Assessment of DNA damage in germ cells of male rabbits treated with isoniazid and procarbazine.

The 2 hydrazine derivatives isoniazid (INH) and procarbazine hydrochloride (P) were injected intravenously into rabbits. Radioactive thymidine was injected into both testicles. Rabbits were ejaculated repeatedly, sperms were counted and incorporation of [3H] thymidine into sperm head DNA was determined by liquid scintillation counting. In P-treated rabbits (5 and 50 mg/kg) radioactivity was significantly increased in sperms that were in late phases of spermatocyte and early phases of spermatid maturation at the time of treatment. This indicates that DNA repair synthesis, (unscheduled DNA synthesis, UDS) occurred following drug-induced DNA damage in these germ cells. Normal DNA synthesis in spermatogonia was inhibited by the high dose only. INH (50 and 125 mg/kg) did not cause UDS in spermatocytes and spermatids and did not affect normal DNA synthesis in spermatogonia. The results are in agreement with literature data indicating that P is a potent mutagen and carcinogen. INH, on the other hand, has weak mutagenic and carcinogenic activities that are most apparent in mice.

The challenge of working together: documentation collaboration.

Duplicating the preoperative assessment kept the staff working parallel, instead of intersecting or collaborating. The process of promoting better communication via collaborative documentation required staff to reflect on their roles--not as members of the PACU or OR team, but as part of the group giving surgical care.

Oxytocin and its receptor in the process of parturition.

OBJECTIVE: To review the potential roles of oxytocin (OT) and its receptor (OTR) in the regulation of parturition. METHODS: We reviewed the literature to describe the molecular aspects of the mechanisms of action and the regulation of OT and its receptor, particularly in relation to changes that occur at parturition. RESULTS: The literature provides strong support for a role for the OT/OTR system in the regulation of human parturition. Paracrine rather than endocrine mechanisms might be more important, and the regulation of the receptor might be more important than the ligand. The system is regulated by a wide variety of chemical and physical factors, including sex steroids, orphan receptors, uterine stretch, and many others. There also might be important interactions with the immune system. CONCLUSION: The OT/OTR system provides a potentially important therapeutic target for regulating the timing of human birth.

Successful bone formation at immediate transmucosal implants: a clinical report.

The aim of this study was to test whether bone could be formed in peri-implant defects at immediate transmucosal implants using guided bone regeneration. Ten patients (median age 48 years) underwent comprehensive dental care including the placement of an implant into an extraction socket immediately following removal of a tooth. An expanded polytetrafluoroethylene membrane and the mucoperiosteal flap were adapted around the neck of the implants, leaving the sites to heal in a transmucosal fashion. During implantation (baseline) and at membrane removal surgery 5 months later, the following clinical measurements from the implant shoulder were assessed at six sites: implant-bone contact (defect depth), level of the alveolar crest, level of the membrane, and distance from the crest to the implant body (defect width). Estimates of the defect volume bordered by the membrane, the implant, and the bony walls were calculated. At baseline, the mean defect depth was 4.7 mm (SD 1.3 mm, range 1 to 14 mm). At membrane removal, the mean defect depth had decreased to 2.1 mm (SD 0.8 mm). Compared to baseline, this decrease was statistically significant (P < .01). The mean increase in bone height at the deepest defect site of each implant was 6.7 mm (SD 3.0 mm), which was significant (P < .01). At baseline, the mean value for the defect volume estimates was 9.45 mm3 (SD 5.75 mm3). At membrane removal, a significant decrease (P < .01) was found. After 5 months, 94% of the area beneath the membrane was filled with new bone. It was concluded that guided tissue regeneration at immediate transmucosal implants is successful in generating bone into peri-implant defects.
