Labelling patterns of carcinomas of the cervix and their precancerous stages after 3H-thymidine incorporation.

1. In-vitro labellings of non-keratinizing squamous cell carcinomas of the cervix uteri yielded greatly differing labelling indices (2.1% to 20.2%) and less greatly varying mitotic indices 0.2% to 1.0%). When the existing limits are taken into account, these values may be indicative of the growth intensity of tumours. 2. The labelling indices obtained for the individual layers of both the normal and atypical squamous epithelium of the portio uteri show that there takes place with the growing degree of dysplasia via the carcinoma in situ to the microinvasive carcinoma a progressive broadening of the zone of growth. Parallel to this, there is an increase in labelling intensity, although it is not possible to derive there from a criterion of malignancy in a given case.

[Evaluation of oral retinoid preventive action on human cutaneous epitheliomas (author's transl)].

Beneficial effects of oral retinoids in prophylaxis of epithelial neoplasias have been demonstrated by experimental works. In this study oral retinoid (RO 10-9359) was used in human dermatosis with high frequency of cutaneous malignancies: xeroderma pigmentosum with or without malignant neoplasias, Mibelli's porokeratosis with multifocal malignant degeneration, basal cell naevus syndrome with basal cell carcinomas, familial epithelioma of Ferguson-Smith and actinic keratosis. This work started in december 1977. Visible epitheliomas have been treated before trial. Initial dose of retinoid was 1 mg/kg daily, decreased depending on individual tolerance. Results were appreciated by comparising number of epitheliomas observed in years preceding retinoid therapy and number of them appearing during treatment; in two familial cases (basal cell naevus syndrom in twins and xeroderma pigmentosum in two brothers) comparison was made between treated and untreated patients. First results are very promising: an excellent response on solar keratosis is noted; epitheliomas occurrence seems actually to be prevented or delayed by oral retinoid therapy. Of course more numerous cases, a longer time, periods without treatment are necessary to confirm these interesting first results. On the other hand drug is not with this dose active on already constituted carcinomas.

[Erythema gyratum repens (Gammel's syndrome)].

A case of erythema gyratum repens is reported in 78 year old woman. The particularly typical eruption, mainly affecting the trunk, was associated with a squamous cell carcinoma of the esophagus. The paraneoplastic dermatosis cleared after radiotherapy of the cancer.

[Studies on lytic enzyme activities of bacteria and streptomycetes against Candida guilliermondii after partial purification with ammonium sulfate from shaking- and fermentation cultures (author's transl)].

Two bacteria and two streptomycetes strains, capable to degrade cell walls of Candida guilliermondii, were isolated from soils. Growth and enzyme production of these strains were investigated on synthetic and complex media in shaking culture and under fermentation conditions. The isolated microorganisms possessed lytic enzyme activities against living and autoclaved yeast cells. The lytic enzyme complex of the culture medium of microorganisms was partially purified by ammonium sulfate precipitation. After this purification step the lytic enzyme activity of the bacteria was higher that of the streptomycetes towards the control. The lytic enzyme activity of the bacteria was thermostable (30 min, 80 degrees C), that of streptomycetes thermolable (inactive - greater than 40 degrees C). The pH-optimum of the bacteria enzymes was in the range of 5.0-7.5, that of the streptomycetes 5.0-8.5.

Comparative Investigation of the Action of Several Chlorosis-inducing Herbicides on the Biogenesis of Chloroplasts and Leaf Microbodies.

Seedlings of Triticum aestivum L. and Secale cereale L. were grown in the presence of six different (five having different chemical structures) chlorosis-inducing herbicides: aminotriazole and its derivative SDR 5175, haloxidine, Sandoz 6706, fluometuron, and EMD-IT 5914. Concentrations were applied which allowed the leaves to grow normally and to reach normal total amino nitrogen contents but evoked a complete chlorosis (less than 6% chlorophyll). The effects of the herbicides on the accumulation of several chloroplast constituents and on peroxisomal and mitochondrial marker enzyme activities were compared. Wheat and rye, in general, gave very similar results, wheat being more sensitive to unspecific inhibitory effects.In dark-grown plants, the herbicides had no or only minor effects on the rRNA pattern and on enzyme activities of the leaves. In the light, all herbicides applied prevented the accumulation of carotenoids and of chloroplastic rRNA. Consequently, ribulose-1,5-bisphosphate carboxylase activity was virtually absent. After all herbicide treatments in light, the leaves contained only rather low catalase activity. In the presence of aminotriazole and haloxidine, the chloroplast-specific NADP-glyceraldehyde-3-phosphate dehydrogenase and the peroxisomal enzymes glycolate oxidase and hydroxypyruvate reductase had high or even normal activities, as in untreated leaves. In leaves treated with Sandoz 6706, fluometuron, or EMDIT 5914, the activities of the latter three enzymes were, in parallel, only very low. Some herbicides interfered with enzyme activities in vitro, particularly with those of catalase and of glycolate oxidase. Among mitochondrial enzymes, cytochrome c oxidase activity was either unaffected or lower, while fumarase had considerably higher activities in the herbicide-treated, as compared to untreated leaves. The specific effects on peroxisomal enzymes cannot be explained by the hypothesis of herbicide-induced photodestructions in carotene-deficient plastids. Alternative explanations for the genesis of the chlorosis are discussed.

[Lupus erythematosus profundus: two cases with ultrastructural study].

Lupus erythematosus profundus (panniculitis) is an unusual clinical variant of lupus erythematosus in which the lesions occur in deeper corium and panniculus. The diagnosis usually causes difficulty. Both reported cases are similar and concern young men with subcutaneous nodules appearing on the face and leaving depressed areas. Histological examination reveals a marked infiltrate in the dermis especially around the skin appendages, reaching the deep dermis and the fat tissue; this infiltarte is composed mainly of lymphocytes and histiocytes. Some moderate epidermal changes are present: liquefaction degeneration of the basal cell layer, epidermal atrophy, thickening of basement membrane (P. A. S.). Direct immunofluorescence of the lesions showed, in one case IgG deposits at the dermo-epidermal junction. Ultrastructural findingd are similar in both cases and may be an important point of diagnosis for isolated forms: -- tubular structures "paramyxo-virus-like-inclusions" are present in dermal infiltrate, endothelial cells and sometimes in keratinocytes of the basal layer; -- the basal lamina has many folds and villous projections;-- vacuoles and concentric lamellar formations are observed inside keratinocytes. No clinical or immunological signs of systemic lupus were found. Antimalaric drugs clear up the nodules.

[Rudiger (E. E. C.) syndrome: report of a case associated with atopic dermatitis (author's transl)].

The syndrome of ectrodactyly, ectodermal dysplasia and cleft lip-palate is described in a boy aged 3; although inherited in most cases as an autosomal dominant trait, it seems here to be sporadic. An atopic dermatitis clinically evident and particularly resistant to treatment is present, but IgE level is not raised. Ophthalmologic changes are prominent. Systematic intravenous pyelogram showed urinary tract strictures.

[Experimental investigations on the regeneration of aortic endothelial cells. Automatic and visual evaluation of autoradiograms (author's transl)].

Autoradiographic studies on plane preparations using 3H-thymidine and pulse cytophotometric investigations on scrape preparations of aortic endothelium from 34 rabbits have brought the following results: By means of multiple 3H-thymidine-injections the number of labelled endothelial cells is higher in young growing animals than in older rabbits; pulse cytophotometrically we did not find any indication of polyploidization processes; from that we conclude a decrease of the growth fraction with advancing age. In comparison to visual counting the automatic image analysis is a highly effective method for the evaluation of autoradiograms. After multiple 3H-thymidine applications the mean blacking intensity of the labelled cell nuclei is lower in 2 months old animals than in 12 months old rabbits; that may mean an increase in the S-phase duration of the proliferating endothelial cells of the older animals. In the area with branching sites there are more 3H-thymidine-labelled endothelial cells than in the neighbouring vascular segments. The significance of these findings for the preferential incidence of arteriosclerosis in this area is discussed.

Granuloma faciale. Ultrastructural study of three cases.

Light and electron microscopic observations were carried out in three cases of granuloma faciale. The dense granulomatous infiltrate of the corium showed: 1) more eosinophils than suspected by light microscopy, most of them with degenerative changes; 2) Charcot-Leyden chrystals; 3) numerous large histiocytes with their cytoplasms filled with lysosomal vesicles containing various inclusions (lamellar figures, myelin bodies, circular concentric organelles, digested membrane structures). The lack of Langerhans granules in dermal cells seems a valid reason to differentiate granuloma faciale from eosinophilic granuloma of the skin, usually considered a variety of histiocytosis X. No evidence of bacterial or viral infections was noticed. Granuloma faciale, despite its uncertain etiology, seems to represent a true entity.

[Onychopathy of mushroom-growers].

Particular nail damage appeared in patients working on mushroom beds. In five cases, all similar, the recent use of plastic bags containing the growing medium, seems to be the origin of this onychopathy. Onycholysis with latero-distal "usure des ongles", koilonychia, longitudinal splitting with sometimes splinter haemorrhages are the most specific changes. Chemical damage by phytosanitary products or fungi infection have no significant part in these abnormalities; their cause is traumatic, representing occupational stigmata mark, and results from repeated rubbing of the nails in workers lifting up heavy plastic bags.

[Lichen aureus or lichen purpuricus; about 5 cases; ultrastructural study].

Clinical, histological and ultrastructural features in five cases of lichen aureus are descirbed. This rare condition with ages ranging here from 6 to 31 consists of a pigmented papular and purpuric eruption. Unilaterally, trunk or limb may be involved, in a possible systematized distribution. The microscopic picture is characterized by a dense histiocytic and lymphocytic infiltration in the upper part of the dermis; extravasation of erythrocytes and iron pigment in the histiocytes are often noticed. In fine structure most cells of the dermal infiltrate are histiocytes (or macrophages) with numerous worm-like structures and even typical Langerhans granules in three cases; histiocytic cells; histiocytes are frequently seen in apposition to lymphocytes. In all these cases, no sign of histiocytosis X is found. Classification of lichen aureus in the group of idiopathic pigmented purpuric eruption is proposed.

[Familial epithelioma of Ferguson-Smith about two familial cases].

Two cases of multiple self-healing epithelioma (Ferguson-Smith) are reported in a French family. This dermatosis has been present since the age of 30 in both father and son. The father aged 66 years, farmer, had suffered since 1939 from recurrent spontaneously healing tumors of the face and neck, with deeply pitted scars on the face. He was treated during fifteen years in a cancer institute for multiple well-differentiated " squamous cell carcinoma " on the neck, face, and tongue; local recurrence did not occur after treatment of the active tumors and there was no clinical evidence of secondary lymphnode involvement. His son, aged 30, teacher, came in July with a papular active tumour on the cheek, which was surgically removed; his face and ears soon became affected. In the two cases, numerous biopsies showed in the early stage a great down growth of the epidermis with a central horny plug, like in keratotic papules or small kerato-acan-thomas and in more active lesions, typical appearances of squamous cell epithelioma grade I. The familial incidence is proved in the majority of the 30 cases reported since the first description by Ferguson-Smith in 1934. Only caucasians are concerned, most of them being Scottish. There is no sex predominance. First changes generally occur in young adults on light exposed areas but mucous membranes can be affected. The individual lesions last for several months, but fresh ones appear continuously, so that the disease remains extensive. The cause of these tumors is still unknown; histological and ultrastructural findings cannot explain this condition. Our ultrastructural study is parallel to previous works and fails to find viral particles in keratinocyte nuclei or cytoplasma.
