Cultured hepatocytes bind and internalize bovine serum amine oxidase-gold complexes.

Cultured hepatocytes express binding sites for bovine serum amine oxidase on their membrane surfaces as evaluated at the electron microscopic level by using enzyme-gold complexes. Hepatocytes show binding sites as small clusters of gold granules, not bound in a specialized region of the plasma membrane. The binding competition of enzyme-gold ligand to cells was achieved by preincubation with uncoupled bovine serum amine oxidase. In addition, N-acetyl-galactosamine, N-acetyl-glucosamine and Mannose, at the final concentration of 80 mM, partially inhibit the binding.

Peculiar effects of temperature and polyvinylalcohol on the activity of bovine serum amine oxidase.

An inflexion point of enzyme activity at 38 - 42 degrees C of the bovine serum amineoxidase was found. This result, associated with non-strict Arrhenius curves and slightly different activation energies in various temperature intervals, suggests some conformational transitions at the mentioned temperatures. The high molecular weight polyvinylalcohol (100,000 Da) generated an activatory effect and a sigmoidal (non-Michaelis) curve of the dependence of the activity on the substrate concentrations, while the low molecular weight polyvinylalcohol (20,000 Da) does not produce this effect. The different ratio of the two types of polyvinylalcohol/enzyme monomer sizes is considered to be responsible for these different effects on the enzyme kinetics.

Pulsed electron paramagnetic resonance studies of types I and II coper of Rhus vernicifera laccase and porcine ceruloplasmin.

Electron spin-echo decay envelopes for types I and II copper of Rhus vernicifera laccase and for type II copper of procine ceruloplasmin have been studied. Nuclear modulation patterns show that imidazole is a ligand for all of them. The linear electric field effect (LEFE) in EPR was studied for type I copper in a laccase preparation from which type II had been removed. The symmetry of the site is near tetrahedral and the magnitude of the LEFE is correlated with the intensity of blue color.
