[Adaptation of the peripheral organs of rats to noradrenaline deficiency following chronic desympathization].

In 3--week guanetidine--desympathized rats, the noradrenaline (NA) concentration in myocardium, small intestine and ductus deferens constituted 1, 6 and 3% of the control, resp., whereas release of the NA from the ductus deferens on transmural stimulation dropped to 3%. From 4 to 16--week age the NA content in myocardium and small intestine reached 30 and 23%, resp., and its release was equal to control. The diminishing of NA content was followed by augmented adrenaline--sensitivity and diminished cholinosensitivity of the organs under study. Significance of local adaptive mechanisms for normalizing of activity of the adrenergic nervous system under conditions of desympathization, is considered.

Acetylcholine↗

[Noradrenaline concentration in peripheral organs and their adrenosensitivity following chemical desympathization].

The rats desympathised with guanetidine developed a 58% decrease of the noradrenaline content in the myocardium, 37% - in the deferent duct, and 82% - in the small intestine. No significant changes of the catecholamines content occurred either in adrenal glands or in the hypothalamus. Sensitivity of the deferent duct's adrenoreceptors increased by 9 times. Sensitivity of the small intestine to noradrenaline increased by 3.4 times whereas the sensitivity to the antagonistic transmitter acetylcholine decreased by 2.4 times. Local adaptation mechanisms maintaining functioning of the organism in chronic desympathisation, are regarded.

[Sensitivity of fragmented and stratified sea urchin embryos to cytotoxic neuropharmacological preparations].

The red half embryos and related quarter embryos (yolk and pigment) of Arbacia lixula, obtained by means of centrifugation of the eggs in sucrose gradient, retain the normal level of sensitivity and supersensitivity to cytotoxic neuropharmaca, antagonists of biogenic monoamines. The white half embryos and clear quarter embryos practically lack supersensitivity whereas the granular quarter embryos restore it to the initial level. The non-pigmented blastomers of stratified embryos are characterized by somewhat weakened supersensitivity. A suggestion is put forward that the supersensitive embryos of A. lixula possess a sensibilizing factor which couples the supersensitivity receptors with the processes of cell division and moves together with the yolk granules upon centrifugation. This factor is not observed in the Strongylocentrotus granularis embryos lacking evident supersensitivity.

[Adrenosensitivity of isolated rat organs after a change in their noradrenaline content].

The 30-day dynamics of noradrenaline contents after a single administration of guanitidine (50 mg/kg) was similar in the heart and small intestine but different in the vas deferens. The decrease of the noradrenaline content in the small intestine and vas deferens duct was followed by an increase of their adrenosensitivity, and the restoration of noradrenaline level--by a decrease of the adrenosensitivity, which reveals an invert relationship between these parameters (the correlation coefficient for small intestine being 0.78 and for vas deferens duct--0.61). The role of the catecholamine level near the adrenoreceptors in regulation of the adrenosensitivity of effectors is discussed.

[Effect of GABA on the synthesis, uptake and release of noradrenaline-H3 from brain slices].

The data obtained showed that GABA exerted no effect on the synthesis and putake of H3-NE while increasing its loss from slices preincubated for 5 min before incubation for 30 min with H3-NE for uptake. When slices were preincubated for 30 before the addition of H3-NE the effect of GABA on NE content in the slices was reversed. A similar reversal of the GABA effect occurred when varying the preincubation period of slices subjected to prolonged electrical stimulation. The uptake and release of H3-NE were not affected by the duration of the preincubation period. The reversal of the GABA effect on H3-NE did not correlate with the depletion of endogenous stores of NE and may be due to the changes in the ionic composition of slices in the course of the prolonged incubation.

[Analysis of the regulatory influence of effector cells on 3H-catecholamine synthesis in an adrenergic neuron].

In isolated organs of the white rats activation of noradrenaline-H3 and dophamine-H3 synthesis during action of the alpha-adrenoblocking agent phentolamine (1.10(-6) M) and inhibition of the amines synthesis under influence of the alpha-adrenoreceptor activator mezaton (1.10(-5) M) were due to a release from cells-effectors of respective humoral factors and to the latters' effect upon the processes of amine formation in the adrenergic neuron. Formation of the humoral factors altering the rate of catecholamines-H3 synthesis is associated with activation of the protein synthesis in cells-effectors and occurs during change of the functional state (excitation or blockade) of the post-synaptic adrenoreceptors. The processes occurring at the time in the adrenergic neuron, are not correlated with protein formation. The possibility of existence of a common mechanism for feed-back transsynaptic regulation of synthesis and neuronal uptake of noradrenaline via adrenoreceptors of cells-effectors by means of changing the intensity of thyrosin and noradrenaline transport into the neuron is discussed.

[Noradrenaline-H3 uptake by isolated rat organs with active and blockaded adrenoreceptors].

The beta-adrenoreceptor blocking agent propranolol was shown to activate noradrenaline-N3 absorption by the rat isolated organs by 30-180%. The alpha-adrenoreceptor blocking agent phentolamine only activated by 30-55% the noradrenaline-N3 absorption in the organs possessing the postsynaptic alpha-adrenoreceptors (ductus semen, spleen and amsll intestine). The beta-adrenoreceptor activator isopropylnoradrenaline decreased the intensity of noradrenaline-N3 absorption in all the organs by 15-50%. The alpha-adrenoreceptor activating agents (urine and mesaton) only depressed the noradrenaline-N3 absorption by 20-45% in the organs possessing postsynaptic alpha-adrenoreceptors. The data obtained suggest existence of reverse transsynaptic regulatory effects via adrenoreceptors of the cell-effector on the processes of noradrenaline absorption by adrenergic neuron.

[Mechanism of the regulatory influence of an effector cell on noradrenaline-H3 uptake by an adrenergic neuron].

In isolated organs of the white rat, activation of neuronal absorption of noradrenaline-H3 under action of alpha-adrenoblocking agent phentolamine (1-10(-6) M) was due to release of a humoral factor from the effector cell and its effect on the adrenergic neuron. Under influence of activating agent for alpha-adrenoreceptors mesatone (1-10(-5) M), formation and release of humoral factor inhibiting the neuronal absorption of noradrenaline-H3, occurrs. Formation of the humoral factors altering the intensity of amine absorption is associated with activation of the protein synthesis in effector cell and occurs on alteration of functional state (excitation and blockade) of postsynaptic adrenoreceptors. A possible way for the reverse transsynaptic control of neuronal absorption of noradrenaline via adrenoreceptors of effector cell, and the role of this regulatory mechanism in activity of adrenergic system, are discussed.

[Several pattern in the synthesis and uptake of H3-catecholamines by isolated rat vas deferens].

In the rat isolated vos deferens, increase of the thyrosine-H3, DOPA-H3, and Dopamine-H3 contents in the incubation medium increases respectively Km and Vmax of the noradrenaline-H3 biosynthesis. Increase of the Dopamine-H3 and noradrenaline-H3 in the medium also increases Km and Vmax of neuronal uptake and reservation of these catecholamines in the adrenergic granules. The simultaneous increase of Km and Vmax of the catecholamines synthesis and uptake leads to that the effectiveness of these processes undergoes much lesser alteration that the main parametres. The interrelationship between the substrate concentration in the medium and the velocity of catecholamines synthesis is, apparently, due to that the adrenergic granules differ from each other by the intensity of synthesis and reservation of noradrenaline. This is one of the factors regulating accumulation and release of the sympathetic mediators.

[The effect of adrenoblockaders on the rate of H3-catecholamine synthesis and uptake in the rat seminal duct].

In the rat vas deferens alpha-adrenoblocking agent phentolamine (1-10(-6)M) and beta-adrenoblocking agent propranolol (1-10(-6)M) activated uptake of exogenous noradrenaline--H-3 by 37 and 47% respectively and synthesis of noradrenaline--H3 from thyrosin-H3 by 72 and 48%. The rate of noradrenaline-H3 synthesis from DOPA-H3 or dopamine-H3 remains unaltered. Regulating effect of the adrenoblocking agents is carried out by means of uptake of tyrosine-H3 and noradrenaline-H3 into adrenergic neuron, i.e. at the main stages sustaining a necessary level of mediator. The rate of noradrenaline biosynthesis is supposed to be independently regulated by means of both types of adrenoreceptors in an adrenergic neuron.
