tested included two anti-Klebsiella K43 sera provided by an Australian group, who have reported them to be specifically cytotoxic for AS+B27+ PBL, four antisera raised against a Klebsiella K43 strain provided by this group, and an antiserum from another group, who have reported it as having increased binding capacity for AS+B27+ and AS-B27+ PBL compared with AS-B27- PBL. The results of other workers who have attempted to reproduce the results of either group are reviewed and the possible reasons for the repeated failure to confirm the reported findings are discussed.

Induction of peroxisome proliferation in hepatocytes transplanted into the anterior chamber of the eye. A model system for the evaluation of xenobiotic-induced effects.

The effect of two hypolipidemic peroxisome proliferators, ciprofibrate and di(2-ethylhexyl)phthalate (DEHP), on hepatocytes transplanted into the anterior chamber of the eye was examined. Young male F-344 rats transplanted with dissociated hepatocytes were fed either a control diet or a diet containing 0.025% ciprofibrate or 2% DEHP. After 4-5 weeks of treatment, all rats were sacrificed and the transplanted liver cells and portions of homotopic liver were processed for light and electron microscopy and for immunofluorescence microscopy. Morphometric analysis of transplanted hepatocytes showed a ninefold and fivefold increase in the volume density of peroxisomes in ciprofibrate and DEHP-fed rats, respectively. Indirect immunofluorescence studies revealed a marked induction of peroxisome-associated enzymes. From these data it is concluded that hepatic peroxisome proliferators cross the blood aqueous humor barrier and the transplanted hepatocytes in the anterior chamber of the eye retain their ability to recognize and respond to peroxisome proliferators.

Effects of an antistress drug on the heat tolerance of broiler breeder hens.

12 broiler breeder hens aged 56 weeks were given 0.2 ml Geriforte/kg body weight via the drinking water. The experiment was carried out between May 25 and July 23. The plasma corticosteroid content of the treated animals was significantly reduced as compared with the 12 control hens (3.86 vs. 5.21 mg/dl during the dry season and 4.0 vs. 5.0 mg/dl during the rainy season). There was no significant influence on the number of eggs, feed consumption, respiration rate, body temperature, protein, glucose, sodium, and potassium content of the blood plasma.

Autoradiographic detection of diphtheria toxin resistant mutants in human diploid fibroblasts.

An autoradiographic procedure for the detection of diphtheria toxin (DT) resistant (DipR) mutants in human diploid fibroblast (HDF) cells has been developed. The assay is based on the observation that when HDFs from confluent cultures are seeded in medium containing 0.01 flocculating units/ml or higher concentration of DT, protein synthesis in sensitive cells is severely inhibited by 4-6 hr. If at this or later time, a radiolabeled protein precursor (eg, 3H-leucine) is added to the culture, it is almost exclusively incorporated into the resistant cells, which are then readily identified by autoradiography. The DipR cells can also be identified by labeling in the presence of 3H-thymidine, although a higher background is observed in these experiments. Reconstruction experiments using DipS and DipR HDFs show that the frequency of heavily labeled cells that are detected by autoradiography show an excellent correlation with the number of DipR cells added and to the number of DipR cells as detected by conventional colony forming assay. These studies provide strong evidence that the labeled cells identified by autoradiography are bona fide DipR mutants. The detection of DipR cells by autoradiography is apparently not affected by the presence of the sensitive cells in the mixtures. The spontaneous frequency of DipR cells in HDFs has been found to be in the range of 1-5 X 10(-6), and this increases in a dose dependent manner upon treatment with the mutagen ethyl methanesulfonate. These results indicate that the autoradiographic assay could be used for quantitative mutagenesis. Since the autoradiographic assay does not depend on cell division, it may prove useful in estimating the incidence of pre-existing mutations in cell populations that either do not divide or have very limited growth potential (eg, lymphocytes, muscle cells, neurons, senescent fibroblasts, etc).

Ambient air concentrations of asbestos fibers near the town of Asbestos, Québec.

Ambient air concentrations of asbestos fibers were measured during the period June 20 to August 12, 1980, at three locations; Danville, Asbestos, and Wottenville in the eastern townships of Quebec. Measurements were done with low-volume samplers and measurement periods extended from 3 to 13 days. Fiber counts were done by means of electron microscopy. Results indicate that overall fiber concentrations are related to atmospheric stability and to the direction of the prevailing wind with respect to the source of emission.

Mapping of regions in the caudal brain stem that produce stimulus-bound synchronization in the cortical EEG.

This study was aimed at filling the lacunae in our knowledge regarding the localization of the regions in the caudal brain stem that bring about cortical EEG synchronization on electrical stimulation and characteristic features of synchronized waves elicited from those regions. Studies were conducted on 40 encéphale isolé cats. Stimulation of ventromedial regions of the caudal brain stem, with low frequency, elicited stimulus-bound synchronized waves in the cortex which were more prominent ispsilaterally. On the other hand, low-frequency stimulation of dorsal and lateral areas produced synchronized waves which were either equally prominent on both sides, or more prominent on the contralateral side. The loci in the brain stem that produce synchronization were very specific. The induced synchronized waves showed amplitude modulation and did not outlast the train of stimuli. The results are further confirmation of the role of caudal brain stem structures in cortical EEG synchronization. They also provide information regarding the nature of cortical synchronization elicited from these brain stem structures.

Alterations in preoptic unit activity on stimulation of caudal brain stem EEG-synchronizing structures.

Effects of stimulation of EEG-synchronizing structures of the caudal brain stem reticular formation with low (6 Hz) and high (100 Hz) frequencies were studied on 42 neurons of the preoptic area, in encéphale isolé cats. Though low-frequency stimulation produced excitation and inhibition, the majority of the influenced neurons of the preoptic area had effects of the former type. Cortical EEG synchronization was also induced by low-frequency stimulation of the caudal brain stem. High-frequency stimulation, on the other hand, produced inhibition in a majority of the influenced neurons and induced, mostly, desynchronization of the cortical EEG. A majority of the neurons that were inhibited on high-frequency stimulation, remained unaffected during low-frequency stimulation. The influence induced on the preoptic area neurons by low-frequency stimulation could be obtained even in the absence of cortical EEG synchronization. Changes induced on preoptic neurons by high-frequency stimulation may be partially related to induced cortical EEG desynchronization.

Tonic activity of medial preoptic norepinephrine mechanism for body temperature maintenance in sleeping and awake rats.

The study was aimed at investigating the possible involvement of tonic activity of the adrenergic receptors in the medial preoptic area (mPOA) for maintenance of body temperature in rats. Differences in drug-induced changes in body temperatures during sleep and wakefulness--during the day and at night--were also investigated. Norepinephrine injected in the mPOA, produced a fall in body temperature, whereas phenoxybenzamine produced an increase. Saline and propranolol produced no alteration. Norepinephrine and phenoxybenzamine produced alterations in sleep-wakefulness also. These changes in sleep-wakefulness did not completely explain the thermal changes. The findings highlight the need for taking into account the basal activity and the time of application of drug in studies on thermo-regulation, using free moving animals.

Isolation of cDNA clones encoding a T-cell receptor beta-chain from a beef insulin-specific hybridoma.

cDNA clones coding for a T-cell receptor beta-chain were isolated from a beef insulin/IAd-reactive T-cell hybridoma, A20.2.15, and its complete amino acid sequence was deduced. This beta-chain gene utilizes the same V beta segment as a thymocyte beta-chain gene (86T1) and rearranges to the 5' proximal J-C locus (J beta 1-C beta 1), thus providing definitive evidence of a beta-chain gene from a functional hybridoma that utilizes C beta 1. The amino acid sequence of the V beta gene in A20.2.15 is identical to 86T1, thus suggesting the absence of somatic mutation in the beta-chain of A20.2.15. Southern blot analysis revealed a somatic DNA rearrangement unique to the A20.2.15 hybridoma. The expression of this gene in the hybridoma was confirmed by RNA dot hybridization. All 24 beta-chain clones so far isolated from the A20.2.15 hybridoma contained C beta 1, suggesting that the beta-chain gene of the fusion partner BW5147 is not expressed in the hybridoma.

32P-postlabeling analysis of peroxisome proliferator-DNA adduct formation in rat liver in vivo and hepatocytes in vitro.

Hepatocarcinogenic peroxisome proliferators, clofibrate, ciprofibrate, Wy-14643 or di(2-ethylhexyl)phthalate, were administered once daily by gavage to groups of three male F344 rats for 3 days and the rats were killed 2 h after the last dose. The DNA isolated from the livers was analyzed for possible carcinogen-DNA adducts, by a most sensitive 32P-postlabeling technique which can detect one adduct in 10(10) nucleotides. No adducts were detected by this assay in the DNA isolated from the livers of rats treated with any of the peroxisome proliferators. Adducts were also not found in the DNA of hepatocytes exposed in vitro to these peroxisome proliferators for 4 h in primary suspension cultures. Failure to detect peroxisome proliferator DNA adducts in hepatocytes under in vivo and in vitro conditions supports the contention that formation of a peroxisome proliferator-DNA adduct is not an essential step in the carcinogenesis by this novel class of carcinogens.

Inhibition of metabolic cooperation by phorbol esters in a cell culture system based on adenosine kinase deficient mutants of V79 cells.

In Chinese hamster V79 cells, stable mutants which are greater than 1000-fold resistant to the adenosine analog, tubercidin (Tubr mutants), and which exhibit high degree of cross-resistance to various other adenosine analogs, viz. toyocamycin, formycin A, 6-methylmercaptopurine riboside and 8-azaadenosine, have been isolated. The inability of the mutant cells to phosphorylate [3H]tubercidin and lack of adenosine kinase activity (AK- phenotype) in their cell extracts provide evidence that the mutant cells are unable to convert adenosine analogs into their toxic phosphorylated derivatives. When AK- cells are co-cultured with increasing numbers of parental V79 (AK+) cells in medium containing tubercidin, then due to metabolic cooperation between AK+ and AK- cells, a cell density-dependent decline in recovery of the resistant cells is observed. However, diphtheria toxin resistant (Dipr) mutants of V79 cells, which are altered in elongation factor-2, showed no similar cell density effect. Addition of 0.01 microgram/ml of phorbol ester, 12-O-tetradecanoyl-phorbol-13-acetate (TPA) to growth medium in these experiments markedly enhanced the recovery of the Tubr mutants, but it had no effect on the recovery of Dipr mutants, which suggests that TPA was enhancing recovery of AK- mutants by inhibiting metabolic cooperation between AK- and AK+ cells. Maximum effect of TPA on the recovery of AK- mutants (3- to 5-fold enhancement) was observed at a density of 6 X 10(5) V79 cells/60 mm diameter dish and it was independent of the particular adenosine analog that was used as selective agent. Studies with a number of different phorbol derivatives show that only those phorbol esters which show tumor promoting activity in the mouse skin system inhibited metabolic cooperation between AK+/AK- cells in a dose-dependent manner. An excellent correlation was observed in these studies between the relative tumor promoting activity of various phorbol esters, their relative binding affinities to the cell surface receptors, and the concentrations at which they inhibited metabolic cooperation in the AK-/AK+ cell system. The AK-/AK+ cell system thus provides a new system for examining the effect of tumor promoters on metabolic cooperation between cells.

Recognition of mos-related proteins with an antiserum to a peptide of the v-mos gene product.

A mos-specific antiserum was generated by injection of rabbits with a peptide predicted from the sequence of the v-mos gene of Moloney murine sarcoma virus (MuSV) strain 124. The peptide is composed of amino acids 37-55 (cyclized at the cysteine residues) conjugated to keyhole limpet haemocyanin. This serum [anti-mos(37-55c)] specifically recognized p37mos in MuSV-124 acutely infected NIH-3T3 cells, P85gag-mos in 6m2 cells, an NRK clone infected with the temperature-sensitive mutant (ts110) of Moloney MuSV, and P100gag-mos in 54-5A4 cells, an NRK clone infected with a spontaneous revertant of ts110. An additional protein of Mr 55000 from uninfected cells was recognized by this serum. Reactivity of the serum toward the v-mos-containing proteins and the 55K protein was completely inhibited by prior incubation with free peptide. The 55K protein was not recognized by antisera made from synthetic peptides prepared from the C-terminal eight or 12 amino acids of v-mos.
