Endogenous factor activating Na,K-ATPase induced by blockade of adrenoceptors.

It was established that the regulatory factor released from the effector cells under the blockade of postsynaptic alpha- and beta-adrenoceptors by phentolamine and propranolol (0.5-1.0 microM) and activating noradrenergic uptake and synthesis increased the activity of Na,K-ATPase of isolated rat organs and partially purified Na,K-ATPase isolated from the rat brain. The regulatory factor is a protein with molecular mass 25-100 kDa, adsorbed on phenylsepharose CL-4B and eluated with 30 mM KCl. It consists of two active components with pI 5.1 and 5.9 which in admixture (1:1) activate Na,K-ATPase by 60%.

[The action of high temperature on beta-adrenoreceptor activity and catecholamine synthesis].

Hyperthermia and overheating of the rat isolated organs induce significant changes of the binding of receptors' specific blockers and catecholamine synthesis. One of the main mechanisms of the organism response to the temperature actions involves a local rearrangement in intensity of different stages of the adrenergic process. Possible significance of these changes in the hypothalamus for synchronisation of the organism's response to extreme temperature effects, is discussed.

[The characteristics of the cholinergic reaction of the portal vein in the rat liver].

The kinetics of M-cholinergic response to acetylcholine and effects of phenylephrine and phentolamine on the functional state of M-cholinoceptors, were studied on the rat isolated vein. Two discrete pools of M-cholinoceptors differing in their sensitivity by a factor of 7.5, were found. Functional interaction between M-cholinoceptors and alpha-adrenoceptors was established as manifested in a change of basic parameters of M-cholinergic reaction (K and Pm) when the alpha-adrenoceptors were activated or blocked. The activation or blockade of specific alpha-adrenoceptors seems to evoke conformational changes of cell membrane thus modulating the affinity to the M-cholinoceptors agonist.

[The characteristics of the adrenergic reaction of the portal vein in the rat liver].

Adrenergic response to noradrenaline and specific agonists of alpha- and beta-adrenoceptors phenylephrine and isopropylnoradrenaline, as well as the effect of the alpha-adrenoceptors blockers on the functional state of beta-adrenoceptors and vice versa, were studied in the rat isolated portal vein. Functional interaction between alpha- and beta-adrenoceptors was manifested in a change of alpha-receptors when beta-receptors were blocked, and a change of reaction of beta-receptors when the alpha ones were blocked. The blockade of specific receptors seems to evoke conformational changes of cell membrane, thus modulating the affinity to the agonist of the receptors of the other type.

[Desensitization of the alpha 1-adrenergic reaction under the action of noradrenaline in increasing concentrations].

The kinetics of alpha 1-adrenergic response to noradrenaline in a wide range of concentrations upon the cumulative and discrete addition of noradrenaline, was studied in isolated epididymal sections of the rat vas deferens. Two pools of alpha 1-adrenoceptors differing in their affinity to noradrenaline were revealed in both cases. The desensitisation of both pools of alpha 1-adrenoceptors is shown during the adrenergic reaction upon the cumulative addition of noradrenaline. The desensitisation is expressed in the decrease of the number of active receptors.

[The effect of acetylcholine and serotonin on the adrenergic reaction of the rat small intestine].

Acetylcholine and serotonine depressed the sensitivity of the rat small intestine's receptors and enhanced the maximal alpha-adrenergic response. Acetylcholine acted as a competitive inhibitor of beta-adrenergic response, the latter being insensitive to serotonine. Acetylcholine even in the threshold concentrations (1.0 x 10(11) M) increased the noradrenaline desensitization of the adrenoreceptors.

[A kinetic analysis of the alpha 1-adrenergic reaction of the smooth muscles in the rat vas deferens].

The kinetics of alpha 1-adrenergic response to noradrenaline was studied in isolated epididymal section of the rat vas deferens. Two discrete pools of alpha 1-adrenoreceptors differing by their affinity to noradrenaline, were revealed. The alpha 1-adrenoreceptors also differed from each other in their ability to interact with one or two molecules of the transmitter. Functional role of heterogeneity of the receptors is discussed.

[Serotonin contents in the hypothalamus of adult males, females and males castrated during the neonatal period].

The aim of the present work was to study the character of the change in serotonin level in the anterior and medial basal hypothalamus of adult rats after the effect of testicular hormones had been switched off on the first day of postnatal life. It was shown in our work that in males serotonin level was significantly lower than that in females by 67 and 46% in the anterior and medial basal hypothalamus, respectively. Castration of newborn males resulted in a significant increase in serotonin level in both anterior and medial basal hypothalamus-up to the level observed in females. It is supposed that the male sex hormones affect differentiation of serotoninergic system of the brain.

[Modulation of the M-cholinergic reaction by the venom of the Central Asian cobra (Naja naja oxiana)].

The effect of the Central Asian cobra's (Naja naja oxiana) poison upon cholinergic response of isolated small intestine was studied in the Wistar rats. The effect involved a significant activation by 30-70 per cent of M-cholinergic response in the type of "noncompetitive" interaction. The modulating effect of the poison is supposed to occur due to changes in cytoplasmatic membrane.

[Analysis of the effect of noradrenaline on the response of the vas deferens of the rat to transmural stimulation].

In the rat isolated vas deferens, exogenous noradrenaline (NA) in concentrations of 5 X 10(-7)-5 X 10(-6) M acting for 10-15 min produced an increase whereas in concentrations above 10(-5) M it produced a decrease of the response to transmural stimulation: the increase being due to raising of the level of its secretion produced by accumulation of NA in nerve endings as well as to augmentation of postsynaptic response to NA resulting from a decrease in its reversal neuronal uptake. The decrease in the organ response to stimulation under the effect of NA at concentrations above 1 X 10(-5) M is due to desensitization of postsynaptic alpha-adrenoceptors.

[Use of Naja oxiana venom for modulating the adrenergic process].

Specific changes of adrenergic responses to the poison of the Middle Asia cobra were found in the rat isolated small intestine. The changes reminded the action of inhibitors upon enzymes and may be characterized as a partial competitiveless interaction. The poison modulating effect upon adrenergic responses seem to be unrelated to its direct action on adrenoreceptors, but mediated through the effector cell's membrane.

[Kinetic analysis of smooth muscle reaction to acetylcholine].

In the holothurian isolated muscle, physiological ambiguity of cholinoreceptors was found in respect to their affinity to acetylcholine. A few discrete pools of receptors were revealed in a single effector system, the pools differing in their main parameters (Pm, K and n) reflecting functional state of specific receptors for biologically active substances.
