Neuropathological studies in a child showing some features of the Rett syndrome.

Clinical and neuropathological data are presented from a girl who died at 14 yrs and who in life displayed some of the characteristics of the Rett syndrome--social withdrawal, progressive loss of locomotor as well as social skills, microcephaly, and a very restricted stereotypy of manipulation. Neuropathological studies showed mild generalised cerebral atrophy with marked unevenness of melanin deposition in nigral neurones. It is suggested that this patient may represent a severe form of the Rett syndrome.

Ornithine carbamoyl transferase deficiency: a neuropathological study.

A detailed autopsy study of three children with ornithine carbamoyl transferase (OCT) deficiency is presented. Although variable in extent, a basic pattern of neuropathological lesions is discernible. Case 1 shows gross cerebral atrophy, cases 2 and 3 milder lesions in the basal nuclei but also multiple cerebellar heterotopias and delayed myelination. We suggest that the findings may provide evidence that OCT deficiency can have a teratogenic effect in utero and suggest that there is a need to monitor the pregnancies of carriers of this disorder.

Computed tomography in acute haemorrhagic leukoencephalitis.

Three patients with biopsy-proven acute haemorrhagic leukoencephalitis are described. All presented with an abrupt onset of focal neurological signs, pyrexia and leukocytosis. In two cases there was disturbance of consciousness. Computed tomography in each case revealed low density virtually confined to the white matter of the cerebral hemisphere; in one case there was central enhancement within the low density. The combined clinical and CT signs should strongly suggest the diagnosis.

Metabolic and secretory processes in nerve-endings isolated from post-mortem brain.

Isolated nerve ending particles (synaptosomes) prepared from rat and human cerebral cortex, after varying conditions of post-mortem storage were shown to possess good structural and morphological integrity and metabolic activity. Respiration, potassium retention, lactate dehydrogenase content, and stimulus-induced release of transmitter candidate amino acids, as well as the neuropeptide somatostatin, were measured. These preparations from post-mortem material showed properties which were closely comparable with similar preparations from fresh material. The relevance of these findings to studies on human post-mortem material is discussed.
